Conclusion The clinically isolated drug resistant Pseudomonas aeruginosa strains are increasing year by year. Therefore, clinical doctors should choose antibiotics rationally in dealing with the P. aeruginosa infected patients according to drug resistance test.

Objective To study the effect of the magnetic field-treated water on metabolism of monoaminergic nerve transmitters in mice. Methods Twenty healthy aged Kunming mice were divided into a control group and an experimental group randomly. The mice in the control group were given tap-water and those in the experimental group magnetic field-treated water for 30 days. The content of monoaminergic nerve transmitters in brain of mice in the two groups were determined. Objective To evaluate the effects of penehyclidine hydrochloride (PHC) on the expression of Toll like receptor-4 (TLR4) and nuclear factor kappa B (NF-κcB) in a rat model of sepsis-induced acute lung injury (ALI).Methods Seventy-two adult male Sprague-Dawley rats,weighing 180-220 g,were randomly divided into 3 groups (n =24 each) using a random number table:shame operation group (group S),ALI group and PHC group.Sepsis-induced ALl was induced by cecal ligation and puncture in anesthetized rats.In group PHC,PHC 0.45 mg/kg was intramuscularly injected immediately after cecal ligation and puncture.At 6,12,24 and 36 h after ligation,the broncho-alveolar lavage fluid (BALF) was collected for detection of TNF-α and IL-6 concentrations and the lungs were removed for determination of the expression of TLR4 (using RT-PCR and Western blot) and NF-κBp65 (using Western blot) in lung tissues and for microscopic examination.The pathological changes of lungs were scored.The wet to dry lung weight (W/D) ratio was calculated and the activity of myeloperoxidase (MPO) in lung tissues was measured.Results As compared with S group,the IL-6 concentrations in BALF at 6,12 and 24 h after ligation,TNF-α concentration in BALF at 6 and 12 h after ligation,and the expression of TLR4 and NF-κBp65,pathological scores,W/D ratio and MPO activity at each time point were significantly increased in ALI group (P ＜ 0.05).Compared with ALI group,the TNF-α concentration in BALF at 6 and 12 h after ligation,and IL-6 concentrations in BALF,the expression of TLR4 and NF-κBp65,pathological scores,W/D ratio and MPO activity at each time point were significantly decreased in group PHC (P ＜ 0.05).Conclusion PHC inhibits NF-κB activation and decreases inflammatory responses through blocking TLR4 expression in lung tissues,thus attenuating sepsis-induced ALI in rats.

Breast cancer is the most common malignant tumor in female,and the recurrence and metastasis is the leading cause of death.Previous image evaluation of breast cancer could not detect the tumor at its early stage timely.The circulating tumor cells in the peripheral blood are like the seeds buried in the body,which cause the later recurrence and metastasis.So we can detect circulating tumor cells in peripheral blood to evaluate breast cancer progression.In present study,circulating tumor cells cau be implied more than three ways in clinical application of breast cancer.They could be used to monitor the therapeutic effect in breast cancer therapy timely,to analyze molecular classification of breast cancer and conduct clinical drug sensitivity tests.Although the value of circulating tumor cells in breast cancer has been widely recognized,many factors limited their application in clinical.Thus,we conduct this review about the current progress of circulating tumor cells in breast cancer.

OBJECTIVE To investigate the methylation status and mRNA expression of the estrogen receptors(ERs) gene promoter in acute leukemic patients and detect the protein expression in leukemia cell lines with or without treatment of 5'-aza-Dc.And to find out the possibility of promoters methylation profile of estrogen receptor gene as an early diagnosis biomarker in leukemia.METHODS With RT-PCR and MSP,evaluating ERs mRNA expression and status of methylation in 40 acute leukemia patients without treatment.With Western-blot,detecting protein expression in leukemic cell lines with or without treatment of 5-azaDc.RESULTS The protein expression was significantly enhanced in all of leukemic cell lines with 5'-Aza-Dc.ER?-A was inactivated and specifically methylated(97.5%;39/40) in most of the acute leukemic patients.CONCLUSIONS The promoter ER?-A is inactivated and specifically methylated(97.5%;39/40) in most of the acute leukemic patients.This study may provide a new direction method to study the pathogenic mechanism of leukemia,and indicates that ER?-A methylation could be a potential reference marker for leukemia diagnosis.

OBJECTIVE To develop new 2?5 type of piezoelectric immunoglobulin microarray immunosensor for determination of immunoglobulin.The energy converters are 10MHz AT-cut quartz crystals with gold-coated(electrodes).The monoclonal antibodies of immunoglobulin are immobilized onto the surfaces of crystals gold(electrodes) by staphylococcal protein A(SPA).METHODS The standard substance and serum were detected to find the detection time,temperature and specificity of the immunosensor.RESULTS The piezoelectric quartz(crystal) immunoglobulin microarray immunosensor could complete the detection in 15 min without nonspecific(response).Under the optimized conditions,the experimental results showed that the piezoelectric immunosensor had good(response) to immunoglobulin whose frequency shifts were linearly dependent on immunoglobulin (concentration) in different range.The piezoelectric microarray immunosensor had been used to detect(immunoglobulin) in serum,the analytical results given by this method were in satisfactory agreement with those given by traditional (immunoassay),with correlation coefficient of 0.94,0.94,0.94,0.92 and 0.91.(CONCLUSIONS) Piezoelectric microarray immunosensor for the determination of immunoglobulin is of high(sensitivity),high specificity,high analysis speed,unnecessary labelling,simple operation,real-time detection,(repeated) use,etc.It is suitable for (detecting) of immunoglobulin and should be used for clinical detection.
