2.Identification of Fritillaria by near-infrared diffuse reflectance spectroscopy

AIM: To establish an identification method of Fritillaria based on near-infrared diffuse reflectance spectroscopy(NIRDRS). METHODS: Seven certified Fritillaria,i.e.,F.przewalskii Matim,F.cirrhosa D.Don,F.unibraacteate Hsiaaoet.K.C.Hsia,F.thunbergii Mig,F.pallidiflora,F.ussuriensis Matin and F.kupehensis Hsiaet K.C.Hsia,and three fake "Fritillaria",i.e.,F.thunbergii Varchekiugensis Hsiaet K.C.Hsia,Tulipa edulis and Iphigeniaindica were dried,grinded and sieved before their NIRDRS spectra were detected.Cluster analysis,convolution transform-visualization-similarity analysis were further employed to(identify) the NIRDRS of the ten herbal medicines. RESULTS: The zero-order NIRDRS spectra of the ten herbal medicines were too similar to be identified respectively.The cluster analysis helped differentiate the three fake "Fritillaria" from seven certified Fritillaria,but the identification between the certified Fritillaria remained dissatisfactory.The convolution transform-visualization-similarity analysis magnified and then quantified the minute differences between the certified Fritillaria,which significantly improved their identification. CONCLUSION: NIRDRS is a direct,non-destructive analytical method of solid samples.Coupled with information treatment techniques,DIRDRS provides a new identification method of crude traditional Chinese medicines.
3.Construction and identification of a double-regulated replicative adenovirus AdTPHre-hEndo

Objective To develop a double-regulated replicative adenovirus carrying the Human endostatin gene(hEndo). Methods The plasmid pTPHre-hEndo was constructed by gene engineering technique, carrying human endostatin gene, in which El A gene and E1B gene were driven by human telomerase reverse transcriptase (hTERT) promoter and hypoxia response element (HRE) promoter,respectively. The pTPHre-hEndo was co-transfected with pBHGE3 in 293 cells to generate recombinant adenovirus AdTPHre-hEndo. Virus titer was measured by the TCID50 method. Virus replication assay was performed to evaluate the selective replication ability of AdTPHre-hEndo. The transgene expression of endostatin was detected by ELISA assay. Results A novel gene-viral therapeutic system AdTPHre-hEndo was constructed by gene engineering technique and its titer was 3. 25 X 1010 pfu/ml.Proliferative test revealed that AdTPHre-hEndo could proliferate selectively in telomerase positive tumors. Furthermore, in comparison with non-replicative adenovirus Ad-hEndo, the transgene expression of endostatin mediated with AdTPHre-hEndo was significantly increased (P < 0. 01).Conclusion The novel gene-viral therapeutic system AdTPHre-hEndo has the capacity to replicate in pancreatic cancer cells and expresses the endostatin efficiently, and may provide a new strategy for pancreatic cancer gene therapy.
4.Effects of mesalazine combined with bifico on the treatment of ulcerative colitis and on TLR4 and MyD88

Objective To investigate the clinical efficacy of mesalazine combined with bifico in the treatment of ulcerative co -litis (UC) and its effect on toll-like receptor 4 (TLR4) and myeloid differentiation factor 88 (MyD88).Methods A total of 86 cases of UC was randomly divided into the observation and control groups with 43 cases in each group .The observation group was given me-salazine combined bifico treatment , and the control group was given mesalazine , and eight weeks as a course of treatment .The clinical efficacy was observed .Serum TLR4 and MyD88 were tested.Adverse reaction in treatment was recorded .Results The total effective rate (95.35%) in observation group was significantly higher than that (81.40%) in control group( P <0.05).The serum TLR4 and MyD88 after treatment of two groups were significantly lower than that before treatment , respectively ( P <0.05 or P <0.01 ) , and the decline in TLR4 of observation group was superior to that of control group ( P <0.05 ) .The difference of adverse reaction rate in two groups was not significant ( P >0.05 ) .Conclusions Mesalazine combined with bifico in the treatment of UC is safe and effective . The effect may be related to regulation of peripheral blood TLR 4 and MyD88 content .
5.Clinicopathologic significance of structural alterations of p53 protein in papillary thyroid carcinoma.

OBJECTIVETo make a thorough study on the clinicopathologic significance of the three-dimensional structural alteration of the p53 protein in papillary thyroid carcinomas and to provide an objective criterion for the evaluation of PTC prognosis.
METHODSA total of 41 PTC cases were enrolled. Techniques including polymerase chain reaction with single strand conformational polymorphism (PCR-SSCP), DNA sequencing, computerized three-dimensional protein modeling by means of international shared resources and related software analysis were used.
RESULTS15 cases with p53 gene mutation defined as Group I were detected in totally 41 PTC cases. No p53 gene mutation was found in the rest 26 cases which were classified as Group II. The differences in lymph node metastatic rate, distant metastatic rate, age, sex, size of the lesion between Group I and Group II were not significant (P > 0.05). The alterations of the amino acid residues of 9 PTC cases out of the 15 p53-gene mutated patients (Group I) were either located in the p53-protein domains, mainly the core domain and the non-specific DNA binding basic domain, or the severely defect cases with the formation of widely divergent structures. It was found that the alterations of the structure of the core domain could directly check the binding of p53 protein to its target DNA molecules. In addition, the alterations of the structure of the basic domain could indirectly prohibit the binding. The ones mentioned above were classified as Group Ib. The rest of six cases with their p53 protein amino acid residues mutated beyond the domains were grouped as Group Ia. The differences in lymph node metastatic rate, distant metastatic rate between Group Ib and Group Ia were statistically significant (P < 0.01, P < 0.05) respectively.
CONCLUSIONSThe alterations of the three-dimensional structure of p53-protein is considered as one of the morphological basis of the progression and heterogeneity of PTC. They render an authentic evidence for the selection of the clinical cases with a poor risk for metastasis.

6.To improve the mechanical function of bovine jugular vein by external stent

Objective To increase the mechanic characteristic of bovine jugular veins conduit (BJVC) by combined knitted stent outside made of nylon fiber. Methods Get rid of the cells of BJVC by enzymolysis, then combined a fabric stent with a acellular bovine jugular vein conduit that cross-linked dealed by photooxidation method to improve the mechanic function of BJVC. To compare the mechanical function and histology change of BJVC combined with fiber stent outside with that of BJVC only without stent. BJVC combined with fiber stent outside was divided into experiment group and the latter into control group. Two roller pumps on cardiopulmonary bypass unit were designed to simulate the pulmonary circulation. One produce the stream, and the other assist to bring a certain pressure by regulating a degree of tightness. The pressure of forward stream was used in messuring extension rate and leakage volume of two BJVC groups, and the pressure of backward stream was used in messuring the regurgitation volume. Results Set the formula to calculate the rate of variation of BJVC diameter under a certain pressure or not. The experiment group extension rate was zero under pressure of 10-20 mm Hg, about ( 7.7 ± 2.3 ) % under pressure of 40 mm Hg, until 60 mm Hg to ( 11.4 ± 3.1 ) %. While the control group reached about ( 14.8 ± 4.7 ) % at 20mm Hg already. There was no regurgitation volume of the experiment group under pressure of 30 mm Hg vs. the control group' s reached 34ml/min at 30 mm Hg while the two groups were same under pressure that lower than 20 mm Hg. When the pressure higher, the difference between the two groups more notable. There was no leakage of the BJVCs with fabric stent till pressure from 40 mmHg to 60mmHg, vs. the control group 1,5, 13 ml/min at pressure 30, 40, 60 mm Hg. Except the regurgitation and leakage volumes under pressure at 10 -20 mm Hg, as the two numerical values were both zero that can not be caculated by the statistic software, the differences of the two groups above had statistics significance. In vivo, 9 dogs received bovine jugular vein conduit transplant procedure to observe the mechanic fnction and tissue reaction. Five adopted B.JVC with stent and four just BJVC only. Six month later after the transplant procedure, there was no significant difference between the two groups but more inflaming cells than the former group, especially at the location round the nylon fibers. Through the observation in HE stain, that was considered as foreign body reaction. Conclusion Fabric stent can improve the mechanical function of BJVC and relatively easy to get.
7.The analysis of blood transfusion and hospitalization duration between the occlusion of ventricular septal defect through transesophageal echocardiography and the repair of ventricular septal defect

Objective To compare the rate of blood transfusion and hospitalization duration between the occlusion of ventricular septal defect through transesophageal echocardiography and the repair of ventricular septal defect.Methods A total of 131 consecutive patients were included in the study from July of 2011 to December of 2011,75 patients of them received the occlusion of ventricular septal defect through transesophageal echocardiography,and 56 patients of them received the repair of ventricular septal defect.Results All of 131 patients had discharged without death.The group of receiving the repair of ventricular septal defect presented the higher rate of blood transfusion(x2 =81.83,P < 0.01) and longer hospitalization duration than the other(t =46.57,P < 0.01).Conclusions The group of receiving the occlusion of ventricular septal defect through transesophageal echocardiography had the lower rate of blood transfusion and shorter hospitalization duration.
8.ISSR Analysis for genetic diversity of Coptis chinensis

Objective To discuss the genetic diversity of Coptis chinensis.Methods The genetic diversity of 78 individuals from seven populations was analyzed by inter-simple sequence repeat(ISSR).Results Twelve primers were selected to produce highly reproducible ISSR bands.Among 106 amplified bands,72 showed polymorphism,the percentage of polymorphic bands reached to 67.92%.Nei's gene diversity index(H)was 0.180 3,Shannon information index(I)was 0.283 2,Gst was 0.681 5.The genetic distance coefficient and genetic similarity were 0.089 4—0.184 6 and 0.832 1—0.912 7,respectively.ConclusionC.chinensis holds high genetic diversity and the majority of genetic variation occurs among the populations.By cluster analysis,the geographical distribution is very obvious.The ISSR marker could be used for the analysis of the genetic diversity and genetic variation of C.chinensis.
9.Construction and tobacco transformation of COR and BBE genes hairpin RNA vector of Papaver somniferum.

The gene expressions of codeinone reductase (COR) and berberine bridge enzyme (BBE) in Papaver somniferum were blocked by RNA hairpin of RNA interference (RNAi). The complete sequences of COR and BBE genes were cloned by reverse transcription-polymerase chain reaction (RT-PCR), the results of homology comparison revealed that the cloned COR and BBE genes had high homology with the other gene family members reported in the GenBank. The target sequences of COR and BBE genes were screened in accordance with the design principle of RNAi, a 643 bp fusion gene was obtained by the method of overlapping PCR, then plant expression vector ihpRNA was constructed based on intermediate vector pHANNIBAL and plant expression vector pCEPSPS. With that 78 transgenic plants were obtained through Agrobacterium-mediated and 17 positive plants were screened by PCR, that could initially indicate that the target fragments of COR and BBE gene had been integrated into tobacco genome.
10.Research progress of micro-RNA in intervertebral disc degeneration

Low back pain is becoming an important factor affecting people's quality of life, while the age of its onset is getting younger and younger, and the social and economic losses caused by low back pain are huge every year. Intervertebral disc degeneration (IDD) is an important cause of low back pain. Due to multiple factors, biomechanical and structural changes occur in intervertebral disc tissue, including rupture of annulus fibrosus, protrusion of nucleus pulposus, which cause compression of spinal cord and nerve root, and lower back pain. Micro-RNA (miRNA) is a kind of single-stranded non-coding small molecule RNA, with 18-24 nucleotides in length, which exists widely in eukaryotes. As one of the important regulatory molecules of gene expression, it has been proved to play a key role in the initiation and progression of many diseases, and it may also play an important role in intervertebral disc degeneration. At present, the clinical treatment for IDD is mainly surgical treatment to alleviate clinical symptoms. Even if surgical treatment can achieve good results, it will bring great physical trauma and economic burden to patients. The role of miRNA in IDD is one of the hotspots in the current academic research. Studies have shown that miRNA has abnormal expression patterns in degenerative intervertebral disc tissues and participates in a variety of pathological processes of IDD. At present, some miRNAs have been proved to be related to a variety of pathological processes in IDD, including nucleus pulposus cell apoptosis and proliferation, extracellular matrix degradation, autophagy, inflammation and cartilage endplate degeneration. The comparative study of gene chip showed that there were significant differences in the expression of some miRNAs between degenerative and normal nucleus pulposus cells. These differentially expressed miRNAs may be involved in the process of nucleus pulposus cell degeneration by regulating their respective upstream or downstream pathways. Most of the regulatory pathways are crossed and parallel, thus constructing a huge miRNA regulatory network. Understanding the target genes and mechanisms of miRNA in the pathogenic process can provide an important reference for the origin, development and prognosis of IDD. In this article, the important role of miRNA in IDD and the potential significance of clinical treatment are reviewed. With the in-depth study of miRNA and the molecular biological mechanism can provide new ideas for the diagnosis and treatment of IDD, which is likely to become a new strategy for biological treatment of IDD.
