1.Effects of TNF? and IL-10 on cytomegalovirus infection in human embryonic lung fibroblasts

AIM: To study effects of tumour necrosis factor alpha(TNF?) and interleukin-10(IL-10) on human cytomegalovirus AD169 (HCMV AD169) infection in human embryonic lung fibroblasts (HEL),and the ability of the infected HEL to produce TNF?. We have attempted to understand the effect of cytokines in the immune of HCMV infection. METHODS: TNF??IL-10 were added separately with different concentrations before 24 h HCMV infection of HEL to study the effect of TNF? and IL-10 on multiplication of HCMV. HCMV was incubated with HEL, TNF? in culture supernatants were measured at 4, 24, 48, 72, 96 h postinfection. The level of TNF? was measured by enzyme-linked immunosorbent assay( ELISA). RESULTS: The addition of TNF? with the concentration of 10-100 ?g/L or IL-10 with the concentration of 1-10 ?g/L before 24 h HCMV infection of HEL could remarkably inhibited the multiplication of HCMV in HEL. Level of TNF? didn't increase in infected cell supernatants. CONCLUSION: TNF? and IL-10 play an important role in the immune of HCMV infection.
2.Study on the Pharmacological Activity of Soybean Stem Soyasaponin

Objective To study the pharmacological activity of soybean stem soyasaponin. Methods Normobaric hyporia and exhaustive swimming in mice and osteoporosis rats model were used to observe the pharmacological effects of soybean stem soyasaponin. Results Soybean stem soyasaponin 150 mg/kg significantly prolonged the survival time in hyporia and the swimming time of mice and 100 mg/kg in rats improved the syomptom of osteoporosis models. Conclusion Soybean stem soyasaponin has obvious effect on anti-exhaustion and increasing tolerance of oxygen deficit in mice and significant therapy effect on osteoporosis rats.
3.Acute Phase Protein in Gynecological Patients with Nosocomial Infection During Chemotherapy

OBJECTIVE To investigate the value of acute phase protein in the diagnosis and therapy for nosocomial infection monitoring. METHODS The levels of serum C-reactive protein(CRP),serum amyloid A(SAA),?_1-acid glycoprotein(AAG),and ?_1-antitrypsin(AAT) were measured in 71 gynecological patients with nosocomial infection during chemotherapy,Thirty normal controls and 33 chemotherapy patient controls were detected by nephelometry and compared with white blood cell counts. RESULTS The nosocomial infection rate of chemotherapy patients was 16.7%.Main pathogens were Gram-negative bacteria(60.6%).Fungi infection rate was 19.7%.The most frequent hospital infection sites were respiratory tract,gastrointestinal tract and wound.Compared with controls,the levels of serum CRP,SAA,AAG and AAT were significantly higher in chemotherapy patients with bacteria infection(P

Experimental course of pathology is a very important part of pathology teaching.Motivation of students was promoted through improving the professional knowledge of teachers,using teacher-student interacted teaching methods,increasing network courscs,using multimedia in teaching and adding extracurricular practice in our university.In the meantime,it was successful and fruitful to use diversified evaluation systems to assess the comprehensive experimental capacity of students,which can provide experiences in reform of pathological experimental teaching.
5.Analysis of psychological tests on newly employed staff in a Chinese comprehensive hospital

Objective This study targeted on 298 newly employed staff in Guang'anmen Hospital,China Academy of Chinese Medical Sciences,who had finished the Minnesota Multiphasic Personality Inventory (MMPI).A retrospective study has been approached to investigate the mental states of the new employers in the Chinese comprehensive hospital.Methods SPSS statistics software 11.5 was used to do descriptive statistical analysis.Results Scores of all factors show no difference compared to the various norm groups studied.Most of the new employers represent calm and gentle personalities.They are confident,optimistic,independent,gentle,kind,decent,rational and objective,have self-control and self-discipline,barely have no doubt about their physical health,trust others; they are mature,able to maintain good interpersonal relationships and to adapt external pressure; they do not harm themselves psychologically; they are calm,stable and pragmatic.Conclusion Subjects are somehow over reporting or exaggerating the prevalence of being nice,which could be subconscious or intentional.However,this does not have impacts on reliability and validity.MMPI is a relatively comprehensive psychometric scale for employment.

8.Association of MK2 and ZFP36 gene polymorphisms with high density lipoprotein cholesterol in Uygur population from Hetian area of Xinjiang.

OBJECTIVETo assess the association of genetic polymorphisms of mitogen-activated protein kinase activated protein kinase 2 gene (MK2) and zinc finger protein 36 gene (ZFP36) with high density lipoprotein cholesterol (HDL-C) in Xinjiang Urgur population.
METHODSNine hundred thirty Uygur individuals were randomly recruited from Hetian area. Single nucleotide polymorphisms (SNP) in MK2 gene (rs44890 and rs45514798) and ZFP36 gene (rs251864 and rs3746083) were determined with Taqman-PCR. All subjects were investigated with questionnaire, physical examination and measurement of lipid levels and plasma tumor necrosis factor-alpha.
RESULTS(1)In Uygur men younger than 50 years old, SNP rs45514798 was associated with HDL-C [dominant model P=0.054, OR(95%CI)0.261(0.082-0.833) after age, smoking, drinking, abdominal circumference, waist/hip ratio and body mass index and tumor necrosis factor were controlled]. (2) For males younger than 50 years old, the concentration of total cholesterol, HDL-C, low density lipoprotein cholesterol were different in dominant model of rs45514798(P< 0.05). Female: total cholesterol, low density lipoprotein cholesterol were different in dominant model of rs45514798(P< 0.05). (3) The distribution of genotype of ZFP36 gene did not differ significantly between the low HDL-C groups and the control group.
CONCLUSIONMK2 gene rs45514798 polymorphisms may be associated with HDL-C in Uygur men younger than 50 years old from Hetian area of Xinjiang. ZFP36 gene is not associated with HDL-C in Uygur people from Xinjiang.

9.Serum calcium is a novel parameter to assess metabolic syndrome in endometrial carcinoma.

OBJECTIVES: To investigate the distribution of serum calcium and the relationship between serum calcium and serum metabolic parameters in endometrial carcinoma (EC) patients. METHODS: Retrospective assessment of patients diagnosed with endometrial cancer from Peking University People's Hospital from 2004 to 2009. Clinical characteristics as well as pretreatment serum calcium, albumin, fasting plasma glucose (FPG), serum triglycerides (TG), high-density lipoprotein (HDL), low-density lipoprotein (LDL), and total cholesterol (TC) value were extracted from patient records. Serum calcium was corrected for albumin. Unpaired t test and analysis of covariance were used to compare serum calcium among categorical variables. Simple correlation analyses and partial correlation analyses were used to assess the associations between serum calcium and continuous variables. RESULTS: Two-hundred twenty patients were included in this study. After adjusting for confounders, postmenopausal patients had higher total serum calcium (p=0.002) and albumin-corrected serum calcium (p=0.012) than premenopausal patients, endometrioid endometrial carcinoma (EEC) patients had higher total serum calcium than non-endometrioid endometrial carcinoma (NEEC) patients (p=0.037). Significant positive correlations were found between total serum calcium and FPG (p=0.017), TG (p=0.043), HDL (p=0.042), LDL (p < 0.001), and TC (p < 0.001) after adjusting for multiple variables, and the corrected serum calcium showed no significant correlation with metabolic parameters. CONCLUSION: Total serum calcium might be a more sensitive parameter for metabolic syndrome in endometrioid endometrial cancer patients than lipids.

10.Phenotypic and functional properties of in vitro-induced Helios+regulatory T cells

Objective To investigate the phenotypic and functional properties of in vitro-induced He-lios+regulatory T cells(Helios+iTreg) and to analyze the differences between Helios+iTreg and Helios+thymic-derived natural Treg (Helios+nTreg). Methods CD4+CD25- effector T cells (CD4+CD25- Teff) that were separated from healthy subjects were cultured with anti-CD3 and anti-CD28 antibodies and stimulated with IL-2 and TGF-β to induce the generation of Helios+iTreg. Flow cytometry and real-time PCR were respectively used to analyze the differences in phenotype and CpG methylation in Treg-specific demethylated region (TSDR) be-tween Helios+iTreg and Helios+nTreg derived from the same donor. Results CD45RA was highly expressed on Helios+iTreg, but not on Helios+nTreg. Additionally, Helios+iTreg expressed significantly higher levels of CD127,ICOS and PD-1,but lower levels of CXCR3,CCR4 and CD25 than Helios+nTreg did. IFN-γ and IL-2 that expressed by Helios+iTreg were more than those by Helios+nTreg. It was also found that the level of CpG methylation in TSDR was much higher in Helios+iTreg than in Helios+nTreg. Conclusion Both nTreg and iTreg expressed Helios,but the phenotypic and functional properties of the two Treg subsets were different. It was suggested that Helios+iTreg and Helios+nTreg might play different roles in the immune system.
