1.The effect of BMSCs/coral composite strengthened by titanium mesh in the repair of mandibular defects

Objective:To observe the effects of bone marrow stromal cells(BMSCs)/coral composite strengthened by titanium mesh in the repair of mandibular defects. Methods:The rabbits' BMSCs were cultured and induced in vitro, and then were combined with coral to form the BMSCs/coral composite, which were implanted into the left mandibular defect in the size of 15 mm?8 mm in 16 rabbits. Titanium mesh was used to strengthen and fix the composites. Coral implant was used as the control in the right mandibular defect.Six and twelve weeks after operation, X-ray observation and histological examination were employed to assess the status of bone formation. Results:In the experimental group, new bone formation was found by radiography and the histological examination 6 weeks after operation. 12 weeks after operation woven bone and lamellar bone were observed in the implanted area and the bone defects were repaired, coral was disappeared, titanium was integrated with surrounding bone tissue. In the control group 12 weeks after operation the bone defects were filled by fibrous tissue, coral was disappeared. Conclusion:Titanium /BMSCs/coral composites can repair mandibular defects effectively.
2.Prevalence of cagA and vacA subtypes of Helicobacter pylori and their relations to gastrointestinal diseases

Objective To investigate the prevalence of cytotoxin associated gene(cagA)and vacuolating cytotoxin gene(vacA)subtypes of Helicobacter pylori(H.pylori)in patients with upper gastrointestinal disease in Shijiazhuang area,and to assess the relationship between cagA,vacA subtypes and H.pylori related gastrointestinal disease.Methods Totally 55 H.pylori strains were isolated from patients with chronic gastrointestinal disease in Shijiazhuang area.Bacterial DNA from all these strains was extractedThe prevalence and genetic diversity in cagA 3′ region,vacA signal sequence and middle-region alleles were studied by the technique of polymerase chain reaction(PCR).Results In Shijiazhuang area,H.pylori cagA positive rate was 89.1%(49/55);cagA positive rate was not significant ly different between chronic gastritis and peptic ulcer(?~2=0.376,P=0.540).There were three vacA subtypes,including s1a/ m2 90.4%(47/52),s1a/ m1b-m2 5.8%(3/52)and s1a/ m1b 3.8%(2/52).The vacA s1a/m2 subtype showed no significant difference between chronic gastritis and peptic ulcer(?~2=0,P=1.000).Conclusion cagA~+and vacA s1a/m2 subtype of H.pylori are predominant in patients with upper gastrointestinal disease in Shijiazhuang area.There is no relationship between cagA,vacA subtypes and H.pylori related gastrointestinal disease.
3.Clinical research for the treatment of temporomandibular joint injury based on three-dimensional digital technology.

OBJECTIVETo investigate the accurate and individual treatment of temporomandibular joint(TMJ) injury based on 3D digital technology.
METHODSMaxillo-mandibular model was made using rapid prototyping technology based on the pre-operation 3D CT results. According to the 3D digital measurement results, TMJ concepts were ordered and the prosthesis was used to simulate the replacement surgery on the model. Then the joint replacement surgery was performed afterward.
RESULTS(1)After total replacement of TMJ, no pain happened and mouth open was not limited. Three months later, the joint position was normal and stable. The month open width was 4 cm. (2)After condyle replacement, primary healing was achieved with complete survival of bone graft. No edema was seen. Symmetric facial appearance was satisfactory.
CONCLUSIONSBilateral individual prosthesis for total TMJ or condyle replacement is an ideal method for TMJ injury.

4.Clinical application of artificial dermis combined with autologous thin skin graft in the repair of burn scar contracture of joint

Objective:To study the clinical value of artificial dermis combined with autologous thin skin graft in the repair of burn scar contracture in joints.Methods:A total of 52 patients with burn scar contractures were enrolled in the No.5 Hospital of Baoding from July 2015 to April 2018. According to different methods of repair, 26 cases were used in each group. The observation group was treated with artificial dermis combined with autologous thin skin graft. The control group was treated with medium-thickness skin grafting. The survival rate of autologous skin was compared between the two groups. The tissue of artificial dermal polyester fiber was taken and HE staining was performed to observe the pathological changes. The Vancouver skin scar assessment score (VSS), functional activity, infection rate, wound healing time, and VSS score after healing of the donor site were compared between the two groups.Results:The survival rate of autologous skin in the observation group (92.31%) was not significantly different from that in the control group (84.62%) ( P>0.05). Compared with preoperative, the VSS scores at 3, 6 months and 1 year after operation in both groups were decreased ( P<0.05). The VSS scores of the observation group were lower than those of the control group at 3, 6 months and 1 year ( P<0.05). The excellent rate of functional activity in the observation group (100.00%) was higher than that of the control group (76.92%) ( P<0.05); There was no significant difference in the infection rate (3.85% vs 7.69%) and healing time of skin grafting area between the two groups ( P>0.05). The healing time of donor site was shorter than that of the control group ( P<0.05). The VSS score of the donor site was lower than that of the control group ( P<0.05). Conclusions:Artificial dermis combined with autologous thin skin graft can be used in patients with burn scar contracture in joints, which can improve the scarring of skin grafting area and donor site, shorten the healing time of donor site and improve the function of burn scar contracture joint.
5.Study on HPLC Fingerprint of Bombyx mori from Different Producing Areas and Content Determination of Multi- component

OBJECTIVE:To establish the fingerprint of Bombyx mori and the method for the content determination of multi- components,and to provide reference for comprehensive quality evaluation of B. mori . METHODS :Using 18 batches of B. mori from different producing areas as samples ,HPLC method was used. The column was Shiseido CAPCELL PAK C 18 AQ S 5 with mobile phase consisted of methanol- 0.05 mol/L potassium dihydrogen phosphate solution (gradient elution )at the flow rate of 1.0 mL/min. The detection wavelength was 260 nm,and column temperature was set at 30 ℃. The sample size was 10 μL. HPLC fingerprint analysis and similarity evaluation were performed by using TCM Chromatogram Fingerprint Similarity Evaluation System(2012 edition),and the chromatographic peak was identified by comparing with the chromatogram of reference substance. The contents of 4 nucleosides as uracil ,guanine,xanthine,uridine were determined . RESULTS :A total of 16 common peaks were identified in HPLC fingerprint for 18 batches of B. mori ,and peaks 3,6 ,7 and 8 were identified as uracil ,guanine,xanthine and uridine. The similarity of sample chromatogram with control fingerprint were 0.912-1.000. The linear range of uracil ,guanine, xanthine and uridine were 5.34-534,5.28-528,5.06-506,5.195-519.5 μg/mL(r≥0.999 8). The limits of detection were 0.032 4, 0.032 0,0.030 7,0.031 2 μg/mL,and the limits of quantitation were 0.106 8,0.105 6,0.101 2,0.103 0 μg/mL. RSDs of precision,reproducibility and stability tests (24 h)were all lower than 1.00%(n=6). Average recoveries were 100.15%-102.95%, and RSD s were all lower than 2.00%(n=9). The content determination results showed that the content of uracil ,guanine, xanthine and uridine of B. mori from different producing areas were 0.41%-2.46%,0.37%-1.98%,0.72%-2.63%,0.94%-3.67%, respectively. CONCLUSIONS :Established HPLC fingerprint and content determination method of 4 nucleosides were specific , accurate and reliable ,which can be used for the quality evaluation and control of B. mori .
