Objective:To assess the surgical outcome of patients with mesenchymal chondrosarcoma (MCS) treated in our insti-tute. This study was also designed to describe the clinical characteristics, treatment, and outcome of MCS to provide a better understand-ing of its clinical management. Methods:A total of 27 patients with MCS were treated in Peking University People's Hospital, Beijing, China from October 1997 to March 2011. Demographic information and follow-up data were obtained and statistically analyzed. Re-sults:Among the 27 patients, 9 were males and 18 were females with a mean age of 30.4 years (ranging from 14 years to 51 years). The median follow-up time was 42.6 months (ranging from 6 months to 104 months). Among the total number of tumor cases, 22 and 5 were detected in bone tissues and extra-skeletal sites, respectively. A total of 25 patients underwent surgery, but only 17 achieved the standard surgical margin of wide excision. Among these patients, 16 and 13 were subjected to chemotherapy and irradiation. The three-and five-year survival rates were 65%and 49.5%, respectively. Conclusion:MCS is a rare tumor resulting in morbidity with local recur-rences and long-term metastases. In this study, standard multimodal regimens were proposed to treat MCS. The results recommended wide resection with suitable surgical margins as the preferred treatment. However, further studies should be conducted because the infor-mation about the benefits of chemotherapy and radiotherapy for the control of local or systemic symptoms of MCS remains insufficient.

BACKGROUND: The source of conspecific heart transplantation organ is very limited and deficiency becomes more and more obvious.OBJECTIVE: To observe the mechanical properties of ascending aorta at one-dimensional loading between healthy persons and swine of different months, so as to provide necessary biomechanical experimental basis for anastomosing blood vessel in heterogeneic heart transplantation from swine to human.DESIGN: Open design SETTING: Staff Room of Anatomy, Yunyang Medical College MATERIALS: This experiment was carried out at the Laboratory of Biomechanics, Yunyang Medical College from April 2002 to July 2003.Ascending aortas of human were obtained from the 6 adult male corpses without cardiovascular diseases, aged 18 to 30 years, who died for accident and donated by Yunyang Medical College. Totally 42 conspecific swine of 1 month old, with certification number of QN0202, were provided by Animal Experimental Center of Yunyang Medical College. They were raised with common foodstuff. The 42 swine were butchered respectively at 1,2,3,4,5,6 and 7 months, 6 swine once. After anatomical isolation and in situ measurement of respective in vivo length, ascending aorta from aorta valve ring base plane to initiation part of innominate artery was taken out (Artherosclerosis was not presented in all the samples) and divided into five equal segments, and the second and fourth segments were used for mechanical test of one-dimensional loading.METHODS: Six adult male corpses and ascending aorta of 42 swine of 1 to 7 months were performed mechanical test of one-dimensional loading. All the blood segments were pre-treated ten times with the same strain rate at room temperature 32 ℃(loading range from 0 to 0.5 N). Hysteresis disappeared after blood vessel was given periodic permanent loading and unloading, and repeated force-deformed data were obtained. Blood vessel was given loading and unloading once with the same loading range and strain rate. The recorded force-deformed data were used for computer analysis. Force and displacement were scaled with standard weight and percentage scale at the end of each time. Constant α and elastic modulus Dt/dλ were fitted from experimental data.MAIN OUTCOME MEASURES: ① Comparison of the mechanical characteristic constant of ascending aorta at one-dimensional loading be tween human and swine of different months. ② Comparison of elastic mod ulus of ascending aorta at one -dimensional loading between human and swine of different months .RESULTS: ① Comparison of the mechanical characteristic constant of ascending aorta at one-dimensional loading between human and swine of different months: With the increase of month, although the material con stant of blood vessel of swine had a little increase, there was no significant difference (P > 0.05). The mechanical material constant of ascending aorta at one-dimensional loading of human was basically familiar to that of swine of 1 to 7 months (P > 0.05). ② Comparison of elastic modulus of ascend ing aorta at one -dimensional loading between human and swine of differ ent months: With the increase of months, elastic modulus of ascending aorta was significantly increased in swine of 7 months old in comparison with that in other months old (P < 0.05); there was no significant difference of elastic modulus of corresponding blood vessel between human and swine of different months (P > 0.05).CONCLUSION: There is no significant difference in mechanical characteristic constant and elastic modulus of ascending aorta between human and swine of 1 to 7 months. Mechanical characteristics of corresponding blood vessel were similar in some segments at least. From the mechanical angle, anastomosis of corresponding ascending aorta in the process of heterogeneic heart transplantation from swine to human has certain feasibility.

Aim To determine whether AngⅡin para-ventricular nucleus (PVN)was involved in the chronic intermittent hypoxia (CIH ) induced-hypertension in rats. Methods Male Sprague-Dawley rats were ran-domly divided into Sham and CIH groups, the Sham rats were exposed to continuous normoxia, while the CIH rats were submitted to CIH (8 h per day for 15 days). The conscious noninvasive method with tail cuff was performed in rats to record the systolic blood pres-sure during establishing the model of CIH induced hy-pertension.Mean arterial pressure (MAP)and heart rate (HR)were recorded in vivo on a PowerLab data acquisition system after CIH.Rats were fixed on the stereotaxic instrument to conduct microinjection in the PVN.We used Western blot to measure Ang Ⅱ level and AngⅡtype 1 receptor (AT1 R)protein expression in PVN.Results The level of PVN Ang Ⅱin CIH rats was significantly higher than that in Sham rats,a-long with increased AT1 R protein expression.Microin-jection of Ang Ⅱ(0.03,0.3,3 nmol)in bilateral PVN dose-dependently increased MAP in both CIH and Sham rats, and this response was significantly augmen-ted in CIH rats.Losartan (50 nmol),AT1 R antago-nist,had no effect on MAP in Sham rats, but caused significant MAP decreases in CIH rats, and prevented Ang Ⅱ-induced increases in MAP in both CIH and Sham rats. Conclusion The results suggest that the increased AngⅡrelease and enhanced AT1 R activation in the PVN contribute to CIH induced-hypertension in rats.

AIM: To study on antisense oligoncleotides as inhibitor of human acute promyelocytic leukemia (HL-60) proliferation and C-myc protein expression. METHODS: Oligonucleotides with different lengths (18-21 mer) complementary to the definite regions of C-myc mRNA, modified groups (with S replaced O in internucleotide phosphate linkage) and unmodified ones (with natural internucleotide phosphate linkage) were designed and synthesized. These olignucleotides were tested for their activity on HL-60 cell and also for their toxicity on normal lymphatic cells of human. RESULTS: It was found that two of the oligonucleotides complementary to 5′-44-61 and 5′-556-576 the regions of C-myc mRNA exhibited great inhibitory effects (59.5 % and 62.7 %) on growth of HL-60 cells for a definite time. And no toxicity of the two antisense oligonucleotides was found on normal lymphatic cells of human. CONCLUSION: The sequence of antisense oligonucleotides complementary to 5′-44-61 of C-myc mRNA was designed newly by us may be turned into inhibitory medicine of HL-60 cells.

Objective To investigate the factors which may affect the secretions of human leptin and free fatty acids (FFA) by measuring plasma glucose, serum insulin, leptin and FFA levels during reduced sample number intravenous glucose tolerance tests (IVGTT) in the subjects with different status of insulin resistance and ? cell function. Methods According to oral glucose tolerance test, 7 normal, 10 overweight/obese individuals, 12 subjects with impaired glucose tolerance (IGT) and 11 with type 2 diabetes mellitus (DM) were recruited. During IVGTT, serum insulin, leptin and FFA levels at 12 time points were simultaneously measured. Results Compared to the stable secretion of leptin, during 180 min in the IVGTT, the FFA secretion showed a "U" shape profile. The correlation analysis showed that the average serum leptin and FFA levels in 3 h were independent to fasting plasma glucose concentration. The partial correlation coefficient of leptin and insulin decreased after being adjusted for FFA (before r=0.77, P0.05). Conclusion The changes of serum FFA levels in IVGTT are more significant than those of leptin after glucose loading. Fasting plasma glucose levels do not significantly affect the average serum levels of leptin and FFA. On the contrary, insulin manifests such action but does not affect the secretion profile of these parameters.
