1.Reliability and validity of Chinese version of Revised Attribution Questionnaire-27 as used in medical students

Objective To develop the Chinese version of Attribution Questionnaire-27 and examine its replicability,reliability and validity.Methods The questionnaire was translated using the standard translation/back-translation method and examine its replicability,reliability and validity in 230 medical students who were selected by convenient method.Results The Cronbach α coefficient for Attribution Questionnaire-27 was 0.82 and ranging between 0.55 and 0.84 for the subscales.The test-retest reliabilities of the total scale was 0.77 and the subscales was between 0.61 to 0.81 after one week.Confirmatory factor analysis confirmed the hypothesized six factors attribution of responsibility model (x2/df=3,GFI=0.925,NFI=0.880,CFI=0.856,IFI=0.761,RMSEA=0.094) and three factors dangerous model (x2/df=1.773,GFI=0.959,NFI=0.941,CFI=0.973,IFI=0.973,RMSEA=0.058).The path analysis results support the existing theoretical assumptions.Condusions The Chinese version of Attribution Questionnaire-27 has good reliability and validity and can be used in medical student.
2.Study on resilience and its related factors of nurses

Objective To investigate the status of nurses' resilience and to explore the related factors.Methods 1356 nurses from 11 level Ⅱ and level Ⅲ hospitals of Guangzhou were assessed by using the general information questionnaire and the Chinese version of Conner-Davidson resilience scale.Results The score of nurses' resilience was(59.99±13.59)points and it was significantly lower than that of the domestic norm.There was statistical difference of the score of nurses' resilience on different genders,educational background,professional titles,hospital level and night shifts.There was a positive correlation among educational background,night shifts and nurses' resilience,and a negative correlation between hospital level and nurses' resilience.The hospital level,night shifts and educational background were the influencing factors of nurses' resilience.Conclusions The level of nurses' resilience is low.The higher hospital level and educational background,and the less night shift,the higher of nurses' resilience.

Objective To explore the resilience and its influencing factors of nurses.Methods I 356 nurses from 11 comprehensive hospitals of Guangzhou were assessed by Conner-Davidson Resilience Scale,General Self-Efficacy Scale,Simplified Coping Style Questionnaire and Chinese Nurse Job Stress Scale.Results ①The score of nurses' resilience was (59.99± 13.59) and it was significantly lower (t=-t4.656,P<0.001) than that of the domestic norm (65.4±13.9,n=560).There were statistic difference (P<0.05) among the total score and 3 dimensions scores of resilience on different educational level and professional rank of nurses.②Nurses' resilience had a obvious positive correlation with the educational level,professional rank,self-efficacy,positive coping(P< 0.01),and a obvious negative correlation with the negative coping and job stress(P<0.01).It can be assumed that educational level,self-efficacy,coping style and job stress had good predictive ability (47.4%).Conclusion The nurses have low resilience than general population.Educational level,self-efficacy,coping style and job stress are influencing factors of resilience.
4.Efficacy Observation and Analysis for Influential Factors of 49 Cases Acid Rebound in Proton Pump Inhibitors

OBJECTIVE:To investigate the acid rebound in proton pump inhibitors (PPIs) and its influential factors. METH-ODS:Totally 109 patients who treated with PPIs for 1 month in our hospital during Jan.-Dec. 2015 were collected,and telephone visit was conducted after 1 week withdrawal to the data of dyspeptic symptoms were input and scored by modified Glasgow Dyspepsia Sere-rity Score,then divided into no phenomenon group(<5 scores)and acid rebound in proton pump inhibitors group(≥5 scores). All patients were classified by basic diseases,age,gender,whether smoking and alcohol drinking,and phenomenon of acid rebound in PPIs were observed and analyzed. RESULTS:Totally 91 patients were observed,49 were classified as acid rebound in PPIs group. χ2 test showed elderly patients(χ2=5.350,P=0.021)and people with smoking and alcohol drinking(χ2=4.351,P=0.037)were associat-ed with the increased risk of acid rebound in PPIs;exclusion of the effects of gender and basic disease,Logistic regression analysis showed the risk of acid rebound in PPIs in elderly patients were 5.708 times to non-elderly patients [OR=5.708,95%CI(1.946, 16.746),P=0.002];people with smoking and alcohol drinking was 15.281 times to non-smoking and alcohol drinking [OR=15.281, 95%CI(2.748,84.965),P=0.002],the difference was statistically significant(P<0.05). CONCLUSIONS:Parts of patients show ac-id rebound after stopping PPIs,while elderly patients and patients with smoking and alcohol drinking are the high-risk population, which should be paid attention to.
5.Effects of Sanren decoction on Th1/Th2 cytokines in rats with spleen-stomach damp-heat syndrome.

OBJECTIVETo investigate the effects of Sanren decoction on the immune function of rats with spleen-stomach damp-heat (DHSS) syndrome.
METHODSFifty male SD rats were randomly allocated into normal control group, DHSS model group, and 3 Sanren decoction groups (high, medium and low doses). The effects of the decoction on the body mass, rectal temperature (RT), water and food intake, histopathological changes of the gastrointestinal mucosa and serum levels of interleukin (IL)-4 and interferon (IFN)-γ were evaluated.
RESULTSThe serum levels of IFN-γ and IL-4 in the model group significantly increased compared with those in the control group (P<0.01), with a slightly increased IFN-γ/IL-4 ratio (P>0.05). Sanren decoction obviously reduced the rectal temperature and significantly decreased the production of both cytokines. High-dose Sanren decoction caused more markedly decreased IL-4 level (P<0.05) to result in a significantly increased IFN-γ/IL-4 ratio (P<0.05).
CONCLUSIONSA shift of Th1/Th2 balance toward Th1 immune response is demonstrated in rats with DHSS syndrome, and Sanren decoction produces a protective effect on the gastrointestinal mucosa by immunoregulation.

6.Analysis of PKD2 gene variant and protein localization in a pedigree affected with polycystic kidney disease.

OBJECTIVE:
To detect the mutation site in a pedigree affected with autosomal dominant polycystic kidney disease (ADPKD) and verify its impact on the protein function.
METHODS:
Peripheral blood samples were collected from the proband and his pedigree members for the extraction of genomic DNA. Mutational analysis was performed on the proband through whole-exome sequencing. Suspected variant was verified by Sanger sequencing. A series of molecular methods including PCR amplification, restriction enzyme digestion, ligation and transformation were also used to construct wild-type and mutant eukaryotic expression vectors of the PKD2 gene, which were transfected into HEK293T and HeLa cells for the observation of protein expression and cell localization.
RESULTS:
The proband was found to harbor a c.2051dupA (p. Tyr684Ter) frame shift mutation of the PKD2 gene, which caused repeat of the 2051st nucleotide of its cDNA sequence and a truncated protein. Immunofluorescence experiment showed that the localization of the mutant protein within the cell was altered compared with the wild-type, which may be due to deletion of the C-terminus of the PKD2 gene.
CONCLUSION
The c.2051dupA (p. Tyr684Ter) mutation of the PKD2 gene probably underlay the pathogenesis of ADPKD in this pedigree.

7.Analysis of gene variant in a Chinese pedigree with preaxial polydactyly.

OBJECTIVE:
To analyze the pathogenic variant of preaxial polydactyly in a Chinese Han pedigree and identify the cause of polydactyly.
METHODS:
The peripheral blood DNA of the proband and her parents was extracted. The polydactyly-related genes were detected by trio whole exome sequencing, and the suspected pathogenic gene was screened out. Sanger sequencing was applied to other members of the pedigree.
RESULTS:
The results of gene sequencing showed that the LMBR1 gene had a heterozygous variant of c.423+4909(IVS5)C>T in 6 patients of the pedigree. The same variant was not detected in family members with normal phenotype. Based on the ACMG guidelines, c.423+4909(IVS5)C>T of the LMBR1 gene was predicted to be pathogenic (PM1+PM2+PP1-S(PS)+PP4+PP5).
CONCLUSION
The heterozygous C>T variant at position 4909 of intron 5 of the LMBR1 gene probably underlies the disease in this pedigree.

8.Identification of a novel SOD1 variant in a Chinese patient with amyotrophic lateral sclerosis.

OBJECTIVE:
To explore the genetic basis for a Chinese patient with amyotrophic lateral sclerosis (ALS).
METHODS:
Peripheral blood samples were collected from the patient and his parents for the extraction of genomic DNA. Genetic variant was identified by whole exome sequencing. Candidate variant was verified by Sanger sequencing of his parents and healthy controls.
RESULTS:
The patient was found to harbor a heterozygous c.420C>G (p.Asn140Lys) variant of the SOD1 gene. The same variant was not detected in his parents and 100 healthy controls. The variant has not been included in HGMD, dbSNP and other databases.
CONCLUSION
The c.420C>G variant of the SOD1 gene may underlie the ALS in this patient. Above finding has enriched the spectrum of SOD1 gene variants.

9.Effect of head acupuncturing therapy combined with massaging key head acupoints on language and cogni-tive function in children with autism

Objective To explore the effect of head acupuncturing therapy combined with massaging key head acupoints on language and cognitive function in children with autism.Methods 60 children with autism who visited the Department of Pediatric Rehabilitation of our hospital from January 2021 to June 2022 were selected as subjects,who were then randomly divided into the conventional group treated with the conventional rehabilitation training and the combination group treated with head acupuncturing therapy combined with massaging the key acu-points,apart from conventional rehabilitation training 30 cases in each group.The cognitive function,developmental status,and parental quality of life before and after the treatment were compared between the two groups,and so were the language function and safety.Results There were no significant differences in the scores by Autism Behavior Checklist(ABC)and Childhood Autism Rating Scale(CARS),adaptability scores,language scores,and individual-social interaction scores between the conventional group and the combination group before the treatment(P>0.05).After the treatment,the scores by ABC and CARS were significantly decreased in both groups,and the scores of the combination group were significantly lower than those in the conventional group(P<0.05);The adaptability scores,language,individual-social interaction scores were significantly increased in both groups,and the scores of the combined group were significantly higher than those of the conventional group(P<0.05).The abnormal rates of symbolic expression,language comprehension,action-oriented subjects,and language development delay in the combined group were all significantly lower than those in the conventional group(P<0.05).There was no significant difference in the scores by the QL-Index between the conventional group and the combined group before the treatment(P>0.05),but after the treatment,the QL-Index scores of the conventional group and the combination group were increased,and the score of the combination group was significantly higher than that of the conventional group(P<0.05).The results of blood routine,urine routine,stool routine,liver function,and kidney function in both groups were not significantly abnormal.There was one case of mild swelling and pain at the acupunctured site in the combination group,which was improved without intervention.Conclusion Head acupuncturing therapy combined with massaging the key head acupoints can improve language function,cognitive function,and development of autis-tic children.In addition,it can enhance parental quality of life and have a high level of safety.
10.Efficacy and Mechanism of Lutongning Granules in Treatment of Trigeminal Neuralgia Induced by Injection of Talc into Infraorbital Foramen of Model Rats Based on P2X7R-mediated Neuroinflammation

ObjectiveTo evaluate the effectiveness of Lutongning granules in the treatment of trigeminal neuralgia in animal models and study its mechanism of action, so as to provide laboratory data support for the clinical application of Lutongning granules and precise treatment. MethodMale SD rats were randomly divided into normal group, model group, carbamazepine group (0.06 g·kg-1·d-1), high-dose Lutongning group (2.70 g·kg-1·d-1), and low-dose Lutongning group (1.35 g·kg-1·d-1) according to the stratified basic mechanical pain thresholds, with 10 rats in each group. A trigeminal neuralgia model of rats was prepared by injecting 30% talc suspension into the infraorbital foramen area of the rat. The drug groups were administered 10 mL·kg-1 of drugs by gavage after 2 h of modeling. The normal group and the model group were administered distilled water by gavage under the same conditions once a day for 10 consecutive days. Von Frey brushes were used to determine the mechanical pain threshold of rats. A fully automated blood and body fluid analyzer was employed to detect the blood routine of rats. Hematoxylin and eosin (HE) staining was utilized to detect the pathological changes in the trigeminal ganglion and medulla oblongata tissue. Transmission electron microscopy was used to scan the ultrastructure of the medulla oblongata tissue. Enzyme-linked immunosorbent assay (ELISA) was used to detect the levels of inflammatory factors interleukin (IL)-1, IL-6, IL-8, tumor necrosis factor (TNF)-α, neuropeptide substance P, and β-endorphins (β-EP) in the serum of rats, and Western blot was used to detect the protein expression levels of IL-1β, purinergic receptor P2X7 (P2X7R), and phosphorylated p38 mitogen-activated protein kinase (p-p38 MAPK). ResultCompared with that in the normal group, the pain threshold of rats in the model group was significantly lower (P<0.01). The absolute value of neutrophils (NEUT#) and the percentage of neutrophils (NEUT) were significantly improved, and the percentage of lymphocytes (LYMPH) was significantly reduced (P<0.01). The serum levels of IL-1, IL-6, IL-8, and TNF-α were significantly increased (P<0.01). SP content in brain tissue was significantly increased, and β-EP content was significantly decreased (P<0.01). The relative protein expression of IL-1β, P2X7R, and p-p38 MAPK was significantly increased (P<0.05). HE staining and transmission electron microscopy results of medulla oblongata tissue revealed neuronal degeneration, mild proliferation of microglial cells, reduction in the number of myelinated nerves, and obvious demyelination. The trigeminal nerve fibers of rats were disarranged, and some nerve fibers showed vacuolization. Axons were swollen, and Schwann cells proliferated. Demyelination was observed. Compared with the model group, each administration group significantly increased the pain threshold of rats (P<0.05, P<0.01), reduced NEUT# and NEUT, and elevated LYMPH (P<0.05, P<0.01). The administration group significantly decreased the levels of IL-1, IL-6, IL-8, and TNF-α in serum and SP in brain tissue (P<0.01) and increased the level of β-EP (P<0.01). They significantly down-regulated the protein expression of IL-1β, P2X7R, and p-p38 MAPK(P<0.05, P<0.01) and significantly ameliorated the pathological changes in medulla oblongata tissue and trigeminal nerves of rats. ConclusionLutongning Granules had significant therapeutic effects on trigeminal neuralgia induced by injection of talc into the infraorbital foramen of model rats, and the mechanism may be related to amelioration of P2X7R-mediated neuroinflammation and inhibition of demyelination of myelinated nerves.
