Objective To investigate the inhibitory effect of ginsenoside Rg3 on herpes simplex virus(type 1(HSV-1) in vitro and) its immunoregulative effect.Methods Crystal violet staining was used to observe the inhibitory effect of Rg3 on the cytopathic effect caused by HSV-1.MTT colorimetry was used to detect the influence of Rg3 on the proliferation of Vero cells and mouse splenocytes.The mouse splenocytes were induced with Rg3,and the activity of IL-2 was detected by lymphoblast proliferation assay and IFN-? by cytopathic effect inhibition.(Results The) A_(570) values of cytopathic effect in the experimental group were significantly higher than those in the control group when doses of Rg3 were 1.56-25 mg?L~(-1) in vitro(P0.05).The secretary levels of IL-2 and IFN-? in experimental group were significantly higher than those in the control group in vitro when doses of Rg3 were(3.13-6.25)mg?L~(-1) (P

Objective To investigate the effect of ulinastin on the gastro-intestinal circulation and systemic inflammatory response during open heart surgery with cardiopulmonary bypass( CPB) .Methods Thirty adult patients undergoing valve replacement with mild hypothermic CPB were randomly divided into two groups: ulinastin group (U ,n = 15) and control group (C , n = 15). In ulinastin group patients received ulinastin 6000 IU?kg-1iv after induction of anesthesia and another 6000 IU?kg-1 was added into the priming solution. In control group patients received equal volume of normal saline, instead of ulinastin. The patients were premedicated with morphine 0.2 mg?kg-1 and scopolamine 0.06 mg?kg-1 .Ranitidine 1 mg?kg-1 was given iv before induction of anesthesia. Anesthesia was induced with midazolam 0.1 mg?kg-1 , fentanyl 10?g?kg-1 and vecuronium 0.1 mg?kg-1 and maintained with fentanyl 50-60?g?kg-1, midazolam, isoflurane and vecuroinum. The patients were intubated and mechanically ventilated. PET CO2 was maintained at 35-45 mm Hg during operation. Gastric intramucosal PCO2 (PiCO2 ) was measured (pHi was calculated) and blood concentrations of TNF-?and IL-6 were determined before CPB (T0), 30 min after aorta was cross-clamped (T1), 60 min after termination of CPB(T2 ) and 6 h after operation (T3 ) .Results The two groups were comparable with regard to age, sex, body weight, ejection fraction, duration of CPB and cross-clamping time. Objective To probe into the proper ratio of doctors and nurses and appropriate nursing programs in community health service. Methods A number of methods were used, including task analysis, use of old handsexperience, sociological survey, and literature review. Results Medical consultation took 1 105 minutes, taking blood pressure took 141.5 minutes, transfusion took 493.5 minutes, intramuscular injection took 44.5 minutes, and drawing blood took 77.5 minutes. According to the above results, the ratio of doctors and nurses was respectively 3.18∶1, 0.94∶1, and 2.02∶1. Also, by analyzing in a comparative way the nursing programs already developed both at home and abroad and taking into consideration the reality, suggestions were put forward as to the kind of nursing program communities ought to start. Conclusion The findings are of referential value to administrative departments of health and medical institutions.
5.Application of monoclonal antibody pool against Bacteroides fragilis and Clostridium perfringens in rapid diagnosis in surgical infection

Objective To prepare monoclonal antibody (McAb) pool against Bacteroides fragilis and Clostridium perfringens and evaluated its effect in rapid diagnosis in surgical infection. Methods After the preparation of monoclonal antibodies against B. fragilis and C. perfringens, 191 specimens collected from infectious patients was detected with indirect immunofluorescent antibody (IFA) and enzyme labeled antibody (ELA). The results were compared with routine cultural method (CM). Results Among the 191 specimens, 53 (27.7%), 55 (28.8%) and 25 (13.1%)strains of B. fragilis were detected with IFA, ELA and CM respectively, and for C.perfringens, 12(6.3%), 11(5.8%), and 6(3.1%) strains were found. The detectable rate of anaerobic bacteria by IFA and ELA was higher than by CM, but no difference was found between these 2. Conclusion The self-made McAb pool against B.fagilis and C.perfringens might be a rapid, specific, sensitive and simple McAb pool for detecting B.fragilis in surgical infection and C.perfringens in wartime.
6.Effects of eukaryotic expressive mature peptide of hCAP-18/LL-37 on expressions of membrane molecules on dendritic cells

Objective To study the effects of eukaryotic expressive mature peptide LL-37 of human cationic antimicrobial peptide(hCAP-18) on the expressions of membrane molecules on dendritic cells(DCs).Methods By gene cloning,the eukaryotic expressive plasmid pcDNA4/Myc-His-LL-37 for the mature peptide LL-37 of hCAP-18 was constructed.Then they were transfected into HEK293 cell lines.After the cell lines were cultivated for 48 h,the supernatant was collected.Then the DCs from peripheral blood mononuclear cells(PBMCs) induced by rh-GM-CSF and rh-IL-4 were cultivated with the supernatant for 48h.The expressions of membrane molecules CD40 and HLA-DR on DCs were detected by flow cytometry(FCM).Results The eukaryotic expressive plasmid pcDNA4/Myc-His-LL-37 was constructed successfully and it expressed in eukaryotic cells HEK293.FCM results indicated that the expressions of CD40 and HLA-DR on the membrane of DCs which were stimulated by the supernatant produced by pcDNA4/Myc-His-LL-37 were higher than those in control group(P

Objective:To establish a mouse fibroblastic cell line stably transfected with HMGA1 gene,and to use the cell line to investigate the role of HMGA1 in tumor development and progression.Methods:Eukaryotic expression vector pcDNA3.0-HMGA1 was transfected into NIH3T3 by lipofectamine-mediation.Stable transfectants were selected by G418.The expression of extraneous HMGA1 gene was analyzed by RT-PCR and DNA sequencing.Cell growth was measured through MTT and the cell cycle by FCM.Soft agar colony formation was employed to analyze the ability of anchorage-independent growth.The expression of the immune inhibition factors of VEGF and FasL mRNA was detected by RT-PCR.Results:NIH3T3 cell lines stably expressing HMGA1 gene were successfully established.Comparing with controls,the HMGA1 gene-transfected cells grew faster,acquired the ability of anchorage-independent growth and expressed the immune inhibition factors of VEGF and FasL mRNA.Conclusion:Extraneous expression of HMGA1 gene can induce malignant transformation of NIH3T3 and modulate mRNA expression of immune inhibition factors.HMGA1 gene plays a key role in tumor development,progression and immune escape.
9.The effect of propofol on isolated human bronchial smooth muscle

Objective Propofol is now the most commonly used intravenous anesthetic. It has vasodilating effect. The purpose of this study is to determine its effect on bronchial smooth muscle. Methods Strips of bronchus (40x2cm2) were taken from the lung resected during lung surgery and suspended a bath filled with K-H solution. One end of the bronchus strip was fixed to the bottom of the bath and the other was attached to a tension transducer through a piece of wire. Bronchoconstriction was induced with acetylcholine added in K-H solution (3?g?ml-1). When peak bronchoconstriction was reached, propofol in fat emulsion was added culmulatively to the bath (1,2,5, 10,20?g?ml-1). The effect of fat emulsion was also examined as control. Results Propofol at concentrations of 1, 2, 5, 10 and 20(?g?ml-1 caused a significant decrease in the acetylcholine-induced bronchoconstriction in a dose dependent manner. Conclusions Propofol at clinically relevant concentrations significantly attenuates acetylcholine-induced bronchoconstriction in human.

Background and purpose:Metabolism change is one of the main characteristics of the tumor de-velopment. Many studies have conifrmed that cytosolic acetyl-CoA synthetase 2 (ACSS2) plays a critical role in hydro-carbon metabolism of cancer cells. This study aimed to explore the effect of ACSS2 on cellular proliferation, apoptosis and migration of A549 cells by RNA interference.Methods:The ACSS2 interference fragment ACSS2-siRNA and neg-ative control were designed and synthesized for RNA interference followed by the transient transfection in non-small cell lung cancer (NSCLC) cell line A549. Real-time lfuorescence quantitative polymerase chain reaction (RTFQ-PCR) was used to detect ACSS2 mRNA expression. Methyl thiazolyl tetrazolium (MTT), lfow cytometry and wound healing assay were used to detect cell proliferation, apoptosis rate and migration.Results:The expression of ACSS2 mRNA was signiifcantly decreased after transfection with the interference fragment ACSS2-siRNA in NSCLC cell line A549. The proliferation and migration activity of ACSS2-siRNA treated cells were decreased significantly compared with the control group. The apoptosis rate, especially the early apoptosis, was increased..Conclusion:Knockdown of the ACSS2 expression in NSCLC cell line A549 can signiifcantly inhibit the cell proliferation, migration ability and pro-mote the apoptosis rate, especially early apoptosis. This study indicates that ACSS2 may contribute to the progression of human lung adenocarcinoma and may have the potential to serve as a novel therapeutic target.
