Herald of Medicine 2017;36(9):982-986
Objective To investigate the protective effect of Jiejiu oral liquid on acute alcohol-induced gastric mucosal lesions in mice. Methods Sixty Knming''s mice were randomly divided into normal control group, model control group, sucralfate group(600 mg·kg-1),the dose of 25.0 g·kg-1 group, the dose of 12.5 g·kg-1 group and the dose of 6.25 g·kg-1 group of the Jiejiu oral liquid (n=10 each).Except for the normal control group, other groups were intragastrically administered with 95% ethanol to create the alcohol-induced acute gastritis model. The effect of pathological histology, injury of gastric mucosa, the mucosal NO,ET-1,TNF-α,IL-6 and PGE2 content of acute alcohol-induced gastric mucosal lesions in mouse were observed. Results The gastric mucosa in model control group appeared obvious bleeding, erosion, and flake ulcer.Compared with the model control group, the acute damage index of the Jiejiu oral liquid therapy groups were obviously reduced (P<0.05 or P<0.01).The content of ET-1,TNF-α and IL-6 in the 25.0 g·kg-1 and the 12.5 g·kg-1 dose group of the Jiejiu oral liquid significantly decreased compared with the model control group (P<0.05 or P<0.01).The content of NO in the 25.0 g·kg-1 group of the Jiejiu oral liquid increased (P<0.05).Jiejiu oral liquid had no significant effect on the content of PGE2 (P>0.05).Histopathologic examination revealed Jiejiu oral liquid can reduce the shedding of gastric mucosa and inflammatory cells infiltration. Conclusion The Chinese herbal mixture Jiejiu oral liquid has a protective effect on acute alcohol-induced gastric mucosal lesions in mice.

Golgi phosphoprotein 3 (GOLPH3 )is closely related to the development and prognosis of cancer, such as non-small cell lung cancer, breast cancer, gastric cancer, esophageal cancer, colorectal cancer, etc. The mainly tumor pathogenisis related GOLPH3 includes modulating the response to DNA damage, vesicle trafficking,mTOR signaling pathway, mitochondrial functions, cytokinesis and Golgi vesicular malignant secretion, and then promoting cell proliferation, invasion and metastasis.GOLPH3 is expected to be a new tar-get for cancer therapy, which may become an important biomarker for the diagnosis, treatment and prognosis of cancer.

OBJECTIVE:To develop an HPLC method for the determination of 5-ISMNconcerntrations in human plasma and study the pharmacokinetics of 5-ISMN orally disintegrating tablets and the reference tablets.METHODS:A single oral dose test capsule(orally disintegrating tablets)or reference capsule of 5-ISMN(60 mg)were administered by randomized crossover way in 20 healthy male volunteers with plasma 5-ISMN concentrations determined by HPLC.The pharmacokinetic parameters were calculated with 3p87 pharmacokinetic program and the bioavailability of the two preparations was evaluated.RESULTS:The main pharmacokinetic parameters of the test preparation vs.the reference preparation were as follows:Cmax:(613.42?73.83)vs.(368.64?38.66)?g?mL-1;tmax:(26.52?2.00)vs.(95.73?4.16)min;AUC0~∞:(73 872.24?543.89)vs.(77 978.47?646.37)ng?min?mL-1,respectively.CONCLUSION:The assay was proved to be sensitive, accurate and suitable for pharmacokinetic study of 5-ISMN.The results of pharmacokinetic study after oral administration of test and reference preparations of 5-ISMN showed that the two preparations were bioequivalent while the test preparation had a higher peak concentration and could reach peak level in less time, and the test preparation showed a rapid drug releasing behavior.

Objective:To explore the characteristics of the ultraweak luminescence of human hepatocarcinoma cells(HepG2). Methods: The intensity of the ultraweak luminescence of HepG2 cells and human hepatocytes(QZG)was examined by an IFFM-D chemiluminescence instrument. The intensity of the ultraweak luminescence of HepG2 cells and human hepatocytes(QZG)were affected by different concentration of Luminol and H_(2)O_(2). Results: In 10~(-4)mol/l Luminol and 0.3% H_(2)O_(2),the intensity of the ultraweak luminescence of HepG2 cells was significantly higher than that of QZG cells(P)0.05). Conclusion:The intensity of the ultraweak luminescence of HepG2 cells is higher than that of QZG cells. The intensity of the ultraweak luminescence is affected by cell number and the concentration of Luminol and H_(2)O_(2).The ultraweak luminescence reflect functional status of tumor cells. It can become an sensitive index of physiology and metabolism of tumor cells.

Objective To describe the caring knowledge training, caring experiences and coping style of nursing attendants in elderly homes of Fujian province. Methods 15 nursing attendants and 3 managers from 4 different geriatric nursing homes were selected and semi-structured in-depth interviewed and the phenomenological analysis method was used. Results Three themes were found as follows. Theme 1: caring knowledge training: (1)training varied owing to different institutions, nursing skills were focused on;(2)caring knowledge accumulated by work, grope for the rich;(3)most nursing attendants wished to learn, paid attention to practical application; (4) part of the nursing attendants were negative to training, less schooling might be the reason. Theme 2:caring experiences:(1)happiness followed;(2)sense of achievement;(3)unavoidable of troublesome and pressure; (4)dedication existing. Theme 3:copying style: (1)negative compromise;(2)positive coping style; (3)varying with each individual. Conclusions The qualities and coping style of nursing attendants in geriatric nursing homes vary, their work is difficult but they work hard, establishing effective support system is recommended.

Chinese Journal of Pathophysiology 2010;26(2):282-286
AIM: To investigate the RNAi effect of the inhibitory member of the ASPP family (iASPP) on the apoptosis of human breast cancer cell MCF-7 which expressed the wild type p53 gene. METHODS: The recombinant plasmid pAd-iASPP-RNAi was transfected into MCF-7 cells. The expression of iASPP mRNA and protein was analyzed by RT-PCR and Western blotting, respectively. The cell apoptosis was detected by FCM, and then the MCF-7 cells were transplanted into nude mice to set up transplantation model. The expression of iASPP RNA and protein in transplanted neoplasm were determined by RT-PCR and Western blotting, the apoptosis index was detected by FCM at the same time. RESULTS: The results showed that the expression of iASPP descended in MCF-7 cells (mRNA 95.4% and protein 96.8%, respectively, P<0.01) and the apoptosis rate and necrosis rate of MCF-7 cells increased (P<0.01) after transfection. As treated with pAd-iASPP-RNAi, the expression of iASPP in transplantation tumor cells descended 87.4% (mRNA) and 89.2% (protein), respectively (P<0.01), and the apoptosis rate and necrosis rate increased accordingly (P<0.01, P<0.05). CONCLUSION: The inhibition of iASPP may resume the ability of p53 to induce apoptosis in breast cancer cells which is able to express wild type p53.
