Chinese Journal of Neurology 2000;0(05):-
Objective To investigate total serum homocysteine (tHcy) concentration in ischemic stroke patients and in carotid atherosclerosis patients in comparison with normal con trol elders and to evaluate whether providing folate and vitamin B 12 coul d decrease serum homocysteine concentration. Methods The sampl e in this study was obtained from the inpatients and outpatients of geriatric d epartment, Huashan Hospital. 160 patients were recruited and were divided int o two groups with smoking controlled. Thyroid gland abnormality and renal f a i lure were excluded. One group consisted of 71 male and 9 female ischemic strok e patients aged 73 years on average; There were 71 male, 9 female carotid ather osclerosis disease patients, aged 7 years on average in the second group. H y pertension and mellitus diabetes were controlled to avoid the influence of the f actors in these patients; the third group consisted of 72 male and 8 female norm al control elders, aged 71 years on average. Besides fast serum total homocyst e ine, every patient took laboratory and physical examinations, including plasma fo liate, vitamin B 12, free plasma glucose, creatinine, cholesterol, triglyce ride, blood pressure and blood mass index. Then each group was randomly subdivided in t o two groups in which one group was provided with folate 1.25 mg qod, and vitam in B 12 8?g qd w hile the other was not provited with folate and vitaminB 12. These patient s were reexamined after 10 months. Results The serum tHcy concentration was (16.1 ?5.3) ?mol/L in ischemic stroke group and (14.2?4.72) ?mol/L in caroti d athero sclerosis group compared with (12.5?3.0) ?mol/L in normal control elders ( P=0.000 1). The plasma foliate was low in ischemic stroke and carotid athe rosclerosis patients, and there were no significantly statistics dif ferences in the other examinations.Correlation analysis showed that tHcy was ne gatively correlated with foliate and vitamin B 12, r=-0.392,-0.36 r e spectively i n ischemic stroke group, r=-0.287,-0.339 respectively in carotid ath eroscleros is group, and r=-0.338,-0.227 in normal control group. The tHcy concent ration was lower in the groups provided with foliate and vitamin B 12 than that in the g roups without foliate and vitamin B 12 (P=0.000 1), even in the normal elders. Conclusions The tHcy concentration was higher in the patients with ischemic st roke and with carotid atherosclerosis. Folate supplement could moderately lowe r tHcy.

Objective:To evaluate the acute toxicity and protective effects of polysaccharide from Schisandrae sphenantherae Fruc-tus in the mice with acute liver injury induced by CCl4 . Methods:The classical acute toxicity tests were used to determine the maxi-mum drug loading (MLD) and observe the acute toxicity. The acute liver injury model was induced by 0. 5% CCl4. Sixty mice were randomly divided into the blank control group, CCl4 model group, bifendatatum model group and polysaccharide group respectively at high (1388 mg·kg-1), medium (694 mg·kg-1) and low (347 mg·kg-1) dose groups with 10 ones in each. After 6-day drug treatment, the levels of aspartate aminotransferase ( AST) and alanine aminotransferase ( ALT) were determined by kits. Histopatho-logical examination was also carried out. Results: General behavior, body weight and organ examination in the acute toxicity tests showed no obvious changes. Compared with those in the model group, the levels of AST in the three polysaccharide groups significantly decreased (P<0. 05 or P<0. 01), the levels of ALT in polysaccharide group at high and medium doses significantly decreased(P<0. 01), and the liver tissue damage was improved to a certain extent as well. Conclusion: Polysaccharide from Schisandrae sphenan-therae Fructus has protective effects on acute liver injury induced by CCl4 in mice with low acute toxicity.

Objective To explore whether the glomerular podocytes can be damaged by aristolochic acid. Methods Thirty-two male SD rats were equally divided into the following 2 groups:model group in which the rats received the extract of Aristolochia manshuriensis Kom (AmK) by gavage; control group only received tap water by gavage.24 h urinary protein excretion was measured at the end of the 1st and 4th week, and SDS-PAGE gel electrophoresis was performed to detect the protein in urine. At the end of the 4th week, all the rats were sacrificed and the glomeruli were isolated by laser capture microdissection technique. The mRNA expression of nephrin,podocin,CDA2P,podocalyxin and podoplanin in isolated glomeruli was determined by RT-PCR,and the average width of glomerular foot process was measured by electron microscopy and image analysis. Results At the end of the 4th week,24 h urinary protein excretion in the model group was significantly higher than that in the control group (P<0.01) and the urinary albumin content in model group was also obviously increased. The average width of glomerular foot process in the model group was significantly larger than that in control group (P<0.01).The mRNA expressions of nephrin,podocin,CDA2P,podocalyxin and podoplanin in glomeruli were significantly down-regulated in the model group compared with the control group, which decreased by 34%,62%,56%,50%(P<0.01) and 27% (P<0.05),respectively. Conclusions Aristolochic acid can damage the glomerular podocytes, resulting in the down-regulation of nephrin,podocin,CD2AP,podoplanin and podocalyxin mRNA expression, the segmental widening of foot process, and increased urinary protein excretion.

Because of the high morbidity and mortality rate,cardio-cerebrovascular diseases, including cerebral embolism, cerebral hemorrhage, cerebral vasospasm and myocardial infarction, have become main diseases threatening human health. Tradition-al chinese medicine(TCM)holds that the basic pathogenesis is Qi imbalances, which could be improved by benefiting Qi and promot-ing blood circulation. The compatibility of Radix astragali and Radix salviae miltiorrhiae are particularly suitable for treating the car-dio-cerebrovascular system diseases by improving Qi deficiency and blood stasis. This paper focuses on the application of Radix astrag-ali,Radix s. miltiorrhiae and their compatibility for treating the cardio-cerebrovascular system diseases. Moreover, our research would offer valuable references for the development of new drugs related to the treatment of cardio-cerebrovascular system diseases.

Aurora-B as an important kinase to adjust the cell normal mitosis is a potent target for cancer treatment. Aurora-B is overexpressed in a broad range of tumor and tumor cells are more sensitive while Aurora-B is inhibited. Due to the key role of the Aurora-B in cell mitosis, the development of its inhibitors is becoming more and more important. Several small molecules inhibit with a similar efficacy both Aurora-A and Aurora-B, however, in most cases the effects resemble Aurora-B disruption by genetic methods, indicating that Aurora-B represents an effective therapeutic target. There were several Aurora-B kinase inhibitors which had entered the clinics and displayed good antitumor activity. In this review, we will outline the functions of Aurora kinase B in normal cell division and in malignancy. We will focus on recent preclinical and clinical studies that have explored the mechanism of action and clinical effect of Aurora-B inhibitors in cancer treatment.
