1.Outcome of the Orientation and Mobility Skills Training Projects in Yunnan

Objective To survey the outcome of the Orientation and Mobility (O&M) Skills Training projects, a communities based rehabilitation which was advancing in Yunnan. Methods 310 blind subjects in model areas were assessed with the Assessment Standard of O&M Training for the Blind. Results 98.7% of subjects improved their mobility skills after receiving O&M skills training, that was 44.5% could walk independently in short distance, 34.5% in long distance, and 19.7% in complex surroundings in detail. There was no difference (P>0.05) among the different gender, educated level, and visual loss level, however, there was significant difference (P<0.05) made by the age level and the causes of disability of vision. Young and middle-aged subjects have better training result than the aged, as well as the congenital blind ones than the postnatal ones. Conclusion O&M skills training in Community Based Rehabilitation Services is effective for visual rehabilitation.
2.Rapid analysis by HPLC in the quality control study of Chinese herbal medicine pseudo-ginseng and its Chinese drugs preparation′s specific chromatogram

Objective To study the HPLC rapid analysis method for the 5 kinds of saponins in pseudo-ginseng herbs and its traditional Chinese medicine preparations Xuesaitong injection at different habitats and analyze the fingerprint characteristics. Methods The Waters Nova-Pak-wat044375 flash chromatography column (4.6 mm×150 mm, 4 μm) was as stationary phase, and acetonitrile-0.5% formic acid solution (v/v) system was as the mobile phase. The gradient elution, column temperature was 30 ℃, and detection wavelength was of 203 nm. Twelve batches of Panax notoginseng in Yunnan, Sichuan, Guangxi, Jiangxi and other places four kinds of Xuesaitong injection from different manufacturers were taken for rapid analysis of HPLC and methodological study. The similarity analysis was carried out by Chinese medicine chromatographic fingerprint similarity evaluation system (2012 edition). Results The establishment of rapid HPLC fingerprints baseline separation occurred within 1 min of detection, and 5 kinds of ingredients were all successfully detected in the 40 min. The similarity of each component was more than 0.9, and with a good precision, stability and reproducibility, RSD within 5%. Conclusions The method of HPLC rapid analysis for Chinese herbal medicine pseudo-ginseng and its Chinese drugs preparation's specific chromatogram is simple, rapid and effective. It can fully reflect the changes of 5 representative components chromatogram in pseudo-ginseng and its preparation, and so it can be as the quality control evaluation of pseudo-ginseng and its preparation.
3.Research on the correlation between uric acid levels and thyroid nodules and gender differences

Objective To explore the correlation between thyroid nodules and uric acid levels and to find their gender differences.Methods A total of 68 056 subjects in a regional medical physical examination center of Shanxi Province from January 2013 to June 2015 were enrolled in this study.All the participants′ general information and parameters were recorded.Thyroid nodules were detected by color Doppler ultrasonography.Results The total prevalence of thyroid nodule was 35.5%, 30.7% in males and 40.0% in females.The prevalence of single nodule was 50.1%, and multiple 49.9%.Compared with no nodule group, thyroid nodule group tended to be older, with higher BMI, and with a worse metabolic status(all P<0.01).The uric acid levels were lower[(352.37±78.14 vs 357.70±77.51) μmol/L, P<0.01] in thyroid nodule group in male and higher[(260.22±61.91 vs 253.91±59.18) μmol/L, P<0.01] in female.Conclusion Thyroid nodules may be associated with metabolism and inflammation.In males, hyperuricemia group had lower, while in females, hyperuricemia ones were with a higher prevalence of thyroid nodules.
4.Comparison of the Effects on Angiogenesis Activity between Whitmania Pigra Whitman and Hirudinaria Manillensis Lesson Based on a Zebrafish Model

Objective:To investigate and compare the effects of water extracts of Whitmania pigra Whitman and Hirudinaria ma-nillensis Lesson on the angiogenesis of Tg (kdrl:mCherry) zebrafish. Methods:The zebrafish embryos 6-8 hours after fertilization (6-8hpf) were transferred to the culture medium containing Whitmania pigra Whitman or Hirudinaria manillensis Lesson extract at different concentrations, and the culture medium containing the drugs was replaced every 24 h. And then, at 72 hpf, the larvalmorphology and intersegmental vessels were observed under a microscope. The hatchability of 48-and 72-hpf embryos, and the number of intersegmen-tal vessels and the heart rate of 72-hpf juveniles were measured. Results:Compared with the control group, when the concentration of Whitmania pigra Whitmanis was higher than 30μg· ml-1 , and the concentration of Hirudinaria manillensis Lesson was higher than 20μg· ml-1, the number of intersegmental vessels was significantly reduced (P<0. 01). Compared with the control group, at 48 hpf, when the concentration of the drug groups was higher than 40 μg· ml-1 , the hatchability of the two groups significantly decreased ( P<0. 01);at 72 hpf, the hatchability of Whitmania pigra Whitman decreased significantly at the concentration of 100 μg·ml-1 (P<0.01), while the hatchability of Hirudinaria manillensis Lesson decreased significantly at the concentration of 80 μg· ml-1(P <0. 01). There was no obvious yolk sac edema, pericardial edema and spine curvature in the two groups. The heart rate decreased sig-nificantly (P<0. 01), while was still within the normal range. Conclusion:Both Whitmania pigra Whitman and Hirudinaria manillen-sis Lesson have notable anti-angiogenic activity, and the anti-angiogenesis activity of Hirudinaria manillensis Lesson is stronger. They both have effects on the development of zebrafish embryos, while the toxicity is not obvious.
5.Characterization of Distinct T Cell Receptor Repertoires in Tumor and Distant Non-tumor Tissues from Lung Cancer Patients.

T cells and T cell receptors (TCRs) play pivotal roles in adaptive immune responses against tumors. The development of next-generation sequencing technologies has enabled the analysis of the TCRβ repertoire usage. Given the scarce investigations on the TCR repertoire in lung cancer tissues, in this study, we analyzed TCRβ repertoires in lung cancer tissues and the matched distant non-tumor lung tissues (normal lung tissues) from 15 lung cancer patients. Based on our results, the general distribution of T cell clones was similar between cancer tissues and normal lung tissues; however, the proportion of highly expanded clones was significantly higher in normal lung tissues than in cancer tissues (0.021% ± 0.002% vs. 0.016% ± 0.001%, P = 0.0054, Wilcoxon signed rank test). In addition, a significantly higher TCR diversity was observed in cancer tissues than in normal lung tissues (431.37 ± 305.96 vs. 166.20 ± 101.58, P = 0.0075, Mann-Whitney U test). Moreover, younger patients had a significantly higher TCR diversity than older patients (640.7 ± 295.3 vs. 291.8 ± 233.6, P = 0.036, Mann-Whitney U test), and the higher TCR diversity in tumors was significantly associated with worse cancer outcomes. Thus, we provided a comprehensive comparison of the TCR repertoires between cancer tissues and matched normal lung tissues and demonstrated the presence of distinct T cell immune microenvironments in lung cancer patients.
