1.Influence of applying food composition table and foodstuff substitution method on nutritional status in patients with diabetic nephropathy undergoing peritoneal dialysis treatment

Objective To study the effect of applying food composition table and foodstuff substitution method on nutritional status in patients with diabetic nephropathy and undergoing peritoneal dialysis.Methods Forty patients with diabetic nephropathy,treated with peritoneal dialysis for over three months was included in this study.Under the guidance of the food composition table and foodstuff substitution method for six months,nutritional status was compared before and after treatment. Results There was a significant difference in HGS,triceps skin foId thickness(TSF),mid-arm circumference(MAC),midarm muscle circumference(MAMC),Hb,albumin(Alb),serum crcatinine after 6 mortths of intervention compared with those before intervention. Conclusions Food composition table and foodstuff substitution method can obviously impwve the nutritional status for patients with diabetic nephwpathy undergoing peritoneal dialysis and increase the patients'diet compliance as well.
2.Value of PET-CT imaging in diagnosing primary breast cancer and axillary lymph node metastases

Objective To investigate the clinical significance of PET-CT to diagnose breast cancer and the axiUary lymph nodes metastasis. Methods 40 patients with breast cancer performed PET-CT were enrolled in the research. Qualitation the primary tumor and accuracy of PET-CT were evaluated on pathological results and compared with axillary lymph nodes dissection. To analyzed the concordance of the size of primary tumor on pathological result with B-mode ultrasonic and PET-CT and the correlation with the SUV. Results The accuracy, sensitivity and specificity of PET-CT in identifying breast cancer were 95%, 94%, 100%. The accuracy, sensitivity and specificity of PET-CT based on the situation of the axillary lymph node dissection were 88.2% 89.2% 83.3%. A significant association was found between tumor weiweihong and PET-CT, furthermore it had not association with SUV. Conclusion The diagnosis of PET-CT in identifying breast cancer and the axillary lymph node metastasis basically conforms to pathology. It possesses high sensitivity and specificity especially in identifying the size of the primary lesion. It offers a beneficial method to determine the axillary lymph node dissection and reasonable therapeutic regimen.

Objective To examine the effect of endotoxin on myocardial lipid metabolism and determine it′s possible mechanism Methods Sixty nine male Wistar rats (350 450g) were randomly divided into 2 groups: in vivo group and in vitro group In in vivo group endotoxin 0 1?g?kg -1 (small dose)or 1mg?kg -1 (large dose) was injected intrabdominally 3h after endotoxin administration the animals were anesthetized Heart was excised and passively perfused in a Laugendorff apparatus with oxygenated Krebs Heuseleit buffer at 37℃ Non esterified fatty acids (NEFA)and triacylglycerols(TAG) were used as energy supplying substance when the isolated heart had been stabilized for 15min NEFA or TAG was slowly added to the perfusate circuit 1 ml of perfusate was drawn every 10 min until the end of experiment (80 min ) for determination of the effects of endotoxin on NEFA or TAG oxidation and utilization by isolated heart At the end of experiment heart was harvested and frozen for measurement of myocardial lipids and lipoprotein lipase activity In in vitro group endotoxin was added to perfusate (1mg/ml) after the heart was excised and passively perfused In control group endotoxin was replaced by normal saline Results Large dose of endotoxin administered in vivo decreased NEFA oxidation rate by 30% in perfused isolated heart and caused increased accumulation of lipid in myocardial tissue By contrast endotoxin administered in vitro increased oxidation rate of TAG accompanied by an increase in myocardial lipoprotein lipase activity Administration of endotoxin in vivo and in vitro also caused accumulation of various exogenous lipids in myocardium in both NEFA and TAG perfused heart Conclusions This study shows that endotoxin has a significant effect on myocardial lipid metabolism The effect of endotoxin could be either direct or indirect according to the different lipids available
4.A Competitive RT-PCR to Study Apolipoprotein E Gene Gxpression in Adipose Cells

Objective To establish the method to detect the expression of apolipoprotein E gene in human adipose cells. Methods Apolipoprotein E gene expression was determined by competitive reverse transcription-PCR in adipose cells of healthy children. Results Apolipoprotein E gene expressed in the adipose cells of healthy children. The mean expression quantity of apolipoprotein E mRNA in adipose cells was (0.41?0.12 ) in healthy children. Conclusions Competitive reverse transcription-PCR is a rapid, simple, sensitive and accurate method to analyze expression of apolipoprotein E gene.
5.Influence of total glucosides of peony combined with leflunomide serum TNF-α,IL-8 in patients with psoriatic arthritis

Objective To observe the influence of total glucosides of peony (TGP) combined with leflunomide on serum level of TNF-α,IL-8 in the patients with psoriatic arthritis(PsA) ,to evaluate its efficacy and safety and to preliminarily study its action mechanism .Methods The open clinical study was adopted .36 patients with PsA were treated by TGP combined with leflunomide for 12 weeks .at the same time 20 healthy control were selected .The double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) was adopted to detect the level of serum TNF-α and IL-8 before treatment and in 12 weeks after treatmention the two groups .The Psoriasis Area And Severity Index (PASI) was adopted as the main effective index and the Health Assessment Ques-tionaire(HAQ) was taken as the secondary effective index .The level of CRP ,ESR ,TNF-α,IL-8 before and after treatment were an-alysed .Results The serum TNF-α,IL-8 levels before therapy in the poriatic arthritis group were significantly higher than those in the normal control group(P<0 .05);after 12 weeks ,PASI ,HAQ ,serum TNF-αand IL-8 levels in the treatment group were signifi-cantly improved than those before treatment ,the difference had statistical significance(P<0 .01) ,and the serum TNF-α,IL-8 levels were similar to those in the healthy control group without statistical difference (P>0 .05) .Conclusion TGP combined leflunomide for treating PsA has better effect and better safety ,which could play the role by influencing serum TNF-α,IL-8 levels .

ObJective To locate the cysteine-rich domains(CRD) of murine 4-1BB binding to its natural ligand. Methods A serial soluble extracellular CRDs of routine 4-1BB and 4-1BBL fusion proteins was constructed and prepared. The binding of purified 4-1BB-Igs to 4-1BBL and 4-1BB monoclonal antibody were tested using ELISA assay and Western blot analysis. Blocking experiment with 4-1BBL and 4-1BB mon-oclonal antibody was performed by ELISA assay. Results All truncated overlapped proteins containing ex-tracellular CRD Ⅱ of murine 4-1BB were able to bind to 4-1BBL by ELISA assay, excepting the CRD Ⅰ do-main alone. A 4-1BB monoclonal antibody proved to block the interaction of 4-1BB and 4-1BBI, was also able to bind to CRD Ⅱ. Conclusion Murine 4-1BBL whose specificity was mapped to CRD Ⅱ of 4-1BB ex-tracellular region with a possible conformational structure.

We investigate the different expression of toxin gene mazF3,6,9 and antitoxin gene mazE3,6,9 in the drug-resistance Mycobacterium tuberculosis,we used quantitative real-time polymerase chin reaction method to detect the expression level of toxin gene mazF3,6,9 and antitoxin gene mazE3,6,9 in M.tuberculosis (20 mono-resistance strains,20 multidrug resistance strains and standard strain H37Rv).The differences of gene expression levels between groups were analyzed by oneway ANOVA.Contrasting with control group,toxin genes mazF6,9 were up-regulated expression levels both in mono-resistance (11.1519±22.31721;8.4306±17.97897) and multidrug resistance (4.6016±1.29018;6.9627±6.92948),had statistical significance (P<0.01),mazF3 expression levels had statistical significance neither in mono-resistance nor in multidrug resistance (P>0.05);antitoxin genes mazE3 was in down-expression level,and had statistical significance both in mono-resistance (0.3606±0.12527) and multidrug resistance (0.2016±0.16542) (P<0.01),mazE6 had no statistical significance (P>0.05)either in mono-resistance or multi drug resistance,mazE9 only in multidrug resistance(0.3989±0.37679) was in downexpression level,and has statistical significance (P<0.001).The toxin gene mazF6,9 and antitoxin gene mazE3,9 may participate in the drug-resistance formation of M.tuberculosis.
9.GM-CSF induces human vascular endothelial cells to form vessel-like structure and the role of VEGF

AIM: To investigate the influence of GM-CSF on human vascular endothelial cells induced to form new blood vessels and the role of VEGF.METHODS: HUVECs were cultured by Matrigel to set up a stable angiogenesis system with the stimulating factors.The rhGM-CSF concentration-dependent and time-dependent effects and the role of VEGF165 were detected.CD34 was measured by immunochemical staining and numbers of vessel formation was calculated under microscopic observation.RESULTS: After treatment with rhGM-CSF at various concentrations and at different time points,the numbers of vessel formation increased in a dose-dependent and time-dependent manner.In the presence of VEGF165,the numbers of vessel formation increased evidently.CONCLUSION: HUVECs were induced to develop tubular structure in vitro cultured with Matrigel.GM-CSF promotes human vascular endothelial cells to form vessel-like structure in vitro in a dose-dependent and time-dependent manner.VEGF also in vitro promotes human vascular endothelial cells to form new vessel-like structure.
10.Evaluation of the efficacy of intensive pulse light on facial beauty

Objective To assess the efficacy and safety of intensive pulse light (IPL) device for facial rejuvenation and the treatment of hyperpigmented lesions, facial telangiectasias, acne vulgaris and hair removal. Methods One hundred females who claim to improve their skin texture, hair re-moval and patients with hyperpigmented lesions, facial telangiectasias and acne vulgaris were treated with IPL device. Patients received five treatments with the time interval of 3 weeks to 1 month. Pho-tographs were assessed 1 month after the last treatment. Results For facial skin texture, the total im-provement were scored 100 %. For hyperpigmented lesions and facial telangiectasias, the total im-provement reached to 90%. For ache vulgaris, the total improvement reached to 75 %. For hair re-moval, the total improvement was 95 %. Conclusion The IPL device is an effective and safe modality for the improvement of skin texture, hyperpigmented lesions, facial telangiectasias and hair removal, and a novel modality for the treatment of acne vulgaris.
