1.Expression of Endothelial Nitric Oxide Synthase and Vascular Endothelial Growth Factor in Human Malignant Melanoma

Objective To investigate the relationship between the expression of endothelial nitric oxide synthase (eNOS), vascular endothelial growth factor (VEGF) and the angiogenesis in human malignant melanoma (MM). Methods Tissue sections from 31 MM patients were examined. The protein expression of eNOS and VEGF was detected using immunohistochemistry technique and morphological quantitative analysis. Microvessel density (MVD) was also counted by immunostaining with anti-FⅧRAg antibody. Results (1)The positive staining of eNOS and VEGF was detected in 71.4% (24 cases) and 83.9% (26 cases) of MM respectively. As negative control there was no staining in the nevus pigmentosus tissue sections. (2)There was a positive correlation between the expression of eNOS and VEGF in MM. (3)The positive correlation was also found between the MVD and the expression of eNOS and VEGF in MM. The number of MVD was higher in MM than that in nevus pigmentosus tissue. (4)The lymphatic metastasis was not correlated with the expression of eNOS and VEGF. Conclusion The number of the MVD increases along with the high expression of eNOS and VEGF in MM.
2.Expression of MDM2 Protein and MDM 2mRNA in Condyloma Acuminatum

Objective To investigate the role of MDM2in the cell proliferation and carcinogenesis in condyloma acuminatum(CA).Methods The technique of immunohistochemistry and in situ hybridization were used to detect the expression of MDM2protein and mRNA in skin lesions of CA and normal skin of genital area.Polymerase-chain reaction(PCR)was used to type HPV DNA.Results Out of32CA specimens,the expression of MDM2protein was found in18(56.25%),the expression of MDM2mRNA was found in22(68.75%),while co-expression of MDM2protein and MDM2mRNA was found in14.According to PCR results,HPV6/11subtype was shown in28out of32CA specimens(87.5%),and HPV16/18subtype was shown in4out of32(12.5%).In eighteen out of positive specimens expressing MDM2protein,HPV6/11subtype was shown in15and HPV16/18subtype3.In twenty-two positive specimens expressing MDM2mRNA,there were HPV6/11subtype in18,and4were HPV16/18subtype.No expression of MDM2protein and MDM2mRNA was observed in normal skin.Conclusion The overexpression of MDM2may be involved in malignant proliferation and carcinomatous change of condyloma acuminatum.
3.Expression of HFGL2/ Fibroleukin in Peripheral Blood Mononuclear Cell in Patients with Systemic Lupus Erythematosus and Its Clinical Significance

Objective To investigate the relationship between procoagulation of HFGL 2 and abnormality of coagulation in patients with systemic lupus erythematosus (SLE) by detecting the expression of HFGL2/fibroleukin in peripheral blood mononuclear cell(PBMC) of SLE patients. Methods A polyclonal antibody against HFGL2 was applied to detecting the expression of HFGL2 protein in 32 SLE patients and 15 healthy volunteers by immunohistochemistry. Semi-quantitative measurement of HFGL2 expression in the blood specimen was done with high multiple image analytical system(HMIAS). Results The expression of HFGL2 in PBMC from 23 active SLE patients was significantly higher than that in the controls, which showed a positive correlation with the disease activity. Conclusion The expression of HFGL2 in PBMC is probably correlated with the pathogenesis and disease activity of SLE.
4.Expression of Candida albicans secreted aspartyl proteinase in acute vaginal candidiasis.

In order to analyze the in vivo expression of Candida albicans secreted aspartyl proteinases (SAP) in human vaginal infection, the vaginal secretion from 29 human subjects was collected by vaginal swab, and the expression of SAP1-SAP6 was detected by reverse-transcriptase polymerase chain reaction using specific primer sets. It was found that Sap2 and Sap5 were the most common genes expressed during infection; Sap3 and Sap4 were detected in all subjects and all 6 SAP genes were simultaneously expressed in some patients with vaginal candidiasis. It was suggested that the SAP family is expressed by Candida albicans during infection in human and that Candida albicans infection is associated with the differential expression of individual SAP genes which may be involved in the pathogenesis of vaginal candidiasis.
5.Effect of cold atmospheric plasma on skin ulcer healing

Objective To investigate the effect of cold atmospheric plasma (CAP) on the healing of skin ulcers using Balb/c mice. Methods Wounds with a diameter 6 mm were created on each side of the backs of BalB/c mice ( n = 150) using a punch bioptome. The mice were assigned randomly into a control group ( wounds healed naturally), a laser group (wounds treated with a He-Ne laser for 10 min daily) and a CAP group (wounds treated with CAP for 10 min daily). Wound healing was evaluated on postoperative days (PODs) 4, 7, 10 and 14 in terms of percent wound closure. Ten mice per group were sacrificed on each of the evaluation days. Both wounds were removed and a histological examination was conducted. A scoring system was used to evaluate the wounds. The expression of vascular endothelial growth factors (VEGFs) in the wounded tissue was detected by using immunohistochemical methods on POD 7. The results were quantified using an HPIAS-1000 system. Results Compared with the control group, the average percentage of wound healing was significantly greater in the CAP group on PODs 7 and 10. The average scores on the histological examination were significantly higher in the CAP group on PODs 7, 10 and 14. Compared with the other two groups, the expression of VEGF was up-regulated significantly in the CAP group.Conclusions CAP can positively affect the wound healing process. This might be related to the up-regulation of VEGF in the wounded tissues.
6.Expression of CD28/B7 in the Lesional Skin and Peripheral Lymphocytes in Psoriasis

Objective To investigate the role of costimulatory molecules CD28/B7 in the pathogenesis of psoriasis. Methods The expression of CD28, CD80 and CD86 was detected in the lesional skin of 22 cases by immunohistochemical technique (ABC), and in the peripheral blood lymphocytes of 17 cases by flow cytometry respectively. As control, normal skin and lymphocytes from peripheral blood of healthy volunteers were also tested. Results The immunohistochemistry study showed that the expression of CD28, CD80 and CD86 in the psoriatic lesions was significantly higher than that in the normal controls (P .05). Conclusion CD28, CD80 and CD86 might play a certain role in the pathogenesis in psoriasis.

Objective To analyze the expression of secreted aspartyl proteinases (Sap) in human vaginal infection in vivo. Methods Vaginal swabs were collected from 9 healthy volunteers and 20 patients with vaginal candidiasis, and the expression of Sap1-6 was evaluated by reverse-transcriptase polymerase chain reaction using specific primer sets. Results The Sap2 and Sap5 were the most common genes expressed in the patients with vaginal candidiasis. The expression Sap3 and Sap4 was detected in all subjects. All six Sap genes were simultaneously expressed in some patients with vaginal candidiasis. Conclusion The data shows that the Sap genes may be involved in the pathogenesis of vaginal candidiasis.

Objectives To study the effects of pU- vascular endothelial growth factor (VEGF)- short interfering RNA (siRNA) on the formation and apoptosis of malignant melanoma in models of nude mice and its mechanism. Objective To study the effect of siRNA targeting survivin gene on the apoptosis of malignant melanoma cell line A375. Methods The eucaryotic expression vector of pU-survivin-siRNA was constructed and transfected into the A375 cells by electroporation. The protein expression of survivin was examined by Western blotting, and cell apoptosis by flow cytometry.
