2.Establishment of human mesangial cell line with high expression of type A scavenger receptor and its function
Basic & Clinical Medicine 2006;0(10):-
Objective To determine the function and binding properties of SR-A in HMC.Methods A human mesangial cell line(HMCL) with high expression of type A SCR(HMCL-SCR) was established after stable transfection of expressive vector with cDNA encoding SR-A.Uptake of fluorescence Dil-labeled acetylated low density lipoprotein(Dil-Ac-LDL) and oxidized LDL(Ox-LDL) by HMC were evaluated by fluorescence microscopy,con-focal microscopy and Oil Red "O" staining respectively.SCR mRNA expression was examined with reverse transcription-polymerase chain reaction(RT-PCR).Results More uptake of Ox-LDL and Ac-LDL was observed in HMCL-SCR than that in the untransfected HMCL.Unlabeled Ox-LDL and Ac-LDL at a 40-fold excess concentration competed significantly with the uptake of Dil-Ac-LDL by HMC.Otherwise this phenomenon was not observed when co-cultured with LDL.Conclusion This results suggest that SR-A mediates the uptake of modified-LDL with high affinity and high specificity in human mesangial cells.

8.The effects of taurine on the expression of Caspase-3 and NF-?B in dorsal root ganglion diabetic rats
Journal of Chongqing Medical University 2007;0(08):-
16.7mmol/L)48hours after injection. The successful diabetic rats models were randomly divided into diabetic mellitus(DM) group and taurine(TA) group.DM and TA groups were subdivided into DM1,DM2 and TA1,TA2 groups according to the duration of diabetes. The expression of Caspase-3 and NF-?B mRNA were studied by reverse-transcriptase polymerase chain reaction(RT-PCR) method. Results:NO obvious expression of Caspase-3 and NF-?B mRNA were found in the nomal groups compared with DM group after the 4th and 8th week.Under the intervention of taurine, the expression of Caspase-3 and NF-?B mRNA were inhibited compared with those in model.group(P
9.CT of sixteen children with intervertebral disc calcification and comparing its diagnostic value with roentgnograph
Journal of Chongqing Medical University 2007;0(10):-
Objective:To explore the CT manifestations of intervertebral disc calcification(IDC)in children patients and its di-agnostic value in comparison with plain film. Methods:CT and plain film data of sixteen children with IDC were collected. The images were analyzed from the following aspects:counting the involved discs of each patient, locating the calcification in each ill disc and nearby tissue, observing the morphological features of calcification, and vertebral tube involvement.Results:①Fifteen patients had one or more than one cervical disc involved, but in only one patient, calcification involved five thoracic discs.Twenty-six discs were calcified in all,in which cervical disc accounted for 77%,and thoracic disc 23%.Most calcifications were nodular or patchy with different size.Sixteen calcification located at the anterior half segment of involved discs;Eight in the central part;only two at the posterior part.②CT and plain film detected calcification respectively of 26 and 25 discs. The for-mer was superior in displaying the location and appearance of calcification, and disclosing vertebral tube and nearby tissue in-volvement.Conclusion:CT is better than plain film in disclosing the details of ill disc.
10.Clinicopathological study of extraskeletal myxoid chondrosarcoma
Journal of Chongqing Medical University 2003;0(05):-
Objective:To investigate the clinicopathological characteristics of extraskeletal myxoid chondrosarcoma(EMC).Methods:Nine cases of extraskeletal myxoid chondrosarcoma were studied.Immunohistochemical analysis was performed in all cases and ultrastructural studies were done in 2 extraskeletal myxoid chondrosarcomas.Follow-up information was available for seven patients. Results:There were 7 males and 2 females whose ages ranged from 31 to 69 years(median 52.78 years).Local pain or tenderness and the presence of a palpable mass were the main complaints of the patients. The tumors were located mainly in the lower extremities(66.7%).Most tumors were deep-seated.They usually had a distinct multinodular configuration delineated by fibrous connective tissue. The tumor cells were arranged in delicate intersecting strands, rings, and garlands for the most part. The myxoid matrix was abundant in most cases.Immunohistochemical analysis was performed in all cases and ultrastructural studies were done in 2 extraskeletal myxoid chondrosarcomas.EMC expressed vimentin(100%,9/9),neuron-specific enolase(77.8%,7/9),S-100 protein(66.7%,6/9),synaptop hysin and chromogranin A(22.2%,2/9),None of the tumors expressed EMA and desmin.Ultrastructurally,EMC was characterized by distinct cords of cells immersed in a glycosaminoglycan rich matrix. The cells were rich in mitochondria, had well-developed Golgi apparatus and there were numerous smooth vesicles. In many cells, there were also prominent glycogen deposits and lipid droplets. Some tumor cells had intracisternal microtubules. In one of 2 extraskeletal myxoid chondrosarcomas there were 140~180 nm diameter membrane-bound dense-core secretory granules in cell bodies. Conclusion:Extraskeletal myxoid chondrosarcoma is a rare soft tissue sarcoma characterized by distinctive morphological and cytogenetical features. It was believed to represent a variant of soft-tissue chondrosarcoma owing to its histological resemblance to chondroblastic tissue in the early stages of cartilage development or chondroid tumors such as skeletal chondrosarcoma. However, the chondroid nature has been a subject of controversy, and its line of differentiation remains to be determined.A substantial proportion of EMC show immunophenotypic and/or ultrastructural evidence of neuroendocrine differentiation.EMC has high potential of local recurrence and metastasis, and a high disease-associated death rate.
