Objective To investigate the effects of taurine on the expression of stem cell factor mRNA of the focal cerebral ischemia in rat. Methods 63 adult male rats were randomly divided into taurine-treated group (Taurine, 80 mg/kg, began at the day of MCAO and continuously intravenous injection for 10 min, daily for 7 days) and controlled group (0.9% normal saline, 1 ml, continuously intravenous injection for 10 min daily for 7 days). The rats received right middle cerebral artery occlusion at 2h and different hours of reperfusion. Expression of SCF was detected by in situ hybridization in the rats subjected to2h,24h,3d,7d, 14 d of reperfusion and sham-operated group (n=3). Results Low level SCF mRNA expressions were found in cortex, striatum and extraventricular zone in sham-operated group. The expression of SCF mRNA in ischemic hemisphere of controlled group increased markedly compared with sham-operated group (t=2.16~25.19, P< 0.05), besides that of 2 h in cortex, 2 h in striatum, 2h and 14d in extraventricular zone. SCF mRNA expression in the treatment group increased markedly in cortex, striatum and extraventricular zone at 3d and 7d(t=5.19~ 26.17,P<0.05). Conclusion Taurine increased the SCF mRNA expression in MCAO rats, therefore it might promote the creation of neural stem cell.

BACKGROUND: The regeneration and repair of skeletal muscle after injury relies on the new nucleus formed through muscle satellite cell proliferation. However, there are few reports on the relationship between the skeletal muscle cell proliferation and the vimentin expression.OBJECTIVE: To explore the relationship between the skeletal muscle cell proliferation and the vimentin expression, as well as the mechanism underlying the repair of exercise-induced skeletal muscle micro-injury. DESIGN, TIME AND SETTING: A randomized controlled animal experiment was done at the Sports Human Science Experimental Center in Hunan Normal University between December 2007 and September 2008. MATERIALS: A total of 50 male SD rats, aged 8 weeks, were randomly divided into a control group and a training group which were subdivided in to four groups at the time points of immediate post-exercise, hour 3, hour 24 and hour 48 post-exercise, with 10 ones in each of the five groups.METHODS: Rats in the training group underwent 3 days of repetitive exhausting eccentric exercise on a treadmill of-16° slope at the speed of 16 m/min. Rats in the control group underwent no running. MAIN OUTCOME MEASURES: Rats in the training group was taken for measurement immediate, 3 hours, 24 hours and 48 hours post-exercise respectively. Immunohistochemistry was adopted to determine the proliferating coil nuclear antigen (PCNA) A expression and the vimentin expression in medial head of triceps brachii muscle cells in each group at different phases of repair.RESULTS: The skeletal muscle cell showed sequentially-changed proliferation. The proliferation index of the training group was significantly higher than that of the control group immediately post-exercise. At hour 24 post-exercise, it reached the peak. Hour 48 post-exercise saw the decreased proliferation. The expression of vimentin also exhibits a sequential change after exercise. Moreover, its immunoreaction score changed with time in the same direction with the proliferation index, but there is no correlation between the two.CONCLUSION: Three days of repeated exhausting eccentric exercise can induce the sequential changes of skeletal muscle cell proliferation and vimentin expressions. Vimentin expression has some kinds of correlations with skeletal muscle cell proliferation, but it is not the only factor that matters.

BACKGROUND: Non-adherence to fluid restrictions is common in patients maintained on hemodialysis prior to and after kidney transplantation, which has a profound influence on the development and transfer of illness of the patients.Mental factors have great influences on adherence to fluid restrictions.OBJECTIVE: To investigate the effects of rational-emotive therapy on adherence to fluid restrictions of patients maintained on hemodialysis prior to and after kidney transplantation.METHODS: In total 100 patients maintained on hemodialysis were randomly assigned to an intervention group(n = 50)and a control group(n = 50).The control group received the conventional therapy. At the same time, the intervention group received the conventional therapy combined with rational-emotive therapy. All cases were assessed with the Symptom Checklist 90(SCL-90)and Medical Coping Modes Questionnaire(MCMQ)prior to and after intervention.Clinical indices related to adherence to fluid restrictions were measured three successive times and the average score of each index was calculated.RESULTS AND CONCLUSION: Forty-eight patients from the intervention group and 44 patients from the control group accomplished the study. After intervention, the scores of the two subscales, confronce and avoidance, were significantly higher than those in the control group(P < 0.05).However,some scores were significantly lower than those in the control group, including the score of acceptance/resignation subscale of MCMQ,total average score of SCL-90,the score of somatization,obsessive-compulsive,interpersonal sensitivity, depression, anxiety, hostility, photic anxiety and additional items, and the score of four clinical indices related to adherence to fluid restrictions(the ratio of interdialysis weight gain to dry weight, mean systolic blood pressure, mean diastolic blood pressure, and mean ultrafiltration volume prior to hemodialysis)(P < 0.05).All these findings suggest that rational-emotive therapy can effectively amend the coping modes and the mental states of the patients, and then improve their adherence to fluid restrictions.
