1.Dynamic changes and significance of peripheral blood Th17/Treg in chronic hepatitis B patients

Objective:To investigate the significance of T helper type 17 cells(Th17)/T regulatory cells (Treg) in the prognosis of chronic hepatitis B(CHB) by comparing the changes of Th17 cells ,Treg cells and their rate under different therapy in chronic hepatitis B patients.Methods: 40 patients infected with chronic hepatitis B(CHB) were divided into chronic hepatitis B without antiviral(CHBWA) group and chronic hepatitis B antiviral(CHBA) group depending on whether nucleoside antiviral was applied,with 20 cases each,10 cases in asymptomatic hepatitis b carriers(AsC) group.10 cases in health control (HC) group.Flow cytometer(FCM) was adopted to detect peripheral blood Th17 and Treg cell frequency in CHB patients at 0,1 month in CHBWA group ,at 0,1,3 month in CHBA group,AsC patients and HC cases ,then calculating Th17/Treg ratio and observing the dynamic changes at different disease states.Results: After the liver-protecting treatment in CHBWA patients,the Th17/Treg ratio at 1 month(0.39±0.11)decreased compared to 0 month(1.20±0.26)rapidly(P<0.01),and the ratio nearly reached the level of AsC and HC group (P>0.05).Ratio of Th17/Treg and ALT had good positive correlation (r=0.709,P=0).In CHBA patients group,after the liver-protecting and antiviral treatment,patients′ Th17/Treg ratio decreased rapidly in the first month(0.73±0.32) and third month(0.76±0.44),and there′s statistical difference compared with 0 month(1.18±0.27,P<0.01),but there was no significant change of Th17/Treg ratio after being treated for 3 months compared with 1 month,and the Th17/Treg ratios at 0,1,3 month were higher than that in AsC group and HC group(P<0.01).Ratio of Th17/Treg and ALT have good positive correlation (r=0.500,P=0.000),also the ratio of Th17/Treg and HBV DNA were positively correlated (r=0.345,P=0.007).Comparison of peripheral blood Th17,Treg cells frequency,Th17/Treg ratio between CHBWA and CHBA group at 0 month had no statistical significance(P>0.05),while the Th17/Treg ratio in CHBA group was higher than CHBWA group(t=4.471,P<0.01) after 1 month of treatment.Conclusion: In the CHB patients,antiviral treatment will influence the Th17,Tregs cells frequency,resulting the change of Th17/Treg ratio,but also the ratio may be associated with virus removal.
2.Detection and clinical significance of serum interferon-inducible protein-10 and interferon-γ in patients with severe hepatitis B

Objective To explore the relationship between serum levels of interferon-inducible protein-10 (IP-10), interferon-γ (IFN-γ) and hepatic inflammatory reaction, disease progression in patients with severe hepatitis B (SHB). Methods Sera of 40 patients with SHB at time of admission,at the beginning of single plasma exchange (PE), at time of PE completion and 5 days after PE. The SHB patients were divided into improved group and aggravated group. And 20 patients with chronic hepatitis B (CHB) and 20 healthy controls were enrolled in this study. Serum levels of IP-10, IFN-γand tumor necrosis factor-α (TNF-α) were determined by enzyme-linked immunosorbent assay (ELISA). Results The serum levels of IP-10 in patients with SHB and CHB on admission were (683.6 174.6)ng/L and (216.1 102.9)ng/L, respectively, which were notably higher than those in healthy controls [(107.6 55.8)ng/L F=9.036, both P<0. 01],and those in patients with SHB was significantly higher than that in patients with CHB (P<0. 01). The serum level of IFN-γ in patients with SHB and CHB on admission were (19. 8 8. 8) ng/L and (16. 7 7. 8) ng/L,respectively, which were significantly higher than those in healthy controls [(2.6 1.2) ng/L F=9. 288, both P<0. 01]. The serum level of IP-10 and IFN-γ were both positively correlated with TNF α (r=0. 366 and r=0. 365, respectively;P<0.05) and both negatively correlated with prothrombinase activity (r=-0.401 and r=-0.350, respectively;P<0.05), but not correlated with serum total bilirubin(r=0. 223 and r=0. 219, respectively;P>0.05). The serum level of IP-10 and IFN-γ were positively correlated ( r= 0. 602 ; P= 0. 000 ). On day 5 after PE, serum level of IP-10 in patients with SHB was significantly decreased compared with that'in patients before PE (t= 8. 947, P<0.01 in improved group;t=4. 121, P<0.05 in aggravated group) and that in aggravated group was significantly higher than improved group (t=7.862, P<0.01). But serum level of IFN-γ was not decreased significantly (t=0. 491, P>0.05). Conclusions IP-10 and IFN-γ are involved in the hepatic immunopathological mechanism. Serum level of IP-10 is correlated with the severity of hepatic inflammatory injury and IP-10 could reflect the progression and development of disease in patients with SHB.
3.The changes of the ratio of T helper 17 and regulatory T cells in the peripheral blood of patients with anti-tuberculosis drug-induced liver injury and its significance

Objective To observe the changes of the peripheral blood T helper (Th)17 cells/regulatory T cells (Treg) ratio in patients with anti‐tuberculosis drug‐induced liver injury ,and to explore the relationship between Th17/Treg ratio and liver function . Methods Ninety inpatients with the diagnosis of secondary pulmonary tuberculosis (TB) from Affiliated Hospital of Zunyi Medical College from Feb .2013 to Mar .2015 were included in this study .All these patients had normal base line liver function .After 2 weeks of anti‐TB chemotherapy ,21 patients with alanine transaminase (ALT) and/or aspartate aminotransferase (AST) ≥2 × upper limit of normal (ULN) were defined as TB with liver injure group ,the other 69 patients with ALT and/or AST<2 × ULN were defined as TB with non‐liver injured group .Before treatment and two weeks after anti‐TB therapy ,the peripheral blood Th17/Treg ratios and liver functions of all patients in both groups were detected in order to compare the changes of Th17/Treg ratios pre and post anti‐TB chemotherapy and to explore the relationship between Th17/Treg ratio and ALT/AST levels .Student′s t test was used for statistical analysis ,and Pearson correlation analysis was used for correlation test .Results Before treatment ,there was no statistical significance of Th17 ,Treg and Th17/Treg ratio between groups (P> 0 .05) .The Th17 levels in TB with liver injured group ([2 .522 ± 0 .388]/mL vs [2 .075 ± 0 .369]/mL ,t=3 .633 ,P<0 .01) and TB with non‐liver injured group ([2 .326 ± 0 .348]/mL vs [1 .929 ± 0 .402]/mL ,t= 6 .468 , P< 0 .01) ,and Th17/Treg ratios in both groups(0.618±0.104vs0.489±0.107,t=3.553,P<0.01 ;0.554±0.108vs0.450±0.098,t=6 .353 ,P<0 .01) were all increased after anti‐TB chemotherapy ,with statistically significant differences . However ,there was no statistical significance of Treg levels pre and post anti‐treatment in both groups (both P>0 .05) .The Th17 level and Th17/Treg ratio in TB with liver injured group were significantly higher than those in TB with non‐liver injured group (t= 2 .203 , P< 0 .05 and t= 2 .345 , P< 0 .05 , respectively) .In liver injured group ,the Th17/Treg ratio was positively correlated with ALT level (r=0 .849 ,P=0 .044) ,but not with AST level (P>0 .05) .Conclusion The changes of Th17/Treg ratio are related with anti‐TB drug‐induced liver injury in patients with secondary pulmonary tuberculosis .
4.The dynamic changes and significance of T helper type 17 cells and cell-related cytokines in the peripheral blood of acute hepatitis B patients

Objective To investigate the expression of T helper type 17 cells (Th17) and cell‐related cytokines ,including interleukin (IL)‐21 ,IL‐22 ,IL‐23 in the peripheral blood of different clinical stages of patients with acute hepatitis B (AHB) .Methods Ten cases of AHB patients were enrolled .The frequency of Th17 cells in the three clinical stages (i .e .acute phase ,convalescent phase and resolved phase) were detected by flow cytometry . IL‐21 , IL‐22 and IL‐23 were measured by enzyme‐linked immunosorbent assay (ELISA) .Control group was composed of ten healthy subjects .The comparison between the two groups was done by t test and the differences among multiple groups were compared by one way ANOVA .Pearson correlation analysis was used for correlation analysis .Results The frequency of Th17 in healthy controls was (0 .68 ± 0 .29)% ,while those in acute phase ,convalescent phase and resolved phase of AHB patients were (18 .22 ± 4 .13)% , (3 .14 ± 1 .90 )% and (3 .31 ± 0 .95 )% , The differences between the two groups were significant (t= 13 .405 ,4 .047 and 8 .342 , respectively ;all P< 0 .01) .The levels of IL‐21 ,IL‐22 and IL‐23 in healthy controls were (42 .00 ± 6 .95) ,(315 .89 ± 96 .16) and (11 .95 ± 6 .95) ng/L ,respectively .Those in acute phase of AHB patients were (575 .39 ± 47 .01) ,(648 .44 ± 47 .12) and (38 .29 ± 4 .68) ng/L ,respectively ,those in convalescent phase were (366 .50 ± 33 .74) ,(405 .04 ± 47 .12) and (25 .10 ± 4 .69) ng/L ,respectively ,while those in resolved phase of AHB patients were (46 .62 ± 8 .28) ,(365 .94 ± 45 .62) and (15 .29 ± 4 .69) ng/L , respectively .Compared with healthy controls ,t values of the levels of IL‐21 in three different phases of AHB patients were 35 .497 ,29 .792 and 1 .354 with P value of <0 .01 ,<0 .01 and 0 .193 ,respectively ;those of IL‐22 were 9 .820 ,2 .632 and 1 .487 with P value of < 0 .01 ,0 .021 and 0 .161 ,respectively ;those of IL‐23 were 9 .944 ,4 .961 and 1 .260 with P values of <0 .01 ,<0 .01 and 0 .226 ,respectively . After comparison of IL‐21 ,IL‐22 and IL‐23 among three different phase of AHB ,F values were 622 .784 , 107 .772 and 60 .743 with all P values less than 0 .01 ,respectively .The levels of IL‐21 ,IL‐22 and IL‐23 were all positively correlated with the serum ALT level in acute phase (r= 0 .655 ,0 .666 and 0 .673 , respectively ;all P<0 .05) .Correlation analysis demonstrated that the frequency of Th17 was positively correlated with the levels of IL‐21 , IL‐22 and IL‐23 in acute phase ( r= 0 .879 ,0 .866 and 0 .879 , respectively ;all P<0 .01) .The frequency of Th17 was positively correlated with the level of IL‐21 in the resolved phase . No correlations between the remaining groups were confirmed . Conclusion The expressions of Th17 and cell‐related cytokines ,including IL‐21 ,IL‐22 and IL‐23 decline with the recovery of A HB .
5.Effects of zinc on carbon and nitrogen metabolism in leaves of Pinellia ternata and their yield

Objective To find out the proper concentration of ZnSO4?7H2O as foliar fertilizer used for Pinellia ternata, and to provide reference for high yield of P.ternate,effects of zinc on the carbon and nitrogen metabolism in leaves of P.ternata and the yield were studied.Methods Seedlings of P.ternata were employed as the experiment material.At the stage of full seedling,ZnSO4?7H2O of different concentration was sprayed on the leaves,the indexes of carbon and nitrogen metabolism were determined on the day 10,the weight of tubers and bulbils were measured at the sprout tumble stage.Results The yield of P.ternata has an exceedingly significant positive correlation with the contents of soluble sucrose and starch in the leaves,a significant positive correlation with the content of photosynthetic pigment and the aldolase activity,and a negative correlation with the amylase activity.In addition,the yield shows both an exceedingly significant positive correlation whith the nitrate reductase activity and a significant positive correlation with the content of protein.At the stage of full seedling,zinc sprayed on leaves can increase the content of photosynthetic pigment and improve the capability of photosynthesis.Meanwhile,spraying zinc can also promote the accumulation of soluble sucrose and starch in the leaves because of its stimulation on aldolase and inhibition on amylase.Besides,spraying zinc on leaves could improve the activity of nitrate reductase and the content of protein,and finally increase the yield of P.ternata.Conclusion The concentration of 600 mg/L is the best for ZnSO4?7H2O used as foliar fertilizer for P.ternate at the stage of full seedling.
6.Age-, period- and cohort-specific incidence of sexually transmitted diseases in Yantian District from 2008 to 2022

Objective:
To investigate the incidence trend of sexually transmitted diseases (STDs) in Yantian District, Shenzhen City, Guangdong Province from 2008 to 2022, and the effects of age, period, and cohort on STDs incidence, so as to provide the basis for formulating control measures for STDs.
Methods:
Data of reported STDs cases (syphilis, gonorrhea, genital Chlamydia trachomatis infections, condyloma acuminatum and genital herpes) among population aged 15 to 64 years in Yantian District from 2008 to 2022 were collected through the Chinese Disease Control and Prevention Information System. The temporal and population distribution of STDs were descriptively analyzed. Trends in incidence of STDs were analyzed using average annual percent change (AAPC). Impacts of age, period and cohort on the incidence of STDs were analyzed by an age-period-cohort model.
Results:
A total of 6 156 cases of STDs aged 15 to 64 years were reported from 2008 to 2022, and the average annual incidence was 229.92/105. The overall incidence showed an upward trend (AAPC=2.409%, P<0.05), and an upward trend among females (AAPC=5.846%, P<0.05), but the trend was not statistically significant among males (AAPC=0.193%, P>0.05). The incidence of STDs showed an increasing trend followed by a decreasing trend with the increase of age. The incidence trend among males was consistent with the overall population, reaching its peak in the age group of 30 to 34 years (371.04/105), while the incidence among females showed an increasing trend with the increase of age, peaking in the age group of 60 to 64 years (442.15/105). Compared with 2013-2017, the incidence risk of STDs showed an increasing trend in 2018-2022 (RR=1.349, 95%CI: 1.217-1.495). Compared with the birth cohort of 1975-1979, the incidence risk of STDs in 1980-2005 birth cohort showed an increasing trend, with the highest incidence risk in 2000-2004 birth cohort (RR=4.178, 95%CI: 2.717-6.423).
Conclusions
The overall incidence of STDs among residents aged 15 to 64 years in Yantian District from 2008 to 2022 appeared a tendency towards a rise. Age, period and cohort all had impacts on the incidence, with a greater effect among females than males.
7.Dynamic change in Th17/regulatory T cell ratio in peripheral blood of patients with acute hepatitis B and its significance

Objective To investigate the dynamic changes in the frequencies ofTh17 (CD3+CD8-IL-17+) and regulatory T cells (Treg,CD4+CD25+CD127low) and Th17/Treg ratio in the peripheral blood in patients with acute hepatitis B (AHB),as well as their association with the outcome of AHB.Methods A total of 10 AHB patients were enrolled as observation group,and according to the outcome of AHB,these patients were further divided into acute stage group,early recovery group,and full recovery group.Another 10 healthy subjects who underwent physical examination were enrolled as control group.Flow cytometry was used to measure the frequencies of Th17 and Treg in peripheral blood,and the Th17/Treg ratio was calculated.An automatic biochemical analyzer was used to measure liver function parameters,and RT-PCR was used to measure HBV DNA.An one-way analysis of variance was used for comparison between groups,and the t-test was used for comparison between any two groups.Results Compared with the control group,the acute stage group showed significant increases in the frequencies of Th1 7 and Treg (18.22%±4.13%/6.46%±2.46% vs 0.68%±0.29%/1.62%±0.18%,P < 0.01) and the Th1 7/Treg ratio (3.37±1.73 vs 0.42±0.20,P < 0.01).Compared with the acute stage group,the early recovery group showed a significant reduction in the frequency of Th 17 (3.14%± 1.90%,P < 0.01),a significant increase in the frequency of Treg (11.73 %± 1.76%,P < 0.01),and a significant reduction in the Th17/Treg ratio (0.27±0.19,P < 0.01).Compared with the early recovery group,the full recovery group showed a slight increase in the frequency of Th17 (3.31±0.95,P =0.888),a significant reduction in the frequency of Treg (6.83%±1.85%,P < 0.01),but which was significantly higher than that in the control group (P < 0.01),and a slight increase in the Th17/Treg ratio (0.52±0.21,P < 0.05),which showed no significant difference between this group and the control group (P =0.286).In the acute stage ofAHB,HBsAg and HBeAg levels were positively correlated with Th17/Treg ratio (r =0.639,P < 0.05;r =0.633,P < 0.05).Conclusion The dynamic changes in the frequencies of Th17 and Treg and the Th1 7/Treg ratio may be associated with the outcome of AHB.
8.Exploring the Intervention Mechanism of Zishen Jianpi Huayu Tablets on Diabetic Retinopathy Based on Network Pharmacology,Molecular Docking and Experimental Validation

Objective To explore the mechanism of Zishen Jianpi Huayu Tablets(Corni Fructus,Notoginseng Radix et Rhizoma,Astragali Radix,Puerariae Lobatae Radix,Spatholobi Caulis,Rehmanniae Radix)in the treatment of diabetic retinopathy(DR)by means of network pharmacology and molecular docking technique,and verified by in vitro experiments.Methods The active components of Zishen Jianpi Huayu Tablets and their corresponding target proteins were screened using the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and the BATMAN-TCM database.Drug target proteins were converted to their corresponding gene names through the UniProt database.DR-related targets were searched using"diabetic retinopathy"as a keyword in GeneCards,DrugBank,OMIM,and TTD databases.Common targets between the disease and the drug were identified using the Venny tool.These common targets were analyzed using the String database,a protein-protein interaction(PPI)network was constructed.Topological heterogeneity analysis was performed using Cytoscape 3.9.1 to select core targets and create a PPI network diagram.These common targets were entered into the Metascape database for Gene Ontology(GO)function analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis to identify potential action pathways.Molecular docking of the main active components and core targets was performed using Auto Dock tools software,followed by further experimental validation.The CCK-8 assay was used to assess the effect of Zishen Jianpi Huayu Tablet medicated serum on the cell viability of Human Retinal Microvascular Endothelial Cells(HRmECs)under high glucose conditions,and RT-qPCR was used to measure the expression of IL-1β,AKT1,VEGFA,and TP53 mRNA in HRmECs.Results(1)The effective components and corresponding target proteins of Zishen Jianpi Huayu Tablets were screened by Traditional Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP)and BATMAN-TCM database.The disease-related targets of DR were searched by GeneCards,OMIM and TTD databases.The use of VENNY platform for drug active components target and DR disease-related target to take intersection(common target),that is,Zishen Jianpi Huayu Tablets in the treatment of DR potential target.The network of"drugs-active components-common targets"was constructed to screen out the key active components of Zishen Jianpi Huayu Tablets in the treatment of DR.Import the common target into STRING database,obtain the PPI network relationship,and screen out the core target.Metascape platform was used to analyze the GO function and KEGG pathway enrichment of the common targets.The key active components and core targets were verified by Autodock 4 software for molecular docking.(2)The drug-containing serum and blank serum of Zishen Jianpi Huayu Tablets was prepared.Human retinal microvascular endothelial cells(HRmECs)were randomly divided into 5 groups:the control group(low-sugar DMEM medium+10%blank serum),high-glucose group(high-sugar DMEM medium+10%blank serum)and Zishen Jianpi Huayu Tablets containing low-,medium-and high-dose serum(high-sugar DMEM medium+10%low-,medium-and high-dose drug containing serum)were detected after 48 hours of culture.The proliferative activity of HRmECs cells was detected by CCK-8 method,and the mRNA expressions of IL-1β,AKT1,VEGFA and TP53 in HRmECs cells were detected by RT-qPCR method.Conclusion Zishen Jianpi Huayu Tablets may act on core targets such as IL-1β,IL-6 and VEGFA,as well as key pathways such as NF-κB signaling pathway,AGE-RAGE signaling pathway and PI3K-AKT pathway through various active components such as quercetin,kaempferol and rehmannia flavonoids,so as to play a therapeutic role in DR.
9.Clinical analysis of 16 cases of connective tissue disease-associated interstitial lung disease complicated with lung cancer

Objective To investigate the clinical characteristics of 16 patients with connective tissue diseases associated interstitial lung disease(CTD-ILD)complicated with lung cancer,and to improve the cognition of the disease.Methods Clinical data of 16 patients diagnosed as CTD-ILD associated with lung cancer,who were admitted to our center,were retrospectively analyzed,including general conditions,clinical characteristics,auxiliary examinations,pathological classification of lung cancer,TNM type,treatment and clinical outcome.Results Among the 16 CTD-ILD patients with lung cancer,there were 12 males and 4 females.The mean age at diagnosis of CTD-ILD was(64.7±9.2)years,and the mean age at diagnosis of lung cancer was(66.6±8.7)years.Lung occupying space on imaging(62.5%)was the most common initial symptom in lung cancer patients,followed by cough and phlegm(12.5%)and chest pain(12.5%).Of patients with lung cancer,adenocarcinoma(8 cases,50.0%)was the most common pathological type,followed by small cell lung cancer(4 cases,25.0%).The diagnosis time of CTD-ILD was earlier than that of lung cancer in 8 cases(50.0%),with a median time of 36.0(11.3,57.0)months,followed by 7 cases(43.8%)of CTD-ILD diagnosed with lung cancer at the same time.The diagnosis time of lung cancer was earlier than that of CTD-ILD in 1 case(6.3%).The most common TNM stage for lung cancer was stage Ⅳ(9 cases,56.25%).Sixteen patients were followed up from 1 to 64 months,with a median of 8.5(1.5,14.3)months.Eleven patients(68.8%)died,including 8 patients(72.7%)died of infection and 3 patients(27.3%)died of end-stage lung cancer.Conclusion For CTD-ILD patients,close follow-up and regular imaging monitoring are necessary to help early detection of lung cancer and improve prognosis.
10.Systematic review on the effect of core stability training for the patients with non-specific low back pain

Objective:To systematically evaluate the rehabilitation efficacy of core stability training versus general exercises or manual therapy in patients with non-specific low back pain.Methods:The Medline, EMBASE, PubMed, Cochrane Central Register of Controlled Trials (CENTRAL) , PsycINFO, China Biology Medicine (CBM) , CNKI, VIP and Wanfang database were searched by computer, and the retrieval time was from the establishment of the database to 20th October, 2018. All randomized controlled trials (RCT) and clinical control studies of the rehabilitation effects of core stability training on patients with non-specific low back pain were collected. According to the inclusion and exclusion criteria, 2 researchers independently searched the literature, screened the literature and evaluated the quality of the included literature. And the data were extracted for Meta-analysis using Revman 5.3 software.Results:A total of 14 articles were included, all of which were RCTs. The results of Meta-analysis showed that the comparison of pain improvement between core stability training group and general exercise training group was ( Z=4.67, P<0.001) [ OR=1.01, 95% CI (-1.43, -0.59) ], and the evidence showed statistical significance ( P<0.05) . The comparison of Oswestry Disability Index scores of patients after core stability training and general exercise training was ( Z=5.03, P<0.01) [ OR=-7.18, 95% CI (-9.98, -4.38) ], and the evidence showed statistical significance ( P<0.05) . The comparison results of other indicators were not statistically significant ( P>0.05) . Conclusions:The core stability training group and manual therapy group can effectively reduce pain in patients with non-specific low back pain, and core stability training can promote the relief of dysfunction in patients with non-specific low back pain, and core stability training should be encouraged as part of musculoskeletal rehabilitation for patients with non-specific low back pain.
