Quantitative calibration strategy as an essential issue in laser ablation ICP-MS plays an important role for the guarantee of analytical accuracy. In this study, uncertainties of reference values in current available glass certified reference materials (NIST, MPI-DING and USGS) as well as the matrix effect among them were systematically evaluated. The results illustrated that NIST610 was better than other reference materials from aspect of reference value uncertainty. The matrix effect among NIST, MPI-DING and USGS reference materials was insignificant under our experimental conditions.A quantification strategy based on two reference materials (NIST610 and StHs6/80-G) and normalization to 100% (w/w) was proposed to avoid the insufficiency of single reference material strategy, which suffered the very low content and large uncertainty for some elements. The comparision of ML3B-G results obtained from three quantification strategies (single reference material NIST610, single reference material StHs6/80-G and two refterence materials) illustrated that the proposed strategy improved the analytical accuracy.Three reference materials (BCR-2G, CGSG-2 and KL-2G) were quantified with the proposed strategy, and almost all data matched well with reference value, meanwhile the data reported here could supplement the reference value database.

Stroke is the disease which caused by many risk factors. The relationship between insulin resistance(IR) and young adult stroke is one of the focuses now in the neurological field.IR refered to the series of pathological and physiological changes,it happens when the response of the target tissue organ to insulin is depressing or losing. There are many ways of detection of IR,the insulin sensitivity index(ISI) is one of the most satisfied indexes to reflect the sensitivity to insulin.IR could cause a series of pathological and physiological changes, which caused hyperinsulinemia,hyperglycemia,hyperpiesia,blood-fat derangement, fibrinolytic system anomaly, blood vessel endothelium change, artherosclerosis,et al,maybe in ways of many metabolic risk factors.Atherosclerosis is the main etiopathogenisis factor in young adult stroke,as a conclusion,IR is also existed in young adult stroke. But,does IR exist in all types of young adult stroke? What is the mechanism? Is the level of IR related with the pathogenetic condition and prognosis? These questions are still not clear at present, and the study on the relationship between IR and young adult stroke is important to prevent young adult stroke.

Objective To investigate the effects of antisense p21 oligodeoxynucleotide (p21 ASODN) on the expression of p21 protein and extracellular matrix in cultured human glomerular mesangial cells (HGMC) under high glucose medium. Methods HGMCs were transfected with 50 nmol/L or 100 nmol/L p21 ASODN or scrambled control oligodeoxynucleotide (SCODN) using lipofectamine 2000. After incubation under normal (5.5 mmol/L) or high (30 mmol/L) glucose media and different times, HGMCs lysates were collected and the expression of p21, fibronectin and laminin was examined by Western blot. The secretion of fibronectin and laminin by HGMCs in supernatants of culture media was also detected with EOSA. Results High glucose media significantly stimulated the expression of p21, increased the syntheses and secretion of fibronectin and laminin in cultured HGMCs in a time-dependent manner. Transfection of HGMCs with p21 ASODN not only decreased p21 protein level caused by high glucose media, but also attenuated the expression and secretion of fibronectin and laminin in supematants of HGMCs lysates and culture media. Meanwhile, SCODN had no significant effects on the expression of p21, fibronectin and laminin. Conclusions High glucose promotes the expression of p21 , fibronectin and laminin in cultured HGMCs. Transfection of HGMCs with p21 ASODN can decrease p21 protein level caused by high glucose media, and attenuate the expression of fibronectin and laminin in supematants of HGMCs lysates and culture media. Modulation of p21 expression in HGMCs may work as an effective way to mitigate the progression of diabetic nephropathy.

PurposeTo evaluate the effect of right ventricular septum (RVS) and right ventricular apex (RVA) pacing on the left atrial systolic function in patients with atrioventricular block by using real-time three-dimensional echocardiography (RT-3DE).Materials and Methods Fifty-one patients with atrioventricular block who were candidates for implanted atrioventricular sequential pacemaker were randomly divided into RVS group (n=31) and RVA group (n=20). The minimum left atrium volume (LAVmin), maximum left atrium volume (LAVmax), and left atrium volume before contraction (LAVprep) were measured by RT-3DE at pre-operation, the 1st month, 3rd month, 6th month, and 12th month after pacemaker implantation. Accordingly, the left atrial total ejection fraction (LATEF) and the left atrial active ejection fraction (LAAEF) were calculated. Results The LAVmin at the 3rd month, 6th month, 12th month after implantation were significantly lower than that at the 1st month and pre-operation in both groups (RVA group:t=2.97 and 2.74,P<0.05; RVS group:t=3.24 and 2.86,P<0.05). LAVprep at the 6th month, 12th month in RVA group, and LAVprep at the 3rd month, 6th month, 12th month in RVS group reduced when compared with that of pre-operation (RVA group:t=3.20,P<0.05; RVS group:t=2.71,P<0.05). LATEF and LAAEF in both groups at the 3rd month, 6th month, 12th month increased when compared with that of pre-operation (RVA group:t=2.87 and 9.68,P<0.05; RVS group:t=3.56 and 8.22,P<0.05). The LATEF and LAAEF in RVS group at the 6th month and 12th month after implantation were significantly larger than that in RVA group at the same time (t=2.90, 5.22, 3.03 and 3.55, P<0.05).Conclusion Atrioventricular sequential pacing is helpful to recovering the left atrial systolic function in patients with atrioventricular block. Compared with RVA pacing, RVS pacing is able to increase LATEF and LAAEF more greatly.

ObjectiveTo investigate the effect of immunoglobulin G (IgG) extravasated from blood circulation on the expression of toll-like receptor 4 (TLR4) induced by peripheral lipopolysaccharide (LPS) in rat brain. Methods The rats were divided into four groups in random, 5 rats in each. Group one received LPS 100μg/kg by intraperitoneal administration, normal saline was given by intravenous injection 6 hours later; group two was injected with adrenalin (AD) 15μg/kg intravenously; group three was treated with LPS intraperitoneally, AD was injected 6 hours later; group four was injected normal saline intravenously as control. For all groups, the animals were sacrificed 30 min after the last injection, and the brains were taken for investigation of the TLR4 expressions by immunofluorescence staining and RT-PCR. Result Immunofluorescence staining showed that IgG immunoreactive product was patch-like, distributed in the brain parenchyma in all the animals that received AD. In the LPS+normal saline group, IgG was found merely around the blood vessels. Meanwhile, in LPS+AD animals, TLR4 immunoreactive product coexisted with microglia marker Iba-1 within the IgG extravasated area. The double-labeled cells dispersed in the brain parenchyma and near to the cerebral vessels. In the LPS+saline group, TLR4 positive cells were endothelial-like. RT-PCR results indicated that the expression level of TLR4 in the LPS+AD group were significantly higher than that in the LPS+saline group or AD group or the saline control (P<0.01). Conclusion Extravasated circulating IgG may enhance the TLR4 expression in the rat brain induced by peripheral LPS.
