4.Screening and identification of oval cells-like stem cells of Thy-1-labeled positive hepatoma carcinoma cells:Are they tumor stem cells?

BACKGROUND:There may be tumor stem cells during metastasis and recurrence of liver cancer.The number of tumor stem cells is very few,so it is difficult to be identified.Thy-1 is a surface marking protein of oval cells.Specific marker can indirectly confirm the existence of tumor stem cells in the tumor,which is a focus in recent studies.OBJECTIVE:To study whether Thy-1 positive marked oval cells was expressed in hepatoma carcinoma cells,and investigate its screening and identification.METHODS:Flow cytometry was used to identify that Thy-1 positive expression in hepatoma carcinoma cells HepG2 and BEL-7402 and normal hepatocytes QSG-7701.Immunomagnetic bead sorting was utilized to select hepatoma carcinoma cells with Thy-1.Immunofluorescence was used to identify selected efficiency.Naked mouse inoculated with hepatoma carcinoma cells in the skin test was assigned to three groups.In the Thy-1~+ cells group,naked mice were inoculated with Thy-1~+ cells (5×10~5/mouse)underneath the skin of the back.In the HepG2 cell positive control group,naked mice were inoculated with HepG2 cells(0.5×10~7/mouse)underneath the skin of the back.In the Thy-1' cells negative control group,naked mice were inoculated with Thy-1~- cells(0.5×10~7/mouse)underneath the skin of the back.Tumorigenicity in each group was observed one month later.RESULTS AND CONCLUSION:Thy-1 expression in hepatoma carcinoma cells HepG2 and BEL7402 was significantly greater compared with normal hepatocytes QSG7701(P< 0.05).Positive rate of immunofluorescence staining was 85% in the Thy-1~+ cell group,1% in the HepG2 cell positive control group,0% in the Thy-1~- cell negative control group.There were nubble of five mice in HepG2 cell positive control and Thy-1~+ groups,and the volume was large.However,there was one nubble in Thy-1~- group,and the volume was significantly small(F=144.568,P< 0.05).Results indicated that hepatoma carcinoma cells contained Thy-1 labeled positive oval cells-like stem cells,which may be tumor stem cells in hepatoma carcinoma cells.

Bronchiolitis obliterans(BO) is chronic airflow obstruction syndrome characterized by small airway obstruction and airflow limitation.In children, BO is secondary to severe infection of respiratory tract, and its development is related with inflammation and immune response.At present, the diagnosis of BO mainly is depended on the clinical manifestation, lung function and examination results of high resolution computed tomography (HRCT) because of difficulty of lung biopsy.Pulmonary HRCT is a non-invasive examination, and worthy of generalization.Currently, BO has no accepted effective treatment,early diagnosis and treatment can block the progression of BO.Due to lack of realization of BO, doctors and parents fail to give enough attention, BO has a prolonged course and very poor prognosis.

7.Systemic Monitoring of Blood Glucose Level in Small for Gestational Age Infants

Objective To investigate the distribution and duration of abnormal blood glucose level in SGA infants. Methods Blood glucose level was systemicly monitored in 37 SGA infants for (133?115) h(range 16~524 h, median 93 h), who were non NICU hospitalized and no feeding or IV infusion when admitted at (2 6?2 4) h (range 0.25~10.0 h, median 2.0 h) of age. Results During monitoring period, there was hypoglycemia in 19 cases (51%),hyperglycemia in 2 cases (5%), and both hypo and hyperglycemia in 3 cases (8%). The median of last time abnormal glucose level appeared was 10 hr after birth, its 95% confidence interval was 5~55hr after birth. Multiple regression analysis shown delivery models bad relationship with glucose level at admition (p= 0.013). The blood glucose level was higher in vaginal delivery babies. Objective: To research the comparability of water vapor transmission of laminated films for pharmaceutical packaging determined by different instruments, methods and experimental conditions to provide reference and advice for the related inspection personnel.Methods: The principle, test parameters, conditions and scope of application for the testing methods for water vapor transmission in the national standard (GB) and international standards (ISO, ASTM) were compared by using three kinds of instruments based on different testing principles.Results: Different temperature and humidity conditions had great influence on the results of the same instrument.Under the same temperature and humidity, the instrument with different principles had little influence on the test results.Conclusion: Testing personnel should regularly use the standard membrane to calibrate the instrument of the sensor method.The temperature and humidity should be strictly controlled during the test.They should take measurement accuracy, personnel operation and economic loss into consideration to select the suitable method.
9.Effects of glutamine on immune function in elderly patients with malnutrition

Objective To evaluate the effects of glutamine on immune function in elderly patients with malnutrition.Methods Twenty-four elderly patients with malnutrition were equally randomized divided into glutamine group and control group.Patients in both two groups were administered with enteral nutrition containing equal nitrogen and calorics for consecutively 7 days.In the glutamine group,patients were also supplied with oral glutamine (7.5g tid).Changes of plasma IgG,IgA,and IgM levels,percentages of T-lymphocyte subsets (CD3,CD4,CD8),and CD4/CD8 ratio were determined before and after treatment.Results Plasma IgG and IgA lev-els,percentages of CD3 and CD4,and CD4/CD8 ratio were significantly higher in glutamine group than in control group (P<0.05 or P<0.01).Conclusion Glutamine can improve the immune function in elderly patients with malnutritiop and promote their recovery.
10.Pathological Changes of auxiliary liver allografts in the rat

Wistar rats randomly divided into groups A,B and C were killed at 2、3 and 12 days after transplantation,respectively.The pathological changes of the donor livers were observed by light microscope and electron microscope.The findings demonstrated that:(1)in the group A there was hepatocellular regeneration including hypertrophy of nuclei and nucleoli,and hyperplasia of mitochondrions and endoplasmic reticula;(2)in the group B the morphologic changes of the allografts involved in distinet fatty degencration and mitochondrial swelling of hepatic cells and peripheral vesscl wall infiltration by monocytes and lymphocytes;(3)in the group C hepatocellular loss with the massive tissue necrosis and fibroplasia occurred in the gratfs.From the above it was concluded that hepatocellular necrosis or loss related to the rejection 12 days after the transplantation might lead to the liver failure in the rats.
