1.Rapid Direct Sampling Detection of Pb in Grain Using Diluted Acid Extraction Coupled with Graphite Furnace Atomic Absorption Spectrophotometry

A mild rapid method for the determination of Pb in grain was established by diluted acid extraction and direct sampling detection with graphite furnace atomic absorption spectrophotometer(GF-AAS). Some factors, such as the grain size, nitric acid concentration, extraction time, solid, liquid proportion, and the instrument conditions were optimized. The results indicated that, at room temperature, the extraction rate of Pb in grain by diluted nitric acid was between 92. 14% and 94. 75%, and the average recoveries of Pb in grain extract reached 91. 65%-94. 58%, the precision of method was less than 5%, and the method detection limits and quantification limits were 0. 64 μg/L and 2. 14 μg/L, respectively, meanwhile, the extraction time was shorten to no more than 30 min. Compared with the results pretreated by microwave digestion and determined by GF-AAS, the detection values obtained by diluted acid extraction had no significance difference (p<0. 1).
2.Comparison study on improving lower limb ischemia by transplantation of antologous peripheral blood and bone marrow stem cells.

Objective Clinical efficacies of angiogenesis and improvement in lower limb ischemia are compared for the transplantation of both the peripheral blood stem cells(PBSC)and bone marrow stem cells(BMSC).Methods A total of 170 patients suffering from lower limb ischemia who received stem cell transplantation were monitored for random comparison study during September 2004 to October 2006.Among them 94 patients were treated with PBSC and 76 were BMSC.All the clinical data and laboratory findings before and after the transplantation in the 1st.12th and 24th weeks were comprehensively evaluated and tested by SPSS 12.0 software package.Results The total number of MNC,CD34+ cells and CD133+ cells in PBSC group was greatly more than BMSC group and had prominent or very prominent variances(P
3.Accelerating personnel training: key to enhancing the competitiveness of special hospitals

4.Transplantation of autologous peripheral blood stem cells for the treatment of diabetic foot in 26 cases

Objective To observe the clinical efficacy of autologous peripheral blood stem cells (PBSC) transplantation in 26 cases of diabetic foot.Methods From 2003-11 to 2004-07,26 patients with diabetic foot received rhG-CSF 450~600ug/d by hypodermic injection for 5 days to mobilize stem cells.On the sixth day,PBSC were collected by COBE 6.1 Spectra Version for 82~148ml.The PBSC were injected into the ischemic lower limbs and foot intramuscularly for 3?3 cm distance.The clinical and laboratory findings were monitored.Results In 26 patients receiving PBSC transplantation,obvious pain-relief was shown in 23 cases for 3~14 days,foot cold-feeling changed in 24 patients after 2~7 days,foot ulcer improved in 4 cases after 4~12 weeks.ABI increased in 6 cases.Digital subtraction angiographic scores were peformed in 2 patients after 8 weeks,and there was new collateral vessels formation.No related complication or adverse effects were observed in all process.Conclusion Autologous PBSC transplantation might be a safe and effctive method for diabetic foot.
5.Transplantation of autologous peripheral blood stem cells for the treatment of arteriosclerosis obliterans of lower limb in 26 cases

Objective To observe the clinical efficacy and interrelated factors of autologous peripheral blood stem cells (PBSC) transplantation in the treatment of arteriosclerosis obliterans (ASO) of the lower extremity. Methodes A total of 26 patients with ASO of the lower limb received rhG-CSF 450-600?g/d by hypodermic injection for 5 days to mobilize stem cells. On the sixth day, PBSC were collected by COBE 6.1 Spectra Version with a total amount of 82-118ml, with the number of mononuclear cells (MNC) (718.2-224.6)?10 9 /L. The MNC were injected intramuscularly into the ischemic limbs with intervals of 3cm apart. The clinical and laboratory findings were monitored. Results In 23 out of 26, pain was relieved after 3-21 days (88.5%), in 24 patients coldness was ameliorated after 2-28 days (92.3%), in 4 patients foot ulcers were better after 4-12 weeks. ABI increased in 6 cases. The total effective rate was 92.3%. No complication or adverse effect was observed in all the patients. Conclusion Autologous PBSC transplantation might be a safe and effective method for ASO of the lower limb ASO.
6.Study of the Relation of Serum Angiotensin Converting Enzyme and Pregnanacy-induced Hypertension

Objective:Our purpose was to investigate the function of angiotensis converting enzyme(ACE) in the occurrence and development of pregnancy-induced hypertension(PIH). Methods:The study group included 30 pregnant women diagnosed with PIH( 10 mild, 10 moderate, 10 severe). Thirty healthy pregnant women were selected as the control group. The serum was assayed for the activity of ACE by using color comparison method, hippuric acid used as the substrate. Results:The activity of ACE of the study group was significantly higher than that of the control group. Significantly positive correlationships existed between mean blood pressure, degree of adema, level of urine protein,and uric acid. Conclusion:The elevation of serum activity of ACE is related to the occurrence and development of PIH.
7.MHC antigen expression on the surface of bone marrow stromal stem cells after directional induction in vitro

BACKGROUND:Alogeneic bone marrow stromal stem cel transplantation for treatment of bone diseases is a hot topic. To seek effective methods for prevention of post-transplantation immune rejection is urgent to be solved.
OBJECTIVE:To explore the expression of MHC antigen on the surface of bone marrow stromal stem cels after osteogenic inductionin vitro.
METHODS:Bone marrow samples were extracted from rabbits to in vitro isolate and culture bone marrow stromal stem cels. Then, the cels were cultured in IMDM medium containing bone morphogenetic protein-2. Flow cytometry was used to analyze the expression of MHC antigen on the osteoblasts differentiated from bone marrow stromal stem cels.
RESULTS AND CONCLUSION:There was highly expressed MHC I antigen but no MHCII antigen on the osteoblasts differentiated from bone marrow stromal stem cels. After osteogenic induction, no immune rejection was found. These findings indicate that alogeneic or xenogeneic bone marrow stromal cel transplantation can be used in the treatment of bone defects.

In order to study the character of periodontal ligament cells (PDLCs) attaching on commercially pure titanium (cpTi) by morphology and metrology on the early stage (24 h), 1 x 10(5)/ml PDLCs in 2 ml culture medium were seeded on cpTi discs fixed in 24-well culture plates. Morphology of cell attachment was observed by contrast phase microscope, scanning electron microscope (SEM) and fluroscence microscopy. Cell adhesion was analyzed by MTT at 0.5, 1, 2, 4 h respectively. PDLCs could attach and spread on cpTi discs. SEM showed that PDLCs had pseudopod-like protuberance. PDLCs showed different attaching phases and reached saturation in cell number at 2 h. It was concluded that PDLCs had good biocompatibility with cpTi, and showed a regular and dynamic pattern in the process of attaching to cpTi.

9.Lactoferrin downregulates the expression of toll like receptor 4 stimulated by lipopolysaccharide in human periodontal ligament cells.

OBJECTIVETo examine the role of lactoferrin (LF) on Toll like receptor 4 (TLR4) stimulated by lipopolysaccharide (LPS) in human periodontal ligament cells (hPDLCs).
METHODSPrimary hPDLCs were cultured by tissue block enzymolytic method. Cells obtained from four passages were identified and used in this experiment. Cells without stimulation served as the controls and cells treated with LPS (0.1 microg x mL(-1)) comprised the LPS group. The LPS + LF group was pretreated with LPS (0.1 microg x mL(-1)) for 2 h, and then treated with LF (10 microg x mL(-1)). Four hours after LF stimulation, the mRNA expression levels of TLR4 were examined by real-time quantitative polymerase chain reaction (RT-PCR). The protein expression of TLR4 was observed by cell immunofluorescence staining after LF stimulation of 24 hours.
RESULTSTLR4 mRNA expression in the LPS + LF group was significantly more decreased than that in the LPS group (P < 0.05), but exhibited no difference with that in the control group (P > 0.05). Cell immunofluorescence staining showed that the protein expression of TLR4 in the LPS + LF group was significantly more decreased than that in the LPS group (P < 0.05), but exhibited no difference with that in the control group (P > 0.05).
CONCLUSIONLF can decrease the expression of TLR4 stimulated by LPS in hPDLCs, thus presenting potential application for controlling the TLR4 immune pathway of periodontitis.

10.APPLICATION OF THE REVERSED ISLAND FLAP BASED ON SURAL NEUROVASCUALR PEDICLE

Objective To report the method for repair of the heel and ankle.Method On the basis of applied anatomy,the reversed island flap based on sural neurovascular pedicle has been designed for repair of heel and ankle defects.Results The flaps have been used in 14 cases,the maximum size 9 cm×15 cm,and all flaps were survival compeletely except partial necrosis in one flap.Conclusion The blood supply of the flap is good.The length of pedicle can reach 12 cm.It is an effective method for repair of heel and ankle defects.
