BACKGROUND: The clinical application of neural stem cells (NSCs) in combination with gene therapy could be used to treat a variety of nervous system hereditary and acquired diseases. However, nervous system diseases are varying. Nerve tissue internal environment of distinctive diseases is also different. All these factors affect the therapeutic effect of NSCs. Moreover, interaction and regulation of a variety of exogenous genes in NSC gene therapy would greatly promote continued regeneration of nerve tissue.OBJECTIVE: To review application of NSCs in the nervous system diseases.METHODS: A computer-based online search of Medline database (2000-01/2009-08) and Tongfang database (2000-01/2009-08) was performed for related articles with key words "neural stem cells, gene, nervous system diseases". RESULTS AND CONCLUSION: A total of 88 English articles were collected. By reading the title and summary, 20 unrelated articles and 28 repetitive articles were excluded. Finally, 40 were reviewed. NSC as a new-type treatment has been used for somatic cell or transgenic vector, and has achieved a certain effect for cerebrovascular disease, brain damage disease, spinal cord injuries, neurodegenerative diseases, brain tumors, and genetic metabolic diseases. However, many key issues such as NSC proliferation, differentiation, migration and control mechanism have not been resolved. In addition, the microenvironment in nerve tissues with different diseases also influences NSC therapy.

Objective To explore the role of ripply1 in zebrafish dorsal-ventral development .Methods Using ze-brafish whole-mount in situ hybridization to examine the ripply1 expression pattern in early embryo development .To analyse the expression pattern changes of dorsal-ventral marker genes at shield stage and the morphological changes at 24 hpf (hours post-fertilization) after overexpression of ripply1 by injecting synthetic mRNA at 1-cell stage.Using Tol2 transposon technology to obtain a ripply1 promoter driven GFP transgenic fish and to identify promoter region that recapitulates endoge -nous ripply1 expression pattern .Results The in situ hybridization results revealed that ripply1 specifically expresses in the future dorsal region at shield stage .Overexpression of ripply1 caused an enhanced expression of dorsal marker genes and a reduction of ventral marker genes .Embryos overexpressing ripply1 also showed severely dorsalized phenotype , with enlarged head, reduced ventral yolk extension , and shortened posterior trunk and tail regions , and the formation of a secondary trunk axis.Transgenic fish revealed the maternal expression of ripply1 and suggested that a 1.2 kb promoter-driven GFP is able to recapitulate the endogenous gene expression pattern .Conclusion ripply1 may participate in the early development of dor-sal-ventral axis in zebrafish embryo .

Notch signaling, a highly conserved pathway, is wide-ly found in invertebrates and vertebrates. By mediating cell com-munication, it can regulate many physiological and pathological processes in various kinds of cells, including cell proliferation, differentiation and apoptosis in multi-cellular organism. Accu-mulating evidence shows that abnormality in Notch signaling is highly related to diabetes mellitus and its complications. Accord-ingly, this paper reviews the molecular basis of Notch signaling and the relations between its abnormal expression and diabetes mellitus, and focuses on the impact of key elements in the Notch signaling pathway on diabetic complications as well as correlative mechanisms.

OBJECTIVE To comprehend the bacterial infection and resistance to antimicrobial agents of the pathogenic bacteria causing chronic prostatitis(CP),so as to provide scientific basis for the diagnosis and treatment of CP.METHODS Bacterial culture and antimicrobial agents sensitivity tests were applied to prostatic fluid in 143 patients with chronic prostatitis.RESULTS A total of 85 strains of bacteria were isolated from 143 clinical specimens and the positive rate was 57.34%.In these strains,Gram-positive cocci were the most predominant accounted for 85.9%,coagulase negative Staphylococcus(CNS) were the highest ones and accounted for 60.0% among Gram-positive cocci.S.aureus and Entercoccus were respectively accounted for 12.9% and 11.8%.The ratio of drug resistance of CNS was high for ?-lactamases,quinolones,erythromycin and tetracycline and they were more sensitive to vancomycin,rifampicin,sulfamethoxazle/trimethoprim and gentamicin.CONCLUSIONS The major pathogens in prostatic fluids were CNS.The chronic prostatitis causing by CNS can be treated by rifampicin,sulfamethoxazle/trimethoprim and gentamicin.It is key to treatment of CP to select the sensitive and infiltrative drug for prostate.

Objective To conclude the results of electrophysiological (EP) mapping and ganglianated plexis (GPs) ablation during the minimally invasive atrial fibrillation (AF) surgery.Methods During the period of Sep 2007 to May 2010 to-tally 185 paroxysmal atrial fibrillation cases were recruited receiving the minimally invasive AF surgery with video assistance.The EP mapping was used to test the dual direction block after ablation and the distribution of GPs was recorded.The sinus rhythm rate was followed up after surgery.Results All the minimally invasive surgery were performed successfully withoutdeath in the hospital.There was no permanent pacemaker implant and other serious complications.EP mapping results showed92％ pulmonary vein (PV) potential to the left atrium (LA) could be isolated with the dual direction block for the paroxysmal atrial fibrillation cases.And there were 89％ GPs positive,higher rate than the persistent AF.There were more GPs positive in the right PV area than the left and 86％ GPs lied at the LA domain,not the PV orifice.At least 5 or 6 ablation lesions were needed to get the dual direction block and denervation of the GPs.The sinus rhythm rate after surgery after 3,6,12 and 24months is 83.7％,82.4％,85.4％ and 83.9％.The 12 months follow up data showed the cases with more GPs positive (＞6) had 81.3％ sinus rate and the less GPs positive (≤2) group with 52.6％.Conclusion The EP mapping and GPsablation techniques during the minimally invasive AF ablation surgery could get good sinus rhythm rate during the long term follow up.

Objective To evaluate the performance parameters of the PET instrument, which has been used for years in our hospital, assuring the stability of image quality and performance parameters. Methods NEMA NU 2-1994 Standards was used as reference for performance measurement of PET, which were published by National Electrical Manufacturers Association(NEMA). The NEMA standard cylinder model, 18F radiation source, and 18F and 68Ge columniform radiation source were used. Results Projects for performance measurement and daily quality control were drawn up. Conclusion This project provides a reference and methods for the performance measurement of PET instrument and daily quality control. It better ensures the stability of image quality and performance parameters of the PET instrument.

Objective To develop a medical equipment management information system in CAPF so as to improve current medical equipment management and maintenance level.Methods Based on a thorough research for current medical equipment management model in CAPF,the platform of NET was utilized to develop the software.The connection,combination and operation among all medical equipment management and maintenance work were realized by hardware construction at last.Results The management information system not only met the requirement in management of medical equipment,but also supplied a data basis to directors.Conclusion The management information system provides a powerful tool for the scientific and standard management of medical equipment in CAPF.

OBJECTIVE To implement the automatic management of medical equipment.METHODS Based on the platform of net,three layers of C/S were applied with database technology to realize one-time importation of the basic information and current information,it made the information function of quantity evaluation,quantity determination,schedule formation,automatic alarm,apparatus distribution list,statistical inquiry,collection analysis,and information feedback coming to true.RESULTS By using one year,the application of management information system of medical equipment could meet the requirement in scientific and standardized supply,and standard,application and management of medical equipment.CONCLUSIONS It can be able to improve the system efficiency,reduce the labor and mistakes,and also achieve the aim that cannot be done by manual labor.

Objective To optimize the formulation of a new kind of ultrasound contrast agents carrying the sensitizer of hematoporphyrin(HP)with[Poly(lactic-co-glycolic acid),PLGA]for material.Methods The technique of double emulsion was applied to produce HP loaded PLGA ultrasound microbubbles,which was optimized through orthogonal test using encapsulation efficiency for the detected index.Then morphology and distribution of HP-PLGA microbubbles were observed through light microscope and scaning electron microscope.The size,Zeta potential and the properties of releasing behavior and ultrasound imaging in vitro of Hp-PLGA contrast agents were detected.Results The optimization parameters were picked out as 10 mg/ml for concentration of HP,40 mg for PLGA,and 1/5 for volume ratio of water inside to dichloromethane.The optimized HP-PLGA contrast agents were spheric with the mean size of 602.3 nm,and Zeta potentiaI of-(17.1±1.6)mV.The drug loading and encapsulation efficiency of HP-PLGA were(2.15±0.15)%and(63.5±2.6)%,respectively.And the releasing behavior of HP-PLGA contrast agents in vitro was that after an obvious release of about 35.1%in former 24 h,there were 86.5%HP-PLGA released within 14 days.The ultrasound imaging of HP-PLGA could be enhanced obviously in vitro.Conclusions The self-made HP-PLGA ultrasound microbubble might be a useful tool for delivering sensitizer and thus provide a novel strategy for sonodynamic therapy on tumor.

Objective To investigate the relationship between β-fibrinogen (β-Fg) gene -455G/A,- 148C/T polymorphisms and atrial fibrillation (AF) with cerebral infarction.Methods Polymerase chain reaction-restriction fragment length polymorphism was used to detect the genotypes of -455G/A and -148C/T in 97 patients with AF with cerebral infarction (AF with cerebral infarction group),80 patients with AF alone (AF group),and 98 health subjects (healthy control group).Results The A allele frequencies of -455G/A in the AF with cerebral infarction group and the AF group were 0.304 and 0.344 respectively.They were significantly higher than 0.179 in the healthy control group (all P＜0.05).The T allele frequencies of - 148C/T in the AF with cerebral infarction group and the AF group were 0.348and 0.369,respectively.They were significant higher than 0.240 in the healthy control group (all P＜0.05).Conclusions The A allele of -455G/A and the T allele of -148C/T were associated with AF with cerebral infarction and AF.
