Objective To investigate the protective role of transient focal ischemic precondition(PC) against permanent focal cerebral ischemia, to define the proper time dose for precondition, and to study the reaction of astroglia in cerebral ischemic tolerance. Methods Temporary middle cerebral artery occlusion(10 minutes, 20 minutes, 30 minutes) followed three day reperfusion before permanent middle cerebral artery occlusion(PMCAO) transcranially was used as precondition in Wistar rats. The protective role was evaluated by observing the neurological deficits, analyzing the infarct volume and studying changes of pathohistology. The reaction of astroglia was observed by using anti GFAP immunohistochemistry.Results Compared with the control group, 20 min ischemic precondition, which did not produce neuronal damage obviously, alleviated the neurological deficits and reduced the infarct volume significantly( P
3.CLONING OF ZNRD1 AND EFFECT OF ITS ANTISENSE VECTOR ON ACCUMULATION OF ADRIAMYCIN IN DRUG-RESISTANT GASTRIC CANCER CELLS

The aim of this study was to construct antisense of ZNRD1 encoding gene, and to transfect it into SGC7901/VCR cells, to seek measures to overcome multidrug resistance of gastric cancer cells. ZNRD1 cDNA amplified by PCR was confirmed by DNA sequencing, and then inserted into the multiple cloning site of the expressing vector pcDNA3 1 + with molecular cloning technique. The recombinant vector was identified by endonuclease digestion. Antisense recombinant vector was transfected into SGC7901/VCR cells using lipofectamine. Flow cytometric analysis was used to detect adriamycin (ADM) accumulation in SGC7901/VCR cells. The results showed that a fragment was obtained by PCR, and its sequence was consistent with ZNRD1 cDNA reported in the literature. After antisense recombinant vector having been transfected into SGC7901/VCR cell, ADM concentration in such cells was increased. The above results indicated that the antisense vector ZNRD1 could enhance the intracellular accumulation of ADM in SGC7901/VCR cells, which might be of potential treatment value.
4.Regulatory effects of thioredoxin on chemosensitivity of gastric cancer cells

Objective To explore the regulatory effects of thioredoxin on chemosensitivity of gastric cancer cells. Methods RT-PCR was employed to clone the full-length cDNA of thioredoxin (Trx) from human drug-resistant gastric cancer cell line SGC7901/VCR. The sense and antisense eukaryotic expression vectors corresponding Trx were constructed and transduced into gastric cancer cells SGC7901 and SGC7901/VCR by lipofectamine, respectively. MTT assay was used to evaluate the sensitivity of gene-transfected gastric cancer cells and control cells to anti-cancer drugs in vitro. Results The results indicated that SGC7901 cells transfected with Trx sense vector showed significantly decreased sensitivity to vincristine, cisplatin and adriamycin, while SGC7901/VCR transfected with Trx antisense vector showed significantly increased sensitivity to these drugs. Conclusion All these data suggested that Trx might participate in the formation of multidrug resistance of gastric cancer cells and alteration of Trx could regulate chemosensitivity of gastric cancer cells.
5.Method and Complications of Direct Fetal Blood Sampling

Objective To study the indications of percutaneous ultrasound-guided direct fetal blood sampling; the methods of fetal blood assessment; and the relationship between the different puncture techniques and the complications. Methods Direct fetal blood sampling were performed on 90 pregnant women whose gestational age were from 16 to 36 weeks for medical indications under the guide of ultrasound. None stress test were performed before and after the puncture. Ulrtrasonographies were done at 2 hours and 24 hours after the procedure. Results There were no severe maternal or fetal complications such as fetal loss and placental abruption. The incidence of bradycardia was 35.6%(32/90), all recovered within 60 seconds; the incidence of bradycardia happened within 4 hours after the puncture was 2.25(2/90). There was a positive relationship among the incidence of bradycardia, the volume of blood taken and the place of punctures,r=0.27 and 0.36 respectively. There was one case with placental changing happened after 2 hours of the puncture, the placenta changing incidence was 1.1%. No emergency cesarean section was done due to the complications of the procedure. All the umbilical cord were normal when the babies were born. Conclusions Direct fetal blood sampling is a safe technique for both fetus and the mother under the guide of real time ultrasound. The data come from fetal blood analysis give reliable informations about chromosome, congenital infections, the status of fetal metabolism, endocrine system and hematologic system directly, which make this technique unreplacable for intrauterine diagnosis.
6.The establishment of x-ray environmental standard calibration quality

Objective:According to the requirement of ISO and National Standard, new X-ray environmental standard for the calibration of environmental dosimeter was established in Beijing SSDL. Methods:Half value layer and air kerma dose-rate was determined in the X-ray environmental qualities by a standard dosimeter. Results: The technical requirement of ISO and National Standard are met within 2% for the X-ray environmental standard qualities in Beijing SSDL. Conclusion:The X-ray standard radiation field can be used for the calibration of environmental dosimeters and experiment of research.
7.Compensation research on energy response for the semiconductor personal dosimeter

Objective:To complete experiments of the energy response compensation for three semiconductor personal dosimeters in order to meet the requirements of IEC and National Standard.Methods: In Beijing SSDL, the semiconductor detectors were covered by the different thicknesses of Pb and Sn additional filters with several diameter holes to compensate the dosimeters’ energy response.Results: At the 50 keV-Cs-137 energy range, the measuring error of dosimeters is met within 30% of energy response.Conclusion:By the compensation of combined Pb and Sn filters with a hole, the semiconductor personal dosimeters can be used for radiation monitoring.
8.Distribution Characteristic and Drug Resistance Analysis of Pathogens in Infective Cases

OBJECTIVE To investigate the distribution and the drug resistance of pathogens in infective cases and to provide a foundation for reasonable application of antibacterials.METHODS The information of all bacterial cultures and susceptibility tests from the inpatients in the year of 2006 was analyzed.RESULTS A total of 336 strains of pathogens were isolated,and the pathogens causing lower respiratory tract infection rated the top(60.7%).The resistance rate of Acinetobacter baumannii was the highest in Gram-negative bacilli.It was 33.3% to imipenem,and more than 66.6% towards the other antibacterials.All Gram-positive cocci were sensitive to vancomycin.The resistance rate of Enterococcus and Staphylococcus aureus was more than 75% to the most antibacterials.CONCLUSIONS Inspecting pathogens and strengthening susceptibility tests are very important in reducing drug abuse,decreasing the resistance rate and raising the cure rate in hospital.

10.Synthesis and anti-tumor activity of oleanolic acid derivatives.

Thirteen novel oleanolic acid (OA) derivatives were designed and synthesized with modification at positions of C-3, C-12 and C-28 of OA. Their structures were confirmed by MS, 1H NMR and elemental analysis. Their in vitro cytotoxicities against various cancer cell lines (SGC7901, MCF-7 and A549) were evaluated by MTT assay. The results indicated that the tested derivatives were found to have stronger cell growth inhibitory activity than OA. Among them, compounds II2 and II3 showed more potent cytotoxicity on MCF-7 and A549 tumor cells than gefitinib (positive control). They are worthy to be studied further.
