1.Treatment of Chloasma by Combination of Acupuncture and Herbs

Purpose: To investigate the therapeutic effect of acupuncture and herbs in chloasma.Methods: Chloasma was divided into three patterns and treated by acupuncture plus oral administration and external application of Chinese herbs. Results: There was an obvious therapeutic effect in chloasma treated by combination of acupuncture and herbal drugs. After two courses of the treatments, the effective rate accounted for 93.3% in 45 cases. Conclusion: There was an obvious therapeutic effect in chloasma treated by combination of acupuncture and herbal drugs.
2.Simultaneous Determination of 4 Components in Compound Metronidazole Clindamycin Cream by HPLC

OBJECTIVE:To establish the method for simultaneous determination of the content of metronidazole,clindamycin hydrochloride,spironolactone and vitamin B6 in Compound metronidazole clindamycin cream.METHODS:HPLC method was adopted.The determination was performed on Diamonsil C18 column with mobile phase consisted of acetonitrile-[0.68% dipotassium hydrogen phosphate solution (containing 1% triethylamine)] (gradient elution) at the flow rate of 1.0 mL/min.The detection wavelength was set at 210 nm,and column temperature was 25 ℃.The sample size was 20 μL.RESULTS:The linear ranges of metronidazole,clindamycin hydrochloride,spironolactone and vitamin B8 were respectively 20.0-120.0 i.tg/mL(r=0.999 9),10.0-60.0μg/mL(r=0.999 1),4.0-24.0 μg/mL(r=0.999 4),10.0-60.0 μg/mL (r=0.999 4).The quantitative limits were respectively 13.06,7.36,2.43,6.42 μg/mL.The detection limits were 4.18,1.82,0.76,1.79 μg/mL.RSDs of the precision,stability,repeatability tests were all less than 2%.The recovery rates were respectively 98.97%-100.91% (RSD=0.79%,n=9),99.02%-101.80% (RSD=1.03%,n=9),98.18%-101.05% (RSD=1.01%,n=9) and 98.75%-101.36% (RSD=0.87%,n=9).CONCLUSIONS:The method is simple,rapid,accurate and can be used for the determination of metronidazole,clindamycin hydrochloride,spironolactone and vitamin B6 in Compound clindamycin metronidazole cream.
3.Clinical Efficacy of Cold Knife Conization and Transcervical Resection of Cervical Lesionin the Treatment of Cervical Intraepithelial Neoplasia Ⅱ, Ⅲ

Objective:To compare the clinical efficacy of cold knife conization and transcervical resection of cervical lesion in the treatment of cervical intraepithelial neoplasia(CIN) Ⅱ,Ⅲ.Methods:The clinical data of 62 patients with CIN Ⅱ,Ⅲ underwent cold knife conization(CKC group) and 114 underwent transcervical resection of cervical lesion(TCRC group)in Zhujiang Hospital Southern Medical University from Jan 1 st 2010 to Jun 1 st 2014 were collected.The patients' age,pregnant frequency,mean operative time,preoperative pathological grading,blood loss during the operation and after the operation,positive resected margin,cervical stenosis and recurrence were compared between the two groups.Results:There was no difference on the patients age,pregnant frequency and preoperative pathological grading between the two groups(P > 0.05).The mean blood loss during the operation (14.16 ± 14.10 ml)and the mean hospital stays(5.73 ± 1.43 days)of CKC group were significantly different from those of TCRC group(29.80 ±20.55 ml,4.75 ±1.23 days) (P<0.05).There was no statistical difference on the mean operation time,postoperative vaginal bleeding rate,rate of positive resected margin,cervical stenosis rate and recurrence rate between CKC group and TCRC group (33.15 ± 11.42 min vs 33.18 ±14.61 min,33.9% vs 21.1%,3.2% vs 5.3%,4.8% vs 4.4%,4.8% vs 3.5%,respectively) (P>0.05).The concordance rate of pre-operation and post operation pathological results of CKC group and TCRC group was 74.2% (46/62) and 72.8% (83/114) respectively,both kappa values were < 0.75.Conclusions;Both CKC and TCRCT are safe as well as effective in the treatment of high-grade cervical intraepithelial neoplasia.Compared with CKC,TCRC has the advantage of short hospital stay,while much intraoperative blood loss,which calls for intraoperative attention.
4.Clinical research in prevention and cure effect of anisodamine gel on phlebitis caused by intra-venous infusion of KLT

Objective To investigate the prevention and cure effect of anisodamine gel on phlebitis caused by intravenous infusion of KLT. Methods 96 patients with intravenous infusion of KLT were divided into the experimental group and the control group, 48 patients in each group, and the skin at the top of transfusion puncture points of the patients of the experimental group was evenly embrocated with 2% anisodamine gel along the vein before 3 to 5 minutes of intravenous infusion of KLT, while the control group was without any treatment. After observing the situation of venous injury, the skin at the top of transfusion puncture points of the positive patients would be embrocated with 2% anisodamine gel. The incidence, degree of phlebitis and treatment effect were compared between the two groups. Results The incidence rate of phlebitis of the control group was 68.7%, of which grade II, III occupied 60.4%, while the incidence rateof phlebitis of the experimental group was 10.4%, in which there was no occurrence of grade II, III phlebitis. All the venous injury symptoms of 33 patients of the control group and 5 patients of the experi -mental group disappeared after treatment, and after statistical processing, the incidence rate of phlebitis in the experimental group and the control group showed significant differences. Conclusions The embrocating of 2% anisodamine gel can effectively avoid the occurrence of the phlebitis caused by intravenous infusion of KLT.

6.Screening and culture of human vaginal mucosa stem cells

BACKGROUND: Nowadays stem cells have been isolated successfully from the epithelial tissue of human and rodents such as skin, hair follicle, cornea, oral mucous, intestinal mucosa dental epithelial cells, and salivary glands by the Chinese or foreign researchers. But according to the vaginal mucosa stem cells (VMSCs), it is not known in the field of isolation and application.OBJECTIVE: To find out the suitable method of isolation, identification, and culture of human VMSCs.DESIGN: Observational basic study. SETTING: Department of Gynaecology and Obstetrics, the Third Affiliated Hospital of Guangzhou Medical College. MATERIALS: This study was performed in the Zhongshan Ophthalmology Center of Zhongshan Medical College from October 2004 to August 2005. Two-week gestation mice were provided by the Experimental Animal Center of Guangzhou University of Chinese Medicine. The sterile human vaginal mucosa was collected from five patients (25-38 years old) after the plastic operation of vagina from the 3rd Affiliated Hospital of Guangzhou Medical College. METHODS: Vaginal mucosa cells could be harvested from human vagina mucosa treated with trypsin-collagenase digesting. The VMSCs were selected by adhesion to type Ⅳ collagen for 20 minutes. The cell cycles of the VMSCs and primary human vaginal mucosa cells were analyzed by flow cytometry. VMSCs were cultured in vitro with 4 different media, including feeder layer cells plus epidermal total culture media, feeder layer cells plus DMEM/F12 (3:1), epidermal total culture media and DMEM/F12 (3:1). At the 12th day, their colony forming efficiency (CFE), CK19 and CK10 positive rates were compared. Then VMSCs were continuously cultured and passaged till they were apoptosis.MAIN OUTCOME MEASURES: ① Positive rate of CK19 and CK10 and G0/G1 proportion; ② Positive rate of CK19 and CK10, the colony forming efficiency (CFE) and the times be passaged when primary VMSCs were cultured in different media after 12 days. RESULTS: The cells at the G0-G1 phase cell cycle, CK19 and CK10 positive rates between the VMSCs and primary human vaginal mucosa cells were statistical different (P < 0.05). When cultured at 12th day, the VMSCs cultured with feeder layer and epidermal total culture media had the highest CFE and CK19 positive rate and lowest CK10 positive rate. They could be passaged over 15 times in vitro. CONCLUSION: It is an effective way that select VMSCs by adhesion to type Ⅳ collagen for 20 minutes. In this research, culturing VMSCs in vitro with feeder layer and epidermal total culture media was the best way to keep VMSC characteristics.
7.Research in prevention and treatment of venous injury caused by intravenous infusion of Delisheng with anisodamine gel

Objective To investigate the effect of prevention and treatment of venous injury caused by intravenous infusion of Delisheng with anisodamine gel.Methods 100 patients with intravenous infusion of Delisheng were randomly divided into the experimental group and the control group,50 patients in each group,and the skin at the top of transfusion puncture points of the patients of the experimental group were evenly embrocated with 2% anisodamine gel along the vein before 3 to 5 minutes of intravenous infusion of Delisheng,while the control group received no treatment.After observing the situation of venous injury,the skin at the top of transfusion puncture points of the positive patients was embrocated with 2% anisodamine gel,and the effect on venous injury was observed.Results The incidence rate of intravenous pain of the experimental group was 4%,there wasn't any occurrence of poor intravenous infusion or swelling of the infusion position.The incidence rate of poor intravenous infusion of the control group was 34% and phlebitis incidence rate was 42%,and intravenous pain incidence rate was 68%.All the venous injury symptoms of 39 patients of the control group and 2 patients of the experimental group disappeared after treatment.Conclusions The embrocating of 2% anisodamine gel can effectively prevent and treat the venous injury caused by intravenous infusion of Delisheng.
8.Evaluation of the effect of family-centered empowerment education of enhancing breastfeeding behavior in primiparas

Objective To evaluate the effects of breastfeeding behavior through the implementation of family-centered empowerment education in primiparas.Methods According to random digit table,128primiparas in our hospitals were divided into the experimental group and the control group with 64 cases in each group from August 2014 to April 2015.At last 58 patients in the observation group and 59 patients in the control group completed the research.Those primiparas in the experimental group were given the familycentered empowerment education.The control group received conventional methods.Results The scores of breastfeeding self-efficacy,perceived social support and breastfeeding behavior of primiparas in the experimental group were 129.64±12.94,58.94±8.97,89.84±11.32,higher than the control group,101.78±78.67,50.27±10.56,71.56±12.03,the differences had statistical significance between two groups,t=9.656-10.487,P < 0.05.Conclusions Using the family-centered empowerment education can effectively improve the breastfeeding self-efficacy,perceived social support and breastfeeding behavior in primiparas.
9.Human umbilical cord blood mononuclear cell transplantation for treatment of radioactive premature ovarian failure in nude mice

BACKGROUND:Stem cel transplantation, in recent years, has become a preferred treatment for premature ovarian failure. Umbilical cord blood mononuclear cel s containing a large number of mesenchymal stem cel s, immature stem/progenitor cel s, and endothelial progenitor cel s can be used as an important source of stem cel transplantation.
OBJECTIVE:To study the therapeutic efficacy of human umbilical cord blood mononuclear cel transplantation on radioactive premature ovarian failure in nude mice.
METHODS:120 female BALB/C nude mice were randomly divided into four groups:blank control group without any intervention;model group, intravenous transplantation group, and in situ transplantation group exposed to 60Coγrays, 0.5 Gry per day, for 30 days. After 30 days, premature ovarian failure models were made in the latter three groups. Then, nude mice in the latter three groups were given bilateral ovary injection of 10 μL DMEM, tail vein injection of 10 μL human umbilical cord blood mononuclear cel s (1×1013/L), and bilateral ovary injection of 10 μL human umbilical cord blood mononuclear cel s (1×1013/L), respectively. Thirty days after cel transplantation, serum levels of estradiol, fol icle hormone, luteinizing hormone, inhibin B and vascular endothelial growth factor were detected, cel apoptosis in the ovary tissue and cel survival were observed pathological y.
RESULTS AND CONCLUSION:After modeling, the serum levels of estradiol, inhibin B and vascular endothelial growth factor were significantly reduced (P<0.01), while fol icle hormone and luteinizing hormone levels increased (P<0.01). After transplantation, these indexes were al improved in the in situ transplantation group (P<0.01), and reduced fol icle hormone and luteinizing hormone levels were visible in the intravenous transplantation group (P<0.05, P<0.01). Compared with the model group, in situ transplantation and intravenous injection of human umbilical cord blood mononuclear cel s could effectively reduce cel apoptosis in the ovary tissue (P<0.05), and transplanted cel s were able to survive in the ovary of nude mice. Al these findings show human umbilical cord blood mononuclear cel transplantation do have curative effects on premature ovarian failure in nude mice through the inhibition of apoptosis and the regulation of hormone secretion.
10.Laparoscopic regular hepatic left lateral lobectomy:A report of 15 cases

1 cm. Postoperative pathological findings showed negative cut surface. No hemorrhage or bile leakage were seen after operation. The abdominal drainage tube was indwelled for 3~4 d. The postoperative hospital stay was 3~5 d. Follow-up checkups for 1~20 months (mean, 12.5 months) revealed no tumor recurrence or incision implantation. All the patients survived. Conclusions Laparoscopic regular hepatic left lateral lobectomy can be carried out safely and effectively in clinically selected patients.
