1.Preventive and therapeutic effects of shenfu injection in rats with renal ischemia reperfusion

AIM: To explore protective and therapeutic effects of the shenfu injection (SF) against renal ischemia reperfusionand and its possible mechanisms by studying the impact of SF on P38 mitogen activated protein kinase (P38MAPK), tumor necrosis factor alpha (TNF-?) and intercellular adhensive molecule-1 (ICAM-1). METHODS: 36 male SD rats were randomly divided into control group, ischemia reperfusion (IR) group and SF group. Rats were subjected to left renal pedicle occlusion followed by reperfusion with contralateral nephrectomy. The expression of P38MAPK and ICAM-1 were evaluated by Imunohistochemistry. The expression of TNF-? in the kidney tissue and plasma were analyzed by enzyme-linked immunoadsordent assay (ELISA). Mammalian gonadotropin-releasing hormone(termed GnRHⅠ) is a decapeptide that plays key role in the process of reproduction.In addition to its well-known endocrine function, it has become evident that GnRHⅠ and the second GnRH subtype(termed GnRHⅡ) are potentially important autocrine and/or paracrine regulators in placental trophoblast cells.The effects of GnRHⅠ and GnRHⅡ on trophoblastic cell invasion were investigated in human choriocarcinoma cell line(JEG-3) by the methods of invasion assay, RNA interference, Western blot and Real-time PCR.The data revealed that exogenous native peptide of these two forms of GnRH significantly induced the expression of typeⅠ GnRH receptor(GnRHRⅠ) in JEG-3 cells.The specific siRNA for GnRHR Ⅰ was capable of blocking the invasion-promoting effect of GnRHⅠ, but did not influence the effect of GnRHⅡ.The data suggested that the two hormones may be elicited by distinct receptors.Interference of the activities of ERK and JNK was capable of attenuating GnRH Ⅰ and Ⅱ-induced MMP-2 expression and trophoblast invasion, indicating that activation of ERK and JNK are required in both GnRH Ⅰ and GnRH Ⅱ-mediated MMP-2 production and invasion-promoting effect in human trophoblastic cells.

0.05).In spinal cord tissues of injury groups,the level of HIF-1? mRNA began to elevate at 3h after injury and reached the maximize on 3d,then it began to decline and returned to the basal level on 14d;the HIF-1? expression showed a time-dependent difference as followed: It began to increase at 3h after injury,peaked on 1d,then gradually decreased,and recovered to the normal level on 14d after injury.Conclusion The result suggested that hypoxia circumstance can enhance the stability of HIF-1? and the transcription of HIF-1? mRNA,and the time-dependent expression of HIF-1? and HIF-1? mRNA can be used to help the diagnosis of SCI and estimation of injury time.
5.Evaluation of reliability and validity of SF-36 Health Survey in old people in community

Objectives To evaluate the reliability and validity of SF-36 in old people in community. Methods 378 old people in community were investigated by SF-36 by face-to-face interview, and the score of items was analyzed.Reliability and validity of SF-36 were evaluated by item-internal consistency,item discriminant vadility,Cronbach's alpha coefficient, et al. Results The clustering and ordering of the item means of SF-36 were the same as that hypothesized, except for items PF2,PF10,RP3,GH4. The correlation between an item and its hypothesized scale was 0.4 or above for all items. The Cronbach's alpha coefficient were above the standard of 0.7 for all except SF scale. Conclusions SF-36 Health Survey Questionnaire is available for the elderly people. The present evaluation proposes some minor word and item changes which is more suitable for old people.

砄bjective:To investigate the consistency of Streptococcus mutans and Streptococcus sobrinus isolates identified by colony morphology,biochemical tests and DNA base content analysis.Methods:46 clinical isolates (30 of S.mutans and 16 of S.sobrinus ) were identified by colony morphology,biochemical tests and determination of DNA base contents with reversed phase high performance liquid chromatography. S.mutans Ingbritt and S.sobrinus 6715 were used as the controls. Results:The DNA contents (G+C mol%) of S.mutans and S.sobrinus clinical isolates, identified by colony morphdogy and biochemical test, were 37.52 and 45.19 respectively,consistent with the expected 36~38 and 44~46 mol percent of G+C.Conclusion: S.mutans and S.sobrinus isolates,identified by colony morphology and biochemical tests ,were consistent with the DNA G+C content analysis. S.mutans and S.sobrinus can be easily and reliably identified by colony morphology and biochemical tests.
8.Study on the levels of tumor necrosis factor-αand anti-ds-DNA antibody in children with systemic lupus erythe-matosus

Objectives To investigate the changes and signiifcance of serum tumor necrosis factor-α(TNF-α) and anti-ds-DNA antibody in systemic lupus erythematosus (SLE) and lupus nephritis (LN) in children. Methods Sixty-seven children with SLE among whom 52 children had LN were selected and 30 healthy children were selected as normal controls. The level of serum TNF-αwas measured by enzyme-linked immunosorbent assay, and the anti-ds-DNA antibody level was measured by radioimmunoassay. Results The serum levels of TNF-αand anti-ds-DNA antibody were signiifcantly higher in children with SLE or LN than those in healthy controls (P all<0.01). Furthermore, the serum levels of TNF-αand anti-ds-DNA antibody were signiifcantly higher in active phase than in stationary phase of SLE and LN (P all<0.01). The serum TNF-αlevel was positively correlated with SLE disease activity index score and serum anti-ds-DNA level in 67 SLE patients (P<0.05). Conclu-sions The serum levels of TNF-αand anti-ds-DNA antibody were increased in children with SLE and LN and were positively correlated with disease activity.

Objective To study the effect of substance P (SP), calcitonin gene-related peptide (CGRP) on the blood flow and the function of endolymphatic sac (ES) in guinea pigs to clarify the pathogenesis of some inner ear diseases further.Methods The guinea pig model of superior cervical ganglion (SCG) resection was set up. The ESs of the model animals were stained by SP and CGRP immunocytochemically. The staining images were analysed by the computer. The ESBF was measured with the biologic microphere method.Results Compared with the control group, the staining of SP decreased sharply in the model group, however, the staining of CGRP remained unchanged. In the model animals, the ESBF was decreased either.Conclusion SP and CGRP could take part in the regulation of ESBF to affect the function of ES further.

Objective:To investigate the role of apoptosis-associated speck-like protein(ASC) in non-apoptotic,non-necrotic cell death of neutrophils induced by ONO-AE-248.Methods:The morphological changes of neutrophil nuclei and DNA fragmentation of the cells were examined by confocal microscopy and terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL).Neutrophils were stimulated with medium alone,LPS,TNF-? or ONO-AE248 respectively.Then expression of ASC was detected by Western blot.Results:ONO-AE-248-induced neutrophil death was negative in TUNEL assay.Western blot showed that the expression of ASC protein was down-regulated by ONO-AE-248.Conclusion:The DNA degradation of the neutrophils when stimulated by ONO-AE-248 is probably different from that of the typical apoptosis.ASC might be a crosslink point between the ONO-AE-248-induced neutrophil death and spontaneous apoptosis of neutrophils.
