1.Protection of Polysaccharide GEP-2 from Gastrodia elata on Carbon Tetrachloride and Alcoholic Liver Injury in Mice

Objective To observe the protection effect of Polysaccharide GEP-2 from Gastrodia elata on carbon tetrachloride(CCl4)and alcoholic liver injury in mice.Methods CCl4 and alcohol were adopted to establish CCl4 and alcoholic liver injury model in mice respectively.Colorimetric method was used to assay the content of serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST),and the content of triglyceride(TG)in alcoholic liver injury,the activity of superoxide dismutase(SOD)and malondiadehyde(MDA)consistent of liver homogenate in mice,and observe liver tissue pathological section.Results Polysaccharide GEP-2 from Gastrodia elata(25,50,100 mg/kg)administration obviously reduced the high level of ALT and AST in serum and the content of TG in alcoholic liver injury,inhibit the high level of MDA and increase the low activity of SOD in liver homogenate.Patholory inspection result indicated that Polysaccharide GEP-2 from Gastrodia elata exert notable influence on CCl4 and alcoholic liver injury in mice.Conclusion Polysaccharide GEP-2 from Gastrodia elata has apparently protective effects on CCl4 and alcoholic induced liver injury in mice.
2.Protection of polysaccharide GEP-2 from gastrodia elata on the immunity-mediated liver injury induced by BCG+LPS in mice

Objective:To observe the protection of polysaccharide GEP-2 from Gastrodia elata on the immunity-mediated liver injury induced by BCG+LPS in mice.Methods:BCG and LPS were adopted to establish BCG+LPS liver injury model in mice. The contents of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), the activity of superoxide dismutase (SOD),glutathion reductase (GSH-Px) and malondiadehyde (MDA) consistent of liver homogenate in mice were measured by colorimetric method. The content of tumor necrosis factor-?(TNF-?)and interleukin-1( IL-1)in serum were measured by enzyme linked immunosorbent assay (ELISA), and T-and B-lymphocyte proliferation were measured by MTT.Results:Polysaccharide GEP-2 from Gastrodia elata (25,50,100 mg/kg) could obviously reduce the high level of ALT and AST,and serum TNF-? and IL-1 either.It inhibited high level of MDA, increased the low activity of SOD and GSH-Px in liver homogenate,and enhanced T-and B-lymphocyte proliferation.Conclusion:Polysaccharide GEP-2 from Gastrodia elata had apparently protective effects on the immunity liver injury in mice induced by BCG+LPS.

Objective To investigate the effect of high glucose on proliferation of rat mesangial cells and reactive oxygen species (ROS) in culture and the intervention of polysaccharides-2b from Mudan Cortex (PSM2b). Methods Cell proliferation was assessed by MTT colorimetric assay. Colorimetric assay was used to detect SOD and MDA in the supernatant of high glucose culture of rat mesangial cells. Results PSM2b could significantly inhibit the proliferation of mesangial cells, the inhibiting effect showed a dose-dependent manner. And SOD increased with PSM2b compared with that only with high glucose, while MDA decreased. Conclusion High glucose contributes to an increase in rat mesangial cells and ROS produced by rat mesangial cells and PSM2b can better control high glucose-induced proliferation of rat mesangial cells and oxidative stress.
5.Anti-aging effects of polysaccharides from rhizomorph of Armillaria mellea

AIM: To study the anti-aging effects of polysaccharides from rhizomorph of Armillaria mellea(ARP) on fruit fly and mouse. METHODS: Regarding fruit files and D-galactose induced aging mice as research objects,observing the fruit flies lifespan;The ability to learn and memorize on model mice were observed by jumping platform test and maze test,the body weight,thymus and spleen index,SOD and GSH-Px activities、MDA and NO levels in serum,liver and brain were measured. RESULTS: ARP could significantly lengthen the lifespan of fruit flies;Obviously improve the ability to learn and memorize,markedly increase the body weight and resist the atrophy of thymus and spleen of model mice,SOD and GSH-Px activities increased obviously,MDA and NO levels decreased dramatically on model mice. CONCLUSION: ARP has anti-aging effects by immune-regulatory and eliminating free radicals.

AIM: To observe the effects of polysaccharides-2b from mudan cortex of Paeonia Suffruticosa Andr on glycocylation hemoglobion(GHb),blood glucose(BG),superoxide dismutase(SOD),the content of malonaldehyde(MDA) in serum and blood rheology of diabetic rats. METHODS: Diabetic rats were induced by associating streptozotocin(STZ) with freund's adjuvant complete(CFA),and then the diabetic rats were divided randomly into five groups: model group,polysaccharides-2b lower dose group(30 mg/kg),middle dose group(60 mg/kg),high dose group(120 mg/kg),positive group.Normal group was established additionally.PSM2b was perfused into stomach in diabatic rats for 60 days.The blood glucose was measured on the 10 th,30 th and 60 th days.The rehologic indexes,GHb,SOD and MDA were measured on the 60 th days. AIM To study the effect of polysaccharides 2b from Mudan cortex (PSM 2b) on type 2 diabetes mellitus (T2DM) rats and explore its mechanism. METHODS T2DM rat model was established by low dosage streptozotocin and high sucrose fat diet. The drugs were administrated by ig for 5 weeks. The serum glucose was assayed with GOD POD method. Insulin was determined by radioimmunoassay. Insulin receptors of the plasma membrane from rat liver were determined with radioligand binding assay of receptors (RBAR). RESULTS PSM 2b could significantly lower fasting blood glucose (FBG), improve impaired glucose telarance (IGT) and dyslipidemia. It could also remarkably raise receptor total (RT2) of the low affinity insulin receptor (InsR) and insulin sensitivity index (ISI) in T2DM rats. CONCLUSION PSM 2b has an obvious therapeatic effect on T2DM in rats. Its mechamsm is relatively improve insulin resistance (IR) in T2DM rats by raising the number of InsR.
8.Polysaccharide from the rhizomorph of Armillaria mella(AMP-1)protects INS-1 cells from alloxan injury

Aim To investigate the protective effects of polysaccharides from the rhizomorph of Armillaria mella(AMP-1) on alloxan injured INS-1 cells.Methods Graded concentrations of AMP-1(2,10,50,100,500,1 000 mg?L-1) were added into the culture medium of alloxan injured INS-1 cells.The survival rate was measured by MTT assay.The amount of glucose-stimulated insulin secretion in different concentrations of AMP-1 was determined by radioimmunoassay(RIA).SOD and NOS activity,NO,MDA and GSH production were assayed colorimetrically.Results AMP-1 could reduce oxidative injuries induced by alloxan in INS-1 cells.The survival rate of cells treated with AMP-1 increased significantly.In the presence of 5.6 mmol?L-1 or 16.7 mmol?L-1 glucose,AMP-1(50,100,500,1 000 mg?L-1)increased glucose-induced insulin secretion in INS-1 cells in a dose-dependent manner.NOS levels and the production of NO and MDA decreased significantly by AMP-1,while SOD levels and the production of GSH increased.Conclusions AMP-1 promoted glucose-induced insulin secretion in INS-1 cells by increasing the abilities of scavenging the free radicals induced by alloxan.
9.Determination of relative molecular weight in polysaccharide-2b of moutan(PSM_(2b))

Objective To determine the relative molecular weight(M_r) of polysaccharide-2b of moutan(PSM_(2b)) to establish the standard of reference substance.Methods PSM_(2b) was separated by a Sepharose 4B column and then PSM_(2b-A) and PSM_(2b-B) were obtained after cryodesiccation.Then HPLC was used to determine their M_r.Ultrapure water was used as mobile phase to compare with 0.7% Na_2SO_4 solution.Results PSM_(2b-A) was pure.The M_r and polydispersity(D) of PSM_(2b-A) obtained by use of dextran as standard were 3.596 2?10~(5) and 4.15 in 0.7% Na_2SO_4 solution phase,respectively,and in ultrapure water phase the M_r and D were 4.758 5?10~(5) and 1.03,respectively.Conclusion Different results are detected by different mobile phases in determination of M_r and D of PSM_(2b-A) by HPLC.And the ultrapure water as mobile phase is more suitable than 0.7% Na_2SO_4 solution.
