1.Model validation and evaluation of developmental toxicity of five drugs using zebrafish embryos

Aim To explore the toxicity and safety of five kinds of known positive drugs, cyclophosphamide, acetyl salicylic acid, tetracycline hydrochloride, dexa-methasone acetate and azacitidine, using zebrafish em-bryos. Methods We selected normally developed 4 hpf zygote, and used water bath infecting method to add the drug to the artificial seawater. Each drug had five concentrating groups, a separate control group and solvent control group. We observed the dead zebrafish embryos after 120 hpf drugs, counted the number of deaths and deformities of zebrafish embryos, and cal-culated mortality abnormal rate, the median lethal con-centration (LC50 ), concentration for 50% of maximal effect (EC50 ), therapeutic index (TI) under 120 hpf condition. We also used the formula TI = LC50 / EC50 to calculate positive drug therapeutic index. Based on measured LC50 we calculated most nonlethal concentra-tion (MNLC) of each drug setting, namely 1 / 10 MN-LC, 1 / 3 MNLC, MNLC,LC10 four concentration, tha-lidomide as a positive control, vitamin C as a negative control, artificial seawater as control, 0. 5% DMSO as solvent control. Put in 28. 5 ℃ environment for 120 hours,embryo development was observed daily for de-velopmental state,mortality,deforming rate and abnor-mal condition. Results The result of five drugs LC50 in descending order: cyclophosphamide > azacitidine> tetracycline hydrochloride > acetylsalicylic acid >dexamethasone acetate. EC50 in descending order: cy-clophosphamide > tetracycline hydrochloride > azaciti-dine > acetylsalicylic acid > dexamethasone acetate. The TI values of cyclophosphamide, acetyl salicylic acid, tetracycline hydrochloride, dexamethasone ace-tate, azacitidine were 1. 92, 1. 11, 1. 05, 1. 44, 2. 99, respectively. Conclusion Zebrafish embryo model can be used in the preliminary evaluation of drugs, and the study of early developmental toxicity and safety.
2.Effect of health literacy on stroke education in patients with ischemic cerebrovascular disease and their family members

Objective To investigate the effect of health literacy on stroke education in patients with ischemic cerebrovascular disease and their family members.Methods Health literacy questionnaire was used to conduct the health literacy questionnaire survey for patients with ischemic cerebrovascular disease and their family members.The respondents received stroke education randomly through neurologists' explanation or self-reading.The corresponding questionnaire was used to evaluate the mastering of the content of stroke education for the respondents.Results The complete data of 42 patients with ischemic cerebrovascular disease (34 ischemic stroke and 8 transient ischemic attack) and 57 family members of the patients with ischemic cerebrovascular disease were collected and enrolled in the analysis.The good health literacy score group (< 10) and the poor health literacy score group (≥ 10) were 61 and 38 subjects respectively.The proportions of younger age (x2 =16.410,P <0.001) and high school or above (68.85% vs.23.68% ;x2 =19.25,P < 0.001) of the former were higher than those of the latter,and the proportion of patients (32.79%vs.57.89%;x2 =6.043,P=0.014) was significantly lower than that of the latter,but there was no significant difference in the proportion of the males between the two groups (55.74% vs.44.74% ;x2 =1.134,P =0.287).Multivariate logistic regression analysis showed that health literacy was independently associated with the age of respondents (≤40 years:odds ratio [OR] 7.753,95% confidence interval [CI]1.208-49.740; P=0.031) and the level of education (high school and above:OR,3.993,95% CI 1.305-12.218; P=0.015),and it was not correlated with gender and role.Those with good stroke education score (≥7)and poor stroke education score (<7) were 57 and 42,respectively.The proportions of younger age (x2 =17.095,P <0.001),high school and above (70.00% vs.23.08% ;x2 =20.836,P < 0.001),and good health literacy score (88.33% vs.20.51%;x2 =45.968,P<0.001) were significantly higher than those of the latter,while there were no significant differences in the proportions of gender,role,and stroke education.Multivariatelogistic regression analysis showed that only the good health literacy was independently associated with the stroke education score (OR 20.147,95% CI6.089-66.655; P<0.001).Conclusions Health literacy can significantly affected the effect of the stroke education in patients with ischemic cerebrovascular disease and their family members.In the development of individualized stroke education program,it should be considered.
3.Investigation of radiological protection status and radiation exposure level at interventional workplace in Zhejiang province

Objective To explore the current status of radiation protection in interventional procedure and to analyze the existent problems.Methods Using the random sampling,70 interventional radiology workplaces were selected from Hangzhou,Ningbo,Wenzhou,Shaoxing and Jinhua.Information on personal radiation protective products and equipment was collected by filling out the form by hospital and the radiation dose was monitored at various body parts of the workers and at the outside of the operating room.Results All hospitals have been equipped with personal radiation protective products and equipment,22 of which were not with these products and equipment as required.Chest,abdomen and lower limb of interventional radiology workers are easy to receive higher radiation dose than in head (F =4.85,4.92,P < 0.05).The acceptance rates among different body parts of the workers were difference significantly (x2 =35.14,14.92,P < 0.05).Using protective curtain can reduce the radiation dose significantly (t =11.61-68.28,P < 0.05).Meanwhile,it also can improve the acceptance rates significantly (x2 =6.09-28.45,P < 0.05).Conclusions It is necessary to strengthen the use of radiation protection equipment among interventional radiology workers to reduce the levels of the individual doses to them.Hospitals must enhance the routine monitoring for improved radiaton protection.
4.Synthesis of Naphthalimide-based Biothiols Probe and Detection of Amino Acids Containing Sulfhydryl Groups

A novel probe (DNSBN) towards biothiols on the basis of 4-hydroxynaphthalimide as fluorophores and 2, 4-dinitrobenzenesulfonyloxy group as specific recognition site was designed and synthesized.The result of absorption and fluorescence spectral analyses indicated that the probe had high sensitivity and selectivity towards cysteine (Cys), homocysteine (Hcy) and glutathione (GSH), and the detection was not affected by other 17 kinds of natural amino acids.Meanwhile, it was confirmed that DNSBN was a ratiometric probe through the fluorescence titration experiment, and the fluorescent intensity at 555 nm had a high linear relationship with biothiols concentration in the range of 0-20 μmol/L.The detection limits (3σ) of Cys, Hcy and GSH were 25.9, 92.0 and 77.9 nmol/L, respectively.The absorption, emission and mass spectra indicated that biothiols could be engaged in nucleophilic substitution reaction with 2,4-dinitrobenzenesulfonate, which induced the sulfonic esters decomposed.With the departure of receptor unit, the d-PeT progress (donor-excited photoinduced electron transfer) was blocked with an obvious colorimetric and fluorescence change.Finally, HeLa cell imaging experiments verified that DNSBN had good biocompatibility and could be used to detect exogenous biothiols.
5.Significant and sustaining elevation of blood oxygen induced by Chinese cupping therapy as assessed by near-infrared spectroscopy

Objective:To objectively evaluate the therapeutic effect of Chinese cupping therapy.Method:Near-infrared spectroscopy (NIRS) was used to evaluate the concentration change in oxygenated hemoglobin ([HbO 2]), deoxy-hemoglobin ([Hb]) and blood volume ([tHb]) in 13 healthy volunteers during cupping therapy for the infraspinatus muscle. Results:During the cupping therapy and its pre- and post-treatment period, it was observed that [Hb] and [tHb] in the tissue surrounding the cupping site decreased significantly (both P<0.05), and [HbO 2] increased significantly ( P<0.05), which showed an increase in oxygen uptake. After the cupping was removed, the above hemodynamic changes could still continue for a period of time. Conclusions:Cupping therapy has potential positive therapeutic effects in promoting the hemodynamics for facilitating muscular functions. Functional NIRS surgery has great application potential in the field of physical rehabilitation therapy such as cupping.
6.Study on rapid microwave-ashing pretreatment method for radioactivity monitoring in food

Objective To establish a rapid,high-efficiency and clean way to monitor and pretreat the radioactivity in foods.Methods Food samples,20 kg each of cabbage,crucian,shrimp,pork and 20 L of milk,were collected around Qinshan nuclear power plant,totaling 10 samples for each food.Each type of foods was arranged in two groups each with 10 samples.Samples were pretreated using traditional ashing method (TAM) and microwave ashing method (WAM) separately.HDEHP were used for 90Sr separation and measurement.Calculations were made for the time spent on food pretreatment.The ash-fresh ratio and the recovery rate of 90Sr by the two different method were calculated,with the quality of ash being evaluated.Results The mean time spent on pretreatment was cabbage 20 h,crucian 54 h,shrimp 46 h,pork 58 h and milk 74 h for TAM group,and cabbage 8.5 h,crucian 26 h,shrimp 23 h,pork 26 h and milk 30 h for WAM group,respectively.The time spent by WAM was half the time less than by TAM,especially for animal edible product.The ash-fresh ratios from WAM group were higher than from TAM group.The recovery rates of 90Sr from WAM group in the analytical process were all above 75%,higher than from TAM group.Ashes produced by WAM were hoary,uniform,carbon residue-free and fully ashed.Conclusions WAM pretreatment can pretreat food rapidly,efficiently,cleanly with less loss and better ashing effect.These can meet the requirement of rapid pretreatment of radioactivity in food.

Objective:To investigate the clinical characteristics and outcome of patients with voltage-gated potassium channel complex (VGKCc) antibody associated clinical syndromes complicated with myasthenia gravis (MG) with thymoma.Methods:The clinical history, examinations and follow-up prognosis of 2 cases of VGKCc antibodies associated clinical syndromes with MG complicated with thymoma in Qilu Hospital (Qingdao), Cheeloo College of Medicine,Shandong University in September 2020 and December 2020 were reviewed. Related literatures were summarized at the same time.Results:Case 1, a 64-year-old female clinically presented with cognitive impairment, psychosis, and epilepsy seizures, whose serum autoimmune antibody testing showed positive leucine-rich glioma-inhibited 1 (LGI1) antibody, was diagnosed as anti-LGI1 encephalitis,and had history of MG with thymoma. Her symptoms were improved by immunotherapy. Case 2, a 67-year-old male, was diagnosed as MG, and developed cognitive impairment, myokymia and autonomic dysfunction later. His serum autoimmune antibody testing showed positive contactin associated protein-like 2 antibody. Therefore, Morvan syndrome complicated with MG with thymoma was definitely diagnosed. After admission, the patient was improved with immunotherapy and thymoma resection.Conclusions:Patients with VGKCc antibody-associated clinical syndromes complicated with MG have the clinical characteristics of the two diseases simultaneously, and there is also crossover. Immunotherapy and treatment for thymoma are generally effective.
9.Clinical features, treatment and follow-ups of Sj(o)gren's syndrome-associated monoclonal gammopathy

Objective To investigate the clinical characteristics,immunological features,treatment and follow-ups of Sj(o)gren's syndrome-associated monoclonal gammopathy (SS-MG).Methods A retro-spective,case-control study was conducted for 18 cases diagnosed with SS-MG and 36 age-and sex-matched non-MG-SS patients from Janurary 2010 to Janurary 2017 in Peking University People's Hospital.The clinical and laboratory features,treatment and follow-ups were recorded and compared.Comparisons between groups were made using t test for normally distributed numerical data,Mann-Whitney U test for non-normally distributed numerical data,and Pearson Chi-square,continuity correction or Fisher's exact tests for categorical data.Results SS patients,when complicated with MG,had significantly increased level of TP [(78± 11) g/L,(71±10) g/L,t=-2.382,P=0.021] and erythrocyte sedimentation rate (ESR) [52.5(45.3) mm/1 h,33.0(42.5) mm/1 h,Z=-2.179,P=0.029],higher prevalence of urine NAG positivity [75%(9/12),28%(7/25),x2=7.298,P=0.007],hypoglobulinemia [33%(6/18),3(1/36),x2=7.407,P=0.006] and thrombotic events [17%(3/18),0%(0/36),P=0.033],and less previous exposure to glucocorticoid [22%(4/18),64%(23/36),x2=8.333,P=0.004],compared to the control group.Primary SS patients complicated with MG had significantly higher ESSDAI [26.0(25.0),12.0 (9.0),Z=-2.724,P=0.006] and Clin EULAR Sj(o)gren's syndrome disease activity index (ESSDAI) [24.0(25.0),10.5 (10.0),Z=-2.523,P=0.011].Among the 18 patients,2 were diagnosed with multiple myeloma,1 was diagnosed with non-Hodgkin lymphoma,and the left were diagnosed as MG of undetermined significance (MGUS).Ten patients were followed up,among whom 2 patients with MGUS experienced increased levels of M protein,newly developed genetic abnormalities,and renal involvement.Conclusion SS patients may be complicated with MG.MGUS is the most common form.However,malignant hematologic disorders are revealed as well.In SS patients with high serum TP and ESR,hypoglobulinemia,tubulointerstitial kidney involvement and unexplained thrombotic events,especially in those with high disease activity,so MG should be an alert for further work-ups,monitoring and treatment.
10.Investigation on levels of gross radioactivity in drinking water and 90Sr in food before and after operation of Sanmen Nuclear Power Plant

Objective:To investigate the levels of gross radioactivity in drinking water and of 90Sr in food around Sanmen nuclear power plant (NPP) before and after operation. Methods:From 2012 to 2019, 7 drinking water monitoring sites were selected around the Sanmen NPP to collect source water, factory water and peripheral water, respectively, in order to measure and analyze the total radioactivity level in drinking water. Based on the dietary habits of local residents, 4 kinds of food such as grains, vegetables, freshwater fish and marine fish grown locally were sampled to measure and analyze the activity concentration of 90Sr. Results:Before the operation of Sanmen NPP, the total α radioactivity and total β radioactivity in drinking water around the NPP were 0.001-0.063 Bq/L and 0.019-0.210 Bq/L, respectively. After operation, the total α radioactivity and total β radioactivity were 0.001-0.084 Bq/L and 0.025-0.079 Bq/L, respectively. Both were lower than the national standard value given in the GB 5749-2006 " Sanitary standards for drinking water" . There was no significant change in the radioactivity level of source water, factory water and peripheral water during the wet and dry periods. The activity concentration of 90Sr in these four types of food ranged from 0.037 to 1.216 Bq/kg before operation and from 0.049 to 0.692 Bq/kg after operation, all lower than the standard value of " Standard for the concentration in radioactive substances in food" (GB 14882-94). Conclusions:Before and after the operation of Sanmen NPP, the total α and total β radioactivity in drinking water and 90Sr activity concentration in food are stable, below the national standard limit.
