1.Lignan glycosides from Saussurea japonica and rules for substituted effects on ~(13)CNMR chemical shifts of (+)-pinoresinol and (+)-lariciresinol

Object To isolate and identify the chemical constituents of Saussurea japonica (Thunb.) DC. and to obtain some rules of chemical shift effects on hydroxylation and O-glycosylation. Methods The constituents were isolated by silica gel column chromatography and HPLC (RP-18), and their structures were elucidated through spectroscopic analysis (NMR and FABMS). The rules were obtained by comparison with those of known analogous lignans. Results Three lignan glycosides were isolated and identifiedcas (+)-1-hydroxypinoresinol-4″-?-D-glucopyranoside (Ⅵ), (+)-lariciresinol-4-?-D-glucopyranoside (Ⅷ), and (+)-1-lariciresinol-4′-?-D-glucopyranoside (Ⅸ), as well as the rules for effects of the hydroxy group at C-1 (upshift about ??-4.2) and O-glucosylation (downshift about ??+3.0) on 13CNMR chemical shifts of (+)-pinoresinol and (+)-lariciresinol. Conclusion All these compounds were obtained from the plants of Saussurea DC. for the first time and the rules should be as some important evidences to assign the position of hydroxylation and glycosylation.
2.Efficacy and safety of ropivacaine combined with lidocaine in caudal block for pediatric laparoscopic hernia surgery

Objective To evaluate the anesthetic effect and safety of ropivacaine combined with lidocaine caudal block in the treatment of pediatric laparoscopic hernia surgery.Methods A total of 70 cases with pediatric hernia surgery in Ningbo Women and Children’s Hospital from May 2014 to October 2015 in this study were divided into control group and experimental group with 35 cases in each group.The patients in the both groups were given routine examination, preoperative anesthesia induction, adopted 7 scalp injection needles to prick into the hiatus sacralis,patients in control group were treated by 1% lidocaine hydrochloride injection mixed liquid 1 mL/kg;patients in the experimental group were treated by 0.25%lidocaine hydrochloride injection and 0.25% ropivacaine hydrochloric injection mixed liquid 1 mL/kg; pediatric hernia surgery, intraoperative continuous pumped into propofol injection 4-6 mL/kg and maintain the mask to the oxygen.The visual analog pain scored( VAS) , visual satisfaction scored ( VSS) and adverse reactions were compared at the end of operation, after operation 0 h,0.5 h,1 h,2 h,4 h,8 h and 24 h.Results The sensory and motor block effective time in the experimental group were later than the control group, the sensory and motor block duration time were shorter than the control group (P<0.05).VAS score of the experimental group was significant lower than control group after operation 2 h ( P<0.05 ) . After operation, postoperative 0.5 h, postoperative 1 h, the VSS score of the experimental group was significant lower than control group ( P<0.05 ) .Lower limbs numbness discomfort duration time of the experimental group was significant lower than control group after operation ( P<0.05 ) . Postoperative agitation rate of the experimental group 8.57% was significant lower than the control group 34.29% (P<0.05).The incidence of adverse reactions of experimental group 5.71% was significant lower than the control group 25.71% ( P<0.05 ) .Conclusion Ropivacaine combined with lidocaine caudal block in the treatment of pediatric hernia surgery has the remarkable effect, with high safety.
3.Different doses of propofol infusion on serum lipid metabolism and pulmonary function in patients with cervical cancer surgery

Objective To investigate effect of different doses of propofol infusion on serum lipid metabolism and pulmonary function in patients with cervical cancer operation.Methods 120 patients with cervical cancer were randomly divided into A, B, C three groups, 40 cases in each group, were treated with laparoscopic cervical cancer radical operation, the 4 mg/(kg· h), 6 mg/(kg· h), 8 mg/(kg· h), and the pulmonary function and lipid profiles were detected.Results Compared with the B and C groups, the recovery time and the the awake time of the patients in B ,C group was shorter (P<0.05).The recovery time and the the awake time of the patients in A group had not significantly different.Compared with before operation, TG, TC and LDL levels were elevated, HDL decreased in theree groups after the operation(P<0.05);compared with the A and B groups, the levels of TC, LDL, TG and were higher, the level of HDL were lower (P<0.05),there were no significant differences in level of TC, HDL, TG and LDL between group A and group B;there was no significant difference between P(A-a) O2 and RI among three groups before surgery, P(A-a) O2 and RI levels were elevated in the three groups after surgery(P<0.05), compared with the A and B groups, P(A-a)O2 and RI levels in group C were lower(P<0.05), there was no significant difference in P(A-a)O2 and RI between group A and group B.Conclusion 4 mg/(kg· h)of propofol can significantly shorten the recovery time and awake time, improve the level of lipid metabolism and pulmonary function.
4.Circadian rhythm changes of PGE2 levels in blood of growing rats after functional mandibular protrusion

Objective:To explore the relationship between wearing of functional appliance and systemic metabolism of PGE2 in the blood of growing rats.Methods:54 male Wistar rats aged 4-weeks were randomly divided into 3 groups(n =18):Daytime group,all-day group and control group.Self-guided functional appliance was made for mandible advancement of the rats in the 2 experimental groups.One week after appliance wearing,rats were sacrificed at the corresponding time points and blood in femoral artery was taken. PGE2 levels in the blood samples were measured,data were analysed by cosine equation.Results:Hypothesis was true in the control group(P <0.05),and was not true in both 2 experimental groups(P >0.05).Conclusion:PGE2 level in rat blood is circadian rhyth-mic,wearing of functional appliance made may disturb the rhythm.
5.Effects of astragaloside IV on experimental ventricular remodeling in mice and its mechanism from matrix metalloproteinase aspect

Aim: To study the effects of astragaloside IV on experimental ventricular remodeling in mice and its mechanism from matrix metalloproteinase aspect.Method: Ventricular remodeling in mice was induced by con-secutively subcutaneous injection of isoproterenol for 14 days.Astragaloside IV(40,80 mg/kg)and propranolol(40 mg/kg,positive control)were administered by gavage to mice.Echocardiography was used to observe the left ventricular end diastolic diameter(LVIDd),left ventricular end systolic diameter(LVIDs),fractional shortening(FS)and ejection fraction(EF).The myocardial pathological pattern was detected by HE staining.Expressions of matrix metalloproteinases(MMP2,MMP9)and tissue inhibitor of metalloproteinases(TIMP1,TIMP2)mRNA in myocardium were detected by RT-PCR.Activities of MMP2 and MMP9 were assayed by zymography.Results:Compared with those of control mice,LVIDd and LVIDs were increased,FS and EF were decreased in the model group.Myocardial injury and fibrosis existed in histop at hological slices of the model group.In addition,the mRNA expressions and activities of MMP2 and MMP9 were increased in the model group.However,there were no markable changes to TIMP1 and TIMP2.Treatment with astragaloside IV reduced LVIDd and LVIDs,increased FS and EF,attenuated myocardial injury,and down-regulated mRNA expressions and activities of MMP2 and MMP9 compared with the model group.Conclusion: Astragaloside IV can greatly improve ventricular remodeling and dysfunction via decreasing MMP2 and MMP9 mRNA expression and activities in cardiac ventricles.
6.Guar gum/ethylcellulose coated pellets for colon-specific drug delivery

The aim of this work was to investigate guar gum/ethylcellulose mix coated pellets for potential colon-specific drug delivery. The coated pellets, containing 5-fluorouracil as a model drug, were prepared in a fluidized bed coater by spraying the aqueous/ethanol dispersion mixture of guar gum and ethylcellulose. The lag time of drug release and release rate were adjustable by changing the ratio of guar gum to ethylcellulose and coat weight gain. In order to find the optimal coating formulation that was able to achieve drug targeting to the colon, the effect of two independent variables (the ratio of guar gum to ethylcellulose and the coat weight gain) on drug release characteristics was studied using 3×4 factorial design and response surface methodology. Results indicated that drug release rate decreased as the proportion of ethylcellulose in the hybrid coat and the coat weight gain increased. When the ratio of guar gum to ethylcellulose was kept in the range of 0.2 to 0.7, and the coat weight gain in the range of 250% to 500%, the coated pellets can keep intact for about 5 h in upper gastrointestine and achieve colon-specific drug delivery. The pellets prepared under optimal conditions resulted in delayed-release sigmoidal patterns with T5% (time for 5% drug release) of 5.1-7.8 h and T90% (time for 90% drug release) of 9.8-16.3 h. Further more, drug release was accelerated and T90% of the optimum formulation pellets decreased to 9.0-14.5 h in pH 6.5 phosphate buffer with hydrolase. It is concluded that mixed coating of guar gum and ethylcellulose is able to provide protection of the drug load in the upper gastrointestinal tract, while allowing enzymatic breakdown of the hybrid coat to release the drug load in the colon.
7.Sequence variability of human cytomegalovirus UL143 open reading frame gene in low-passage clinical isolates

Objective To explore the relationship between ULI43 sequence variability and clini-cal disease. Methods UL143 from samples obtained from suspected congenitally human cytomegalovirus (HCMV) infected symptomatic infants were PCR amplified and sequenced. Results There were not too much sequence variability of UL143 compared with Toledo. But no one was completely identical to Toledo, and all UL143 ORFs were shorter than Toledo for frame-shift. Conclusion HCMV-UL143 existed in moat of low passage isolates and sequences were variable. No obvious linkage was observed between UL143 poly-morphisms and outcome of suspected congenital HCMV infection.
8.Blockade of CLC-3 chloride channel inhibited the viability and invasion of colorectal cancer cells

Objective:To examine the expression of CLC-3 in colorectal tissues and the effect of CLC-3 on the viability and invasion of colorectal cancer (CRC) SW480 and SW620 cells. Methods:The mRNA levels of CLC-3 in CRC cell lines were determined by RT-PCR. CLC-3 expression was inhibited by adding DIDS or NPPB to the CRC cells. Subsequently, cell viability and invasion were assessed by CCK-8 assay and Transwell assay, respectively. In addition, the effects of DIDS and NPPB on the Wnt orβ-catenin signaling pathways were de-termined by Western blot analysis. Results:The mRNA level of CLC-3 was remarkably increased in the CRC tissues compared with that in normal colorectal tissues (P<0.05) and was positively correlated with the T stage of CRC. The blockade of CLC-3 inhibited the viability and invasion of CRC cells (P<0.05). The expression ofβ-catenin, C-myc, cyclin D1, Ki-67, and survivin were evidently reduced by the in-hibition of CLC-3 (P<0.05). Conclusion:The inhibition of CLC-3 decreases the cell viability and invasion of CRC cells by reducing the ex-pression of the proteins related to the Wnt orβ-catenin signaling pathway.
9.Study on Intervention of Zi-He-Che on E2, LH, FSH among Perimenopausal Rat Model

This study was aimed to explore effects of Zi-He-Che (ZHC) on estradiol (E2), follicle stimulating hor-mone (FSH), luteinizing hormone (LH) among perimenopausal rat model. The 4-month-old young female SD rats with vaginal smear of normal estrous cycle and 18-month-old aged female SD rats with vaginal smear of no estrous cycle were selected as the normal control group. The 11- and 12-month-old female SD menopausal rats with vaginal smear of estrous cycle disorder were selected and randomly divided into the model group, treatment group and posi-tive control group. The normal control group and the model group were given physiological saline. And the treatment group was given ZHC. The positive control group was given prescription from the textbook. Observation was given on effects of ZHC on E2, LH and FSH among perimenopausal rat model. The results showed that compared with the young group, the level of E2 in the old and model group was significantly decreased (P<0.01), the level of FSH and LH were significantly increased (P<0.01). Compared with the model group, the level of E2 in the treatment group and positive control group was significantly increased (P<0.01), the level of FSH and LH were significantly decreased (P<0.01). It was concluded that both ZHC and prescription from the textbook were able to reduce FSH and LH levels, and increase the estrogen level among perimenopausal rats.
10.Determination of ammonium glycyrrhizinate in Shufei Mixture by RP-HPLC

Objective To establish a RP-HPLC method for determination of ammonium glycyrrhizinate in Shufei Mixture .Methods The sample was analyzed on an Agilent Zorbax SB-C18 column (4.6mm ×150 mm, 5μm), and acetonitrile-0.0025 mol/L sodium heptanesulfonate solution–0.05 mol/L potassium dihydrogen phosphate solution(20:45:45) ( adjusted pH value of 7.2 ±0.05 with 20% sodium hydroxide solution) was used as mobile phase.The flow rate was at 1.0 mL/min.The detective wavelength was at 250 nm.The column temperature was 30 ℃.Results With this chromatographic condition, the ammonium glycyrrhizinate peak in Shufei Mixture sample chromatogram could be separated with other ingredient peaks completely.The negative sample had no interference.The calibration curve was linear at a ranges of 23.6-118.1μg/mL for ammonium glycyrrhizinate, and equation of regression was Y=0.1133X–0.00110,r=0.999 8.The average recovery from sample was 97.8% and RSD was 0.88%(n=6).The content range of ammonium glycyrrhizinate in three batch Shufei Mixture sample was 0.2532-0.2865 mg/mL, and average content was 0.2721 mg/mL. Conclusion This method is simple, accurate, and useful for control method of this preparation.
