Chinese Journal of Bases and Clinics in General Surgery 2003;0(03):-
Objective To study the apoptotic induction effect of Thapsigargin on estrogen receptor positive human breast cancer cell lines MCF 7. Methods Cells were treated with Thapsigargin and 5 FU in vitro . The rate of cell apoptosis and distribution of cell cycle were detected on flow cytometry. The cell viability was measured by MTT assay and ultrastructural changes in apoptotic cells were confirmed by transmission electron microscopy. Results Thapsigargin could increase the rates both of cell apoptosis and growth supression of MCF 7 cells induced by 5 FU and alter the distribution of cell cycle. Under electron microscope, apoptotic bodies in MCF 7 cells considerably increased. Conclusion Thapsigargin apparently enhances the effect of apoptotic induction of 5 FU on MCF 7 cells, it is worthy of being further studied.

Chinese Journal of General Surgery 1994;0(05):-
Objective To study the effect of rising [Ca 2+ ]i on the expression of Caspase 3, Bax and Bcl 2 in human breast cancer MCF 7 cell lines. Methods After the MCF 7 cells treated with different dose of thapsigargin(TG), intracellular calcium concentrations in MCF 7 cells were measured by calcium sensitive fluorescence indicator Fura 2/AM technique; and immunohistochemical method was used to detected the expressions of Caspase 3, Bax and Bcl 2. Results Bax was over expressed and Bcl 2 was down expressed in a [Ca 2+ ]i concentration dependent way;nevertheless, the expression of Caspase 3 was negative in all the TG treated cells. Conclusions The increase of intracellular calcium concentration may have an inductive effect of apoptosis on MCF 7 cells, which is independent to Caspase 3.

International Journal of Laboratory Medicine 2014;(24):3300-3303
Objective To investigate the effects of c-myc promoter binding protein(MBP-1)gene expression silencing on the pro-liferation in vitro in human gastric cancer cell line SGC-7901.Methods The cells divided into three groups:blank control group (cells without transfecting gastric cancer cell),negative control group(cells transfecting missense sequence)and experimental group (cells transfecting MBP-1 shRNA).Two MBP-1 shRNA sequences and one negative control shRNA sequence were designed,syn-thesized and cloned into pSIREN-retroQ plasma. Then the recombinant plasmids were constructed and transfected into human gas-tric cancer SGC-7901 cells by Lipofectamine 2000.After selection of cells with puromycin for two weeks, stable cell line SGC-7901 expressing gene of MBP-1 specific shRNA was obtained. The expressions of MBP-1 mRNA and protein in SGC-7901 were deter-mined by the real time PCR and Western blot, respectively. The effects of altered expression of MBP-1 on the cell proliferation were measured by MTT cell proliferation assay. Results PCR and sequencing indicated that the recombinant plasmids pSIREN-retroQ was constructed. Then the recombinant plasmids were transfected into human gastric cancer SGC-7901 cells by Lipofectamine 2000. After selection of cells with puromycin for two weeks, stable cell line SGC-7901 expressing gene of MBP-1 specific shRNA was ob-tained.The relative expression level MBP-1 mRNA in the MBP-1 siRNA transfection group was significantly decreased compared with the blank control group(P <0.05).Compared with the blank group, the expression levels of MBP-1 protein in the experimental group also significantly decreased. The proliferation abilities of SGC-7901 cells at 48,72,96,120 h after MBP-1 siRNA transfection were significantly increased compared with the blank control group (P < 0.05 ).Conclusion Down-regulating the expression of MBP-1 can obviously promote the proliferation of human gastric cancer cell line SGC-7901.MBP-1 gene may become the new target of gene therapy for gastric cancer.

Objective To study the effects of Duliang capsule on the expression of calcitonin gene‐related peptide(CGRP) and cholecystokinin(CCK) in the midbrain of a rat migraine model ,and explored treatment effects and mechanisms of Duliang cap‐sule on rats with migraine .Methods A total of 24 rats were randomly divided into four groups :normal control groups(A) ,migraine model groups(B) ,Duliang capsule control groups(C) and Duliang capsule treatment groups(D) .C and D were intragastrically per‐fused with Duliang capsule(0 .5 g · kg -1 · d-1 ) .After 7 days ,nitroglycerin was subcutaneously injected into the buttocks of the B and D to induce migraine .Two hours after nitroglycerin injection ,the midbrain were isolated CGRP and CCK expression in midbrain were determined using SYBR Green I real‐time quantitative PCR .Results CGRP mRNA expression was significantly lower in mid‐brains of rats in the Duliang capsule treatment groups compared with migraine model groups(P<0 .05) .CCK mRNA expression was significantly lower in midbrains of rats in the Duliang capsule control groups compared with normal control groups(P<0 .05) . Conclusion Duliang capsule can exhibit the expression of CGRP and CCK in the midbrain of the migraine rats .weaken the CGRP and CCK‐induced inhibition of the analgesic effects of opioid peptides .

Objective To investigate the effects of hydroxyethyl starch(HES 130/0.4)on leukocyte activation, mast cell degranulation and vascular permeability in rat mesentery during the early phase of endotoxemia. Methods Thirty-six male Wistar rats weishing 200-250 g were randomly divided into 3 groups(n=12 each):Ⅰ normal control group received only normal saline(NS);Ⅱ LPS group and Ⅲ HES group received LPS 2 mg/kg iv followed by iv HES infusion at 16 ml·kg-1·h-1 for 60 min(HES group)or equal volume of NS(LPS group). The animals were anesthetized with intramuscular 20% urethane 1 ml/100 g.The abdomen was opened and the mesentery of small intestine was pulled out and placed on transparent observation plate at constant temperature and moisture and examined under microscope for microcirculatory changes.Leukocytes rolling along, adhering to and emigrating from mesenteric venules, mast cell degranulation and FTTC-albumin effux from venules were examined before(baseline)and during the 90 min after LPS administration. The expression of CD11b and CD18 on the leukocytes was determined using flow cylometry. Results The number of leukocytes rolling along, adhering to and emigrating from venules and degranulated mast cells were signitlcanfly increased in LPS group as compared with control group. These LPS-induced changes were significantly inhibited in HES group. The albumin effiux Was enhanced and CD11b/CD18 expression upregulated in both LPS and HES groups. Conclusion Hydroxyethyl starch(130/0.4)can ameliorate the increased vascular permeability and microcirculatory disturbance in rat mesentery during the early phase of endotoxemia.

International Journal of Biomedical Engineering 2008;31(2):115-119
The conformity between bone implant and bone tissue plays an important role in osseointegration.Protein molecules such as extracellular matrix proteins, cytoskeletal proteins, and attachment proteins, which produced by osteoblasts, promote the attachment of osteoblasts to bone implant and the surrounding ceils. In vitro studies show that nano-plated implant material bears excellent biocompatibility and superior physical and chemical features.Nano-scale material has the advantage of high surface roughness, wettability and electrochemistry features which erdaance the adherence and interaction of extracellular matrix proteins and promotes the attachmerit, proliferation and differentiation of osteoblasts. The expression of cytokines, such as prostaglandin E2(PGE2)and transforming growth factor-β(TGF-β)are also increased in nano-structured bone implants which in turn promote the new bone formation.Further studies need to be done to explain the mechanism of the effect of nano-tructured bone implant on osteoblasts.
