1.Effects of ethyl pyruvate on myocardial apoptosis and expression of Bcl-2 and Bax proteins after ischemia-reperfusion in rats.

In order to study the effects of ethyl pyruvate on cardiomyocyte apoptosis following ischemia/reperfusion (I/R) in vitro and the expression of Bcl-2 and Bax proteins, isolated rat hearts were perfused in a Langendorff model. Twenty-four rats were randomly divided into 3 groups (n=8 in each group): control group was perfused for 120 min. In the I/R group, after 30 min stabilization the injury was induced by 30 min global ischemia followed by 60 min reperfusion. Ethyl pyruvate (EP) group was set up with the same protocol as I/R group except that it was supplied with 2 mmol/L EP 15 min before ischemia and throughout reperfusion. Myocardial malonaldehyde (MDA) content was measured. Myocardial apoptotic index (AI) was tested by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) method. The expression of anti-apoptotic protein Bcl-2 and pro-apoptotic protein Bax in cardiac myocytes was detected by immunohistochemistry. As compared with control group, the content of MDA, myocardial AI and the expression of Bcl-2, Bax proteins were increased significantly in I/R group, but the content of MDA, myocardial AI and the expression of Bax protein were decreased obviously and the expression of Bcl-2 protein was up-regulated in EP group (P<0.05). These results demonstrate that EP could inhibit apoptosis of cardiac myocytes possibly via alleviating oxidative stress, up-regulating Bcl-2 and down-regulating Bax proteins.

2.Related Factors about the Quality of Life in the Elderly with Knee Osteoarthritis

Objective To explore the factors related with the quality of life in the elderly patients with knee osteoarthritis. Methods 92elderly patients with knee osteoarthritis were tested with short-form 36 health survey scale (SF-36), visual analogue pain scale (VAS), 5times' movement of sit-to-stand, 10 m walking speed, 3 m timed-up and walking test. At the same time, the radiological grades of knees andthe total amount of chronic diseases were collected. Results The VAS of knee pain, the course of knee osteoarthritis, the total amount ofchronic diseases and 10-m walking speed were related with SF-36 score (P<0.01). Conclusion Knee pain, course of knee osteoarthritis, totalamount of chronic diseases and walking ability are related with the quality of life in the elderly patients with knee osteoarthritis, especiallyfor knee pain.
3.Effect of TGF-βRⅡ siRNA on AngⅡ induced proliferation of glomerular mesangial cell

Objective To investigate the effect of transforming growth factor β receptor-Ⅱ(TGF-βRⅡ) on AngⅡ inducing glomerular mesangial cell(GMC) proliferation and the expressions of TGF-β1 and psmad3 through transfecting TGF-βRⅡsmall interfering RNA(siRNA)into GMC. Methods Through transfecting fluo-rescence control siRNA into GMC ,we observed the transfection efficiency under fluorescence microscope after 6 hours. Transfecting TGF-βRⅡsiRNA and negative control siRNA into GMC respectively ,we detected the expres-sion of TGF-βRⅡ by western blot after 24 hours. The cells were divided into four groups:control group,AngⅡgroup,negative siRNA control group and TGF-βRⅡ siRNA group. Each group was stimulated by AngⅡ except the control group. After 24 hours,we detected the TGF-β1 and psmad3 protein levels by western blot and detected GMC proliferation by CCK8 kit. Results (1) Comparing with the scrambled control group,the expression of TGFβRⅡin the TGF-βRⅡsiRNA group was significantly decreased(P<0.05).(2)AngⅡcould accelerate the expression of TGF-β1. Transfecting TGF-βRⅡsiRNA into GMC decreased the expression of TGF-β1 protein(P<0.05).(3)AngⅡ could stimulate the phosphorylation of smad3. Transfecting TGF-βRⅡ siRNA into GMC de-creased the expression of psmad3 protein(P < 0.05).(4)Transfecting TGF-βRⅡ siRNA into GMC relieved the GMC proliferation AngⅡ-promoted(P < 0.05). Conclusions The AngⅡ stimulates the GMC proliferation,de-pending on the expression of TGF-βRⅡ. It is related to the expressions of TGF-β1 and psmad3.
4.Treatment effect of blood purification combined with fasudil in elderly cardiac surgery patients with postoperative acute kidney injury

Objective To explore the clinical effect of blood purification combined with fasudil in elderly cardiac surgery patients with postoperative acute kidney injury.Methods Fifty elderly cardiac surgery patients with postoperative acute kidney injury were divided into control group and study group by random digit table method with 25 cases each.The 2 groups were treated with routine drug and blood purification,the study group was additionally given fasudil injection 30 mg + 0.9% sodium chloride injection 50 ml vein pumping,1 time/12 h,for 7 d.The urine volume,urine N-acetyl-β-D-glucosaminidase (NAG),urine γ-glutamyl transpeptidase (γ-GTP),urine α 1-microglobulin (α 1-MG),serum creatinine (SCr),blood urea nitrogen (BUN) and creatinine clearance rate (CCr) were observed,and the acute physiology and chronic health evaluation (APACHE) Ⅱ score was computed.Results There were no statistical differences in the indexes before treatment between the 2 groups (P> 0.05).The urine volume after treatment 3,5,7 d in study group was more than that in control group [(38.72 ± 2.68) ml/h vs.(31.68 ± 2.52) ml/h,(47.24 ±3.73) ml/h vs.(40.24 ± 2.52) ml/h、(63.80 ± 2.50) ml/h vs.(56.60 ± 3.30) ml/h],urine NAG,urine α 1-MG,urine γ-GTP,SCr and BUN were lower than those in control group [NAG:(25.05 ±5.44) U/L vs.(28.04 ± 5.21) U/L,(24.06 ± 3.43) U/L vs.(27.23 ± 6.43) U/L,(22.08 ± 3.25) U/L vs.(26.23 ± 4.41) U/L; α 1-MG:(24.05 ± 3.65) mg/L vs.(26.74 ± 6.74) mg/L,(22.98 ± 3.58) mg/L vs.(25.57 ± 3.58) mg/L,(20.95 ± 3.78) mg/L vs.(25.48 ± 3.45) mg/L; γ-GTP:(8.2 ± 0.4) U/L vs.(10.8 ± 3.8) U/L,(7.3 ± 0.2)U/L vs.(10.5 ± 2.5) U/L,(6.5 ± 1.4) U/L vs.(9.7 ± 2.6) U/L; SCr:(206.52 ± 6.72) μ mol/L vs.(255.16 ±6.75) μmol/L,(182.98 ±6.26) μmol/L vs.(252.23 ±9.53) μmol/L,(33.25 ±7.95) μmol/L vs.(170.75 ± 7.94) μ mol/L; BU N:(19.61 ± 3.23) mmol/L vs.(20.25 ± 3.25) mmol/L,(16.76 ± 2.06) mmol/L vs.(18.32 ± 4.84) mmol/L,(12.28 ± 2.26) mmol/L vs.(14.27 ± 4.54) mmol/L],CCr was higher than that in control group [(18.66 ± 3.89) ml/min vs.(13.28 ± 3.25) ml/min,(27.76 ± 4.36) ml/min vs.(16.23 ± 4.18)ml/min,(33.79 ± 5.58) ml/min vs.(22.12 ± 4.65) ml/min],there were statistical differences (P < 0.05).The APACHE Ⅱ score before treatment and after treatment 5,7 d in control group were (32.20 ±4.51),(26.38 ±5.28) and (21.43 ±4.22) scores,in study group were (33.05 ±3.82),(22.15 ±3.42) and (13.25 ± 2.15) scores.There was no statistical difference in the APACHE Ⅱ score before treatment (P > 0.05),the APACHE Ⅱ score after treatment was improved,furthermore APACHE Ⅱ score after treatment 5,7 d in study group were better than those in control group,there were statistical differences (P < 0.05).Conclusions The treatment effect of blood purification combined with fasudil is remarkable in elderly cardiac surgery patients with postoperative acute kidney injury.At the same time,it has high security and very important clinical significance.
5.Effect of Vestibular Sense, Proprioception, and Vision on Fall Risk in Elderly

ObjectiveTo assess the effect of the vestibular, visual, and somato-sensation on the fall risk in elderly individuals. Methods56 subjects aged 60 or over were measured with the static postural stability and fall risk, including stability index (ST), the body weight distribution index (WDI) and the fall index (FI) under 4 conditions: standing on solid surface with eyes open (NO),standing on solid surface with eyes closed (NC), standing on pillows with eyes open (PO),and standing on pillows with eyes closed (PC). ResultsThe mean of fall index of all subjects was (48.21±26.47). The ST was different among groups, which was PC>PO>NC>NO (P=0.000),but no significant differences were noted between PO and NC groups (P=0.705). The WDI was not significantly different among the 4 groups (P>0.05). The FI was correlated with ST (P<0.05), but not correlated with the WDI(P>0.05). ConclusionAll of the vestibular, visual, and somato-sensory systems play an important role in the balance, postural stability and falls. The ST is sensitive to evaluate the static balance function and associated with the fall risk in an elderly population. The WDI is not sensitive enough to evaluate the static balance function and fall risk.
6.Characteristics of DC subsets in chronic HIV-1 infected patients during antiretroviral therapy

Objective To understand the dynamics of DC subsets in chronic HIV-infected patients during antiretroviral therapy (ART).Methods Seventeen treatment-naive chronic HIV-infected patients were enrolled in our study,and blood was collected at week 0,4,12,24,48 and 60 of ART.Additional 15 HIV-uninfected age-matched healthy adults and 15 long term non-progressors(LTNP) were recruited as controls.CD4+T cell counts and viral loads were examined routinely.The counts of DC subsets were measured by flow cytometry and IFN-α plasma levels were measured by ELISA.Data analysis was performed using SPSS version 16.0.Results ( 1 ) Before ART,mDC ( percentage and absolute count) in ⅢⅤ-infected group were significantly lower than those in healthy group and LTNP group,P<0.001 in both groups.After 60 weeks of ART,mDC in HIV-infeeted group were significantly increased,and there were no significant differences compared with healthy controls and LTNP group.(2) pDC and IFN-α plasma levels remained relatively stable during ART,and similar to those in healthy controls and LTNP group.(3)Significant associations were found for DC subsets and CD4+ T cell counts before ART.mDC subsets at week 12,24,60 post ART were positively correlated with CD4+ T cell counts,and negntively correlated with virus load.Changes in mDC at week 8 post ART positively correlated with the changes in CD4+T cells at week 60 post ART,and negatively correlated with the changes in virus load at week 60 post ART.Conclusion The counts of mDC significantly reduced in HIV-infected patients,increased after ART,and positively correlated with CD4+ T cell counts.The findings suggest that mDC may play an important role in controlling HIV infection,mDC may serve as an early predictor for immune reconstitution in the initiation of ART.
7.STUDY ON KINDS AND ACTIVITIES OF PROTEOLYTIC ENZYMES IN THE DIGESTIVE SYSTEM OF GRUS GRUS

Objective The kinds and activities of proteolytic enzymes of organs in the digestive system of Grus grus were studied to provide basic information for the taxonomy and evolvement study of wild birds. Methods The experiment was carried out at 4℃ by using gelatine polyacrylamide gel electrophoresis(G-PAGE) ameliorat. Results 1.Altogether 26 kinds of proteolytic enzymes were found in the digestive system of Grus grus in all pH value conditions.2.The kinds and activities of proteolytic enzymes in the digestive system of Grus grus were affected by pH.The activities of proteolytic enzymes were weakest in the acid environment and strongest in the neutral environment,which shows that the activities of the neutral proteolytic enzymes were stronger than those of the alkaline proteolytic enzymes,and those of the acid proteolytic enzymes were weakest.3.The kinds and activities of proteolytic enzymes of the same organ varied tremendously in different pH value conditions.The proteolytic enzymes of different organs had different optimal pH values.4.Proteolytic enzymes with 19?kD molecular weight existed in every organ of the digestive system except glandular stomach in all pH conditions.Proteolytic enzyme with 66 and 22?kD molecular weight was found in every organ of the digestive system in the neutral environment.Conclusion The optimal pH value of proteolytic enzymes in the digestive system of Grus grus is 7.0.The distribution,activity and pH-dependency of proteolytic enzymes of 66?kD,22?kD,19?kD molecular weight are the important characteristic of proteolytic enzymes in the digestive system of Grus grus.

Methods The experiment was carried out at 4℃ by using gelatine polyacrylamide gel electrophoresis(G-PAGE) ameliorated by Xu Cunshuan. Results 1.At least 26 kinds of proteolytic enzymes were found in digestive system of Buteo buteo.2.The kinds and activities of proteolytic enzymes in the digestive system of Buteo buteo were affected by pH.The activities of proteolytic enzymes was the weakest in the acid environment and the strongest in the alkaline environment,showing that the activity of the alkaline proteolytic enzymes were stronger than that of the neutral proteolytic enzymes,and that of the acid proteolytic enzymes were weakest.3.Duodenum and pancreas had more kinds and stronger activities of proteolytic enzymes than those in tongue,esophagus and craw in any pH value condition.4.The proteolytic enzymes with 75 and 70?kD molecular weight existed in every organ of digestive system except pancreas and liver in any pH condition.Conclusion The similarity of the kinds and the activities of proteolytic enzymes exist in the organs with similar structure and similar functions. The proteolytic enzyme of 75 and 70?kD molecular weight,whose pH-dependent activity is not obvious,exists widely in digestive system of Buteo buteo.The activity of proteolytic enzyme with 198?kD molecular weight is found in every organ of digestive system under pH 8.5 condition.
10.Host Cell Protein C9orf9 Promotes Viral Proliferation via Interaction with HSV-1 UL25 Protein

In light of the scarcity of reports on the interaction between HSV-1 nucleocapsid protein UL25 and its host cell proteins,the purpose of this study is to use yeast two-hybrid screening to search for cellular proteins that can interact with the UL25 protein.C9orf69,a protein of unknown function was identified.The interaction between the two proteins under physiological conditions was also confirmed by biological experiments including co-localization by fluorescence and immunoprecipitation.A preliminary study of the function of C9orf69 showed that it promotes viral proliferation.Further studies showed that C9orf69 did not influence viral multiplication efficiency by transcriptional regulation of viral genes,but indirectly promoted proliferation via interaction with UL25.
