1.Expression of mannose-receptor in cultured bovine iris pigment epithelial cells in vitro

Objective To verify whether iris pigment epithelial cells (IPECs) possess the similar potential of specific phagocytosis to retinal outer segments (ROS) with retinal pigment epithelial cells (RPECs). Methods IPECs were isolated from neonatal bovines with Hu′s method, and were cultured. The cultured cells were identified by immunohistochemical methods with antibodies to cytokeratin and S 100. Total RNA of IPECs was extracted by Trizol. The specific primers for mannose receptor and ? actin were designed according to their sequence from Genbank. The mRNA expression of these proteins in the IPECs was analyzed by reverse transcription polymerase chain reaction (RT PCR). Results The cultured IPECs have no contamination of other cells. The extracted RNA was ideal and had no degradation. RT PCR analysis showed that mannose receptor′s mRNA was expressed in cultured IPECs in vitro. Conclusions Cultured IPECs may express the mannose receptor, and may have similar potential of phagocytosis to ROS with RPECs.

3.The protective effect of exercise on osteopenia in ovariectomized rats

Objective To investigate the protective effect of exercise on osteopenia female rats caused by ovariectomy and its mechanism. Methods Twenty-four female rats aged 6 months were randomly divided into 3 groups: control group(CON),ovariectomized model group(OVX) and exercise group(Ex).The ovariectomy operation was performed with rats in OVX and Ex groups. Rats in Ex group were trained on a treadmill at a velocity of 18m/min for 1h, once daily for 5 days a week. Each group of rats were sacrificed at 12 weeks postoperatively and Dual-energy X-ray absorptiometry was carried out to measure the bone mineral density(BMD) of the femur. IGF-1 proteins in trabecular bone of proximal femur were examined by using immunochemical stain and the intensity of IGF-1 was determined by use of imaging analysis techniques. The apoptotic osteocytes in the trabecular bone of proximal femur were detected by using terminal-deoxynucleotidyl mediated nick end labeling (TUNEL) technique. Results The BMD of CON, Ex and OVX group was (0.170?0.011)g/cm2,(0.154?0.013)g/cm2 and(0.167?0.013)g/cm2, respectively. The difference of the BMD level between Ex and CON groups was not significant, however, the BMD level in OVX group was significantly lower than that of the Ex and CON groups (P
4.Construction of a subcutaneous matrigel angiogenesis model in diabetic rats

Objective There are different degrees of angiogenesis in the tissues and organs of the patient with diabetic chron -ic vascular disease , and it is somehow difficult to observe angiogenesis .This study was to construct an experimental model of angiogen-esis in the diabetic rat in vivo. Methods Twelve SD rats were equally randomized into a normal control and a diabetes mellitus ( DM) group, the DM model constructed by intraperitoneal injection of streptozotocin .At 8 weeks after modeling , matrigel was planted under the abdominal skin of the control and model rats and removed 2 weeks later .Then the structure of the new vessels was observed by HE staining and the expressions of aminopeptidase P (JG12) and alpha-smooth muscle actin (α-SMA) detected by immunohisto-chemistry and immunofluorescence . Results Compared with the control rats , the DM models showed significant increases in the blood glucose level ([6.15 ±3.02] vs [35.08 ±6.92] mmol/L), 24-hour urine volume ([5.33 ±1.53] vs [105.00 ±20.82] mL), and 24-hour urinary protein level ([7.78 ±2.12] vs [47.70 ±9.63] mg/24 h) (P<0.01).The matrigel plug was white and transparent in the controls but red with a visible tiny vessel-like structures in the model rats .The vessel-like structures were found oc-casionally in the matrigel plug of the control animals but in large numbers with many erythrocytes in that of the DM models .In compari-son with the normal controls , the model rats exhibited significantly upregulated expressions of JG12 (7.76 ±1.85 vs 27.36 ±3.75, P<0.01) and α-SMA (10.22 ±2.64 vs 36.85 ±4.28, P<0.01) in the matrigel plug , and a vessel-like reticular distribution was ob-served. Conclusion Hyperglycemia may induce angiogenesis in the matrigel plug .The DM rat model of subcutaneous matrigel angio-genesis may contribute to the study of angiogenesis in diabetes .
5.Construction of Antisense GA cDNA Recombinant Plasmid and Its Effect on the Growth of Colon Cancer Cells

Objective To construct the recombinant plasmid carrying antisense GA cDNA, and evaluate the effect of antisense GA gene on the biological characteristics of colon cancer cells. Methods The recombinant plasmid pcDNA3.0/GA containing antisense GA gene was constructed by gene recombination technique. The recombinant plasmid was transfected into colon cancer cells by lipofectamine. The effect of antisene GA gene on the biological characteristics of colon cancer cell line DH52 was evaluated by MTT and electron microscopy. Results The recombinant plasmid pcDNA3.0/GA was successfully constructed. The growth rate of colon cancer cells decreased, and microstructure of the cells appeared typical apoptotic changes after transfection. Conclusion Antisense GA gene could inhibit the growth of colon cancer cells possibly by suppressing GA gene expression and decreasing GA activity in the colon cancer cells.
6.Value of red blood cell distribution width-to-platelet count ratio in predicting the prognosis of children with sepsis.

OBJECTIVE:
To study the value of red blood cell distribution width (RDW)-to-platelet count (PLT) ratio (RPR) on the first day of admission into the pediatric intensive care unit (PICU) in predicting the prognosis of children with sepsis.
METHODS:
A retrospective analysis was performed for the clinical data of 186 children with sepsis who were hospitalized in the PICU. According to their prognosis, they were divided into a survival group with 151 children and a death group with 35 children. Clinical data were compared between the two groups. The Cox proportional-hazards regression model analysis was used to investigate the factors influencing the prognosis. The receiver operating characteristic (ROC) curve was plotted to evaluate the value of RPR in predicting death. The children were divided into a high RPR group and a low RPR group according to the optimal cut-off value, and the Kaplan-Meier method was used to compare the 28-day survival rate between the two groups.
RESULTS:
Compared with the survival group, the death group had significantly higher RDW, procalcitonin (PCT) and RPR (P<0.05) and significantly lower PLT and albumin (ALB) (P<0.05). The Cox regression model analysis showed that low ALB, high PCT and high RPR were independent risk factors for the prognosis of children with sepsis (P<0.05). The ROC curve analysis showed that RPR had a certain value in predicting the prognosis of children with sepsis (P<0.05), with an area under the ROC curve of 0.937, an optimal cut-off value of 0.062, a sensitivity of 94.29%, and a specificity of 77.48%. The Kaplan-Meier survival analysis showed that the high RPR group had a significantly lower 28-day survival rate than the low RPR group (P<0.05).
CONCLUSIONS
RPR on the first day of admission into the PICU is closely associated with the prognosis of children with sepsis and has an important value in predicting the prognosis of children with sepsis.

7.The effect of telmisartan on APN,hs-CRP and TNF-α in hypertensive patients with unstable angina pectoris

Objective To investigate the effect of telmisartan on the expression of APN,hs-CRP,TNF-α and the prognosis of hypertensive patients with unstable angina pectoris. Methods Totally 100 hypertensive patients with unstable angina pectoris were randomly divided into the telmisartan group(n =50) and the losartan group(n =50),each group with oral administration. The blood pressure controlled condition, APN, hs-CRP and TNF-α content changes and the occurrence of cardiovascular accident after treatment were observed in both two groups. Results In both groups, blood pressure returned to normal in 8 weeks,with no significant difference(P > 0.05). After treatment,APN level in the telmisartan group increased faster than that in the losartan group(P <0.05) ,while the hs-CRP,TNF-αdecreased significantly(P < 0.05). After 8 weeks, the occurrence of cardiovascular accident in the telmisartan and losartan groups was 10.0% (5/50) and 24. 5 % (12/49) respectively in the following 4 months (P < 0.05). Conclusion Telmisartan has significant effect in regulating APN, hs-CRP, TNF-α expression in hypertensive patients with unstable angina pectoris, which is able to decrease the short-term occurrence of cardiovascular accident.
8.Evaluation of application effect of evidence- based nursing in reducing ventilator - associated pneumonia of patients with severe head injury

Objective To evaluate the application effect of evidence-based nursing in reducing ventilator-associated pneumonia (VAP) of patients with severe head injury. Methods 100 patients with severe head injury using mechanical ventilation were randomly divided into the control group and the observation group with 50 patients in each group.The observation group explored evidence from nursing of ventilator tube,oropharyngeal and sound sects care,airway care,balloon sleeves care to prevent biofilm on the endotracheal tube(ETT-BF) to fall off,enteral nutrition care,decubitus care,ward environment and the hand disinfection of medical staff and was given evidence- based nursing.The control group took routine care.The incidence of VAP,off-line time and mortality rate of the two groups were compared during the same time period. Results The incidence of VAP,off-line time,mortality rate showed significant difference between the two groups.The observation group showed better effect. Conclusions Implementation of evidence-based nursing can significantly reduce the off-line time of patients with severe traumatic brain injury using mechanical ventilation.It can decrease the incidence of VAP and mortality rate,improve the prognosis and reduce medical cost.
10.The MRI findings of mitochondrial encephalomyopathy,lactic acidosis and stroke-like episodes

Objective To analyze retrospectively the MR features of MELAS patients,in order to improve the early diagnosis of MELAS.Methods MR data of 1 6 MELAS patients confirmed by clinical diagnose and muscle biopsy were retrospectively analyzed. MR features of plain scan(n=1 6),ASL(n=3),MRA(n=1 5),DWI(n=14)and MRS(n=3)were analyzed.Results MRI data demonstrated brain lesions mainly distributed in posterior cerebral hemisphere extending to subcortical area,which was not consistent with the vascular supply territories.The lesions were observed as low signal on Spin-echo sequence of T1 WI,while high signal on T2 WI, FLAIR and DWI.The focal lesions showed increasing Lac peak on MRS.Fifteen of 16 patients underwent MRA and one showed increased arterial branches in lesion zones.Three patients had ASL scanning demonstrating high irrigation in lesion zones which would wander once relapsed.Meanwhile,old lesions showed encephalatrophy and cerebromalacia.All patients’muscle biopsy pathology showed ragged red muscle fibers.Under electron-microscopic,bioblasts were bigger and more than average level and muscle fibers atrophied.Mitochondrial DNA sequence of 3 patients showed mtDNA A3243G transgenation.Conclusion The DWI,MRS and ASL sequences show good ability in MELAS diagnosis and differential diagnosis.
