Objective To investigate the inhibition of proliferation and the regulation of histone acetylation modification in Jurkat cells treated by sodium valproate(VPA). Methods Jurkat cells were treated with VPA.Cell proliferation was determined by CCK-8 assay, and cell cycle were analyzed by flow cytometry (FCM). mRNA of HDAC1 was detected by semi-quantitative RT-PCR, and protein expression of HDAC1 and acetylation of histone H3, H4 was examined by Western blotting. Results VPA inhibited the proliferation of Jurkat cells in concentration-and time-dependent manners. After exposure to VPA in different concentrations for 48h,cell cycle was arrested obviously at G0/G1 phase (P <0.05), and with increasing concentration, the percentage of G0/G1 phase cells was increased and that of S phase were decreased. HDAC1 mRNA expression were inhibited with the increasing concentration of VPA. The protein level of HDAC1 was down-regulated, while acetylation of histone H3、H4 was up-regulated in Jurkat cells by VPA. Conclusion VPA can inhibit proliferation of Jurkat cells and induce G0/G1 phase arrest. The mechanism may be that VPA increase acetylation of histone H3/H4 by inhibiting expressions of HDAC1 gene.

Objective In our previous study, we had generated various zebrafish mutant lines with tissue?specific GFP expression by Tol2 transposon?mediated insertional mutagenesis. Among these mutants, the Tol2:20141221t line ex?presses GFP in nervous system, while the position within zebrafish genome where transposon inserted has not yet been iden?tified. The aim of this study was to identify and analyze this genetically modified mutant. Methods The transgenic inser?tion loci in the genome of Tol2:20141221t line was identified byTAIL?PCR and the spatial and temporal expression profile of the affected gene was examined by in situ hybridization. Homozygous mutant of Tol2:20141221t was generated for explo?ring related developmental defects. Results Tol2 transposon was inserted into the 8th intron region of sat1.a gene, and in?duced premature transcription termination. The maternal and zygotic mutants of Tol2:20141221t was generated, while with?out apparent developmental defects. Conclusions We have generated and identified the zebrafishsat1.a mutant mediated by Tol2 transposon. This gene insertion mutant exhibits no obvious developmental abnormalities, but may serve as a power?ful tool to study the development of nervous system.

Journal of Clinical Neurology 2024;37(2):149-152
Stroke is the main cause of disability in adults. With the progress of stroke diagnosis and treatment technology, the mortality rate of stroke patients has decreased significantly, but its incidence keeps increasing, and there is an increasing number of stroke survivors develop chronic neurological disorders. At present, there is no clear drug to promote functional repair after stroke.Several studies have shown that gut microbiota can improve stroke prognosis by regulating neuroactive molecules and immune cell functions, enhancing neural network plasticity, and reducing neuroinflammation.Based on a review of previous studies, this paper describes the mechanism of action of gut microbiota on neural network plasticity and neuroinflammation after stroke and its impact on functional recovery after stroke and explores its clinical value and feasibility in improving neurological dysfunction after stroke.

Background:
Classical swine fever virus (CSFV), the causative agent of classical swine fever (CFS), is a highly contagious disease that poses a serious threat to Chinese pig populations.
Objectives:
Many provinces of China, such as Shandong, Henan, Hebei, Heilongjiang, and Liaoning provinces, have reported epidemics of CSFV, while the references to the epidemic of CSFV in Yunnan province are rare. This study examined the epidemic characteristics of the CSFV in Yunnan province.
Methods:
In this study, 326 tissue samples were collected from different regions in Yunnan province from 2015 to 2021. A reverse transcription-polymerase chain reaction (RT-PCR), sequences analysis, and phylogenetic analysis were performed for the pathogenic detection and analysis of these 326 clinical specimens.
Results:
Approximately 3.37% (11/326) of specimens tested positive for the CSFV by RTPCR, which is lower than that of other regions of China. Sequence analysis of the partial E2 sequences of eleven CSFV strains showed that they shared 89.0–100.0% nucleotide (nt) and 95.0–100.0% amino acid (aa) homology, respectively. Phylogenetic analysis showed that these novel isolates belonged to the subgenotypes 2.1c and 2.1d, with subgenotype 2.1c being predominant.
Conclusions
The CSFV was sporadic in China’s Yunnan province from 2015 to 2021. Both 2.1c and 2.1d subgenotypes were found in this region, but 2.1c was dominant.
