1.Analysis of Dangguibuxue decoction on rabbit schistosomiasis hepatic fibrosis through histopathological evaluation

Objective To evaluate the treatment effects on schistosomiasis hepatic fibrosis with Dangguibuxue decoction.Methods Thirty-four rabbits weighing 2.0-2.5 kg were selected.The procedure for the model of schistosomiasis liver fibrosis was as follow.Thirty-four snails infected by schistosoma japonicum were placed in triangle flask to escape cercariain.Abdominal skin of each rabbit wag infected by 100 schistosoma japonicum cercariaes.All rabbits were housed in conventional cages at room temperature.After 13 weeks schistosoma japonicum infected,the rabbits'livers were examined by B-ultrasound examination every week.After 18 weeks of infection,B-ultrasound showed images of schistosomiasis liver fibrosis,the rabbits were thus identified as schistosomiasis liver fibrosis.Praziquantel Was as insecticide to treat rabbits.All rabbits were divided into treatment and control groups according to body weigh.The high and low dosage group were treated with Dangguibuxue decoction at 50 g/kg and 5 g/kg(n=11),respectively,and the control group with no treatment(n=12).Two groups were treated with Dangguibuxue decoction every day for 10 weeks.After 10 weeks of treatment,the rabbits were killed and the livers were obtained to go HE staining and histopathological examination under microscope.Results After 18 weeks of infection,B-ultrasound showed that the liver parenchyma Was coarse and net-shaped.having hyperechoie spots with lightbrand and facula.The liver's shape Was abnormal,portal and splenic vein diameter was widened.Histopathological examination showed that the high dosage group presented fibrosis,which was formed by collagen fiber extending to peripherM regions,low dosage group showed mild fibrosis,mild collagen fiber present with extension without compartment formation, while the control group presented severe fibrosis and severe collagen fiber present with a thickening of the partial compartments and the distribution of eggs nodules. The liver histopathological ranking results from different groups showed that the difference was significant (χ2= 13.585,P < 0.01), the difference between the treatment groups of high and low dosage and control group was significant,respectively(all P < 0.01 ). Conclusions The histopathology results showed that both high and low dosage Dangguibuxue decoction can reverse or eliminate schistosomiasis liver fibrosis in rabbits.
2.EFFECT OF TAURINE ON MODULATING CALCIUM OF CARDIAC MUSCLES IN RATS

The effect of Taurine ( Tau)on 45Ca influx of isolated left ventricular muscles in rats has been investigated. The 45Ca influx of the preparations untreated with Tau, are affected by the concentrations of calcium in KH solution. when the concentration of calcium is 0.62; 1.25; and 1.87mmol/L respectively, the 45Ca influx is 1.02? 0.25; 1.37?0.14 and 1.45?0.14 ?mol/g wet tissue, respectively. After adding Tau, the situations above were changed significantly. It was shown that is 1.11 ? 0.11, 1.45?0.12, and 1.48 ? 0.09 ?mol/g wet tissue in low Ca2?; 1.19?0.07, 1.14 ? 0.23, and 0.97?0.24 ?mol/g wet tissue in normal Ca2?; and 1.12?0.05,0.58?0.18 and 0.53?0.10 ?mol/g wet tissue in high Ca2? treated with Tau 10, 20, and 40 mmol/L, respectively. The results show that the varied ,Ca2? concentrations in KH solution can affect 45Ca influx of cardiac muscles, but Taurine has a bisphasic modulation o,f calcium influx in the same tissue. The effects of modulating calcium may play an important role for anti-arrhythmic mechanism of Taurine.
3.EFFECT OF RETINOIC ACID ON DIFFERENTIATION,MATURATION AND FUNCTION OF INTESTINAL DENDRITIC CELL

Objective To investigate the influence of retinoic acid (RA) in differentiation,maturation and function of intestinal dendritic cells (DC),and probe into the effect of Vitamin A (VA) on intestinal mucosal immunity and pathway. Method Rat intestinal mucosa was cultured in vitro,supplemented with all-trans RA and/or retinoid acid receptor ? (RAR?) antagonist (Ro 41-5253). After cultured for 24h and 48h,the surface markers OX62、OX6 and CD86 on DCs were detected by flow cytometry,to observe the effect of RA on differentiation and maturation of DC. The cytokines and RAR? were measured in mRNA levels by RT-PCR to analyze the influence of RA on mucosal immunity and the pathway. Results RA promoted the maturation of DC in mucosa cultured in vitro,and up-regulated RAR? mRNA levels,the changes were marked at 24h. Moreover,when RA was present in the culture,IL-12 and IFN-? (Th1 cytokine) mRNA were reduced,and IL-10 was increased significantly compared with control . However,the actions of RA above can be inversed by Ro 41-5253. Conclusion The modulation of RA on DC could be one of the important mechanisms that VA influences intestinal mucosal immunity. RAR? participates in the regulation of RA on DC.

Liver fibrosis can be caused by chronic liver injury arising from various etiological factors.The activation of the hepatic stellate cell(HSC) is the central event in liver fibrosis,the apoptosis of which was the initiating factor for the reversal process.The apoptosis of HSC is very complicated.The signaling pathways lack of enough research in regulating and controlling and activating HSC's apoptosis.The apoptosis pathways as known currently mainly include mitochondrial pathway,death receptor pathway and non-death receptor pathway.Based on these signaling pathways,selectively inducing cell death of activated HSCs is becoming the new antifibrotic treatment.This review focuses on the new progress of HSC's apoptosis signaling pathways,and summarizes the drugs that have the effect of inducing HSC apoptosis.
5.Surface Electromyography of Lumbar Muscles in Non-specific Low Back Pain Patients during Different Bridging Exercise

Objective To investigate the features of surface electromyography of lumbar muscles in non-specific low back pain (NLBP) patients when they were practicing different types of bridging exercise. Methods 18 healthy subjects (control group) and 18 NLBP patients (NLBP group) were included and asked to practice bridging exercise with single and double feet. The root mean square (RMS) of surface electromyography of both sides of lumbar muscles were recorded. Results No significant difference was found in the control group. The RMS of erector spinae was more in the supported side than in the contra side for NLBP patients in bridging exercise with single foot. There was no significant difference among different time points in the same muscle. Conclusion The contractions of erector spinae in both sides are not equal in asymmetric bridging exercise in NLBP patients.
7.Laparoscopic splenectomy for idiopathic thrombocytopenic purpura:a report of 21 cases

Objective To evaluate the therapeutic efficacy of laparoscopic splenectomy (LS) for patients with idiopathic thrombocytopenic purpura (ITP) and to investigate the change of parameters of platelets.Methods Twenty-one patients with ITP after LS were studied, including the following paramenters: the operation time, intraoperative blood loss and postoperative complications, and compared perioperative platelet count(PC), packed platelet volume(PPV), mean platelet volume(MPV), and platelet distribution width (PDW) with the postoperative results.Results The mean operation time was 1.5 h. Intraoperative mean blood loss was 60 mL, postoperative hospitalization time was 5 d and with no case of delayed healing of incision. Accessory spleen was found in 19 % of patients . The incidence of complication was 4.8 %, and with no deaths. Platelet count and packed platelet volume increased with fluctuation, but mean platelet volume and platelet distribution width decreased with fluctuation. The parameters were in multipoint dynamic equilibrium with fluctuation and coordinated restoration. All patients were followed-up. No case had recurrence at 1.5~5.5 y, and platelet-associated antibody was in normal range. Bone marrow examination was normal.Conclusions The treatment of ITP with LS is safe and effective. The selection of therapeutic plan should depend on the changes of platelet parameters.
8.Effect of tumor necrosis factor-alpha on acyl coenzyme A: cholesteryl acyltransferase activity and ACAT1 gene expression in THP-1 macrophages.

In order to explore the effect and mechanisms of tumor necrosis factor-alpha (TNF-alpha) on the activity of the acyl coenzyme A: cholesteryl acyltransferase (ACAT), THP-1 monocytes were cultured and induced to differentiate into macrophages with phorbol ester. TNF-alpha (60 ng/mL) was added at different time points into the macrophage-containing medium and the ACAT enzyme activity was measured by quantifying the incorporation of [1-(14)C] oleoyl CoA into cholesteryl esters. The expression of ACAT-1 protein and mRNA was respectively detected by Western blotting and RT-PCR in THP-1 macrophages 24 h after treatment with TNF-alpha (60 ng/mL). The results indicated that ACAT activity in THP-1 macrophages treated with TNF-alpha was increased in a time-dependent manner. The expression levels of ACAT-1 protein and mRNA were significantly increased in THP-1 macrophages after treatment with TNF-alpha (P<0.05). It was suggested that TNF-alpha could increase the activity of ACAT in THP-1 macrophages by up-regulating the expression of ACAT-1 gene.

The work was proceeding orderly. Good results have been achieved. The clinical pharmacists of TCM participate in clinical rational use of drugs to change the service mode ofindirect service for the patients in the past with the patients, doctors and nurses so as to make better service for people's life and health of Chinese medicine.
10.MRI Features of Neuropsychiatric Abnormalities in Systemic Lupus Erythematosus

Objective To evaluate the MRI features of neuropsychiatric abnormalities in systemic lupus erythematosus(NPSLE).Methods Brain MRI images of 21 cases with NPSLE confirmed by clinic were analyzed retrospectively.ADC values of the lesions and normal brain tissue were measured.Results Brain abnormal MRI findings in 20/21 cases were found(95%)including:(1)High signal intensity on DWI in 14 cases,11 were diffuse lesions,3 were focal lesions,4 of 14 combined with brain atrophy.The lesions mostly localized in white matter.The ADC values of the lesions in gray matter were decreased(t=2.513,P=0.019),while the ADC values of the lesions in white matter were increased(t=2.877,P=0.007).(2)2 cases only showed brain atrophy.(3)Leukoaraiosis presented in 2 cases.(4)Encephalomalacie presented in 2 cases,1 accompanied with brain atrophy.(5)No enhancement(3/5)and little patchy enhancement(2/5)were observed at contrast MRI study.Conclusion MRI plays an important role in displaying cerebral lesions and the location of it,progression and succession of NPSLE.
