Objective:To investigate the influential factors on the formation of mammospheres from MCF-7,MDA-MB-231 and primary breast cancer epithelial cells,and to detect the proportion of CD44+/CD24-/low among them.Methods:MCF-7,MDA-MB-231 and primary breast cancer epithelial cells were cultured in suspension to generate primary mammospheres.After 7 days of cultivation,the proportion of cancer stem cells was measured in cells derived from mammosphere cells or monolayer culture cells by flow cytometry.Results:MCF-7 had the highest efficiency of mammosphere formation(2.1%?0.3%),while MDA-MB-231 cells hardly had any mammosphere formation,and no mammosphere formation was observed in breast cancer epithelial cells.MCF-7 tended to adhere under stem cell culture conditions without B27.Flow cytometry analysis indicated that,compared with 2.0%?0.1% for the MCF-7 monolayer culture cells,11.8%?0.3% and 8.2%?0.8% of the MCF-7 mammosphere cells from MCF-7 under different conditions were CD44+/CD24-/low(P
2.Effects of the Acute Sodium Salicylate Injection on the Distortion Product Otoacoustic Emissions of Awake Guinea Pigs

Objective To study the change and the characteristics of distortion product otoacoustic emissions (DPOAE) of awake guinea pigs with the acute injection of sodium salicylate, and to investigate the ototoxicity of sodium salicylate to OHC. Methods With CELESTA 503 otoacoustic emission analyzer, DPOAE including DP-gram and DP-I/O function of awake guinea pigs were recorded.DPOAE were measared before and 2,4,8 h after acute injection of sodium salicylate or saline respectively. The data were analysed with SPSS 10.0.Results Acute sodium salicylate injection mainly caused the DPOAE amplitude and the I/O slope to reversibly decrease and increase respectively. Objective To explore the inhibition effects of epidermal growth factor receptor (EGFR) antagonist monoclonal antibody cetuximab (C225) on breast cancer stem cells in human breast cancer cell line MCF-7.Methods The effects of C225 on the proliferation of breast cancer cell line MCF-7 were detected by MTT assay. MCF-7 cells were cultured to generate primary mammospheres, and were divided into control group, C225 group, epidermal growth factor (EGF) group and EGF + C225 group according to whether or not the culture media contained exogenous EGF and C225.Thirteen days after culture, the volume and number of mammospheres of these four groups were observed, and mammosphere-forming efficiency ( MFE) was calculated. The percentages of CD44~+ CD24~- cells in mammospheres of these four groups and in routinely cultured MCF-7 cells were determined by flow cytometry. Results The inhibition rate on MCF-7 cells increased with the concentration of C225.Compared with control group, the volume of mammospheres in C225 group significantly decreased, and MFE and percentages of CD44~+ CD24~- cells in mammospheres significantly decreased [(0.61 ±0.04)% vs (1.44±0.09)%, P<0.01; (3.50±0.29)% vs (9.07 ±0.52) % , P<0.01]. Compared with EGF group, the volume of mammospheres in EGF + C225 group significantly decreased, and MFE and percentages of CD44~+ CD24~- cells in mammospheres significantly decreased [ (0.68 ± 0.04) % vs (1.61 ± 0.05) % , P < 0.01; (4.00 ± 0.58) % vs (10.47 ± 0.79) % , P < 0.01]. The percentage of CD44~+ CD24~- cells in routinely cultured MCF-7 cells was (2.03 ±0.15) % , and was significantly different from those in EGF group and control group (P < 0.01). There was no significant difference in volume of mammospheres, MFE and percentage of CD44~+ CD24~- cells in mammospheres between EGF group and control group (P >0.05). Conclusion C225 has significant inhibition effects on CD44~+ CD24~- cells in MCF cells.
4.The value of CT combined electronic level guided percutaneous transthoracic needle biopsy in diagnosis of small pulmonary nodules

Objective To discuss the clinical value of CT combined electronic level guided percutaneous transthoracic needle biopsy in diagnosis of small pulmonary nodules.Methods 44 cases of lung mini-nodule underwent CT combined electronic level guided percutaneous transthoracic needle biopsy.After operation the tissue was sent for pathologic examination.The nodules were divided into two groups,including Group A (diameter 0.5-1.0 cm,N=10) and Group B (diameter 1.1-2.0 cm,N=34),according to the different sizes of diameter.The excision of postoperative pathological and diagnosis result of clinical diagnosis and treatment of more than 12 months follow up were taken as the final result.The sensitivity,specificity,veracity and complications were compared and analyzed using paired x2 test,taking P<0.05 as significant difference.Results (1)The arrival rate of target puncture were 100 % in 44 cases,with the sensitivity rate of 90.9%,specificity rate of 100 %,veracity rate of 88.6 %,positive predictive value of 97.5 %,and negative predictive value of 100 %.The sensitivity,specificity and veracity of Group A were 77.8%,100% and 70.0% respectively.And the sensitivity,specificity and veracity of Group B were 94.1%,100% and 94.1% respectively.The sensitivity,veracity of Group A was higher than Group B (P<0.05),while there was no difference in specificity.(2)Complication:the incidence rate of pneumothorax,pulmonary bleeding symptoms and hemothorax were 9.1%,11.4%,2.3%,respectively.The incidence rate of total complication was 22.7%,and all be cured after the treatment.The lesions with emphysema around is a risk factor for pneumothorax and pulmonary bleeding symptoms (x2 =11.2,10.2,P<0.05).However,the position,size and deepness was not correlated with the complications mentioned above (P >0.05).Conclusion CT combined electronic level guided percutaneous transthoracic needle biopsy is a safe,effective and accurate diagnostic method for lung mini-nodule lesion.
5.A Traditional Chinese Medicine Research on the Principal Antidepressive Effect in Kaixin Jieyu Prescription Based on Uniform Distribution

Objective To pick up the Chinese materia medica with principal antidepressive effect in Kaixin Jieyu prescription based on uniform distribution. Methods 11 groups of concerted application with different dosages of six simplex Chinese materia medica (X 1-X 6) of Kaixin Jieyu prescription were arranged by the designlist and sustentacular table of U11(1110). Positive and negative control groups were also formed. Mice were randomized into 13 groups according to body weight and fed with medicine 14 d and estimated by tail suspension test, forced swim test and autonomic activities test. Results Relationship of percentage of immovability time (Y 1) in the tail suspension test and X 1-X 6 were analyzed with multivariant non-linear regression analysis and resulted in Y 1=1.133 1-0.842 3×X 1+0.438 2×X 3-0.712 9×X 6. It indicated that both Radix Ginseng and Fructus Aurantii Immaturus had obvious antidepressive function, and Radix Paeoniae Rubra may facilitate depression. Relationship of immovability time (Y 2) in forced swim test and X1-X6 were analyzed with multivariant non-linear regression analysis and resulted in Y 2=167.69-2.99×X 12-2.14×X 1-1.65×X 6. It also indicated that both Radix Ginseng and Fructus Aurantii Immaturus had obvious antidepressive function. Relationship of autonomic activities index (Y 3) in autonomic activities test indicated that X 1-X 6 had no obvious influence on Y 3 and antidepressive function of KaixinJieyu prescription was not concerned with excitability to the central nervous system. Conclusion Radix Ginseng and Fructus Aurantii Immaturus were the Chinese materia medica with principal antidepressive effect in Kaixin Jieyu prescription.
6.Barium-based Colonic Transit Detection and Individualized Treatment for Chronic Constipation

Background:Chronic constipation is a major cause of impaired quality of life in modern society. Reasonable and effective management of chronic constipation could be achieved based on the principle of evidence-based medicine and the modern concept of constipation,and this is a challenge facing the clinicians. Aims:To investigate the role of barium-based colonic transit detection in diagnosis and treatment of chronic constipation. Methods:Fifty patients with chronic constipation from Apr. 2013 to Oct. 2014 at the First Hospital of Shanxi Medical University were recruited and randomly allocated into two groups,control group and individualized treatment group. Patients in individualized treatment group received 20 barium markers orally and abdominal plain radiography was performed 48 and 72 hours later,respectively for calculating the colonic transit index. According to the type of colonic transition and the characteristics of colonic motility estimated by colonic transit index and clinical manifestations,an individualized therapeutic regimen was formulated and the efficacy was evaluated. Patients in control group were treated empirically according to the clinical manifestations. Results:Mosapride and lactulose or polyethylene glycol were administered orally in control group;when abdominal pain or abdominal distension was predominant,pinaverium bromide or trimebutine was used respectively instead of mosapride. Barium-based colonic transit detection revealed that 9 patients in individualized treatment group were slow transit constipation,6 were outlet obstructive constipation and 8 were the mixed type. After 2 weeks of empirical or individualized treatment,the defecation rates of the two groups were 24. 0%(6 / 25)and 52. 2%(12 / 23)within 24 hours and 64. 0%(16 / 25)and 87. 0%(20 / 23)within 48 hours,respectively(P all < 0. 05). Conclusions:Barium-based colonic transit detection is a simple,economical and practical modality for guiding the individualized treatment in patients with chronic constipation.
7.Effects of Kaixin Jieyu Pills on monoamines and BDNF in rats with vascular depression

AIM To aim at investigating the function of Kaixin Jieyu Pills (Ginseng Radix et Rhizoma,Bupleuri Radix,Morindae officinalis Radix,etc.) to vascular depression and their mechanism of action.METHODS A vascular depression model of rat was established by chronic unpredictable mild stress and separation after ligation of the bilateral common carotid arteries.The rats were treated with Kaixin Jieyu Pills and fluoxetine hydrochloride (FLU) for 28 days,respectively.The sucrose preference test and open-field test were performed.Cerebral perfusion was investigated with a Perfusion Speckle Imager.The monoamines including serotonin,dopamine and noradrenaline,and brain-derived neurotrophic factor (BDNF) were determined with ELISA and Western bolt,respectively.RESULTS Kaixin Jieyu Pills significantly increased sucrose preference,moving distance and cortical blood flow.Kaixin Jieyu Pills could upregulate,to different extents,the expressions of monoamines,including serotonin,noradrenaline and dopamine,and BDNF.Kaixin Jieyu Pills had a function similar to FLU in behavior,upregulating monoamines and BDNF,but it is superior to FLU in cortical blood flow.CONCLUSION Kaixin Jieyu Pills has the advantage of reducing the depression-like behavior and improving cerebral hypoperfusion,which might be mediated by the upregulation of the serotonin,noradrenaline and BDNF.
8.Effect of Kai-Xin Jie-Yu Prescription on Behavior and Serum Inflammatory Cytokines in Rat Model of Depression

This study was aimed to explore the antidepressant effect of Kai-Xin Jie-Yu (KXJY) prescription on depressive model of rats,and to explore the level of inflammatory cytokines in peripheral blood.According to sucrose preference and weight,male SD rats were randomly divided into the normal group,model group and traditional Chinese medicine (TCM) group.The model of depression was established by chronic unpredictable mild stress and separation for @@6 weeks.Body weight,sucrose preference and forced swimming test were used to evaluate the depressive state.The levels of IL-1β,IL-2,IL-6 and TNF-α in the serum of rats were measured by ELISA.The results showed that compared with the normal group,the weight,sucrose preference and total drinking water of rat model gToup were significantly reduced.The immobility time of forced swimming was significantly prolonged.The level of IL-1β,IL-2,IL-6 and TNF-α expression in serum was significantly increased.There was no significant difference between the body weight of rats in the model group and TCM group.The total amount of drinking water and sucrose preference were significantly increased.The immobility time of forced swimming was reduced.Levels of IL-1 β,IL-2,IL-6 and TNF-α expression were decreased significantly.It was concluded that KXJY prescription can improve depression symptoms in depression model of rats.It may be related to regulate the expression level of serum inflammatory cytokines.
9.Expression of miR-665 and Bioinformatic Analysis on Its Target Genes of Kai-Xin Jie-Yu Decoction Based on Depression Model Rats

This study was aimed to detect the expression of miR-665 in the hippocampal of depression rat model treated with Kai-Xin Jie-Yu (KXJY) decoction and bioinformatic analysis of target genes of miR-665,in order to investigate the role of miR-665 in the pathogenesis of depression and the antidepressant mechanism of KXJY decoction.The rat model of chronic stress depression was established and then treated with KXJY decoction for 42 days.The total RNA from hippocampus tissues was extracted.And the relative expression of miR-665 in hippocampus of rats in each group was detected by RT-qPCR.TargetScan and microRNAorg databases were used to predict target genes for miR-665.DAVID database was used to classify GO function and to analyze KEGG signaling pathway of target genes.The results showed that compared with the normal group,the expression level of miR-665 in hippocampus of the model group was significantly higher with significant difference (P < 0.01).Compared with the model group,the expression level of miR-665 in hippocampus of Chinese medicine group and western medicine group decreased significantly with significant difference (P < 0.01).The biological functions of miR-665 target genes were mainly concentrated in the response to organic substance.The signal pathway was mainly concentrated in N-Glycan biosynthesis.It was concluded that miR-665 may be involved in the pathological process of depression,by correcting the abnormal expression of miR-665,which may be one of the antidepressant mechanisms of KXJY decoction.Through the analysis and prediction of the target genes,it provided a certain direction and theoretical basis for further study on the specific mechanism of KXJY decoction intervention on miR-665.
10.Effects of self breathing exercise on heart rate and systolic blood pressure in adults:a Meta-analysis

Objective To review the effects of self breathing exercise (SBE)on heart rate and systolic blood pressure in adults.Methods We electronically searched databases including CENTRAL (Cochrane Central Register of Controlled Trials),MEDLINE,EMbase,PEDro,OVID,CNKI,VIP,WanFang Data and CBMfrom the establishment of database to November 2014 to identify randomized controlled trials (RCTs)of VBE and references.Two researchers assessed data according to PEDro,and the effective data was used by Meta analysis which met the including criteria.The statistical analysis used RevMan 5.0 software.Results A total of 5 RCTs were included.The results of meta-analysis showed that the effects of VBE on heart rate and systolic blood pressure in adults was statistically significant compared with control group (P <0.05).Conclusions The self breathing excise can decline adult heart rate and systolic blood pressure with low adverse effects,and it is easy to operate and is a favorable physical therapy method,which can be recommended to self health care and disease rehabilitation nursing.
