2.Study on the Pharmacological Activity of Soybean Stem Soyasaponin

Objective To study the pharmacological activity of soybean stem soyasaponin. Methods Normobaric hyporia and exhaustive swimming in mice and osteoporosis rats model were used to observe the pharmacological effects of soybean stem soyasaponin. Results Soybean stem soyasaponin 150 mg/kg significantly prolonged the survival time in hyporia and the swimming time of mice and 100 mg/kg in rats improved the syomptom of osteoporosis models. Conclusion Soybean stem soyasaponin has obvious effect on anti-exhaustion and increasing tolerance of oxygen deficit in mice and significant therapy effect on osteoporosis rats.

Objective To investigate the role of ERK1/2 signal pathway in the apoptosis of rat mesangial cells inhibited by TIMP-1.Methods The human TIMP-1 sense and antisense plasmid were transferred into rat mesangial cells through liposome,while mesangial cells transfected with pcDNA3 and non-transfected mesangial cells were used as control.After being stimulated with high glucose(HG 25mmol/L D-glucose)and PD98059(50 ?mol/L)for 24 hours,the apoptosis rate was measured by flow cytometry.The expression of ERK1mRNA was assayed by RT-PCR.The protein expression of p-ERK1/2 in the cell was analyed by ELISA.Results(1)The apoptosis rates of the non-transfected group,transfect pcDNA3 group,sense group and antisense group in the culture medium of high glucose withact PD98059,were(12.10?2.21)%,(11.90?3.34)%,(5.50?0.50)%and(20.70?3.41)%,respectively.The apoptosis rate of the sense group was significantly lower than that without transfection(P
4.Role of mPTP in hepatocytes in reduction of hepatic ischemia-reperfusion injury by limb ischemic postconditioning in rats

Objective To evaluate the role of mitochondrial permeability transition pore (mPTP) in hepatocytes in reduction of hepatic ischemia-reperfusion (I/R) injury by limb ischemic postconditioning in rats.Methods Forty-eight healthy male Sprague-Dawley rats,weighing 240-280 g,were randomly divided into 4 groups (n =12 each) using a random number table:sham operation group (group S);I/R group;limb ischemic postconditioning group (group LIPC);limb ischemic postconditioning and atractyloside group (group LIPC + APC).In I/R,LIPC and LIPC + APC groups,I/R injury was induced by 30 min occlusion of the hepatic artery and portal vein entering the left and middle lobes of the liver,followed by 120 min of reperfusion.The animals underwent 10 min of bilateral limb ischemia starting from 20 min after liver ischemia in group LIPC.In group LIPC+APC,the animals underwent 10 min of bilateral limb ischemia starting from 20 min after liver ischemia,and atractyloside 5 mg/kg was infused simultaneously via the penile vein.Blood samples were collected from inferior vena cava at the end of reperfusion to detect plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities.The rats were sacrificed,and the left lobe of the liver was removed to detect the degree of mPTP opening and to examine the ultrastructure of hepatocytes under electron microscope.Results Compared with group S,the plasma ALT and AST activities and degree of mPTP opening were significantly increased,and the damage to the ultrastructure of hepatocytes was obvious in the other three groups.Compared with group I/R,the plasma ALT and AST activities and degree of mPTP opening were significantly decreased,and the damage to the ultrastructure of hepatocytes was reduced in LIPC and LIPC + APC groups.Compared with group LIPC,the plasma ALT and AST activities and degree of mPTP opening were significantly increased in group LIPC+APC.Conclusion Limb ischemic postconditioning reduces hepatic I/R injury through inhibiting mPTP opening in hepatocytes of rats.
5.Selection and analysis of targeted cell-binding mimicry peptides in endometrial adenocarcinoma cell line through random peptide phage display

AIM: To isolate targeted the peptides that binding and internalizing into endometrial carcinoma cell line EAC. METHODS: The tumor cells were screened five rounds of whole cell screen through the Ph.D.-12 phage display library. Analysis of the displayed peptides specific binding efficiency to the tumor cells was proceeded. The DNA of phages was extracted, sequenced and translated to the sequences of amino acid and analysis using computer software. RESULTS: After five biopannings, the isolated phages showed high specificity and strong affinity for their cognate cell types relative to different cell lines. Through sequencing, the sequences of displayed peptides were obtained. CONCLUSIONS: Whole cell screening against EAC cells through random phage peptide library can obtain phage peptides with a highly tissue specific binding and internalizing ability. The peptides do provide a basis for tumor's targeted delivery as therapy vector.
6.Evaluation of the efficacy and usefulness of low temperature hydrogen peroxide gas plasma sterilization system for endoscopies

Objective To evaluate the efficacy of STERRAD (r) 100S (low temperature hydrogen peroxide gas plasma sterilization system) by comparing with formaldehyde steam. Methods From 2005.1 to 2005.3, totally sterilization of 2 760 pieces of operating tools were carried out with formaldehyde steam. From 2005.1 to 2005.3, totally sterilization of 4 950 pieces of operating tools in 180 circles were carried out with STERRAD (r) 100S. The instrument damage rate, operating infection rate and satisfaction rate of medical workers of 2 groups were compared. Results All data were in favor of the efficacy of STERRAD (r) 100S. Conclusions STERRAD (r) 100S is a safe, effective, low temperature, and cost-effective sterilization system..
7.The prenatal diagnosis of fetal formations by two-dimensional associtated with three-dimensional ultrasonic inspection

Objective To investigate the clinical application value of three-dimensional ultrasonic inspection in prenatal diagnosis.Methods 90 cases of fetal formations were Tetrospecthely analysed,which were diagnosed by two-dimensionul associtated with three-dimensional ultrasonic inspection.We compared the ultrasonic inspection with the clinical diagnosis.Results The all coincidence rate was 100%by combining the two measures,three-dimensional ultrasonic inspection can make up the defect of two-dimemional ultrasonic inspection.Conclusion Two-dimensional combined with three-dimensional ultrasonic inspection can accurately diagnose fetal formations,and increase the detection rate.
8.Effect of motivational interviewing health education based on trans-theoretical model on healthy diet cognition and behavior change of patients with cerebral infarction

Objective To investigate the intervention effect of motivational interviewing health education based on trans-theoretical model to hospitalized patients with cerebral infarction on their healthy diet cognition and behavior change.Methods 60 patients with cerebral infarction were selected with convenient sampling method,and in accordance with the incorporating order randomly divided them into the intervention group and the control group,each with 30 cases.The intervention group received the motivational interviewing health education based on the trans-theoretical model,which mainly focused on a healthy diet,while the control group received regular health diet education.The intervention effect was compared between two groups.Results The comparative difference of baseline data between the two groups before intervention had no statistical significance,while after intervention the change on knowledge and attitude of these two groups both had statistical significance,but the intervention group improved more obviously than the control group; In regards to behavior change phrase,comparing the healthy eating behavior phase of the intervention group patients before and after the intervention,it was found that before the intervention only 4 cases were on the stage of about-to-change,accounting for 13.3%,but after the intervention the proportion was up to 40.0% with 12 cases.Using chi-square test to compare the behavior phrase difference of the patients before and after the intervention,the difference was statistically significant.Conclusions Motivational interviewing technique based on the trans-theoretical model is an effective method in terms of cognitive and behavioral change,and the health education will improve the cognition on healthy diet and the behavior of the hospitalized patients with cerebral infarction.

Problem-based learning (PBL) teaching method can improve students' ability of study,analysis and problem-solving.Network platform based PBL teaching mode combines the network education and PBL teaching mode; it has clear superiority in information acquisition,communication and transmission.Furthermore,it can also solve the problem of inadequate teaching sources.Network platform based PBL teaching mode was applied in neurology teaching to investigate the best scheme and form for teaching plan compilation,network platform building-up and teaching process implementation.At the same time,teaching effect was evaluated and summarized in an aim to improving neurology teaching quality and speeding up the reform of network-based PBL teaching.
10.Studies on Callus Induction and Plantlets Regeneration of Pogostemon cablin(Blanco)Benth.

Objective Callus induction and plantlets regeneration of Pogostemon cablin(Blanco)Benth.are studied to establish a high-efficiency regeneration system for tissue culture.Methods The leaf segments,nodular stem segments,stem segments and root tips from Pogostemon cablin(Blanco)Benth.were cultured in vitro.The factors which affect the induction,multiplication and differentiation of callus were studied.Results Benzylaminopurine(BA) or thidiazuron(TDZ) combined with ?-naphthylacetic acid(NAA) was helpful to induce callus.The optimal concentrations of BA were 0.2~0.5mg/L.Nodular stem segments and stem segments were the most suitable explants for callus induction.The medium containing BA and NAA promoted the multiplication of callus.The multiplication rate and differentiation rate of 30-day callus were much higher than those of 60-day callus.BA was helpful to induce callus differentiation.The callus from leaf segments exerted a higher differentiation rate.Conclusion A high-efficiency regeneration system of callus induction and plant regeneration from Pogostemon cablin(Blanco)Benth.has been established.
