Chinese Journal of Practical Nursing 2008;24(18):11-12
Objective We discussed the anxiety alleviation effect of nursing interview and health education on patients after radical mastectomy. Methods We divided 60 patients underwent radical mastectomy into the observation group and the control group with 30 cases in each group. The observation group was given nursing interview and health education based upon perioperative nursing. The control group received routine perioperative nursing. Self-rating anxiety scale (SAS) was adopted to assess the psychological status on the second day after admission, one day before operation and the 4th day after operation. Results Evident anxiety existed in the two groups compared with normal control. The SAS score one day before operation decreased compared with that of two days after admission in the observation group. But the control group showed reverse results compared with the observation group (P < 0.05). The SAS score 4 days after operation dropped evidently in the two groups. But difference still existed in the two groups (P<0.05). Conclusions Nursing interview and health education could decrease patients' anxiety after radical mastectomy and made them to receive the operation in a good psychological status.

3.Investigation in the incidence of sharp- appliance wound of trainee nurses and its preventive countermeasures
Chinese Journal of Practical Nursing 2009;25(15):66-67
Objective To acknowledge the incidence of sharp- appliance wound of trainee nurses and explore the relative countermeasures. Methods A retrospective investigation was carried out in 140 trainee nurses of grade 2003 about the incidence of sharp- appliance wounds, then the corresponding pre-ventive countermeasures were formulated and applied in the clinical practice of trainee nurses of grade 2004 and the incidence of sharp- appliance wound was investigated and compared with the results of those grade 2003.X2 test was adopted. Results The incidence rate of sharp- appliance wound in trainee nurses of grade 2004 was obviously lower than that of grade 2003. Conclusions Due to the weak safety con-sciousness, unfamiliar with operation and lack of clinical experience, the sharp- appliance wound will easily occur among trainee nurses. If the relative professional preventive knowledge is strengthened, the opera-tional procedures are observed strictly and the sense of responsibility of tutor teachers is facilitated, then the incidence of sharp- appliance wound of trainee nurses will drop.

Objective To investigate the clinical effect of high dose of calcitriol on end-stage renal disease (ESRD) in patients with secondary hyperparathyroidism (SHPT). Methods The clinical data of patients with ESRDcomplicated with SHPT were retrospectively analyzed. According to the treatment method were divided into control group and observation group, the control group were given conventional dose of calcitriol treatment, the observation group were treated with high dose of calcitriol. The differences of calcium and phosphorus levels, parathyroid hormone (PTH) level and quality of life were observed between the two groups before and after treatment. Results The serum phosphorus, calcium and calcium-phosphorus product in two groups of patients before treatment had no significant difference. After treatment, the above indicators decreased in two groups of patients compared with those before treatment, and the observation group was significantly lower (P<0.05). Before treatment, there was no difference in the levels of PTH between the two groups. After treatment, the level of PTH in the observation group was lower than that in the control group, and the difference was statistically significant (P<0.05). There was no significant difference in quality of life between the two groups before treatment. After treatment, the quality of life in the two groups were higher than before treatment, and the observation group was significantly higher (P<0.05). Conclusion High dose of calcitriol has good therapeutic effect for ESRD patients with SHPT and can significantly improve the patient's calcium and phosphorus and PTH levels, improve the quality of life of patients.

Objective To investigate the role of LncRNA ROR in Ad36-induced browning of human adipose-derived stem cells(hADSC). Methods After hADSC was induced by cocktail and Ad36 for 2,4,6,and 8 days,Oil red O staining was performed for observing the adipogenic status. The mRNA expressions of LncRNA ROR, uncoupling protein 1(UCP1),and PRDM16 were detected by real-time PCR and the protein expressions of UCP1 and PRDM16 were detected by Western-blot. After LncRNA ROR was knocked down by siRNA, UCP1 and PRDM16 mRNA and protein expression levels in the process of Ad36-induced adipocyte differentiation were detected by real-time PCR and Western-blot. Results Oil red O staining showed that fat droplets in the cocktail-induced group were larger than those in the Ad36-induced group. Compared with the cocktail group, the mRNA expressions of LncRNA ROR, UCP1 and PRDM16, and the protein expressions of UCP1 and PRDM16 in Ad36 group were significantly increased(P<0.05). The mRNA and protein expressions of UCP1 and PRDM16 in LncRNA ROR knockdown group were significantly lower than those in control group(P<0.05). Conclusion In the process of Ad36-induced hADSC differentiation, the up-regulation of LncRNA ROR may stimulate UCP1 and PRDM16 expression, and thus promote the browning of hADSC.

Objective To investigate the expressions of RNA-binding protein human antigen R(HuR), fatty acid binding protein type 4(FABP4),fatty acid synthetase(FASN),and lipoprotein lipase(LPL)during the differentiation of human adipocytes, and to explore their possible roles. Methods Human adipose-derived mesenchymal stem cells were induced by adipogenic differentiation, and the adipogenesis of cells was observed by oil red O staining. The expressions of HuR,FABP4,FASN,and LPL mRNA and protein were detected by real-time PCR and Western blotting. After HuR was silenced by siRNA, the change of adipogenesis for human adipose-derived mesenchymal stem cells was observed and the expressions of adipogenic genes were detected. Results The expressions of HuR,FABP4,FASN,and LPL mRNA and protein were significantly increased after human adipose-derived mesenchymal stem cells were induced to differentiate into adipocytes(all P<0.01). After HuR expression was down-regulated by siRNA,the adipogenic level of human adipose-derived mesenchymal stem cells was reduced, with decreased protein levels of FABP4,FASN,and LPL(all P<0.05),which were without changes for their mRNA levels. Conclusion HuR promotes the differentiation of human adipocytes mainly via regulating the changes of FABP4,FASN,and LPL protein levels.
