Stem cells are crucial for embryonic development and in the maintenance of adult cellular homeostasis.Understanding the regulatory network of stem cells,including embryonic and adult stem cells,will allow us to learn the pathogenesis and possibly design novel approaches to treat many diseases (such as cancer and degeneration).The retinoblastoma (Rb) pathway controls cellular proliferation,differentiation and death.More and more evidences support an important role of Rb activity in the biology of stem and progenitor cells.Transiently inactivating Rb pathway might favor the expanding of functional stem cell populations,thus have values in the future stem cell applications.
2.Application of adipose-derived stem cells in cartilage tissue engineering

Cartilage is one of the earliest reconstructed tissues used in tissue engineering. Due to the lack of appropriate seeding cells, cartilage tissue engineering is, however, relatively lagged behind. With the emergence of stem cell research, adipose stem cells(ASCs) are introduced as seeding cells into tissue engineering for possessing many advantages such as wide spreading, large amount of cells available and easy to obtain. However, the outcome of tissue engineered cartilage construction by ASCs is not as ideal as that by bone marrow stem cells (BMSCs) yet. Low efficiency of ASC chondrogenesis is considered the major cause. This review summarizes the purification of adipose-derived cells, maintenance of sternness and optimization of ehondrogenie induction, which play vital roles in improving ASC s chondrogenesis.
3.Study on Brain Pharmacokinetics of Tetramethylpyrazine Phosphate After Intranasal Administration

Objective To evaluate the rat brain pharmacokinetics of tetramethylpyrazine phosphate (TMPP) after intranasal administration.Methods The pharmacokinetic behavior of TMPP in rat brain was investigated after intranasal or intravenous administration (10mg?kg-1) of TMPP.Brain samples were collected by intracerebral microdialysis.The concentration of TMPP in the sample was measured by high performance liquid chromatography (HPLC).Main pharmacokinetic parameters were calculated and analyzed by Das 2.0 pharmacokinetic software.Results The results showed that two administration methods were all fitted to one compartment model.The absorption of TMPP from the nasal cavity to brain was rapid.The concentration-time curve showed a prolonged duration of TMPP concentration in the brain after intranasal administration.AUC0-∞values were (698.26?122.59)?g?min?mL-1 in intranasal group and (446.25?107.44)?g?min?mL-1 in intravenous group,and the difference was significant between the two groups (P
4.Study on In-vivo Nasal Mucosa Absorption of Tetramethylpyrazine Phosphate Liposome in Rats

Objective To study the characteristic of in-vivo nasal mucosa absorption of tetramethylpyrazine phosphate(TMPP) liposome in rats. Methods Nasal circulatory perfusion method in rats was used to study the effects of different circulatory rates and TMPP concentrations on nasal mucosa absorption of TMPP liposome. Nasal mucosa absorption rate constants of TMPP liposomes and TMPP solutions at the same concentration were also compared. Results The absorption rate constant of TMPP liposomes was increased with the increase of circulatory rates at the range of 1.2~2.4 mL/min and with the increase of TMPP concentrations at the range of 0.245~0.98 mg/mL. Both the absorption rate constant and absorbed amount of TMPP for TMPP liposomes were higher than those for TMPP solutions at the same concentration. Conclusion The nasal mucosa absorption of TMPP liposome was complete as compared with that of TMPP solution.
5.Ultrasonographic diagnosis of benign,borderline and malignant ovarian epithelial tumors

Objective To study the value of ultrasonographic diagnosis of benign,borderline and malignant ovarian epithelial tumors.Methods In the 203 patients of ovarian epithelial tumors,the substantial tissues′ area of 128 cases with cyst-solid mass was measured through ultrasonography and compared with pathology.Results The area of substantial tissues in ovarian epithelial tumors was benign 5 cm 2 were regarded as optimal threshold values for differentiation of the benign,the borderline and the malignancy,their sensitivities were 94.7% , 80.0% and 90.7% ,their specificities were 96.3% , 92.4% and 97.6% ,accuracies were 96.1% , 90.0% and 92.9% .Conclusions It provides an effective method for diagnosis of benign, borderline and malignant ovarian epithelial tumors.If combined with conditions of color Doppler flow imaging, and comprehensive consideration on greater omentum and lymph node, the accurate diagnostic rate will be increased.
6.Oncocytic papilloma in nasal cavity and paranasal sinuses: 3 of cases.

Oncocytic papilloma (OP) of nasal cavity and paranasal sinuses are uncommon. We report 3 cases of oncocytic papilloma arising in the nasal cavity and paranasal sinuses, and review the relevant literatures. Unilateral sinonasal lesions are the main clinical manifestation. Pathological feature is multiple layers of epithelial cells. The cytoplasm contains abundant eosinophilic granular and microcapsules filling with mucus. The neutrophils aggregate in the microcapsules which form microabscess. The tendence of recurrence and malignant transformation are the biological character. The best treatment choice is surgical excision completely, and long-term postoperative follow-up is necessary to prevent relapse.

7.Study on Purification of Sini Decoction with Macroporous Resin D_(101)

Objective: To study the purification of Sini Decoction (SND) with macroporous resin D 101 . Methods: SND was isolated and purified by macroporous resin D 101 . The purification of SND was evaluated by chemical and pharmacological methods. Results: The thin layer chromatograms, high performance liquid chromatograms of the SND purification and those of SND were almost the same; also the SND purification can significantly promote the content of nitric oxide (NO) in serum of mice treated by Metaraminol. Conclusion: The SND purification can keep the active ingredients of SND.
8.Pharmacokinetical study on Sini Decoction

Objective: To study the correlation of pharmacokinetics(PK) and pharmacodynamics(PD) of Sini Decoction (SND) (Radix Aconiti Praeparata, Rhizoma Zingiberis, Radix Glycyrrhizae, etc.) and reveal the compatibility of SND. Methods: Using the drug serums from the same source, we determined the plasma concentration in vivo in dog and the pharmacological effects at the same time inspected by the contents of aconitine type alkaloids and nitric oxide (NO). Results: The data of PK and PD can be both described by one compartment open model with first order absorption in vivo in dog and the parameters of two methods are similar. The results show that the effective fraction of SND gives a prompt and lasting effect in accordance with characteristic function of SND. Conclusion: A good correlation exists in SND of the parameters between pharmacokinetics and pharmacodynamics.
9.Effect of profound hypothermia on function of mitochondria in hippocampus after global ischemia in rats

Objective To detect the effect of deep hypothermia on the function of mitochondria in hippocampus after global ischemia in rats and to explore the protection mechanism. Methods The animal model of cardiopulmonary bypass (CPB) was established in rats that were then randomly divided into three groups,ie,control group,normothermia ischemia group and hypothermia ischemia group,eight rats per group.The mitochondria was extracted from the hippocampus of each rats for observing the mitochondrial respiratory function,the activities of succinate dehydrogenase (SDH),the cytochrome oxidese(CCO),the lnembrane fluidity and the content of intramitochondria free calcium and MDA. Resuits The content of intramitochondria free calcium and MDA in the normothermia ischemia group was increased significantly compared to the control group and that in the hypothermia ischemia group wag decreased significantly compared with the normothermia ischemia group(P<0.05).Respiratory state Ⅲ (R3),respiratory state IV(R4),P/O ratio and oxidative phosphorylation (OPR) in the normothermia ischemia group were decreased significantly compared to the control group (P<0.05).R3,R4,P/O ratio and OPR in the hypothermia ischemia group were increased significantly compared with the normothermia ischemia group (P<0.05).Membrane fluidity in the normothermia ischemia group wag decreased significantly compared to the control group (P<0.01),while that in the hypothermia ischemia group was increased significantly compared with the normothermia ischemia group(P<0.05).The activities of SDH and CCO in the normothermia ischemia group were decreased significantly compared to the control group (P<0.01),while those in the hypothermia ischemia group were increased significantly compared with the normothermia ischemia group (P<0.05). Conclusion Profound hypothermia exerts a protective effect on the function of mitochondria in the hippocampus after global ischemia in rats.
10.Study on the In- vitro Recovery of Tetramethylpyrazine Phosphate in Microdialysis Probe

Objective To detect the in-vitro recovery of tetramethylpyrazine phosphate (TMPP) in microdialysis probe and to study its stability. Methods The recovery of TMPP was detected by concentration difference method (increment method and decrement method ). The effect of flow rates and concentrations on recovery was studied. Results The recovery detected by increment method was as the same as that detected by decrement method; TMPP recovery was independent of TMPP concentration in the external medium. TMPP recovery had good stability. Conclusion Microdialysis sampling can be used for the pharmacokinetic study of TMPP and decrement method (ie. retrodialysis) can be used for the determination of TMPP recovery.
