To compare the anxiolytic effects of reduced metabolite of progesterone and benzodiazepine.Methods The effects of allopregnanolone and diazepam on spontaneous locomotor activity and on exploration in the elevated plus-maze were studied in C57 mice 20 min after vehicle or drug intraperitoneal administration.Results Allopregnanolone (0.1 mg.kg-1,ip) elicited marked anxiolytic effects in terms of significantly reducing the latency to enter the open arm from (31.30±8.39)s to (8.80±6.00)s,(P<0.001),and significantly increasing both the number of open arm entries from 1.20 ± 0.42 to 4.80 ±1.75,(P<0.001) and the proportion of total time spent on the open arm from 7.13% to 32.50%,(P<0.001).Meanwhile,the diazepam (0.25 mg·kg-1) produced a lower anxiolytic effect comparing to that of the allopregnanolone.Analysis of spontaneous locomotor activity showed while 0.5 mg·kg-1 of diazepam decreased the locomotor activity (P<0.01),neither 0.1 mg·kg-1 of allopregnanolone nor 0.25 mg·kg-1 of diazepam affect the locomotor activity score.Conclusion Together,these results provide evidence for differential behavioral actions of the neurosteroids and benzodiazepines.Since the allopregnanolone produce a selective anxiolytic effect without affecting the spontaneous locomotor activity,the allopregnanolone may be a better alternative for diazepam in treating anxiety.
3.Effects of Epidermal Growth Factor on Cell Proliferation and Cell Cycle of Cultured Bovine Corneal Endothelial Cells in vitro

Objective To observe the effects of epidermal growth factor (EGF) on cell proliferation and cell cycle of cultured bovine corneal endothelial cells. Methods Bovine corneal endothelial cells were cultured with different concentrations of hEGF (1, 10 and 100 ng/mL). MTT test was performed to evaluate the cell proliferation. The bovine corneal endothelial cells were divided into two groups:control group (cultured in DMEM) and EGF-stimulated group (cultured in DMEM with EGF). Flow cytometry was performed to determine the cell cycle phases on the third and seventh day. Results Compared with the control, EGF enhanced the cell proliferation in a dose-related response. 10 ng/mL and 100 ng/mL EGF were much more effective than 1 ng/mL.On the third day, S phase cells accounted for 24.5% and G_2-M phase cells 0.08% in the control group,while 24.6% and 0.06%, respectively in the EGF-stimulated group. However, on the seventh day, those came to 20.8% and 0.41% in the control group, and 18.2% and 1.55% in the EGF-stimulated group,indicating a significant change in the cell cycle (P

5.Urticaria among children aged 2-6 years in Nanjing city: a cross-sectional investigation

Objective To investigate the prevalence of urticaria among children aged 2-6 years in Nanjing city.Methods A questionnaire survey was carried out in children aged 2-6 years in 5 main districts of Nanjing City.Results The number of valid questionnaires was 13 610,accounting for 92.8% of total respondents.Among the 13 610 respondents,1 116 were diagnosed as urticaria,including 565 boys and 551 girls,with the total prevalence rate being 8.54%.No significant difference was observed in the prevalence rate of urticaria between boys and girls (8.49% vs.8.60%,x2 =0.04,P =0.832) or among children of different age groups (x2 =1.84,P =0.17).The prevalence of urticaria increased with age,and positively correlated with parents' educational level (rs =0.96,P =0.000).Children born in urban areas showed higher prevalence of urticaria than those in rural areas (8.79% vs.5.56%,x2 =13.98,P =0.000).Conclusions The prevalence of urticaria has increased among children aged 2-6 years in recent years.More studies should be carried out to investigate into the pathogenesis and relevant factors of urticaria,so as to facilitate its prevention and treatment.
6.A survey of the current status of atopic dermatitis among children aged 2-6 years in Nanjing city

Objective To investigate the prevalence of atopic dermatitis (AD) among children aged 2-6 years in Nanjing city.Methods A questionnaire survey was carried out in children aged 2-6 years in 7 main districts of Nanjing City.Results A total of 13 061 children were investigated,among which,768 were diagnosed as AD,including 410 boys and 358 girls.The prevalence rate of AD was 5.88% (95% CI,5.48%-6.28%) in total,6.16% (95% CI,5.59%-6.74%) in boys and 5.59% (95% CI,5.02%-6.15%) in girls.Higher prevalence rate of AD was observed in boys compared with girls,and in children whose place of birth was in urban areas compared with those in rural areas.In addition,the prevalence rate of AD increased with age and with the rise in parental educational level.Conclusions Rapid economic development has dramatically changed life habits,dietary structure and residential environment of people,which can influence the prevalence of AD.

Objective To investigate the effect of the inhibitor SB203580 of p38 mitogen-activated protein kinase (MAPK) signal transduction pathway on biopterin (BH 4)/nitric oxide (NO) expression and elucidate the potential mechanism of MAPK in biopterin-mediated NO induction after endotoxic shock. Methods A total of 56 male Wistar rats were randomly divided into normal control group (n=8), endotoxic shock group (n=32) and SB203580 treatment group (n=16). After animals were sacrificed, tissue samples from the liver, the lungs as well as the kidneys were harvested to determine the expressions of guanosine triphosphate cyclohydrolase I (GTP-CHI) and inducible nitric oxide synthase (iNOS) mRNA and observe the changes of BH 4 and NO levels in blood and tissues. Results With endotoxin challenge, GTP-CHI mRNA expression and BH 4 levels were significantly elevated in various tissues and maintained at high levels up to 24 hours. Similarly, the iNOS mRNA expression and NO levels in the tissues significantly increased too, especially in the liver and the lungs.
