1.Research on the Induction of Apoptosis in MCF-7 Cells Enhanced by Thapsigargin

Objective To study the apoptotic induction effect of Thapsigargin on estrogen receptor positive human breast cancer cell lines MCF 7. Methods Cells were treated with Thapsigargin and 5 FU in vitro . The rate of cell apoptosis and distribution of cell cycle were detected on flow cytometry. The cell viability was measured by MTT assay and ultrastructural changes in apoptotic cells were confirmed by transmission electron microscopy.Results Thapsigargin could increase the rates both of cell apoptosis and growth supression of MCF 7 cells induced by 5 FU and alter the distribution of cell cycle. Under electron microscope, apoptotic bodies in MCF 7 cells considerably increased.Conclusion Thapsigargin apparently enhances the effect of apoptotic induction of 5 FU on MCF 7 cells, it is worthy of being further studied.

Objective An intensive analysis on the recent advances in cardiovascular medicine both in China and abroad was made,so as to provide an orientation of developing cardiovascular medicine in the future.Methods Literature of cardiovascular medicine published domestically and abroad in recent 5 years was reviewed.The advances in the use of new drug and techniques,clinical diagnosis and therapeutic methods were analyzed.Results Cardiovascular medicine in basic,clinical and practical fields had made rapid advances in the military institutions lately,especially in the interventional diagnosis and treatment of coronary heart disease,arrhythmia and peripheral vascular disease,and in the treatment of hypertension,where outstanding successes had been achieved.Conclusions The general level of cardiovascular medicine of military medical establishments could be considered foremost in our country.During the coming years of "the 12th Five-Year Plan",the emphasis of research work should be placed upon the medical care,basic and clinical research of common cardiovascular diseases existing in military officers and soldiers,such as coronary heart disease,peripheral vascular disease and arrhythmias,both in peace time and military conflicts.
4.Effect of raised intracellular free calcium on the expression of Caspase-3,Bax and Bcl-2 in human breast cancer MCF- 7 cell lines

Objective To study the effect of rising [Ca 2+ ]i on the expression of Caspase 3, Bax and Bcl 2 in human breast cancer MCF 7 cell lines. Methods After the MCF 7 cells treated with different dose of thapsigargin(TG), intracellular calcium concentrations in MCF 7 cells were measured by calcium sensitive fluorescence indicator Fura 2/AM technique; and immunohistochemical method was used to detected the expressions of Caspase 3, Bax and Bcl 2. Results Bax was over expressed and Bcl 2 was down expressed in a [Ca 2+ ]i concentration dependent way;nevertheless, the expression of Caspase 3 was negative in all the TG treated cells. Conclusions The increase of intracellular calcium concentration may have an inductive effect of apoptosis on MCF 7 cells, which is independent to Caspase 3.

The freshmen column in 39 985 Projectacademic libraries was investigated , which showed that 15 out of the 39 985 Projectacademic libraries have established freshmen column on their library website .Its posi-tion, contents and forms were analyzed.The typical freshmen column in libraries of Qinghua University, Shanghai Jiaotong University , and Sun Yat-sen University was described with suggestions put forward for improvement of its position, contents and forms.

7.Risk factors of anastomotic leakage and its significance on the survival of rectal cancer patients after low anterior resection

Objective To identify risk factors for anastomotic leakage after low anterior resection (LAR) in rectal cancer patients and study its impact on patients'long-term prognosis.Methods Chnical data were analyzed from 720 patients who underwent low anterior resection(LAR) for rectal cancer between 1996 and 2006.Results Anastomotic leakage after anterior resection occurred in 44 cases(6.1%).The median time of anastomotic leakage after operation was 5.6 days.Muhivariate analysis showed male patients.history of preoperative chemoradiation therapy,diabetes,cancer distance from anus less than 7 cm and hand-sewed anastomosis were independent risk factors predisposing anastomosis leakage (P<0.05).Tumor local recurrence rate was 13.6% in patients suffering from leakage and 5.9% for those without leakage (χ2= 4.116,P<0.05).The distant metastasis rates were 25.0 and 14.1 percent for the leakage and noaleakage groups,respectively(χ2=3.943,P<0.05).The survival rates were 56.8 and 72.5 percent in the leakage and nonleakage groups,respectively(χ2=4.979,P<0.05).Conclusion Sex,preoperative chemoradiation therapy,diabetes,cancer distance from anus less than 7 cm and hand-sewed anastomosis were found to be independent risk factors for anastomotic leakage after rectal cancer surgery.Anastomotic leakage was associated with poor prognosis.

By studying medical works during the Sui and Tang dynasty,we discussed the factors that doctors of that period believed to affect the dosage of medicine.We found that doctors of that period developed the dosage theory in terms of compatibility of medicines,onset time a disease,dosage form of medicine,constitution and age of patients,and toxicity of medicine.

Objective To investigate the recent studies on the biocharacters of keratin family (e.g. genetic mutations and abnormal expressions) and their relationships with the malignant tumors. Methods The literatures of recent years on the biocharacters of keratin family (e.g. genetic mutations and abnormal expressions) and their relationships with the malignant tumors were reviewed. Results Keratin family is a kind of structural proteins in cell which plays an important role in cytomechanics and regulates cell-cycle. The mutations of keratin genes (mRNA) or the overexpression of keratin proteins would interfere with the order of cell-cycle or the integrity of cytomechanics, and lead to some diseases and malignant tumors finally. Conclusion The studies on biocharaters of keratin family (e.g. genetic mutations and abnormal expressions) are helpful in the diagnosis, staging and the evaluation of prognosis of some diseases and cancers, e.g. liver cirrhosis, breast cancer, rectum carcinoma, etc.
10.Effects of hydroxyethyl starch on leukocyte activation and vascular permeability in endotoxic rats

Objective To investigate the effects of hydroxyethyl starch(HES 130/0.4)on leukocyte activation,mast cell degranulation and vascular permeability in rat mesentery during the early phase of endotoxemia.Methods Thirty-six male Wistar rats weishing 200-250 g were randomly divided into 3 groups(n=12 each):Ⅰ normal control group received only normal saline(NS);Ⅱ LPS group and Ⅲ HES group received LPS 2 mg/kg iv followed by iv HES infusion at 16 ml·kg-1·h-1 for 60 min(HES group)or equal volume of NS(LPS group).The animals were anesthetized with intramuscular 20% urethane 1 ml/100 g.The abdomen was opened and the mesentery of small intestine was pulled out and placed on transparent observation plate at constant temperature and moisture and examined under microscope for microcirculatory changes.Leukocytes rolling along,adhering to and emigrating from mesenteric venules,mast cell degranulation and FTTC-albumin effux from venules were examined before(baseline)and during the 90 min after LPS administration.The expression of CD11b and CD18 on the leukocytes was determined using flow cylometry.Results The number of leukocytes rolling along,adhering to and emigrating from venules and degranulated mast cells were signitlcanfly increased in LPS group as compared with control group.These LPS-induced changes were significantly inhibited in HES group.The albumin effiux Was enhanced and CD11b/CD18 expression upregulated in both LPS and HES groups.Conclusion Hydroxyethyl starch(130/0.4)can ameliorate the increased vascular permeability and microcirculatory disturbance in rat mesentery during the early phase of endotoxemia.
