1.Effect of Rehabilitation Training on Motor Function and Activities of Daily Living of Subacute Stroke

Objective To explore the effect of rehabilitation guidance combined with family training on motor function and activities of daily living of subacute stroke.Methods 89 patients with subacute stroke were divided randomly into two groups,control group(43 cases)and rehabilitation group(46),receiving routine neurological medication.In addition,rehabilitation group received rehabilitation guidance and training in the meantime.Results After 60 days of rehabilitation training,the scores of Fugl-Meyer Motor Function Assessment(FMA)and Activities of Daily Living improved significantly(P<0.01;P<0.001).Conclusion Rehabilitation guidance combined with family training is effective to improve motor function and activities of daily living of subacute stroke.
2.Effect of losartan on myocardial ischemia-reperfusion induced activation of NF-?B in rats

Objective To investigate the effect of angiotensin Ⅱ type Ⅰ receptor antagonist losartan on the infarct size and ultrastructure of myocardium and activation of NF-?B induced by myocardial ischemia and reperfusion (I/R) and the possible underlying mechanism. Methods Forty-two healthy male SD rats weighing 215-285 g were randomly divided into 4 groups: in control group ( n = 6) the anterior descending branch of left coronary artery (LCA) was exposed but not occluded; in I/R group (n = 12) the anterior descending branch of LCA was occluded for 30 min followed by 60 min reperfusion; in group LOS1 ( n = 12) and group LOS2 ( n = 12) losartan 5 mg (LOS1) or 10 mg (LOS2) was injected i.v. 15 min before myocardial ischemia. At the end of 60 min reperfusion the animals were killed and the hearts were harvested for determination of (1) myocardial infarct size (by TTC) , (2) ultrastructure of myocardium (electron microscope) and (3) activation of NF-?B.Results The infarct size in group LOS1 (34.6%?2.2%) and LOS2 (21.8%?4.0%) was significantly smaller than that in I/R group (45.4%?2.4%). Myocardial ultrastructure was well preserved in group LOS1 and LOS2 as compared to I/R group. NF-?B activity was significantly increased in I/R group and losartan pretreatment significantly reduced the increase in NF-?B induced by I/R. Conclusion Angiotensin Ⅱ type Ⅰ receptor antagonist losartan has cardioprotective effect on myocardium against I/R in a dose-dependent manner. Modulation of NF-?B activated by I/R may be involved in the mechanism.

4.Improved primary culture system increases the yield of human umbilical cord mesenchymal stem cells

BACKGROUND:At present, a lot of research about culture methods for umbilical cord mesenchymal stem cels, but not for the waste of primary system. OBJECTIVE:To explore the best culture method of human umbilical cord mesenchymal stem celsin vitro. METHODS:Human umbilical cord mesenchymal stem cels were prepared by tissue explants method, recorded as initial culture group. The centrifugal fluid and tissue of the primary culture flask were centrifuged and divided into three groups for secondary culture: tissue group, mixed group and pure liquid group. Cel morphology, time for cel acquisition, and yield of primary cels in the four groups were observed; the cel growth curve was analyzed by MTT assay; and cel cycle and phenotype were detected by flow cytometry. RESULTS AND CONCLUSION: The average time for cel acquisition in the initial culture group, tissue group, mixed group and pure liquid group were (15.00±0.45), (7.0±0.3), (8.00±0.25) and (8.00±0.25) days, respectively. The number of cels at first generation was (4.0±0.5)×105, (9.0±0.55)×105, (15.0±0.2)×105 and (7.0±0.33)×105 markers of the four groups had no significant differences. The human umbilical cord mesenchymal stem cels can be obtained rapidly and largely through the secondary culture to the primary culture system. T75 culture bottle, respectively. Under the inverted microscope, cels in the four groups were fusiform-like adherent cels, which were in paralel or circinate arrangement. Growth curve, proliferative activity, surface markers of the four groups had no significant differences. The human umbilical cord mesenchymal stem cells can be obtained rapidly and largely through the secondary culture to the primary culture system.
5.Apelin-13 promote mesenchymal stem cells isolated from Wharton’s jelly to differentiate into vascular networks

Objective To explore the role of apelin-13 in regulating stem cell differentiation into vascular net. Meth?ods Mesenchymal stem cells were isolated from human umbilical Wharton’s jelly using tissue adherence method.Their immunophenotypes were detected by flow cytometry . Passage 3 of WJ-MSCs (Wharton’s jelly-mesenchymal stem cells) were inoculated in 4 flasks, denoted as A1, A2, A3, A4 group. TwentyμL of apelin-13 at concentrations of 0, 1×10-6, 10 × 10-6 and 100 ×10-6 mol/L were added to A1, A2, A3 and A4 respectively each day. After being induced for 7 days, cell mor?phology and viability were observed under inverted microscope. Von Willebrand factor (vWF) was examined by immunofluo?rescence and CD31 was identified by flow cytometry. Upon incubating with three dimensional culture medium of hydrogel, those cultured A1, A2, A3 and A4 were renumbered as S1, S2, S3, S4. Again, twentyμL of apelin-13 at concentrations of 0, 1×10-6, 10 ×10-6and 100 ×10-6 mol/L were used to treat S1, S2, S3 and S4 respectively. After 7 days, cell morphology, via?bility and vas-like networks were observed with inverted microscope. Results Our study showed that WJ-MSCs can be in?duced by apelin 13 to differentiate into endothelial cells lineage indicated by positive of vWF staining. Moreover, CD31 expres?sion increases significantly upon apelin-13 addition in a dosage dependent manner. The endothelial cells line formed vas like networks when cultured with three-dimensional medium containing hydrogel. Conclusion This study demonstrated that ape?lin-13 could promote human umbilical cord-MSCs to differentiate into endothelium lineage then to form vascular networks.
6.Regulation of rhIFN-? and rhIL-2 on vascular endothelia l cell in vitro

AIM: To study the effects of rhIFN-? and r hI L-2 on human umbilical vein endothelial cell (HUVEC) proliferation, migration, cell cycle and vascular endothelial growth factor (VEGF). METHODS: Cultured HUVECs were used as model to determine the cel l proliferation with MTT method. Cell cycle was analyzed by cytometry. The cell migration was investigated by agarose scraping method and vascular endothelial g rowth factor (VEGF) content in supernatant of cultured HUVEC was determined by e nzyme-linked immunosorbent assay (ELISA). RESULTS: The growth and migrating number of endothelial cells in rhIFN-? group were 0.199?0.009 and 75.750?23.330, in rhIL-2 group was 0 .217?0.005 and 49.250?8.140, and in combined group was 0.183?0.080 and 40.500?17.230, respectively. In comparison with control group (0.248?0.005 and 160.500?13.220), the effects showed more significant (all P0.05). CONCLUSIONS: rhIFN-? and rhIL-2 show inhibitory effect on vasc ular endothelial cell proliferation, migration and DNA synthesis. When used in c ombination, synergistic effect of rhIFN-? and rhIL-2 is observed, suggesting th at these cytokines play an important role in angiogenesis diseases.
7.PTEN inhibits cell growth and increases chemosensitivity to doxorubicin of human breast cancer cells

Objective To study whether transient overexpression of tumor suppressor gene PTEN could lead to growth suppression and up-regulate the sensitivity to doxorubicin of human breast cancer MCF-7 cells. Methods The eukaryotic expression plasmid pEGFP-C 1-PTEN containing whole cDNA of PTEN was constructed and transfected into MCF-7 cells by Lipofectamine 2000 in vitro. Growth inhibition of the cells was observed by phase contrast microscope and flow cytometry. The clonogenic cell survival ability was studied by clony forming assay. MCF-7 cells′ chemosensitivity to adriamycin was studied with MTT assay. Results PTEN overexpression led to morphological changes characteristic of apoptosis of MCF-7 cells. PTEN overexpression also resulted in a significant increase in G 0/G 1 cell population (14.79%) and apoptosis (10.60%) detected by flow cytometry. The clonogenic survival rate of cells transfected with PTEN was significantly decreased after doxorubicin treatment compared with control. 8.Advantage of controlled-release tramadol in treatment of chronic musculoskeletal pain

BACKGROUND: Chronic musculoskeletal pain can cause disability and decreased quality of life of patients. Probing into the therapeutic methods of musculoskeletal disease and choosing proper treatment drugs are important for patients.OBJECTIVE: To evaluate the efficacy and tolerance of controlled-release tramadol in patients with moderate to severe chronic pain of musculoskeletal origin.DESIGN: A self-control study was conducted.SETTING: Arthrtosis Institute, People' s Hospital Affiliated to Peking University.PARTICIPANTS: Patients who were in the Orthopaedic Outpatient Service,People' s Hospital of Peking University were involved. This observational study was conducted in 40 adults who experienced moderate to moderately severe chronic musculoskeletal pain that was not controlled by routine nonsteroidal anti-inflammatory drugs(NSAID) therapy.INTERVENTIONS: Any NSAIDs and other analgesics were prohibited. Patients initially took 50 mg of controlled-release tramadol (Tramcontin) every 12hours, supplemented by every 50 mg if insufficient pain relieved. Recommended daily maximum dose was 400 mg per day. The observation lasted for 4 weeks. The patients were asked for document therapy-relevant data every day and were interviewed every week to doctor assessment.MAIN OUTCOME MEASURES: Pain intensity before and after treatment, pain remission degree, all side effects and their severity.RESULTS: All cases were finished the study and no one withdrew from follow-up. No eligible patients exited from the study because of therapeutic failure or intolerance. In comparison with the initial visual analogue scale (VAS) mean value 6. 80 ± 1.84, the average VAS value at the end of observation was 1.00 ± 1.46, which had significant difference( P < 0. 001) . An average pain reduction of (85. 50±5.35)%was obtained, among which 78% were absolutely and obviously lessened. Side effects were reported in 9cases (22%).Drowsiness and dizziness(6 cases) was the most common, followed by nausea (3 cases), vomiting (1 case), somnolence (2 cases), constipation and urinary retention (1 case) and blurred vision (1 case).CONCLUSION: Tramcontin, as slow-release tramadol, proved to be an effective, safe, and easy-to-use central acting analgesic has an important role in the management of chronic musculoskeletal pain. Light side effect and good tolerance has provided a new choice for treatment of chronic pain in the orthopaedic department.
9.Value of ultrasonography in predicting the birth weight discordance of twin pregnancy

Objective·To identify the relationship between the estimated fetal weight (EFW) discordance and birth weight (BW) discordance, and evaluate the accuracy of ultrasonography in predicting birth weight discordance (△BW) of twin pregnancy. Methods·The ultrasound was used to detect the growth of twins at different gestational weeks and the data were analyzed to compare the difference between EFW discordance (△EFW) and ?BW. Results·About 15.9 percent of twins (totally 277 cases) had severe △BW (△BW ≥20%) in our study, and the percentages of dichorionic-diamniotic twin (DCDA) and monochorionic-diamniotic twin (MCDA) were 68.2% and 31.8%, respectively (P>0.01). Compared with △EFW in other gestational weeks,△EFW last time before parturition was most consistent of △BW. Furthermore, the negative predictive value of MCDA was highest (94%) at 22-24 gestational weeks. Conclusion·Twin △BW was relatively better predicted by △EFW last time before parturition. Twin pregnancy without severe △EFW (△EFW<20%), especially MCDA with △EFW<20% at 22-24 weeks, had low probability of selective intrauterine growth restriction in the future.
10.Comparison of physicochemical properties of five kinds of cancellous bones

BACKGROUND:Existing evidence has shown that xenogeneic bone derived graft materials have the ability to repair bone defects, but little report is on the comparison of composition of xenogeneic bone and human bone.
OBJECTIVE:To analyze the chemical composition, mineral phase, calcium and phosphorus contents, total protein contents, and appearance of the cancel ous bones of the tibia from pig, bovine and human.
METHODS:Cancel ous bones of the tibia from pig, Banna miniature pig, bovine, young and old people were cut into bone particles of 0.5 cm×0.5 cm×0.5 cm. After degreasing or drying treatment, chemical composition, mineral phase, calcium and phosphorus contents, total protein contents of bone particles were detected using Fourier transform infrared spectroscopy, X-ray diffraction, plasma emission spectroscopy, scanning electron microscopy, and Kjeldahl method, respectively. Nano Measurer 1.2 and Photoshop cs6 were used to measure pore diameter and porosity rate based on the results of scanning electron microscope.
RESULTS AND CONCLUSION:The main inorganic phase of five kinds of cancel ous bones was hydroxyapatite and the main chemical composition was phosphate and carbonate. The calcium content of the bovine cancel ous bonewas lower than that of the other cancel ous bones (P<0.05) and the phosphorus content of the cacel ous bone of young man was higher than that of the pig and bovine cancel ous bones (P<0.01). The total protein content in the cancel ous bones of normal pig and Banna miniature pig was higher than that in the other three kinds of cancel ous bones (P<0.05). The pore diameter of five kinds of cancel ous bones was 400 to 600μm and the porosity rate was 60%to 70%. Moreover, the porosity rate of the bovine cancel ous bone was the highest. Therefore, we conclude that there are some certain differences in the organic and inorganic components of the pig or bovine cancel ous bones as compared with humans.
