Objective:To investigate the potential mechanism of miR-1298-5p in non-small cell lung cancer(NSCLC)to regu-late the MSH2 gene and its effect on the biological behavior of tumor cells and the tumor immune microenvironment.Methods:Bioin-formatics was used to identify the key genes and miRNA involved in NSCLC.Cell proliferation was detected by CCK-8 assay, and cell invasion and migration were detected by Transwell assay.Levels of inflammatory factors were detected by ELISA assay.Western blot was used to measure the expression of MSH2 in cells, and fluorescence quantitative polymerase chain reaction(RT-qPCR)was used to detect the expressions of miR-1298-5p and MSH2 gene in NSCLC cells.Dual luciferase reporter gene assay was performed to verify the targeting relationship between miR-1298-5p and MSH2.Spearman correlation analysis was performed to correlate miR-1298-5p with immune cells and immune factors in the tumor immune microenvironment. Results:The level of miR-1298-5p was down-regulated in NSCLC cells compared with normal lung tissue cells.miR-1298-5p overexpression inhibited the proliferation, migration and inva-sion of NSCLC cells.MSH2 was confirmed to be a target gene of miR-1298-5p using luciferase reporter gene assay.Furthermore,down-regulation of miR-1298-5p in NSCLC cells could be reversed by silencing MSH2.miR-1298-5p expression levels were negatively corre-lated with the levels of Treg,IL-10,and TGF-β,and positively correlated with the levels of CD3+T,CD4+T,CD8+T,NK cells,IL-2,and IFN-γ.Conclusion:miR-1298-5p negatively regulates MSH2 to inhibit the proliferation, invasion and migration of NSCLC cells and improve the tumor immune microenvironment.
2.A Preliminary Framework for Syndrome Differentiation of Prostate Meridian, Qi And Blood Based on the “Conception Vessel, Governor Vessel and Thorough Vessel Sharing the Same Origin” Theory
Journal of Traditional Chinese Medicine 2023;64(24):2588-2592
The “conception vessel, governor vessel and thorough vessel sharing the same origin” theory refers to the concept that the conception vessel, governor vessel and thorough vessel all originate from the same source within the uterus and emerge from the perineum. Based on the modern research of McNeal's prostate zonal anatomy theory, it is believed that the conception vessel, governor vessel and thorough vessel have the closest relationship with the prostate in terms of their origin and running course. It is proposed that the essential meaning of the ‘conception vessel, governor vessel and thorough vessel sharing the same origin’ theory in relation to the prostate is that the conception vessel, governor vessel and thorough vessel respectively connect to the transition zone, peripheral zone, and central zone of the prostate. Moreover, the differences in the yin-yang attributes of the conception vessel, governor vessel and thorough vessel exhibit distinct functional characteristics in different zones of the prostate, which serve as an important basis for the physiological and pathological differences in various prostate zones. Based on this premise, a prostate meridian, qi and blood syndrome differentiation method is further proposed, wherein the differences in the meridian characteristics of the conception vessel, governor vessel and thorough vessel determine the differences in the yin-yang aspects of qi and blood in different prostate zones. When clinical diagnosis is conducted, the inherent physiological differences of the different prostate zones should be fully taken into account, as well as the pathological characteristics of the disease, in order to guide the diagnosis and treatment of related clinical conditions.

Chinese Journal of Biotechnology 2016;32(10):1455-1464
A mammalian nonclassical secretion sequence derived from mouse Engrailed2 homeoprotein (En2) was used to direct the secretion of the enhanced green fluorescent protein from Pichia pastoris. This signal peptide conferred the transport of enhanced green fluorescent protein into periplasm through an endoplasmic reticulum-golgi independent pathway, without inducing severe unfolded protein response as compared with Saccharomyces cerevisae α-factor preprosequence. This study implies that this mammalian nonclassical signal peptide could be developed as a useful tool for delivering cargoes to the cell surface of yeast.
