Chinese Journal of Medical Science Research Management 2016;29(2):-
Objective A survey was conducted to explore the relationship between research capacity and clinical practice in local public hospitals. Methods A third grade class A hospital from Yangzhou city was selected as an example;and 357 medical staff were surveyed through a designed questionnaires. Results Data collected by questionnaires was analyzed to understand the relationship between research ability and clinical practice as well as the awareness of the hospital staff. The results showed that the hospital staff realized that such relationship should be positively associated with each other, and mutual penetration. Conclusions Recommendations have been raised to promote research ability, and then enhance the quality of clinical practice.

Objective To recognize hereditary IgM glomerulopathy. Methods Three patients with hereditary IgM glomerulopathy were report first time in China. Genealogic investigation and light, immunofluorescence and electron microscopy were performed. Literatures concerned were reviewed. Results Kidney biopsies from three patients all revealed diffuse mesangial proliferative glomerulonephritis with intense IgM and C3 deposits. These siblings might be suffering from a hitherto undescribed clinico-pathological entity. Genealogic investigation enabled us to discover 10 additional affected members with clinical glomerulonephritis in the kindred of the family. The clinical picture was that of hematuria and/or proteinuria, meanwhile one patient died of uremia. Conclusion Genetic factors may be involved in the mechanism of IgM nephropathy .

Objective To explore the expression of the Pendrin gene (SLC26A4) and protein in multinodular goiter. Methods Thyroid tissues were obtained from 40 multinodular goiter patients undergoing surgery while the control group were obtained from 40 nomal thyroid tissues.RT-PCR was used to test SLC26A4 gene while western blot and immunohistochemistry were used to test Pendrin protein expression and distribution. Results SLC26A4 mRNA expression in multinodular goiter tissue was significantly increased in comparison with normal nodular tissues (t=2.663,P=0.011).Pendrin protein expression in multinodular goiter group was higher than that in normal tissue (t=2.286,P=0.026).The immunohistochemistry results showed that the Pendrin protein in multinodular goiter was mainly located in cytoplasm. There was positive expression in 24 patients (60%) in multinodular goiter group, while it was in 14 patients (35%) in the normal control group. The difference was significant (X2=5.013,P=0.025).Pendrin protein mainly expressed in cytoplasm in multinodular goiter tissue while it was mainly in cytomembrane in the normal control group. Conclusion SLC26A4 mRNA and its coding protein Pendrin expression are increased in multinodular goiter group, and mainly located in cytoplasm, indicating that iodide transporter function may be damaged when multinodular goiter occurs.

Objective:To investigate the significance of regulatory T cells in peripheral blood of chronic renal insufficientce. Methods:The peripheral blood samples were collected from 40 patients with chronic renal insufficient. The ratios ot CD4+T cell in lymphocyte and CD4+CD127-Treg and CD4+CD25+CD127-Treg in CD4+T were detected by flow cytometry. Results:The number of CD4+T in lymphocyte of chronic renal insufficient was higher than in healthy control group and there wasn’t significantly difference of the CD4+CD25+CD127-Treg ratios in CD4+T between chronic renal insufficience and healthy central. The ratio of CD4+T cells in lymphocytes of chronic renal insufficience was lower than in healthy control group except compensatory stage. There was no correlation between CD4+T cell ratios in lymphocytes,CD4+CD127-Treg or CD4+CD25+CD127-Treg ratios in CD4+T cells and the values of BUN,Cr among the hypertension patients. Conclusion:The number of CD4+T cells increases, and CD4+CD127-Treg decreases in the patients with chronic renal insufficience and their immune functions are shown in disoroler .

Objective To study the killing effects of fractional proteins from Microtus fortis (Mf) serum on schistosomula of Schistosoma japonicum. Methods Mf serum proteins were separated into albumin and globulin by means of salt out of ammonium sulfamate. The globulin was then separated into 4 big and 12 small fractional proteins through Sephacryl S-300 column chromatography and electrophoresis elution. The killing effects were observed in vitro in cultivation in which the purified fractional proteins and schistosomula of S. japonicum were incubated together for 48 h. Results The mortality rate of schistosomula acted by Mf globulin was 59.2% and when added with complements was 68.4%. The killing effects of the 2nd and 3rd big fractional proteins were the same as that of Mf globulin. Three small fractional proteins (3.2, 3.3, 3.4) showed the higher killing effects which made the mortality rate of schistosomula 45.1%, 57.6% and 67.2%, respectively. The fractional protein of 100-135 kDa also showed the same killing effect as that of Mf globulin. Globulin from BALB/c mice sera had no significant effect on schistosomula. There was no significant difference in the mortality rate of schistosomula acted by both albumins. Conclusions Mf globulin has significant killing effects on schistosomula of S.japonicum in vitro and 100-135 kDa fractional protein may be an important effective molecule.

Chinese Journal of Parasitology and Parasitic Diseases 1997;0(06):-
Specific antibodies purified from sera of rabbit infected with Toxoplasma gondii were used to immun-oscreen the 12-mer and 7-mer phage random peptide libraries. By 3 rounds of screening, 70 clones were picked out ra-ndomly to test the specificity and 43 were found positive by ELISA. 23 of the 43 positive clones showing strong reaction in ELISA were picked out for sequencing. Clone A7 was selected as mimic antigen in ELISA test after analysis of the 23 short peptide sequences. 23 out of 47 sera from rabbits infected with Toxoplasma gondii were ELISA positive by A7 mimic antigen with a positive rate of 68.1%, and 10 of 155 sera from healthy persons showed false positive with a sp-ecificity of 93.5%. The primary result suggests that mimic antigen may have a potential use in diagnose of toxoplasmosis.

Chinese Pharmacological Bulletin 2003;0(12):-
Aim To study the mechanisms of protective effects of extract of astragalus against focal cerebral ischemia/reperfusion injury in rats. Methods Focal cerebral ischemia was induced by intraluminal thread occlusion of middle cerebral artery. Immunohistochemistry was used to determine expression of TNF-?,IL-1? was measured by radioimmunity, and the apoptosis was observed by TUNEL. Results EA(20、40、80 mg?kg -1,ig)could decrease the expression of TNF-? and the level of IL-1? and reduce cell apoptosis. Conclusion EA has inhibitory action on the increase in the expression of TNF-?,the level of IL-1? and the apoptosis of neurons induced by focal cerebral ischemia/reperfusion.

Objective:To investigate the effects of a stannous-containing sodium fluoride dentifrice and crisscross bristle manual tooth-brush in the management of dental plaque and gingivitis.Methods:249 cases with gingivitis were enrolled in an office-based study. The study was unsupervised and single-centered with open-label and self-control.At baseline, gingival health and plaque coverage were assessed by dentists using categorical scales.Participants were given stannous containing sodium fluoride dentifrice and crisscross bristle design manual brush, and were instructed to use the products by manufacturer's usage instructions twice daily for 30 days.At the end of 30 days, plaque and gingivitis were reassessed using the same categorical scales.Results:232 participants(1 78 females and 54 males) completed the study.Gingivitis data of 5 cases and dental plaque data of 3 cases were not judgable. After 30 days of product use,226 cases(99%)showed noticeable improvement in their gingival health;227(96%)cases showed improvement in overnight plaque cover-age.Conclusion:Stannous-containing sodium fluoride dentifrice in combination with crisscross bristle toothbrush is effective in the management of gingivitis and dental plaque.
