2.PRELIMINARY STUDY ON SCREENING OF PHAGE RANDOM PEPTIDE LIBRARY USING RABBIT SERUM FROM A NEW MODEL INFECTED WITH SCHISTOSOMA JAPONICUM

Objective To screen the 12 mers-phage random peptide library using the serum from the new model rabbit and to identify the immuno-protection of the positive phages. The new model infected with Schistosoma japonicum was proved that has a high protection against the challenge infection. Methods After being absorbed by E.coli antibody, the serum of the new model rabbit was used to screen the peptide library. Through three rounds of biopaning and enriching, lots of positive phages were obtained and their antigenic ability was tested. Every mouse was immunized by subcutaneously injecting 1?10 14 pfu positive phages from the new model rabbit serum respectively at 0-2-4 th week. After 4 weeks of the last immunization, the challenge infection was performed. At the same time, several control groups including the group immunized with the phages from the rabbit serum of the normal model infected with Schistosoma japonicum, the group immunized with the original 12 mers-phage random peptide library and the control group of challenge infection were arranged. Results ①The positive clones of phage(1?10 14) from the new model rabbit serum were strongly recognized by the rabbit serum of the new model, weakly recognized by the rabbit serum of the normal model infected with Schistosoma japonicum,but not recognized by the serum of healthy rabbit. ②The reduction rate of adult worms and liver eggs induced by phages screened with the rabbit serum of the new model group and the nomal model group and that induced by the original peptide library were respectively 27 2% and 38 8 %, 17 8% and 35 0%, 4 5% and 6 0% Conclusion The new model group obtained a higher reduction rate of adult worms than the nomal model group (P
3.Study on screening simulated epitopes of protective antigens of Schistosoma japonicum and their immuno-protective effect

Objective To study the immuno-protective effect against Schistosoma japonicum challenge of the positive monoclonal phages which were screened from the 12 mers-phage random peptide library by the new model rabbit serum. Methods The new model was established by injecting the Schistosoma japonicum infection rabbits with inhibitor of phenol oxidose. Positive clones immunoscreened with the new model rabbit sera were absorbed by SEA immune rabbit sera, and 14 clones selected randomly from them were compared and their antigenic ability was identified by ELISA. The two best positive monoclonal phages (No.8, No.13) recognized by the new model rabbit sera were selected to immunize Kunming mice by subcutaneously injecting 1?10~15 pfu positive phages at 0, 2nd, 4th week respectively. After 4 weeks of the last immunity,each mouse was challenged with 40?1 S.japonicum cercariae. All mice were sacrificed after 42 days and the reduction rates of adult worms and the liver eggs were investigated. Results The positive phage clones after immune absorption were weakly recognized by the SEA immune rabbit sera. The 14 monoclonal phages were recognized by the rabbit sera of the new model and the normal model. Especially No.8, No.13 were strongly recognized by the rabbit sera of the new model,while weakly recognized by the SEA immune rabbit sera. The reduction rates of adult worms and liver eggs induced by the monoclonal phage No.13, the monoclonal phage No.8 and the original peptide library were 35.81% and 63.32%, 32.09% and 52.02%, 14.90% and 30.64%, respectively. Conclusion Most clones of simulated epitopes of SEA can be removed by absorbing positive clones with SEA immune rabbit serum .The 14 monoclonal phages from the new model contain the simulated S.japonicum epitope. The two monoclonal phages have higher reduction rates of adult worms and eggs than original 12 mers-phage random peptide library and the positive polyclonal phages.[
4.Changes of calcium handling protein after acute myocardial infarction in rats and effect of carvedilol

AIM: To observe the changes of sarcoplasmic reticulum Ca~(2+)-ATPase (SERCA), phospholamban (PLB) during heart failure after acute myocardial infarction (AMI) in rats and the effect of carvedilol. METHODS: Rats were randomly assigned to normal control group, sham-operation group, AMI group and carvedilol (CAR) group. 6 weeks later, in vivo hemodynamic, morphometry and SERCA, PLB mRNA and protein expression of myocytes were measured in all animals. RESULTS: In comparison with sham-operation group, LV end diastolic pressure (LVEDP) and weight of ventricles were increased, while maximal rate of rise and fall (?dp/dt) of LV pressure were decreased in AMI group. After treatment with carvedilol, these parameters were all improved. The mRNA and protein expression of SERCA were downregulated (P0.05). CONCLUSIONS: The changes of SERCA and PLB may be the important mechanism of contractile dysfunction in heart failure after AMI. Carvedilol is effective in preventing LV dysfunction after AMI. The molecular mechanism may be related with normalization of SERCA expression.
5.Outpatient experiences of caring in the outpatient clinic:a qualitative research

Objective To explore the outpatients′experience of caring and provide references for improving their experience of medical care.Methods The phenomenology research method was adopted in this study,and twenty-one outpatients were subject to an in-depth interview to learn their outpatient experiences.Results Ten themes of experience with caring were extracted as follows:passionate and patient,careful and considerate,respect for others,offers to help,encouraging and comforting,apologies and courteously,humor,truthful considerations,conscientious and responsible,and a caring environment setup.Five themes of experience without caring were extracted as follows:impatient communication, careless seeing of outpatients,delayed service,passive reminders,and facilities without enough details. Conclusions The outpatients may have abundant experiences of caring at the outpatient clinic,also lack of caring sometimes as well.Outpatient managers should keep an eye on outpatients′needs and satisfaction of caring from time to time,to improve the quality of outpatient service.
6.Research on army cadres population weight scientific intervention management mode and effect

Objective To look for suitable short-term weight loss and long-term maintenance method for army cadres. Method A total of 142 patients with simple overweight and obesity were randomly selected as the research subjects, and were followed up for 2 years, 130 of them were valid for analysis, 97 were male, and 33 female, age 27-59 years, average(47.41±7.55)years. SPSS 16.0 was used for analysis. Repeated measurements analysis of variance was used for data analysis, K-related samples nonparametric test was used for enumeration data. Result The army cadres population weight intervention management model was divided into three stages. They were respectively intensified weight loss stage for 6 months, the consolidation of weight loss for 6 months, and self-management stage maintained for 12 months, totally 24 months. Variance analysis results showed that after two years of focused intervention and health management, the body mass index(BMI)fell from(27.42 ± 2.81)kg/m2 to(24.69 ± 2.63)kg/m2 (F=2 649.945, P<0.05), waist circumference decreased from(91.09±8.24)cm to(85.26±7.76)cm(F=1 207.248, P<0.05). Nonparametric test results showed that after two years intervention management the behavior patterns concerning physical exercise became better. Exercisers increased from 33.8%to 73.3%(H=68.448, P<0.05); proportion of smoking decreased from 38.5% to 20.0%(H=33.692, P<0.05); drinking alcohol decreased from 50.8%to 20.8%(H=59.128, P<0.05);high salt diet decreased from 39.2%to 23.1%(H=31.722, P<0.05);high-fat diet decreased from 46.2%to 27.7%(H=41.571, P<0.05). Conclusion The practice showed that the short-term intensified weight loss, long-term maintenance, exercise and psychological intervention, changed the cadres population from passive to active on the weight control and self-management with remarkable effects, therefore such intervention is worthy of promoting.
7.Application of the inpatients' humanized nursing needs survey in the wards of whole-process humanistic care mode

Objective To investigate the requirements of hospitalized patients on humanized nursing in the ward of whole-process humanistic care mode and provide scientific references for continuous improvement of nursing quality.Methods A total of 408 patients from 2 wards of whole-process humanistic care mode.Patients were applied the self-designed questionnaire in combination with interview to understand the patients’ requirements of humanized nursing.Survey was carried out before the inpatients were discharged from the hospital adopting the “Evaluation Table of Patients’ Satisfaction Degree about the Nursing Service” produced and researched by our hospital independently.Results After carrying out the whole-process humanistic care service mode,the inpatients’ overall satisfaction degree and satisfaction rate with humanistic care were respectively 98.2%,97.5%.Conclusions Carrying out the quality care demonstration project and enhancing the concept of humanistic care can promote each other.Carrying out the inpatients’ care needs survey in the ward of whole-process humanistic care mode,integrating humanistic into every aspect of clinical nursing,are the crux of deepening high-quality nursing care as well as a new subject that clinical nurses should confront conscientiously.
8.Standardized management and effects for hospital′s humanistic nursing care

Objective To explore the methods of the standardized management used in humanistic nursing care and to assess its effects.Methods Measures of the standardized management began to be used in such care at the hospital since January 2013.Questionnaires of patient satisfaction with nursing care for inpatients and those discharged were used to assess the effects of the standardized management measures periodically and synthetically.Results The patient satisfaction with humanistic nursing care keeps increasing year by year from 2013 to 201 5.The differences among three sets of data are statistically significant(P < 0.05 ).Conclusions Humanistic nursing care calls for promotion of standardized management.It is high time to build such a standardized management system at the hospitals.
9.A Comparative Study on the Mutagenicity of Natural Borneol and Synthetic Borneol

Objective&Methods The mutagenicity of natural Borneol and synthetic Borneol was studied by using Ames test.Results Natural Borneol and Synthetic Borneol in the dose range of 0.4~250.0 ?g/plate had no effect in reducing the reversion frequencies of Salmonella Typhimurium histidine auxotrophic mutant(TA97,TA98,TA100,TA102)whether if metabolic activation system(S9mix)existed or not.Moreover,the backward mutant colony had normal background.Conclusion Natural Borneol and Synthetic Borneol do not possess evident mutagenicity under experimental condition.
10.Roles of transforming growth factor-β1 gene polymorphisms in severe acute respiratory syndrome and the interstitial lung fibrosis after recovered

Objective To investigate the roles of transforming growth factor(TGF)-β1 gene polymorphisms in severe acute respiratory syndrome-coronavirus(SARS-CoV)infection and the interstitial lung fibrosis after recovered.Methods Sixty-five recovered SARS patients,37 health care workers and 66 healthy controls were enrolled in this case-case study.The association between genetic polymorphisms of TG F-β1 and suscept ibility to SARS or interstitial lung changes after SARS reco,vered was carried out.Polymerase chain reaction-sequencing based typing(PCR-SBT)method was used to determine the polymorphisms of TGF-β1 gene at locus+869 and+915.Data were analyzed using t test and chi square test.Results There was no significant association of TGF+β1 gene polymorphisms at locus+869 and+915 in recovered SARS patients,health care workers and heahhy controls.And gene linkage of this two loci was not related with SARS-CoV susceptibility.Furthermore,no association between interstitial lung changes in recovered SARS palients and TGF-β1 gene polymorphisms or genetic linkage of this two loci.Conclusions It may not be related between TGFβ1 gene polymorphisms at locus+869 and+915 and SARS-CoV susceptibility.And interstitial lung changes in recovered SARS patients may not be influenced by TGF-β1 gene polymorphisms.
