Chinese Journal of Nosocomiology 2006;0(07):-
OBJECTIVE To establish a method of real-time PCR for detection of mecA gene in meticillin-resistant Staphylococcus aureus(MRSA).METHODS The experiment conditions including Mg2+ concentrations, primers concentrations and probe concentrations were optimized for real-time PCR in the light of factorial design principle.Totally 109 strains of S.aureus were collected and detected respectively in oxacillin disk diffusion method and real-time PCR and MRSA detection rate in the two methods were compared and analyzed.RESULTS Thirty seven MRSA isolates were detected by real-time PCR from the 109 S.aureus strains. The MRSA isolating rate by real-time PCR was conspicuously higher than oxacillin disk diffusion method,by which 27 MRSA isolates were detected.CONCLUSIONS The technology of real-time PCR to rapid identification of MRSA is superior to common PCR and it can make up the deficiency of detecting borderline-resistant strains in oxacillin disk diffusion method.
3.Detection of Meticillin-resistant Staphylococcus aureus by Fluorescence in situ Hybridization and Flow Cytometry
Chinese Journal of Nosocomiology 1994;0(01):-
OBJECTIVE To establish and evaluate detection method of meticillin-resistant Staphylococcus aureus(MRSA) by fluorescence in situ hybridization(FISH) and flow cytometry.METHODS The experiment conditions including bacteria suspension concentrations, probe concentrations, and hybridization temperatures were optimized for FISH test in the light of orthogonal design principle. The fluorescence signals were detected by flow cytometer, and detection results by FISH and flow cytometry were compared with that of real-time PCR.RESULTS The optimal hybridization conditions of FISH detecting MRSA were 40?105 of bacteria suspension concentrations per microliter of hybridization buffer solution, and 4 ng of probe concentrations per microliter of hybridization buffer solution and 50 ℃ of hybridization temperatures. The sensitivity and specificity of detecting MRSA by FISH-FCM were 97.3% and 100.0%,respectively,and detection results between FISHFCM and real-time PCR had not significant deviation.CONCLUSIONS Fluorescence in situ hybridization and flow cytometry, which detect directly mecA gene in MRSA, are a fast and accurate methods for MRSA detection and can apply in clinical laboratory.

Objective To investigate the expression of insulin receptor substrate-2(IRS-2)in the liver of nonalcoholic fatty liver(NAFL)mice. Methods Ten female C57BL6J mice fed with normal diet were served as controls, while fifteen fed with high fat and sugar diet for either 8 weeks(group-A)or 16 weeks(group B)were as NAFL mice models.Abdominal fat mass, total cholesterol(TC),triglycerides(TG),high density lipoprotein cholesterol(HDL-C),fasting blood glucose(FBG)insulin(INS)and liver function, liver weight were measured. And the expression of IRS-2 and the content of lipid in the liver were also detected. Results The abdominal fat mass serum TC,and FBG were significantly higher in high fat and sugar diet fed mice than that in controls(P

Objective To optimize the extraction process by determing the volatile oil yield of steam distillation and supercritical CO2 extraction from flowers of Elaeagnus angustifolia L..Methods Orthogonal test was carried out to compare two extraction effects between steam distillation and supercritical CO2 extraction by detecting the extraction ratio from flowers of Elaeagnus angustifolia L..Results Steam distillation time on volatile oil extraction has the greatest impaction, and the best condition of supercritical CO2 extraction was extraction pressure of 20 MPa,extraction temperature of 40 ℃,and flux of CO2 is 10 L/h.Conclusion Compared with steam distillation, supercritical CO2 extraction spends less time, has higher efficiency and less harm to the materiel after extracting. It adapts to batch process and is convenient for subsequent extraction technology.

Objective To investigate the laws of the treatment of diabetic retinopathy with traditional Chinese medicine. Methods By searching domestic literature of treating DR with traditional Chinese medicine in CNKI from 2000 to 2010, made primary summary and analyzed. Results 85 eligible published documents were found involving 147 medicinal herbs. Herbs with the functions of tonifying, activating blood circulation and eliminating stasis and heat were mostly seen.Cases reported in literature were mainly non-proliferative lesions, occupying a ratio of 58.82%;drug observation time was mainly 12 weeks, occupying a ratio of 44.17%. Conclusion Asthenia, statis and heat were pathological factors existing throughout the whole process of DR. Invigorating, romoting blood circulation, and, cooling blood should be the basic therapeutic methods.

Objective To optimize the technological conditions of polysaccharide extraction from Elaeagnus angustifolia L. by enzymatic-assisted ultrasonic. Methods The effects of different enzymes, enzymolysis temperature, enzyme dosage, enzymolysis time, material-water ratio, crushing degree, ultrasonic time and ultrasonic temperature on the extraction rate of Elaeagnus angustifolia L. polysaccharide was analyzed. Results The optimal technological condition was as follows:1.5% cellulase, 1.5% pectic enzyme, enzymolysis time 40 min, enzymolysis temperature 55 ℃, ultrasonic time 25 min, ultrasonic power 400 W, material- water ratio 1∶30, and temperature 60 ℃. In these conditions, the extraction efficiency of polysaccharide was 12.35%. Conclusion The process is simple and practicable, and can be used for the extraction of Elaeagnus angustifolia L. polysaccharide.

Chinese Journal of Immunology 2000;0(09):-
Objective:To explore the effects of myofibroblast in the pathogenesis of oral submucous fibrosis(OSF).Methods:The expressions of ?-SMA in the tissues of OSF and normal control were detected by immunohistochemistry. Fibroblasts were isolated from oral mucosa of OSF patients and healthy controls and were cultured under stimulation by arecoline. The expressions of ?-SMA in OSF fibroblasts and normal fibroblast were detected by immunocellchemistry and RT-PCR.Results:(1)No positive expression was found in normal tissues except in the blood vessel, but in OSF tissue,?-SMA positive expression was found in many spindle cells in the oral submucousal layer.(2)Few fibroblasts in vitro from normal controls were ?-SMA positive.(3)Most of the second passage of OSF fibroblasts were ?-SMA positive(85.80%?3.56%). (4)The expression of ?-SMA in fibroblasts could not be increased by the stimulation of arecoline. Conclusion:Myofibroblasts may play an important role in the pathogenesis of OSF,and the appearrence of myofibroblasts in OSF may not be directly induced by the stimulation of arecoline.

Chinese Journal of Immunology 2001;0(10):-
Objective:In order to elucidate the derivation and differentiation mechanisms for derivation of myofibroblasts in oral submucous fibrosis. Methods:Oral keratinocytes and fibroblasts were isolated and cultured, and a co-cultur system composed of the keratinocytes and fibroblasts was established in vitro.Expressions of ?-SMA protein and mRNA in fibroblasts were detected by immunohistochemistry and RT-PCR. The level of TGF-?1 in conditioned medium were evaluated by ELISA.Results:The expressions of ?-SMA mRNA and protein in fibroblasts co-cultured with keratinocytes preprocessed by arecoline was higher than that in fibroblasts co-cultured with keratinocytes and without preprocessed by arecoline. The level of TGF-?1 in conditioned medium of fibroblasts-keratinocytes preprocessed by arecoline co-cultures was higher than those of fibroblasts cultured alone. Conclusion:The derivation of myofibroblast from FB can be promoted by keratinocytes preprocessed by arecoline and TGF-?1 may play a role in the process.
