4.Application of Structural Equation Model in the Study of Therapeutic Effect of Chinese Medicine

Objective The questionaires of the ONFH aegers who were hospitalized in Kanggu hospital,during 2005-2008 ,were collect-ed to evaluate the therapeutic effects of acupotomy by proper statistical methods. Methods Four indexes according to the criteria of Therapeutic effect of adult ONFH recovery and reconstruction (ZHAO Dewei, 1996) were measured to evaluate the effect of the treatment. Results Monofac-torial analysis showed that the treatrnent was effective,but some phenomena could not be correctly explined in specialized field. This problem was re-solved appropriately by structural equation model(SEM). Conclusion By the method of SEM, the confounding factors could be better controlled and the evaluation was more reliable.
5.Apoptotic mechanisms of pirarubicin against bladder cancer and its clinical use for preventing postoperative tumor recurrence

Objective To study the antitumor mechanisms of pirarubicin (THP) and its effect on preventing postoperative recurrence of superficial bladder cancer. Methods MTT assay, flow cytometry and transmission electron microscope were used to assess the effects of different concentration of THP on T24 cell line. THP 40 mg in 50 ml distil water was used intravesically in 60 patients with transitional cell carcinoma after TURBT to prevent tumor recurrence. Results T24 cells were suppressed significantly by THP in concentrations of 10 mg/L and 100 mg/L,the suppressive rates being 80% and 94% respectively. Apoptosis peak was evident before G1 phase. The characteristics of cell apoptosis, such as bubble formation in cytoplasm and condensed chromosome, were typically manifested. Cells became necrotic when the concentration of THP was 1 000 mg/L. One course of THP intravesical administration was completed in 58 cases. All patients were followed up for a mean of 18.8 months (range 6~24 months), and tumor recurred only in 5 cases (8.6%). Conclusions Suppression of tumor cell growth and inducing apoptosis, even necrosis, might be the main antitumor mechanisms of THP. Clinically, intravesical administration of THP after TURBT was effective and safe for preventing tumor recurrence.
6.Study of digestive diseases diagnosed by M2A capsule endoscopy

Objective To evaluate the application of M2A capsule endoscopies ( CE) in the diagnosis of digestive tract diseases. Methods Forty - five patients (26 males, 19 females) , suffered from gastrointestinal bleeding, abdominal pain or chronic diarrhea were referred to this study and underwent capsule endoscopies. Their average age was 47. 8 years old (a range of 15 - 78). Capsule endoscopies were performed with the Given M2A video capsule system. The conventional diagnostic procedures (including small bowel radiograph, gastroenteroscopy, colonoscopy, etc. ) were done in 31 patients before or after the CE, thus the results of both procedures were compared. Results Among 45 patients, 44 have completed CE successfully. Any complication never experienced. The quality of photos was excellent. The significant pathological findings were revealed in 37 patients, diagnostic yield was therefore 84. 1% . Patients with esophagogastric diseases, small bowel diseases and colon diseases were found in 5,29 and 3 cases respectively. Thirty -one patients received a conventional diagnostic procedure before or after CE. Twenty one of them suffered from GI bleeding, pathological changes were found in 17 patients by capsule endoscopies, only 4 patients by conventional procedure; five of them suffered from abdominal pain, pathological changes were found in 5 patients by CE, only 2 patients by conventional procedure; 4 of them suffered from chronic diarrhea, pathological changes were found in 4 patients by CE, only 1 patient by conventional procedure. The pathological sites located by CE were quite similar to those by conventional diagnostic work-up. Conclusion CE has the advantages of high safety, excellent visualization and precise localization. Its efficiency in diagnosis is much better than that of conventional procedure, and is indispensable in the diagnosis of digestive tract diseases.
7.Various Amounts of rhGM - CSF cDNA Transfer Directly into Mice's Muscle in vivo

We have transferred the different amounts of the plasmid DNA pCGl, which was constructed by cloning rhGM-CSF gene into the eukaryotic expression plasmid pCDS, directly into the BALB/c mice's quadriceps pretreated with bupivacaine. The result of ELISA confirmed that rhGM - CSF was expressed and secreted in mice of group B injected 60?g of plasmid pCGl, but not in mice of group C injected 30?g of plasmid pCGl. The time for the highest level of rhGM-CSF expression was during the fifteenth to twentieth days after plasmid DNA pCGl was injected. Meanwhile, the results of biological activity detection showed that mice's serum of group B (injected 60?g) had the function of retaining the growth of TF - 1 cells. This demonstrates that the rhGM-CSF in mice's serum of group B has certain biological activity. This Model of animal will blaze a new trail in the treatment and prevention of the leukyopenia.
8.Determination of serum insulin-like growth factor and their binding protein-3 in liver cirrhosis and their clinical significances

Objective To investigate the relationship between insulin like growth factor (IGF Ⅰ)、IGF Ⅱ and IGFBP 3 and Child Pugh classification in patients with liver cirrhosis and to determine the potential clinical markers of functional hepatic reserves. Methods Forty four patients with posthepatitic cirrhosis were divided into 3 groups according to disease severity (Child Pugh Score) and 38 healthy subjects severed as controls. Serum levels of IGF Ⅰ, IGF Ⅱ and IGFBP 3 were measured in these groups by immunoradiometric assay. Results Baseline IGF Ⅰ, IGF Ⅱ and IGFBP 3 levels were significantly lower in patients with cirrhosis than in controls, and the serum concentrations of IGF Ⅰ, IGF Ⅱ and IGFBP 3 were associated with the marked changes of liver function due to Child Pugh score. They all showed a significant correlation with the degree of hepatic dysfunction and dropped markedly during the progression of liver failure. The sensitivity of IGF Ⅱ is much higher than that of IGF Ⅰ and IGFBP 3. It was found that IGF Ⅰ
9.Diagnosis and Treatment of Malignant Brain Tumor in Children

Objective To explore the diagnosis,treatment and pathology of malignant brain tumor in children.Methods The clinical data of 61 children with malignant brain tumor were reviewed,including their age distribution,diagnosis,operation route(according to the position of the brain tumors),chemotherapy [using bischloro-nitrosourea(BCNU),teniposide(VM-26) and temozolomide]and radiotherapy (part and whole brain irradiation,three diamensions conformal therapy,gamma knife and X-kinfe).Results Of 61 cases,which including 37 boys and 24 girls whose mean age was 11.3 years,24 cases were supratentorial tumors and 37 cases were infratentorial.Forty-five tumors received total or subtotal recession,10 cases with major recession,and 6 cases with partial recession.After operation,39 cases (63.9%) received radiotherapy,and 29 cases (47.5%) with chemotherapy.Follow-up of 49 cases were achieved,in which 22 cases died and 27 cases survived.Conclusions Surgical removal is still the chief treatment for malignant brain tumor in children.The exairesis profect should be formulated according to the specificity of each case.Radiotherapy methods should be related to post-operational images(CT or MRI).And it was different from the brain tumors in adult.It is good to prolong survival duration for some chosen cases with optimal chemotherapy.
