Chinese Journal of Ultrasonography 2001;10(1):19-21
Objective To examine the left ventricular diastolic function of patients with coronary artery diseases by noninvasive echocardiography and invasive coronary angiography. Methods Mitral and pulmonary venous Doppler signals were recorded by the continuous wave Doppler technique in 62 patients, both coronary angiography and left ventricle pressure were measured. Patients were divided into normal and abnormal groups based on coronary angiography. Results Left ventricular end diastolic pressure (LVEDP) (17.6 mm Hg vs 13.2 mm Hg,P<0.05),the left ventricular negative Δp/Δt before atria contraction [-(224.63±52.18),-(87.81±24.41),P<0.05],and the elevated value of left ventricular pressure derived from atria contraction [(6.66±0.92)mm Hg vs (0.96±1.87)mm Hg,P=0.0094] in abnormal group were more than those in the normal. The ratio of slope in pulmonary vein and mitral A wave peak was less in normal group than that in the abnormal (5.10±0.31 vs 4.12±0.20,P=0.0106). But the isovolume relaxation parameters had no differences between the two groups. Conclusions Coronary arter ischemia is an important cause of left ventricular impaired relaxation aggravation and stiffness, the diastolic abnormalities existed in the whole diastolic duration. Except ischemia, other factors also could cause relaxation disfuction and even elevate the LVEDP.

Objective The purpose of this study was to evaluate the effects and mechanism of ACEI(Fosinopril),AT1 receptor antagonism (Irbesartan),and combination of these two drugs on ventricular remodeling in rats with myocardial infarction. Methods Rats were randomly divided into four groups at twenty-four hours after MI,and treated for 6 weeks.Mean blood pressure and left ventricular end diastolic pressure(LVEDP) were evaluated,as well as ventricular weight(VW)/body weight(BW) ratio. The total, type I and type Ⅲcollagen and nonmyocyte cellular proliferation were quantified by histomorphometry. The expression of transforming growth factor-beta 1(TGF-? 1)mRNA within the noninfarction zone was determined by in situ hybridization. Results Irbesartan and combination therapy decreased total collagen more significantly compared with fosinopril.Combination therapy decreased typeⅠcollagen more significantly than fosinopril.Nonmyocyte cellar proliferation in MI-combination group was more significantly suppressed than in MI-fosinopril group.Treatment with irbesartan or combination therapy normalized TGF-? 1 mRNA level within the noninfarction zone. Conclusion Fosinopril or irbesartan alone ,and combination of these two drugs can limit myocardial hypertrophy, attenuate the development of myocardial interstitial fibrosis, and prevent nonmyocyte cellar proliferation in the noninfarcted left ventricle. The use of irbesartan, especially combined with fosinopril was more effective than fosinopril alone in the suppression of histopathologic changes resulting in ventricular remodeling after MI.Irbesartan and combination therapy was more effective than fosinopril alone in suppressing TGF-? 1 mRNA expression.

Objective To investigate the action and the mechanisms of nerve growth factor(NGF) and its receptor(p75、TrkA) on the pathological alterations of the enteric nervous syst em in the rats cathartic colon. Methods The experimental animal model of cathartic colon was established by using rhubar b and phenolphthalein. Thirty Sprague Dawley (SD) rats were randomly divided int o the control group(10 rats) and cathartic colon model groups(10 rats with rhuba r b group; 10 rats with phenolphthalein group). The model group rats were fed with stimulant laxatives rhubarb and phenolphthalein respectively, and the control g roup rats were fed with distilled water. The rats were killed and the colon samp les were taken after 3 months. The expressions of the mRNA of NGF, p75 and TrkA in the colonic tissues were detected by semi-quantitive reverse transcription- polymerase chain reaction(RT-PCR) technique. Results The expressional levels of NGF mRNA were lower in the rhubarb group and phenolph thalein group than that in control group (P

Objective To study the expression of Akt and MAPK in the stomach and colon of slow transit constipation (STC) in rats, as well as the effect of exogenous glial cell line-derived neurotrophic factor (GDNF) on it. Methods Forty-four SD rats were divided into control group and model group randomly. The STC model group was established by gastric irrigation of rhubarb for 3.5 months. The control group was received normal saline. After model building, each group was equally divided into 2 subgroup randomly, administrated with exogenous GDNF and normal saline by vein injection for one week respectively. The expression of Akt and MAPK in stomach and colon was detected by immunohistochemistry. Results ( 1 ) The expression of Akt in the stomach tended to weaker in STC rats comparing with the normal rats ( P > 0. 05 ), but it was stronger in STC plus GDNF group than in STC group ( P < 0. 05 ). ( 2 ) The expression of Akt and MAPK in the colon was weaker in STC group than in the normal group ( all P <0. 05 ), and was stronger in STC plus GDNF group than in STC group ( all P < 0. 05 ). ( 3 ) The expression of MAPK in the stomach in STC group was weaker than in normal group (P < 0.05 ), and was stronger in STC plus GDNF group than in STC group (P <0.01 ). There was no significant difference among STC plus GDNF group, normal group and GDNF group (P > 0. 05 ). Conclusions Long term consumption of rhubarb could induce STC by down-regulating the expression of Akt and MAPK in digestive tract. Exogenous GDNF may have a potential role on the etiology of STC.
