1.Study on the Apoptosis of Leukemic Cell Induced by Antisense Oligodeoxynucleotides Complementary to c-myc

Purpose To observe the effect of Anti-c-myc on the apoptosis of HL60 cells and leukemicpatients' leukocytes. Methods HL60 cells and leukemic patients, leukocytes were treated with anti-c-mycoligomer-5' d(CTT CTC GAG GCA GGA GGG)3'-complementary to the transcriptional initiation of c-mycmRNA. After 10 hours, we detected the apoptosis by several methods: DNA agarose gel electropboresis,ELISA(enzyme- linked immunosorbent assay), Flow Cytometric Analysis, and Accounting the number ofapoptotic cells after stained with Wright' s. Results The apoptosis of HL60 cells were induced by theanti-c-myc oligomer, and the induction of apoptosis was dose-dependent, i. e. more HL60 cells vereapoptotic as we treated them with higher dose of the antisense oligodeoxynucleotides. Furthermore, the antic-myc oligomer also induced the apoptosis of leukemic patients, leukocytes, but had no effect on theapoptosis of normal leukocytes. Conclusions These results imply that we can treat acute myeloidleukemia with the anti-c-myc oligomer through apoptosis-inducing, and it has good specificity. This studyprovides basis for leukemia treatment.

Besides cyclooxygenase and nitric oxide synthase, another distinct endothelial pathway, endothelium-dependent hyperpolarization factor (EDHF), is involved in relaxation of the vascular smooth muscle cells. EDHF has been demonstrated unequivocally in various blood vessels from different species, including human, and is likely to play an important role in cardiovascular physiology and pharmacology.
3.Effects of total flavones of rhododendra (TFR) on bioelectricity of ventricular myocytes in guinea pig

Aim To study the effect of total flavones of Rhododendra(TFR)on bioelectricity changes of ventricular myocytes in guinea pigs. Methods Conventional microelectrode technique was used to record action potentials (AP) , resting potentials (RP) and effective refractory period (ERP). Results TFR 25,50 mg?L-1 decreased the action potential duration at 50%,90% repolarization (APD50,APD90) while TFR 100,200,400 mg?L-1 prolonged APD50 and APD90Significantlg. TFR 200,400 mg?L-1markedly decreased action potentials amplitude (APA) and velocity maximum of depolarization (Vmax). TFR 400 mg?L-1decreased resting potential. TFR 200 mg?L-1 prolonged ERP. Conclusion TFR may antagonize Ca2+ inward flow at lower concentration and block K+ inward flow at higher concentration.
4.A Case Report of Lymphomatoid Papulosis Followed By Mycosis Fungoides:Derived from a Com-mon T Cell Clone

Objective To report a case of lymphomatoid papulosis(LyP)followed by mycosis fun-goides(MF)during his27years course and determine whether these diseases are clonally related.Methods Characteristics of clinicopathology and immunophenotype of skin tissues of different phases of the patients were compared.Simultaneously,clonal T-cell receptor gene rearrangement of skin lesion tissues and blood specimens were detected by means of polymerase chain reaction and Southern blot analysis.Results A dis-tinct clinical and pathological feature was shown in this case of LyP followed by MF.The proliferating lym-phocytes in LyP and MF were mature T helper lymphocytes,and with same T-cell clone.Conclusion LyP followed by MF displays a same T-cell clone.The clonal T-cell is irrelevant to the biological behavior of these diseases.
5.Cutaneous Type Adult T-cell Leukemia/Lymphoma: The First Case Report in China

Objective To report the first case of cutaneous type adult T-cell leukemia/lymphoma(cATLL) in China. Methods The skin lesion was examined by histopathology and direct immunofluorescence (DIF), and the immunophenotype was also studied. ELISA and Western blot were used to test the serum antibodies to HTLV-Ⅰ, and HTLV-Ⅰproviral DNA of lymphoid cells was detected by PCR. Results The patient had polymorphic skin lesions including papules, plaques, and bullae with tense or flaccid walls. Histopathogical examination showed subepidermal bullae, and there were small-to-medium-sized atypical lymphoid cell infiltrations in the dermal papilla in the bottom, and border of the bulla. CD45+ and CD45RO+ staining, and negative DIF were observed in the atypical lymphoid cells. The serum antibodies to HTLV-Ⅰ, and proviral DNA of HTLV-Ⅰin the blood lymphoid cells were detected. The patient died with a disease course of one year and ten months. Conclusion ATLL is not extremely rare in China, and cATLL may also exist.
6.Effects of bFGF and insulin on proliferation of mice cartilage tissue cells

AIM: To explore the effects of basic fibroblast growth factor ( bFGF) and insulin on the cell proliferation and differentiation in primary cartilage cells. METHODS: After induction with different concentrations of bFGF and insulin, cell proliferation was measured with WST-1 method and fluoroscope methods. RESULTS: bFGF and insulin exerted their related action on primary cartilage cells in 0.4% fatal bovine serum at different concentrations. 25 ?g/L bFGF and 5 mg/L insulin promoted cell proliferation significantly. CONCLUSION: bFGF and insulin prolong the survival time and promote cell proliferation in primary cartilage cells.
7.Study on antisense oligoncleotides as inhibitor of human acute promyelocytic leukemia proliferation and protein expression

AIM: To study on antisense oligoncleotides as inhibitor of human acute promyelocytic leukemia (HL-60) proliferation and C-myc protein expression. METHODS: Oligonucleotides with different lengths (18-21 mer) complementary to the definite regions of C-myc mRNA, modified groups (with S replaced O in internucleotide phosphate linkage) and unmodified ones (with natural internucleotide phosphate linkage) were designed and synthesized. These olignucleotides were tested for their activity on HL-60 cell and also for their toxicity on normal lymphatic cells of human. RESULTS: It was found that two of the oligonucleotides complementary to 5′-44-61 and 5′-556-576 the regions of C-myc mRNA exhibited great inhibitory effects (59.5 % and 62.7 %) on growth of HL-60 cells for a definite time. And no toxicity of the two antisense oligonucleotides was found on normal lymphatic cells of human. CONCLUSION: The sequence of antisense oligonucleotides complementary to 5′-44-61 of C-myc mRNA was designed newly by us may be turned into inhibitory medicine of HL-60 cells.
8.Evaluation of the scientific research efficiency of 100 tertiary hospitals and the influencing factors analysis based on Bootstrap DEA-OLS model

Objective To evaluate the scientific research efficiency of tertiary hospitals and analyze the influencing factors, providing reference for the decision-making of scientific research managers in China. Methods Evaluation indicators were collected from 100 tertiary hospitals in China, and their research efficiency was evaluated using Bootstrap data envelopment analysis (DEA) method. The influencing factors were analyzed using ordinary least square ( OLS) regression model. Results The averaged scientific research efficiency of these hospitals using Bootstrap DEA method was 0. 5224, lower than that using conventional DEA method (0. 0676), yet with a great variation (from 0. 1103 to 0. 8790) among them. Linear regression analysis showed that factors such as R&D input and output, and hospital types has statistical significance on the saentific research efficiency (P < 0. 05). Conclusions These hospitals are inefficient in scientific research with sizable room for improvement. Conventional DEA method should be used with caution, as deviations should be corrected with Bootstrap DEA method. The research development level and hospital types of their province can influence the research efficiency the hospitals significantly.

After more than six years of practice, the National Metabolic Management Center(MMC) has developed distinctive approaches to manage metabolic patients and has achieved a series of positive outcomes, continuously advancing the standardized diagnosis and treatment model. Since December 2019, the outbreak of coronavirus disease 2019 (COVID-19) has occurred in China. Higher risk of COVID-19 infection and worse prognosis were observed among patients with diabetes. There are rigorous challenges existing in terms of diabetes prevention and glycemic control in primary medical care during period of COVID-19. Here, expert recommendations were developed by Chinese diabetologists, healthcare providers, and public health administrators to improve the ability of primary health facilities and provide standardized basic public health and medical services throughout country. The main contents include basic requirements for management, workflow of health management, referral, treatment, and long-term follow-up.
