1.ISOLATION OF FEMALE- SPECIFIC SEQUENCE OF SCHISTOSOMA JAPONICUM

Aim To isolate the female-specific sequence of Schistosomajaponicum S. J. in order to provide a basis for researching vaccines of anti-fecundity. Methods Representational difference analysis (RDA) was used to obtain a female-specific sequence of S.j. by the male S.j. genomic DNA substractive hybridization with the female. The fragment was about 562bp. Results This sequence was labeled as Dig probe by PCR technique to hybridize with the restriction fragments of each sex and the tester as well as the driver on the nylon membranes.The antiboly of Dig、 NBT、 and X-phosphate solution were utilized to immunoassay the result of hybridization. The result showed that the probe can hybridize to the tester but can not hybridize to the driver. Conclusion The result suggested that the fragment of approx. 562bp was the female-specific sequence of Schistosoma japonicum.
2.Different densities of autologous bone marrow mesenchymal stem cell transplantation for repairing Beagle canine teeth class Ⅱ furcation defects

BACKGROUND: Regeneration of type Ⅱ furcation defects of periodontal tissues is still a great clinical challenge. OBJECTIVE: To compare different densities of autologous bone marrow mesenchymal stem cells (auto-BMSCs) for repairing canine experimental class Ⅱ furcation defects of periodontal tissues. DESIGN: A randomized controlled trial. SETTING: Laboratory in Stomatological Hospital Affiliated to Fujian Medical University and Department of Animal Experiment in Fuzhou General Hospital. MATERIALS: Experiments were performed at the Laboratory in Stomatological Hospital Affiliated to Fujian Medical University and Department of Animal Experiment in Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA from July 2005 to September 2006. Six 18-month Beagle dogs were provided by Department of Animal Experiment in Fuzhou General Hospital of Nanjing Military Area Command of Chinese PLA. Animal intervention met animal ethical standards. Bio-Gide collagen membrane and BME-10X collagen membrane were used in the study.METHODS: Class Ⅱ furcation defects were induced surgically on the buccal side of canine mandibular second and third premolar (P2, P3) and first molar (M1). The ex vivo expanded auto-BMSCs from six 18-month Beagle dogs were seeded in BME-10X collagen membranes at cell density of 5×108 L-1,5×109 L-1,5×1010 L-1, and delivered into experimental class Ⅱ furcation defects, underneath a Bio-Gide membrane. Bio-Gide membrane alone was used as a control. The percentage of new cementum length and percentage of new alveolar bone area were measured on OLYPUS IX 71 inverted research microscope and OLYSIA BioAutoCell software in a computer.MAIN OUTCOME MEASURES: Each specimen was stained with hematoxylin and eosin. The lengths of new cementum and the area of new alveolar bone were calculated.RESULTS: The percentage of newly formed cementum length and the percentage of newly formed alveolar bone area were (51.5±5.6)% and (27.1±7.7)% in the control group,(84.8±8.9)% and(30.6±7.7)% in the 5×108 L-1 BMSCs group, (91.8±5.2)% and (68.3±11.4)% in the 5×109 L-1 BMSCs group and (88.8±7.2)% and (78.5±12.7)% in the 5×1010 L-1 BMSCs group. There were significant differences when comparing the BMSCs groups to the control group (P < 0.01), but there was no significant difference in each BMSCs group. There were significant differences in the percentage of newly formed alveolar bone when comparing the 5×109 L-1 and 5×1010 L-1 BMSCs groups to 5×108 L-1 BMSCs group and control group (P < 0.05), but there was no significantly difference between the first two groups, and neither was the later.CONCLUSION:Periodontal regeneration can be induced by BMSCs transplantation. The mechanism of regeneration is associated with inoculated density.

Trautv. var kulingensis Rehd as a basis for the rational exploitation and utilization of this medicinal plant Methods The characteristic features were studied by macroscopic and microscopic observations and its chemical costituents identified qualitatively by TLC Results Macroscopic and microscopic characteristics of this crude drug were described 4 chemical compositions, such as lupeol, were found by TLC Conclusion The distinct characteristics revealed in the studies could provide a basis for the identification of this crude drug
4.Cloning of Alpha gene segments from t(6;11) translocation renal cell carcinoma and analysis of their promoter activities

To construct recombinant reporter plasmids containing different Alpha gene segments and Alpha1-TFEB fusion gene and to evaluate the promoter activity of the Alpha gene. Methods:Promoter regions of the Alpha gene were predicted using a software Primer 0.5. Five Alpha gene segments with different lengths and a normal TFEB gene promoter (pTFEB) were amplified via polymerase chain reaction, and recombinant reporter plasmids containing different Alpha gene segments and a normal TFEB gene pro-moter were constructed. Liposome transfection was used to transfect these vectors into the human embryo kidney 293T cells. The pro-moter activity of the Alpha gene was evaluated via luciferase assay. Meanwhile, the recombinant Alpha1-TFEB plasmid was construct-ed and transfected into the 293T cells. The TFEB expression of the recombinant Alpha1-TFEB plasmid was then detected via Western blot. Results: Recombinant reporter plasmids containing different Alpha gene segments and pTFEB were constructed successfully. Compared with the luciferase activity of pGL3-Basic, that of the groups with Alpha1, Alpha2, Alpha3, Alpha4 and Alpha5 significantly increased (P<0.01). The luciferase activity also increased significantly in the groups with Alpha1, Alpha2 and Alpha5 compared with that of the pTFEB group (P<0.01). The TFEB expression of the pGL3-Enhancer-Alpha1-TFEB was significantly higher compared with that of the pGL3-Enhancer group. Conclusion:In t(6;11) translocation RCC, the Alpha gene has a strong promoter activity and it en-hances TFEB expression. The strongest promoter activity region is in Alpha5 with a sequence from 643 bp to 693 bp.

Objective To investigate the histopathology of liver,spleen and heart in mice model with Chlamydia pneumoniae ( C pneumoniae ) pneumonia Methods The Icr mice were inoculated with C pneumoniae ,strain CWL 029,by the intranasal or intravenous routes After a single inoculation,mice were killed on the 1st,3rd,7th,14th,21st,28th and 60th day separately Specimens of spleen,liver and heart were obtained from the acute stage of C pneumoniae pneumonitis and stained with hematoxylin eosin Results After inoculation of C pneumoniae ,there were infiltration of neutrophils and macrophages in the spleen and necrosis of hepatocytes The histopathology of iv inoculation group was more serious than that of intranasal inoculation group No pathology changes were observed in heart Conclusions After iv inoculation of C pneumoniae ,the histopathologic changes appeared in the tissue of liver and spleen correspondingly,and the changes were more serious than that of iv inoculation group This study demonstrated that C pneumoniae was mainly limited to lungs and dissemination of C pneumoniae was seldom
6.Effects and mechanisms of punicosides on acute alcoholic liver damage in mice.

OBJECTIVETo evaluate the protective effects of punicosides on alcohol induced acute liver injury in mice and its possible mechanisms as well.
METHODThe 60 mice were randomly divided into normal control, model group, three dose groups of punicosides with low, medium and high, then there is silibinin group. Three dose groups of punicosides and silibinin were given in advance by gavage for 4 weeks, then the mouse model of alcoholic acute liver injury was established. The serum levels of ALT, AST and TG were determined, and the mice were killed to calculate somatic index of liver, thymus as well as spleen. MDA, SOD, GSH-Px and GSH-ST were detected in the liver homogenate. Histopathological changes of the liver were observed by HE staining. The expression of MCP-1 and NF-kappaB in the liver tissues were detected by immunohistochemistry.
RESULTMid and high dose of punicosides reduced the liver index in mice significantly, improved liver steatosis, decreased the level of ALT, AST and TG in serum and the content of MDA in liver homogenate, furthermore the two dose groups increased the activity of SOD, GSH-Px and GSH-ST, inhibited the expression of MCP-1 and NF-kappaB in liver tissue.
CONCLUSIONPunicosides can protect the acute liver damage induced by alcohol.

7.Clinical Study on Medicinal-cake-separated Moxibustion Treatment for Mild-to-moderate Ulcerative Colitis of Spleen Deficiency and Dampness Type

Objective To explore clinical efficacy and mechanisms of medicinal-cake-separated moxibustion treatment for mild-to-moderate ulcerative colitis (UC) of spleen deficiency and dampness type.Methods Totally 60 cases were randomly divided into observation group and control group according to a random digits table, with 30 cases in each group. The control group was inserted into the anus with mesalamine suppository 1 g once a day, while the observation group was given medicinal-cake-separated moxibustion treatment once a day based on the treatment for the control group. Medicinal cake (Aconiti Lateralis Radix Praeparata: Codonopsis Radix: Coptidix Rhizoma: Salviae Miltiorrhizae Radix et Rhizoma: Corydalis Rhizoma: Aucklandiae Radix=3:3:2:2:2:1) was put on the acupoints (Pishu, Zhongwan and Zusanli for the first group, Dachangshu, Tianshu and Shangjuxu for the second group, one group for each time, and 2 groups were used interchangeably). One wormwood cylinder was put on medicinal cake, 5 min each time, 3 doses each time, once a day. The treatment course for all was 8 weeks. The main symptom scores (diarrhea, bellyache, pus and blood stool) and total scores, colonoscopy Baron score and serum CC chemokine ligand 20 (CCL20) expression level were compared between before and after treatment in the two groups. The clinical efficacy was evaluated and the adverse reactions were observed.Results The total effective rate in observation group was 93.33% (28/30) and significantly higher than that of the control group of 76.67% (23/30). After the treatment, the main symptom scores and total scores, colonoscopy Baron score and serum CCL20 expression level were significantly lower than those of pre-treatment in the same one group (P<0.05), which of the observation group were remakably lower than those of the control group (P<0.05); there was one case occured adverse reaction in the observation group and two cases in the control group, with no statistical significance between the two groups (P>0.05).Conclusion Medicinal-cake-separated moxibustion treatment is effective and safe in patients with mild-to-moderate UC of spleen deficiency and dampness type, which effects might be involved in regulating the expression of serum CCL20.
8.Drug Release Comparison of Nifedipine Sustained-release Tablets from Four Manufacturers

Objective:To compare the drug release of nifedipine sustained-release tablets from four manufacturers to evaluate the intrinsic quality. Methods:The drug release rate was determined by UV respectively with pH 1. 2 HCl solution, pH 4. 0 sodium ace-tate buffer, pH 6. 8 phosphate buffer and water as the dissolution medium. The dissolution curves were compared by f2 factor. Results:The drug release rate of nifedipine sustained-release tablets from the four manufacturers all met quality standard of our country, al-though the dissolution curves in the different dissolution medium was various. Conclusion:There are differences in intrinsic quality a-mong the nifedipine sustained-release tablets from the four manufacturers. The dissolution examination standard should be improved fur-ther.
9.The nursing practice in the work of civil cardiac death organ donation

Objective To investigate the role of nurses in civil cardiac death organ donation work.Methods Cooperating,propagating and promoting in civil cardiac death organ donation; building a bridge between the families of organ donation,donor coordinators,physicians and hospital ethics committees; participating in organ donation medical ethics assessment work; cooperating with the doctors do intend to maintain organ donation,access and preservation; protecting donors remains.Results During the time of July 2011 to November 2013,we successfully completed 52 cases of civil cardiac death organ donation cooperating with doctors,and got 41 liver,which entered the national organ allocation system for distribution.Conclusions The nurses will play a more and more important role in work of civil cardiac death organ donation cooperating with doctors.
