Journal of China Medical University 2016;45(7):596-600
Objective To investigate the influence of different kinds of post?core materials on fracture resistant ability of different walls during weakened root restoration. Methods Forty?eight human single root mandibular first premolars were randomly assigned to two groups(groupⅠwas repaired with quartz?fiber post?core and mental crown ,and groupⅡwas repaired with Co?Cr cast metal post?core and mental crown). Each main group was then divided into four subgroups of 6 specimens. All samples were tested on the universal test machine. We observed the fracture load and the fracture mode of the specimens. Results Cr?Co ally post?core group exhibited significantly higher resistance to fracture than quartz fiber post?core in the same number weakened root walls(P<0.05). With the increase in the quantity of weakened root walls ,the fracture resistance was significant decreased in the same post?core material(P<0.05). The possibility of repairable fractures in groupⅠwas significantly higher than that in groupⅡ(P<0.05). Conclusion Cr?Co alloy post?core can bear a higher bite ,but quartz fiber post?core has the most favorable fracture modes.

Objective To evaluate the usefulness of DiversiLab system for genotyping of Acinetobacter baumannii. Methods Fifty-eight non-duplicated clinical Acinetobacter baumannii isolated from 15 cities in China in 2005 were typed by rep-PCR-based DiversiLab system. The results were compared with those of PFGE and multilocus sequence typing. Simpson's index of diversity was used to compare the discriminatory power among DiversiLab system, PFGE and MLST.Results Fifty-eight Acinetobacter baumannii isolates were differentiated into 5 clusters and 25 unique types by DiversiLab system. MLST identified 35 distinct sequence types, which fell into one clone complex of CC22 and 35 singletons, while PFGE resolved 5 pulsotypes and 34 unique types.Simpson's diversity indices for DiversiLab system, MLST and PFGE were O.876, O.944 and 0.961, respectively. Conclusions The discriminatory power of DiversiLab system is lower than that of PFGE and MLST.But as a simple, fast and reproducible typing method, it could be used as a first-line typing tool for the analysis of a large number of isolates.

Objective To compare the difference of the microleakage repaired by direct method and indirect method of the funnel shaped root canal in the anterior teeth. Methods A total of 22 middle teeth at the similar size were collected, prepared into funnel root canal after finishing the root filling. The teeth were divided into A and B groups randomly, and there were 11 test pieces in each group. The root canal was repaired by direct method in group A and indirect method in group B. After 500 thermal-cold cycling and 240 000 dynamic cyclic loading, one tooth was selected in each group randomly. The microstructure were observed by scanning electron microscope (SEM). One piece of each group was randomly selected and the image of the core and the dentin was scanned by electron microscope. The remaining 20 pieces were dyed by India ink, dealt with transparent, compared and observed under the microscope. The degree of microleakage was recorded and analyzed in two groups. Results Under electron microscope, a gap can be observed in the bonding interface of group A. The gap was significantly bigger than that of group B. The leakage was significantly serious in group A than that in group B (P<0.05). Conclusion The level of microleakage of anterior teeth with flared canals, which is restored by fiber post core with the indirect method, is significantly better than that with the direct method. The indirect method has a good edge seal action and is more suitable for clinical application.

Objective To perform mutation analysis of survival motor neuron gene 1 (SMN1 in two spinal muscular atrophy (SMA) patients and their parents to evaluate the effects of the two SMN1 gene mutations on the transcript levels of the gene and preliminarily predict their effects on the structure and function of SMN protein. Methods Mutation analysis of SMN1 gene was carried out by multiplex ligationdependent probe amplification, reverse transcript-polymerase chain reaction (RT-PCR) and cloning sequencing.Transmission of the mutations was confirmed by the mutation analysis in patients' parents. The full-length SMN1 (SMN1-fl) transcript levels of the patients carrying these subtle mutations were detected using quantitative RT-PCR.Results The two patients were diagnosed as SMA Ⅱ and SMA Ⅲ.They carried p.Val19GlyfsX21 and p.Ala2Gly SMN1 mutations in SMN1 gene, respectively. Both of the two mutations were originated from their fathers.Compared with the healthy individuals (23.5 ± 4.9),the two patients had a significant reduction in the level of SMN1-fl transcripts (t =3.322,P =0.011 (p.Ala2Gly) ;t =6.964,P =0.000 (p.Val19GlyfsX21)).However,compared with the healthy carriers (14.1 ±4.5),the patient with p.Ala2Gly mutation had no significant reduction in the level of SMN1-fl transcripts (13.9 ±3.6,t =0.058,P =0.955) ; however, the patient with p.Val19GlyfsX21 mutation had a significant reduction (4.9± 2.4,t =3.725,P =0.004).Conclusions Two SMN1 gene mutations are identified in our study. The mutation p.Val19GlyfsX21 is a novel mutation and p.Ala2Gly is firstly reported in Chinese SMA patients.p.Val19GlyfsX21 may possibly lead to decreased SMN1-fl mRNA by nonsense-mediated messenger RNA decay,however,p.Ala2Gly has no obvious effects on the amount of the SMN1-fl transcripts, indicating that its deleterious effect may be occurring at SMN protein level or the function of SMN protein.

T,P=0.008; Genotype:P=0.031)of DISC1 gene were significantly associated with schizophrenia. The haplotypes constructed by these two markers were significantly associated with schizophrenia, such as AT(?2=7.065,P=0.008,OR=1.42,95%CI=1.10~1.83)and GA(?2=6.009,P=0.014,OR=0.80,95%CI=0.68~0.96).When the subjects examined with the positive and negative syndrome scale(PANSS),the risk haplotype AT was not significantly correlated with positive, negative, excitement, depression and cognitive impairment factors of PANSS. Conclusion:These findings provide further evidence for DISC1 as a predisposing gene involved in schizophrenia in the Chinese Han Population. However,no positive association is found between DISC1 polymorphisms with schizophrenia clinical symptoms.
