2.Family factors influencing dietary behavior of primary and secondary sch ool students in Guangzhou city

Objective The purpose of the study was to explore influence of family factors on dietary behavior of primary and secondary studen t s in Guangzhou city, in order to provide suggestions for intervention of dietary behavi or. Methods The study population included 1 539 student-parent s pairs selected from Guangzhou city by using the method of stratified multistage cluster random sampling, and data were collected through questionnaires. Results The ratio of parents who often reminded and forced the ir children to eat some fo od they thought of as nutrition were 56.0% and 7.7%, and 39.7%, 29.2% of ch ildren complied with their parents, respectively. At dinner, 14.8% and 27.5% of parents often and sometimes criticized their children, and the ratio of childre n couldn't eat anything and only eat a little food were 5.9%,19.3%, respectively . Forty-two point three of primary and secondary school students watched televi sion wh en they had a dinner, which made 39.4% of students not eat seriously. C onclusions Dietary behavior of primary and secondary school students is influenced strongly by their parents and family environment, it is necessary for parents and themselves to grasp enough nutritional knowledge.
3.Therapeutic effects of leaf extract from Cyclocarya paliurus on rats with type 2 diabetes mellitus

AIM To speculate the hypoglycemic mechanism for rats with type 2 diabetes by exploring the therapeutic effects of leaf aqueous extract from Cyclocarya paliurus on liver insulin receptor (InsR) and insulin receptor substrate 2 (IRS-2).METHODS The diabetic rat model was established through intraperitoneal injection of streptozotocin and fed with high-fat diet.The moleled rats were equally assigned into the control group and leaf aqueous extract from Cyclocarya paliurus group (extract group).After the test extract was orally administrated for four weeks,body weight,urine output,food intake,water intake and fasting blood-glucose (FBG) were measured,and the levels of serum insulin,InsR and IRS-2 mRNA in liver tissue were investigated in rats.RESULTS Compared with the control group,the extract group showed a reduction in urine output,food intake,water intake,FBG and insulin levels.Meanwhile,the rats' body weights in extract group were presented a trend to increase.The gene expressions of InsR and IRS-2 in liver tissue were up-regulated.Moreover,the insulin sensitivity was improved.CONCLUSION The leaf aqueous extract from Cyclocarya paliurus can reduce FBS,improve insulin sensitivity,which may be associated with the increase of InsR and IRS-2 gene expression in liver tissue.
4.Shujin Jiannao Prescription alleviates inflammation in the cerebral cortex of rats with hypoxic-ischemic cerebral palsy

BACKGROUND:Shujin Jiannao Prescription is an empirical formula for the treatment of cerebral palsy in Dongzhimen Hospital,Beijing University of Chinese Medicine,with clear clinical efficacy,but the specific mechanism needs to be elucidated. OBJECTIVE:To explore the possible mechanism of Shujin Jiannao Prescription in treating cerebral palsy. METHODS:Sixty-four 7-day-old Sprague-Dawley rats were randomly divided into a normal group(n=12)and a model group(n=52).An animal model was established by the Rice-Vannucci method.After successful modeling,52 model rats were randomly divided into control model group(n=12),minocycline group,and the low-,medium-,and high-dose groups of the Shujin Jiannao Prescription(n=10 per group).Rats in the minocycline group were given 40 mg/kg·d minocycline by gavage;rats in the low-,medium,and high-dose groups were given 4,8,and 16 g/kg·d Shujin Jiannao Prescription granules by gavage,respectively;and rats in the normal group and control model group were given an equal dose of normal saline by gavage.Medication in each group was given once a day for 1 week.The rats in each group were evaluated behaviorally using suspension test,abnormal involuntary movement score,and Bederson score.The pathological changes of the cerebral cortex were observed by hematoxylin-eosin staining.The levels of tumor necrosis factor α,interleukin 1β,and interleukin 10 in the cerebral cortex were determined using ELISA.The positive expressions of Janus kinase 2(JAK2),phosphorylated Janus kinase 2(p-JAK2),phosphorylated signal transducer and activator of transcription 3(p-STAT3)in the cerebral cortex were detected using immunohistochemistry.The protein expression levels of JAK2,p-JAK2,and p-STAT3 were detected using western blot. RESULTS AND CONCLUSION:Compared with the normal group,the suspension test score and involuntary movement score were decreased in the control model group(P<0.01 or P<0.05).The pathological results showed structural disruption of nerve cells,formation of large numbers of vacuoles,cell swelling,and increased intercellular space in the control model group.In addition,the expressions of tumor necrosis factor α and interleukin 1β in the cerebral cortex were significantly increased(P<0.01),the expression of interleukin 10 was decreased(P<0.05),and the protein expressions of JAK2,p-JAK2,and p-STAT3 in the cerebral cortex were significantly increased(P<0.01)in the control model group compared with the normal group.Compared with the model group,minocycline and Shujin Jiannao Prescription at each dose could improve the behavioral indexes of rats(P<0.01 or P<0.05)and ischemic-hypoxic pathological changes were attenuated,with only a small amount of necrotic nerve cells and a few vacuoles,and reduced intercellular space.Moreover,the expressions of tumor necrosis factor α and interleukin 1β in the cerebral cortex were decreased in each drug group compared with the control model group(P<0.05),while the protein expressions of JAK2,p-JAK2,and p-STAT3 in the cerebral cortex were significantly decreased(P<0.01).The most obvious improvement was observed in the high-dose Shujin Jiannao Prescription group.To conclude,Shujin Jiannao Prescription can inhibit inflammation in the cerebral cortex of rats with hypoxic-ischemic brain injury.The mechanism may be related to the regulation of the JAK2/STAT3 signaling pathway.
5.Effects of Shujin Jiannao Prescription on cell apoptosis in rats with hypoxic-ischemic brain injury

BACKGROUND:Perinatal hypoxic-ischemic brain injury is one of the most common causes of cerebral palsy.Shujin Jiannao Prescription is an experienced formula for treating cerebral palsy and improving blood supply to the brain developed by the Dongzhimen Hospital,Beijing University of Chinese Medicine. OBJECTIVE:To explore the possible mechanism of Shujin Jiannao Prescription in treating hypoxic-ischemic cerebral palsy. METHODS:Sixty-four 7-day-old Sprague-Dawley rats were randomly divided into six groups.There were 12 rats in each of the control and model groups as well as 10 animals in each of the minocycline group,and the low-,medium-,and high-dose groups of Shujin Jiannao Prescription.The neonatal rat ischemic-hypoxic cerebral palsy model was established in all groups except for the control group.After successful modeling,rats in each drug group were respectively gavaged with minocycline and Shujin Jiannao Prescription at a dose of 4,8,and 16 g/kg per day for 1 week.Body mass of rats was measured and behavioral changes were detected before and after drug administration.Hematoxylin-eosin staining was used to observe the histomorphology of hippocampal CA1 region of rat brain tissue,and immunohistochemistry and western blot were used to detect the expression levels of Bcl-2,Bax,and Caspase-3 in the brain tissue of rats. RESULTS AND CONCLUSION:Compared with the model group,medium-and high-dose Shujin Jiannao Prescription significantly increased the body mass of rats(P<0.05).Compared with the model group,minocycline effectively prolonged the suspension time of ischemic-hypoxic cerebral palsy rats(P<0.05),while medium-and high-dose Shujin Jiannao Prescription significantly prolonged the suspension time,shortened the inclined plane test time,and increased the Longa score of rats(P<0.05).The pathological results showed that after drug intervention,only a small number of neuronal cells in the brain tissue of rats were necrotic,the cells were more neatly arranged,the cell structure was more complete,and only part of the cell nuclei became smaller.Compared with the model group,minocycline and medium-and high-dose Shujin Jiannao Prescription reduced the expression of Bax Caspase-3(P<0.05),medium-and high-dose Shujin Jiannao Prescription increased the expression of Bcl-2(P<0.05),and Bcl-2/Bax protein expression was increased in minocycline and three Shujin Jiannao Prescription groups(P<0.05).In addition,the protein expression was increased in a dose-dependent manner after intervention with Shujin Jiannao Prescription,and there was no significant difference between the minocycline and three Shujin Jiannao Prescription groups(P>0.05).To conclude,the mechanism by which Shujin Jiannao Prescription treats ischemic-hypoxic cerebral palsy in rats may be to enhance the expression of anti-apoptotic protein Bcl-2,inhibit the expression of pro-apoptotic protein Bax,and reduce the expression of Caspase-3,ultimately inhibiting the apoptosis of hippocampal neuronal cells in rats with cerebral palsy.Within a certain range,the higher dose of Shujin Jiannao Prescription indicates the better therapeutic effect,and the high-dose Shujin Jiannao Prescription is as effective as minocycline.
6.Establishment of Fingerprint and Multivariate Statistical Analysis of Compatible Herb Pairs with Polygonum multiflorum as the Core

OBJECTIVE:To establish UPLC fin gerprint of 32 compatible herb pairs with Polygonum multiflorum as the core , and to conduct multivariate statistical analysis. METHODS :UPLC method was adopted. Using P. multiflorum and single decoction pieces of compatible herb as reference ,UPLC fingerprints of 32 compatible herb pairs of P. multiflorum were drawn. Common peaks were confirmed by relative retention time and UV absorption spectrum. Non-supervised PCA and supervised OPLS-DA were conducted by using SPSS 19.0 software and SIMCA 13.0 software. RESULTS :There were totally 12 common peaks in UPLC fingerprints of 32 compatible herb pairs of P. multiflorum . The results of non-supervised PCA showed that the accumulative variance contribution rate of primary 6 principal components was 84.633%. The results of cluster analysis of PCA comprehensive score showed that single decoction piece of P. multiflorum ,compatible herb pairs of P. multiflorum with Lycium barbarum ,Rehmannia glutinosa,Paeonia lactiflora ,Codonopsis pilosula ,Eclipta prostrate ,Angelica sinensis ,Glycyrrhiza uralensis ,Astragalus membranaceus and Ophiopogon japonicus were clustered into one category ;others were clustered into one category. Results of supervised OPLS-DA analysis showed that eigen values of 4 principal components were 2.32,2.61,1.58 and 0.90,respectively. There were differences in the contents of 12 common components in the compatibility of P. multiflorum with tonic medicine and non-tonic medicine. The changes of the content of the components after compatibility with tonic medicine were similar. Common peak 7,4,6,3 were main reasons for the differences (variable importance projection value were all higher than 1). CONCLUSIONS:Established fingerprint is simple in operation ,and can be combined with multivariate statistical analysis to evaluate the content changes of common components of 32 compatible herb pairs with P. multiflorum as the core.
7.Effect of miR-203a-3p overexpression on pulmonary fibrosis after LPS-induced acute lung injury in rats and its mechanism

Objective:
To investigate the effects of miR-203 a-3 p overexpression on pulmonary fibrosis after acute lung injury induced by lipopolysaccharide(LPS) in rats and its possible mechanism.
Methods:
The rat model of pulmonary fibrosis after acute lung injury was established by multiple low-dose injections of LPS. The successfully modeled rats were randomly divided into model group, agomir-negative control group(agomir-NC group) and miR-203 a-3 p agomir group(agomir group), and another control group was set, with 15 rats in each group. One day before modeling and one week after modeling, miR-203 a-3 p agomir was injected by caudal vein for intervention. Two weeks after modeling, the wet/dry specific gravity(W/D) and hydroxyprolinic acid(Hyp) content of lung tissue were determined. The pathological changes and the degree of pulmonary fibrosis of lung tissue were observed by HE staining and Masson staining. The levels of interleukin-1β(IL-1β), interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in broncholveolr lavage fluid(BALF) were determined by ELISA. The expression levels of miR-203 a-3 p and follicle-statin like 1(Fstl1) mRNA in the lung tissues of rats were detected by qRT-PCR. The protein expression level of Fstl1 in lung tissue of rats was detected by Western blot. The targeting relationship between miR-203 a-3 p and Fstl1 was detected by dual luciferase reporting system.
Results:
Compared with control group, the degree of lung injury and pulmonary fibrosis in model group were relatively serious, the W/D value and Hyp content of lung tissue increased(P<0.05), the levels of IL-1β, IL-6 and TNF-α in BALF increased(P<0.05), the expression of miR-203 a-3 p in lung tissue decreased(P<0.05), while the mRNA and protein expression levels of Fstl1 increased(P<0.05). Compared with model group, the degree of lung injury and pulmonary fibrosis were improved in agomir group, the W/D value and Hyp content of lung tissue decreased(P<0.05), the levels of IL-1β, IL-6 and TNF-α in BALF decreased(P<0.05), the expression of miR-203 a-3 p in lung tissue increased(P<0.05), while the mRNA and protein expression levels of Fstl1 decreased(P<0.05). The luciferase reporter gene assay confirmed that Fstl1 was the target gene of miR-203 A-3 p.
Conclusion
Overexpression of miR-203 a-3 p improves pulmonary fibrosis after LPS-induced acute lung injury, and the mechanism may be related to the targeted down-regulation of Fstl1 expression.
8.Transcriptome analysis of human neuroblastoma cells infected with Zika virus

Objective To analyze the transcriptome data of Zika virus(ZIKV)-infected human neuroblastoma cells SH-SY5Y by bioinformatics method,and to identify the potential genes involved in the pathogenic mechanism of ZIKV.Methods SH-SY5Y cells were infected with ZIKV.The total RNA was extracted and the differentially expressed genes(DEGs)were screened by transcriptome sequencing.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analyses were performed to predict the biological processes,molecular functions,and signaling pathways mainly involved in the DEGs,and the results were verified by quantitative polymerase chain reaction(qPCR).Results A total of 259 DEGs were identified,including 172 up-regulated genes and 87 down-regulated genes.GO functional enrichment analysis showed that the DEGs were mainly related to extracellular matrix,response to stimulus,antimicrobial humoral response,and developmental process.KEGG pathway enrichment analysis revealed that the DEGs were predominantly associated to inflammatory reaction and immune response.The qPCR validation results of DEGs were basically consistent with the transcriptome sequencing results.Conclusion The expression of genes involved in extracellular matrix,response to stimulus,and regulation of inflammatory reaction is significantly altered in SH-SY5Y cells after ZIKV infection,suggesting that ZIKV may cause neurological lesions by remodeling the extracellular matrix and regulating inflammatory reaction.

Ameloblastoma is a benign tumor characterized by locally invasive phenotypes,leading to facial bone destruction and a high recurrence rate.However,the mechanisms governing tumor initiation and recurrence are poorly understood.Here,we uncovered cellular landscapes and mechanisms that underlie tumor recurrence in ameloblastoma at single-cell resolution.Our results revealed that ameloblastoma exhibits five tumor subpopulations varying with respect to immune response(IR),bone remodeling(BR),tooth development(TD),epithelial development(ED),and cell cycle(CC)signatures.Of note,we found that CC ameloblastoma cells were endowed with stemness and contributed to tumor recurrence,which was dominated by the EZH2-mediated program.Targeting EZH2 effectively eliminated CC ameloblastoma cells and inhibited tumor growth in ameloblastoma patient-derived organoids.These data described the tumor subpopulation and clarified the identity,function,and regulatory mechanism of CC ameloblastoma cells,providing a potential therapeutic target for ameloblastoma.

Postoperative pain seriously affects the recovery process of patients, resulting in prolonged hospital stay and increased care costs. Appropriate application of patient-controlled analgesia devices can effectively relieve perioperative acute pain. In 1994 patient-controlled analgesia began to be used in Peking Union Medical College Hospital, and the Acute Pain Service Working Group was established in 2004. With the cooperation of anesthesiologists and specialist nurses, the group jointly has implemented the whole process and standardized management based on patient-controlled analgesia, and constantly improved and innovated working methods, laying a solid foundation for the development of postoperative pain management. This paper systematically reviews and summarizes the work from the aspects of clinical focus, nursing management experience, promotion and dissemination of pain treatment concepts, and development of acute pain service model under the new situation, with the hope of providing valuable reference for comprehensively strengthening pain management in the process of diagnosis and treatment, and enhancing patients' satisfaction with perioperative analgesia services.
