1.Cell cycle arrest and cell apoptosis induced by HIV-1 Vpr in cancer cells in vitro

Objective To investigate the G2 cell cycle arrest and apoptosis induced by HIV-1 Vpr gene on Hela,Lovo and HepG2 cancer cells.Methods Recombinant vector pcDNA4-Vpr was constructed by DNA recombination technology and was then transfected into Hela,Lovo and HepG2 cells.Meanwhile,pcDNA4-EGFP plasmid group,FUGENE group and blank group were also set up as control.Ratio of cytostasis was evaluated by MTS assay 24 h,48 h and 72 h later,cell cycle arrest examined by flow cytometry and apoptosis detected by staining with ANNEXIN V and PI double dyes.Results Compared to the control group,the value of inhibition ratio,G2 arrest and apoptosis of Hela,Lovo and HepG2 cells increased obviously 24 h,48 h and 72 h after the transfection ( P < 0.05 ).72 h after the transfection,the inhibition ratio of Hela,Lovo and HepG2 was 29.67%,27.35% and 31.67% respectively.Percentage of G2 phase cells was 24.9%,18.8%and 32.1% respectively.Apoptosis percentage of Hela,Lovo and HepG2 ceils was 15.46%,7.7% and 41.5% correspondingly.Conclusion HIV-1 Vpr gene can induce cell cycle G2 arrest and apoptosis of Hela,Lovo and HepG2 cell lines in vitro.
2.Research progression on relationship between angiosteosis and matrix vesicle

Angiosteosis is associated with increased vascular stiffness and leads to clinical manifestations such as high blood pressure and heart failure,which increases the risk of cardiovascular morbidity and mortality.Angiosteosis is considered to be ectopic deposits of calcium phosphate crystallization in the form of hydroxyapatite in cardiovascular tissue.However,the mechanism of angiosteosis is not yet clear.The pathophysiologic process of angiosteosis is similar to normal bone mineralization.Matrix vesicles (MV) play an important role in bone mineralization.Researches confirmed the existence of MV in the calcified arteries under the microscope.Angiosteosis is widely found in clinical diseases,so the studies of the mechanism of angiosteosis and the role of MV in angiosteosis are of great significance.
3.Relationship between change of dynamic electrocardiogram and coronary artery disease

Objective: To explore diagnostic value of dynamic electrocardiogram (DCG) on coronary artery disease. Methods: A total of 100 inpatients with “angina pectoris” history from our hospital were enrolled and received 24h DCG measurement. The DCG examination results were compared with those of coronary angiography (CAG) and analyzed then. Results: Among the 100 patients, there were 85 patients with positive DCG, including 70 cases with positive CAG and 15 cases with negative CAG; and 15 patients with normal (negative) DCG, including 10 cases with negative CAG and five cases with positive CAG. With CAG as golden standard, sensitivity, specificity, positive predictive value (accuracy) and negative predictive value of DCG in diagnosis of coronary heart disease were 93.3%, 40.0%, 82.4% and 66.7% respectively. Partial patients had “angina pectoris”, no stenosis in CAG but there’s slow blood flow. Conclusion: Dynamic electrocardiography is one of important measuring methods diagnosing coronary heart disease. It is simple, practical, safe, economic and appropriate for wide application in clinic, but its specificity is just 40.0%, so coronary heart disease cannot be excluded in negative patients.
4.PRESENT SITUATION OF BREAST-FEEDING AND COMPLEMENTARY FEEDING AND THEIR INFLUENCE ON GROWTH OF CHILDREN UNDER 36 MONTHS IN WESTERN CHINA

Objective To find out the present situation of complementary feeding, breast-feeding and children nutrition, and further explore the relationship between them. Methods Random sampling was adopted and 7302pairs of mothers and their children under 36 months were investigated by means of questionnaires. Results Breastfeeding rate was 96.8% ,but exclusive breast-feeding rate was only 37.6% . Breast-feeding rate over 12 months was 34.7% ,breast-feeding rate over 24 months was only 15.8%. Timely sucking rate was 8.4% ,19.2% of children had too early complementary feeding, 26.8% had too late,and 43. 1% had appropriate feeding . To the children over 6months, the feeding rates of meat and eggs per week were 36. 3% and 49. 3 % , respectively. The stunting-rate was 24.0%, underweight-rate 22. 1 % ,and wasting-rate 8. 9 %. The incidence rate of iron deficiency anemia (IDA) was 23.3 %. Multivariable analysis showed that growth of children was associated with diet diversity,intake frequency of eggs and meat, too early complementary feeding,prolonged breast-feeding and diarrhea, etc. Conclusion In rural areas of western China ,breast-feeding has been fundamentally popularized ,however ,the unfit complementary feeding and malnutrition of children under 36 months are common and serious ,important complementary feeding recommendations and good hygiene condition are strongly needed.
5.Determination of Trace Flow Injection-Catalytic Spectrophotometry

The color fading reation on potassium bromate with Victoria green stand G by the catalysis of nitrite in hydrochloric acid medium was studied. The experimental condition offlow injection spectrophotometric method for the determination of trace NO2- was optimized. The linear range for the determination of nitrite is 0.00~0.30 mg/L and 0.30~ 2.00 mg/L, the solpes of standard curves are 0.708 and 0.339 respectively, the analytical speed is 80/h. It has been used todetermine trace nitrite in the collanae lake water, fishpond water, power plant waste water and well water. The results are satifactory.
7.Comparison of the influence of different HBV genotypes and subgenotypes to patients 'immune function

Objective To compare the influence of different genotypes of hepatitis B virus to the body's immune function.Methods 377 patients with chronic hepatitis B virus (serum HBV DNA are all positive)were selected.The serum specimens were collected.Nest PCR method was used to detect all entrants subtype HBV genotypes,ELISA method was used to detect serum levels of IL -10 and IL -17,fully automatic biochemical instrument was used to test all entrants liver function.Results Of 377 patients with chronic hepatitis B,C genotype in 323 cases,C2 type in 321 cases (85.1%),C1 type 2 cases (0.5%);Genotype B 41 cases (10.9%),were B2 subtype;B and C mixed 12 cases (3.2%);Type D in 1 case (0.3%).The sex,age,ALT,AST,TBIL of B2,C2 subtypes,BC mixed infection had no statistically significant differences.B2,C2 subtypes,BC mixed infection had no statistically significant difference in serum IL -17 level.And B2,C2 subtype infection had statistically significant difference in IL -10 level (F =0.444,P <0.05).Conclusion Different genotype HBV subtypes may have different effect to the body's immune function.
8.Research progress in immunomodulatory properties of mesenchymal stem cells

BACKGROUND:Mesenchymal stem cel s are multipotent progenitor cel s that can be isolated from the bone marrow, adipose tissue, umbilical cord blood and so on. Mesenchymal stem cel s possess strong immunosuppressive effects on both innate and adaptive immunity.
OBJECTIVE:To summarize the immunomodulatory properties of mesenchymal stem cel s for T lymphocytes, B lymphocytes, natural kil er cel s and dentritic cel s and prospect its therapeutic implication.
METHODS:PubMed and CNKI databases were searched for relevant literatures published from 2000 to 2015. The key words were“mesenchymal stem cel s, immunomodulator, T cel s, B cel s, NK cel s, dendriticc cel s”in Chinese and English, respectively. Then, 61 papers were included and further analyzed.
RESULTS AND CONCLUSION:The immunomodulatory effects of MSCs on lymphocytes, B lymphocytes, natural kil er cel s and dentritic cel s were elicited by cel-cel contact and soluble cytokines. But mesenchymal stem cel s from different sources hold different immunomodulatory effect on immune cel s. When we use mesenchymal stem cel s in clinic, some important factors, such as isolation methods, cel sources, colonization sites, should be taken into account. In order to ensure the clinical safety and effectiveness, there are stil many problems to be further studied before mesenchymal stem cel s can be widely used in clinic.
9.Distribution and antibiotic resistance profile of the pathogenic isolates in a gynecology and obstetrics hospital during 2009-2014

Objective To analyze the distribution and antimicrobial susceptibility of the pathogenic isolates during 2009-2014 for rational use of clinical antibiotics.Methods The clinical data were reviewed and analyzed retrospectively to examine the distribution and resistance profile of the pathogenic isolates.Strain identification and antimicrobial susceptibility test were conducted by BioMerieux VITEK-2 automatic microbiology analyzer and the associated identification cards.All results were interpreted according to CLSI standards.Results A total of 6 393 strains of pathogenic bacteria were isolated during the six-year period.The top three pathogens were Escherichia coli (33.6 %),Enterococcus faecalis (19.4 %),and Streptococcus agalactiae (13.7 %).In addition,8,1,and 10 strains ofListeria spp.were isolated in 2009,2010,2014,respectively.A total of 40 strains of anaerobic bacteria had been isolated since 2012.The isolates were mainly from genital tract (53.7 %) and uterine cavity (21.3 %),followed by blood stream (8.2 %),including venous blood samples (6.1%) and umbilical cord blood samples (2.2 %).There were 165 (2.6 %) cases of surgical site infections.Escherichia,Enterococcus,and Streptococcus were the top three bacteria isolated from both uterine cavity and venous blood samples.The top three bacteria isolated from surgical sites were Escherichia,Enterococcus,and Staphylococcus.The bacteria isolated from umbilical cord blood samples were mostly Streptococcus,Escherichia,and Enterococcus.Anaerobic bacteria were mainly isolated from blood samples,which accounted for 82.5 % of all anaerobic bacteria.Listeria monocytogenes isolates were mainly from venous blood samples or uterine cavity.Overall,the antimicrobial susceptibility profile of the pathogenic isolates was relatively stable during the period from 2009 to 2014.Most antibiotics showed good activity against these isolates.Conclusions The pathogens isolated from this gynecology and obstetrics hospital during 2009-2014 period are mainly Escherichia coli,Enterococcusfaecalis,and Streptococcus agalactiae.Most isolates were susceptible to the commonly used antimicrobial agents.In addition to a few MRSA strains,ESBLs-producing Escherichia coli isolates were the main multidrug resistant organisms.
10.Effect of curcumin on cardiac remodeling and heart function change induced by adriamycin in rats

Objective: To study the effect of curcumin on cardiac remodeling and heart function change induced by adriamycin in rats. Methods: A total of 50 SD rats were randomly divided into blank control group (n=10), adriamycin group (n=20) and curcumin intervention group (n=20). General conditions of rats, including weight, hair color, diet and activity capacity, were observed. Left ventricular ejection fraction (LVEF), left ventricular fractional shortening (LVFS) and left ventricular end-diastolic dimension (LVEDd) were observed by echocardiography. Rat heart was weighed and levels of cardiac collagen type Ⅰ and Ⅲ were observed using immunohistochemistry method. Results: Compared with blank control group after experiment, there were significant increase in heart weight [(168.53±26)mg vs. (208.34±31)mg], heart weight/body weight [(0.36±0.06) vs. (0.52±0.08)], and significant decrease in body weight [(469.27±34)g vs. (428.47±45)g]; significant increase in LVEDd [(4.87±0.19)mm vs. (7.49±0.83)mm], significant decrease in LVEF [(69.53±6.25)% vs. (41.46±4.38)%] and LVFS [(45.83±3.79)% vs. (17.25±2.92)%] in adriamycin group, P<0.05 all, significant increase in expression rates of collagen type Ⅰ and Ⅲ, P<0.01.Compared with adriamycin group, there were significant increase in body weight (452.46±38)g, significant decrease in heart weight (178.73±38)mg, heart weight/body weight (0.39±0.07); significant decrease in LVEDd (5.96±0.65)mm and significant increase in LVEF (53.12±5.43)% and LVFS (36.57±3.66)%, P <0.05 all; significant decrease in expression rates of collagen type Ⅰ and Ⅲ in curcumin group, P <0.01. Conclusion: Curcumin can improve cardiac remodeling and heart function induced by adriamycin in rats.
