Objective To explore the correlations between emotions and self-concept in patients with traumatic brain injury. Method The Chinese version of the Positive and Negative Affect Scale and the Tennessee Self-Concept Scale was used to test 178 patients with traumatic brain injury. Results The mean scores on positive and negative emotions were (1.93±0.53) and (2.76±0.61), respectively. The mean score on self-concept was (2.45±0.31). The positive emotions of patients were positively correlated with the level of self-concept and the negative emotions negatively with the level of self-concept (r=0.437and -0.363respectively,bothP<0.01). Conclusions For patients with traumatic brain injury, the negative emotions outnumber the positive emotions and their self-concept was at lower level. Nurses should take measures to improve their positive emotions and reduce their negative emotions in order to improve their self-concept level.

AIM To study the early cardioprotective effects of pharmacological preconditioning of nitroglycerin (NG) and buprenorphine (BU) used alone and in combination on myocardial ischemia in rats. METHODS Male Wistar rats were randomized to 5 groups. The ischemia and reperfusion injury was induced by ligation of the left arterior descending coronary arrery for 30 min and followed reperfusion for 2 h. The rats were subjected to different treatments before I/R. Normal saline (NS) was infused intravenously with the same volume of NG and BU in Sham group. Ischemia/preconditioning (IP) was performed by three cycles of 5 min I/R in EIP group. BU (1 0 mg?kg -1 ) and NG (0 3 mg?kg -1 ) was administered intravenously alone in BU and NG group or given together in B+N group to mimic the effects of IP, respectively. Heart rate, blood pressure, ST segment and arrhythmias were recorded continuously throughout the whole test. Plasma LDH and CK were measured on 30 min after ischemia and 2 h after reperfusion, HE and TTC staining were performed to determine myocardial necrosis at the end of test. RESULTS Compared with Sham group the onset of arrhythmias was delayed and the duration of ventricular premature contraction (VPC) was shortened remarkably ( P

Journal of Medical Postgraduates 2004;0(01):-
Objective RNA interference refers to post-transcriptional gene silencing caused by double strands RNA.To investigate the effect of EGFR receptor on esophageal carcinoma, the expression vector of HER4 gene targeted small interfering RNA was constructed to observe its silencing effect in human esophageal carcinoma cell line Eca-109,in order to find a promising method for the gene therapy of this disease. Methods Two complementary oligo DNA strands targeting HER4 gene were designed and synthesized according to the principles of designing siRNA.After annealing, oligo DNAs were inserted into SUPER.neo+gfp vector, then enzyme digestion analysis and DNA sequencing were applied. After transfecting it into human esophageal carcinoma cell line,we detected the level of expression of HER4 gene through real-time quantitative PCR and Western Blot. Results The enzyme digestion analysis and DNA sequencing show that HER4 gene targeted small interfering RNA and its expression vector were constructed successfully, and after transfection, the expression of HER4 gene in esophageal carcinoma cell line was suppressed greatly. Conclusion HER4 gene targeted small interfering RNA and its expression vector were constructed successfully, and could decrease the expression of HER4 gene in Eca-109 cell line, which laid the foundation for the following experiment.

Objective: To explore the sensitization effects of resveratrol on CNE2 nasopharyngeal carcinoma cell line with hypoxia-induced chemotherapy resistance and the potential mechanism. Methods: Human CNE2 nasopharyngeal carcinoma cell line was cultured under hypoxic conditions (37 degrees centigrade, 5% CO(2), 2% O(2)) in vitro. The cultured cells were treated with different concentrations of resveratrol for 48 h. Reversal fold (RF) of reseratrol to chemotherapeutic drugs in CNE2 cells was measured by methyl thiazolyl tetrazolium (MTT) assay. Apoptotic rate of CNE2 cells was observed by flow cytometry. Reverse transcription-polymerase chain reaction (RT-PCR) method and Western blotting were used to investigate the expressions of multidrug resistance gene 1 (mdr1), multidrug resistance-associated protein 1 (MRP1) and hypoxia inducible factor 1alpha (HIF-1alpha) in CNE2 cells. Results: Resveratrol combined with chemotherapeutics produced a synergistic effect. The RF of 200 alphamol/L resveratrol to paclitaxel was 2.58. Combined with paclitaxel, 25, 50, 100 and 200 alphamol/L of resveratrol increased the apoptotic rate of CNE2 cells from (22.14+/-1.09)% to (23.24+/-1.37)%, (27.57+/-2.01)%, and (30.36+/-2.31)%, respectively. Resveratrol could down-regulate the expressions of HIF-1alpha, mdr1 and MRP1 significantly. After being treated with resveratrol at different concentrations separately, the expressions of HIF-1alpha, mdr1 and MRP1 in CNE2 cells decreased significantly as compared with paclitaxel alone or paclitaxel plus verapamil (P<0.01). Conclusion: Resveratrol can enhance the sensitivity of CNE2 cells to chemotherapeutic drugs under hypoxia. The potential mechanism is partly attributed to inhibiting the gene expressions of HIF-1alpha, mdr1 and MRP1.

Objective To investigate the protective effects of (-)-epigallocatechin gallate ( EGCG ) on daunorubicin ( DNR)-induced cardiotoxicity in mice. Methods The qualified mice were randomly divided into four groups:normal control group, myocardial injured model control group, high dose group (80 mg·kg-1 ) and low dose group (40 mg·kg-1 ) of EGCG. EGCG was administered intragastrically once daily for 7 days, followed by a single intraperitoneal injection of DNR (15 mg·kg-1 ) except in the normal control group. The electrocardiogram, myocardial enzymes and TNT-Hs in serum, cardiac ultrastructure of mice were detected after 48 h. Results In DNR model control group, the incidence of arrhythmia was 64. 7%. The activity of serum cardic enzymes including CK,CK-MB,LDH,α-HBDH and ALT,AST, level of TNT-Hs were significantly higher than those in the normal control group(P<0. 01),and myocardial ultrastructure was injured remarkably. The incidence of arrhythmia was 44. 4% in mice treated with high dose of EGCG and 31. 6% in mice with low dose of EGCG. Compared to the model control group, the activity of CK,CK-MB,LDH,α-HBDH and ALT,AST, level of TNT-Hs in serum decreased remarkably in EGCG groups( P<0. 05 or P<0. 01). Low can EGCG alleviated the injury to the ultrastructure of myocardium compared to the model control group. Conclusion EGCG can prevent the cardiac toxicity induced by DNR in mice.

Traumatic brain injury (TBI)has been classified as mild, moderate,or severe,on the basis of the Glasgow coma scale (GCS)score.Mild TBI is estimated to account for 90% of all cases of TBI, and it has become a serious public health problem, with morbidity increasing year by year.At present, there is a lack of accepted uniform definition of mild TBI.Clinically, mild TBI and concussion are interchangeable terms. In recent years, advances in brain imaging, biomarkers determination, and neuropathology have encouraged people to revise and update their knowledge about mild TBI.In view of the high prevalence of mild TBI in the emergency and community, and the absence of the data concerning the long-term effects of mild TBI, further research is needed about how to reduce morbidity and costs, alleviate delayed consequences, and develop evidence-based interventions to improve outcomes.
