Objective Studying risk factors of carotid atherosclerosis in metabolic syndrome(MS),and the relative factors with progression of carotid atherosclerosis. Methods A total of 401 elders were included in the present study.MS was defined by the criteria of IDF(219 MS,182 NMS).All subjects were required to have blood pressure monitored.Venous blood was collected after an overnight fast.Total serum cholesterol,triglycerides,high-density lipoprotein cholesterol(HDL),low-density lipoprotein cholesterol(LDL),apolipoprotein B,lipoprotein a,high-sensitive C-reactive protein, and fasting glucose and insulin were tested;insulin sensitivity index and insulin resistant index were estimated.Uria sample was also collected to test microalbumiuria, creatine and Alb/Cr.Carotid atherosclerosis was determined by B-mode ultrasound, and number and diameter of plaques,if present, were measured to obtain an atherosclerosis score. Results The occurrence of carotid atherosclerosis was significantly higher in MS(P
2.The total homocysteine concentration in the patients with ischemic stroke or carotid atherosclerosis disease, and intervention study
Chinese Journal of Neurology 2000;0(05):-
Objective To investigate total serum homocysteine (tHcy) concentration in ischemic stroke patients and in carotid atherosclerosis patients in comparison with normal con trol elders and to evaluate whether providing folate and vitamin B 12 coul d decrease serum homocysteine concentration. Methods The sampl e in this study was obtained from the inpatients and outpatients of geriatric d epartment, Huashan Hospital. 160 patients were recruited and were divided int o two groups with smoking controlled. Thyroid gland abnormality and renal f a i lure were excluded. One group consisted of 71 male and 9 female ischemic strok e patients aged 73 years on average; There were 71 male, 9 female carotid ather osclerosis disease patients, aged 7 years on average in the second group. H y pertension and mellitus diabetes were controlled to avoid the influence of the f actors in these patients; the third group consisted of 72 male and 8 female norm al control elders, aged 71 years on average. Besides fast serum total homocyst e ine, every patient took laboratory and physical examinations, including plasma fo liate, vitamin B 12, free plasma glucose, creatinine, cholesterol, triglyce ride, blood pressure and blood mass index. Then each group was randomly subdivided in t o two groups in which one group was provided with folate 1.25 mg qod, and vitam in B 12 8?g qd w hile the other was not provited with folate and vitaminB 12. These patient s were reexamined after 10 months. Results The serum tHcy concentration was (16.1 ?5.3) ?mol/L in ischemic stroke group and (14.2?4.72) ?mol/L in caroti d athero sclerosis group compared with (12.5?3.0) ?mol/L in normal control elders ( P=0.000 1). The plasma foliate was low in ischemic stroke and carotid athe rosclerosis patients, and there were no significantly statistics dif ferences in the other examinations.Correlation analysis showed that tHcy was ne gatively correlated with foliate and vitamin B 12, r=-0.392,-0.36 r e spectively i n ischemic stroke group, r=-0.287,-0.339 respectively in carotid ath eroscleros is group, and r=-0.338,-0.227 in normal control group. The tHcy concent ration was lower in the groups provided with foliate and vitamin B 12 than that in the g roups without foliate and vitamin B 12 (P=0.000 1), even in the normal elders. Conclusions The tHcy concentration was higher in the patients with ischemic st roke and with carotid atherosclerosis. Folate supplement could moderately lowe r tHcy.

Objective Study on the clinic value of the application of two-dimensional and three-dimensional sonography on observing the fetal facial structure. Methods Applying Voluson 730 three-dimensional probe surface imaging mode and two-dimensional probe to make sonography on 16~48 weeks fetus in 1805 cases, emphasized on observing the fetal facial structure, saving the image information, making three-dimensional reconstruction and comparing the result of post partum and the result after induction of labor. Results In 1805 cases, applying two-dimensional and three-dimensional sonography to observe the facial structure of the fetus of 23~35 weeks, the display rate is 93.1% and 95.1% respectively, and there is no obvious difference between the two modes. The three-dimensional is higher than two-dimensional for the fetus less than 23 weeks, with the display rate 69.6% and 84.8% respectively, and the two-dimensional is a bit higher than three-dimensional for the fetus more than 35 weeks, with the display rate 90.4% and 88.7% respectively. One cleft lip case is excluded. Conclusion As long as mastering the optimum examination time and operation skill, there is no obvious difference between the display rate of two-dimensional and three-dimensional sonography on observing the fetal facial structure, the two modes can be complementary in the work to improve the accuracy of the diagnosis and be favorable for the prepotency.

Objective To explore the association of T cell immunoglobulin- and mucin-domain-containing molecule-3 (TIM-3) gene polymorphism and IFN-γ levels with the incidence of primary nephrotic syndrome (PNS) in children. Methods A case-control study was conducted and 21 Han patients with PNS were selected and included in case group. Meanwhile, 20 each from Mongolian and Han were selected and assigned into control group and at least three generations of their family members were from the same nationality. PCR-restriction fragment polymorphism analysis was used to detect and analyze single nucleotide polymorphisms of exon -574A/C in TIM-3 gene in PNS children and controls. Also the genotype and allele frequencies between the two groups were compared. Enzyme linked immunosorbent assay (ELISA) was used to detect the level of serum IFN-γ and its changes was analyzed. Results There was no significant difference in the distribution of genotypes (AA, AC, CC) of exon -574A/C in TIM-3 gene between the Han and Mongolian subgroups in control group (P=0.741). Neither did the allele frequency between the two groups (P=0.655). Compared with control group (Han and Mongolian), the frequencies of AA, AC and CC genotypes were 9.52%, 28.57% and 61.90% respectively in -574A/C loci of the Han nationality children with PNS. There was significant difference in genotypes distribution between the two groups (P=0.017). The frequency of C allele in PNS children of Han nationality was 76.2% which was higher than that in normal control group (50%), and the difference was statistically significant (P=0.005). Compared with A allele carriers, the risk of PNS in C allele carriers increased by 3.20 times (95%CI: 1.39~7.37). There were no significant differences in serum IFN-γ among the Han nationality with PNS, Han and Mongolian normal control groups (P>0.05). Conclusion The single nucleotide polymorphism of the exon -574A/C of TIM-3 gene may be related to the pathogenesis of PNS in children. In addition, IFN-γ is not associated with the incidence of primary nephrotic syndrome in children.

Chinese Journal of Gastroenterology 2016;21(3):138-143
Background:Recent studies have shown that long non-coding RNAs(lncRNAs)play important roles in carcinogenesis and cancer biology and the related context has attracted more and more attentions. PVT1,which encodes a lncRNA,is reported to be up-regulated and exhibit pro-oncogenic activity in a wide variety of human cancers. Aims:To investigate the expression of PVT1 in human pancreatic cancer cells and its effect on proliferation and apoptosis of HPAF-Ⅱ cells. Methods:One target siRNA against PVT1 was synthesized and transfected into HPAF-Ⅱ cells by using lipofactamine technique. PVT1 mRNA expression was detected by real-time PCR;capability of cell proliferation was examined by MTS and colony formation assays;cell cycle progression and apoptosis were measured by flow cytometry;and Western blotting was performed to determine the expressions of apoptosis-related proteins and proto-oncogene protein c-Myc. Results:The mRNA expression of PVT1 in several human pancreatic cancer cell lines, especially HPAF-Ⅱ cells was significantly higher than that in H6c7,a human immortalization normal pancreatic ductal epithelial cell line. Compared with HPAF-Ⅱ cells transfected with negative control siRNA or without transfection, silencing of PVT1 by siRNA-PVT1 resulted in remarkable reduction in cell proliferation, cell cycle G1 phase arrest, and notable apoptosis;meanwhile, the expressions of apoptosis-related proteins(cleaved-caspase-3 and cleaved-PARP)were up-regulated,the ratio for Bcl-2 / Bax was decreased, and the expression of c-Myc protein was down-regulated. Conclusions:LncRNA-PVT1 is highly expressed in human pancreatic cancer cell line HPAF-Ⅱ. It may affect the proliferation and apoptosis of HPAF-Ⅱ cells partially through regulating c-Myc expression.
