1.Efficacy of lived combined Bacillus subtilis and Enterococcus faecium enteric-coated capsules on diarrhea predominant irritable bowel syndrome with bacteria overgrowth in small intestinal

Objective To explore the corrective role of lived combined Bacillus subtilis and Enterococcus faecium enteric-coated capsules on the bacteria overgrowth in small intestinal (SIBO)and assess the efficacy and possible mechanism of it in the treatment of diarrhea predominant irritable bowel syndrome (IBS-D).Methods From May to August 2013,IBS-D patients were collected.Screened by lactulose hydrogen breath test (LHBT),and a total of 60 SIBO positive patients were enrolled in the study and divided into experimental group and control group.Live combined Bacillus subtilis and Enterococcus faecium enteric were given to the experimental group and starch given to the control group,both were capsules,each 250 mg,three times per day,two capsules each time,and the treatment course was four weeks.Before and after administration a questionnaire was conducted in the study objects,including basic information (age,gender,disease type,family history and accompanied diseases) and the severe degree of the symptoms (abdominal pain,bloating,abdominal discomfort,increased frequency of defecation,defecation urgency,defecation endless,defecation hard,bowel and exhaust).Fasting peripheral venous blood was taken from the study individuals before and after administration.The level of interleukin (IL)-1β,IL-6,IL-10,IL-12 and tumor necrosis fator-α(TNF-α) in serum were detected by enzyme linked immunosorbent assay(ELISA).t-test was performed for measurement data comparison between two groups,and chi-square test was for count data comparison.Results Among 60 SIBO positive IBS-D patients,30 patients were in experimental group and 30 in control group.The SIBO negative conversion rate of experimental group and control group was 63.3 %(19/30) and 30.0% (9/30),the difference was statistically significant (x2 =6.696,P<0.05).Before and after administration,in control group there were no significant differences in severe degree scoring of abdominal pain,bloating,abdominal discomfort,increased frequency of defecation,defecation urgency,defecation endless,defecation hard,bowel and exhaust or the level of TNF-α,IL-1β,IL-6,IL-10 and IL-12 (all P>0.05).In experimental group,before and after administration there were statistically significant differences in severe degree scoring of abdominal floating (2.97 ± 1.45 vs 2.61±1.28),level of IL 1β ((1390.81±103.95) ng/L vs (1209.76±113.26) ng/L)and IL-10/IL-12 (4.94±0.74 vs 5.30±0.43),the differences were statistically significant (t=2.052,5.279 and 2.282,all P<0.05).There were no signinficant differences in other items (all P>0.05).Conclusions Lived combined Bacillus subtilis and Enterococcus faecium enteric coated capsules could alleviate the symptoms of abdominal floating.The possible mechanism may related with correcting SIBO,inhibiting low degree of inflammation reaction and regulating the inflammatory cytokines.
2.Clinical significance of CUL4B and interleukin-6 levels in patients with multiple myeloma

Objective To investigate the clinical significance of CUL4B and interleukin?6( IL?6) levels in patients with multiple myeloma(MM).Methods The mRNA of CUL4B and IL?6 were detected by real?time relative quantitative PCR and enzyme?linked immunosorbent assay( ELISA) in 46 cases of MM and 40 cases of non hematologic cancer patients.Results CUL4B concentrations in patients with MM were 3.455(2.098,4. 768).IL?6 concentrations in patients with MM were significantly higher than those of the normal control group ((23. 985(23. 015,26. 878) ng/L vs. 6. 205(5. 405,10. 215) ng/L,Z=-8. 262,P=0. 000). The expression of CUL48 mRNA in Stage Ⅰ,Ⅱ,Ⅲ was 2. 424 ( 1. 881, 3. 583 ) , 3. 594 ( 2. 093, 5. 738 ) , 4. 300 ( 2. 928, 7. 272) respectively, and IL?6 was 23. 115 ( 22. 723, 23. 568 ) , 23. 630 ( 22. 860, 26. 625 ) , 26. 35 ( 24. 995, 30. 550) ng/L respectively,with the increase of clinical stage,the level of CUL4B and IL?6 showed increasing trend( P<0. 05) . CUL4B concentrations were significantly decreased in MM patients after treatment than before treatment(1. 665(1. 420,2. 298) vs. 3. 455(2. 098,4. 768),Z=-4. 955,P=0. 000). IL?6 concentrations were significantly decreased in MM patients after treatment than before treatment( 15. 160( 11. 705,17. 195) ng/L vs. 23. 985(23. 015,26. 878) ng/L,Z=-7. 981,P=0. 000). CUL4B level in patients was positively correlated with IL?6( r=0. 386,P=0. 008 ) . Conclusion It is of great significance to detect CUL4B and IL?6 in patients with MM on clinical stage,the condition of judgment and efficacy observation.
3.Experimental study on renal interstitial fibrosis of chronic aristolochic acid nephropathy in rats

Objective To establish a model of chronic aristolochic acid nephropathy (CAAN) in rats and to investigate the pathogenesis of its renal interstitial fibrosis.Methods Male Sprague-Dawley rats were randomly divided into two groups. One group received extract of Aristolochia manshuriensis Kom by gavage intermittently as model group. Another group received only tap water by gavage as controls. Six rats in each group were sacrificed at the end of 4th, 8th and 12th week respectively and the kidneys of each rat were separately harvested. The mRNA and protein expression of type I collagen (Col I ), transforming growth factor-?1 (TGF-?1), connective tissue growth factor (CTGF), plasminogen activator inhibitor-1 (PAI-1) and tissue inhibitor of metalloproteinase-1 (TIMP-1) was detected by real-time quantitative RT-PCR and immunohistochemical staining respectively. 5.Proteome analysis of indomethacin treated human colon cancer cells

Objective To study the effect and mechanism of indomethacin(IN) on protein maps of human colon cancer cell line (HCT116). Methods Proteome profiles of HCT116 in IN treated and control group were separated by immobilized pH gradient based two dimensional gel electrophoresis (2 DE). Differential expression of protein and proteome analysis were identified and analyzed by silver scan using Image Scanner and ImageMaster software. Peptide mass fingerprint (PMF) was based on matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI TOF MS) and database searching. Results Clear background, well resolution and reproducible 2 DE patterns of HCT116 cells were acquired in IN treated and control group. The protein spots and match rate in control group and IN treated group were 1299?55 (94.2%) and 1208?47 (91.0%) . There were totally 45 differential protein spots, of which the expression of protein in IN treated group was down regulated in 34 spots and up regulated in 9 spots, while 2 spots of protein was expressed only in control group. Twelve differential proteins were identified by PMF, and HCT116 cells apoptosis was through BfL 1 signaling pathway. Conclusion IN can induce HCT116 cells apoptosis though BfL 1 signaling pathway.
7.Combined impacts of blood glucose level and glucose metabolism-related factors on liver 18F-FDG uptake

Objective To evaluate the combined impacts of blood glucose and its related metabolic factors on 18F-FDG uptake by liver.Methods A total of 544 subjects (384 males and 160 females, age range 24-73 years) undergoing 18F-FDG PET/CT were recruited in this retrospective study.SUVmean of the right lobe of liver was calculated.Two-sample t test and one-way analysis of variance were performed to compare SUVmean between patients with different genders and BMI levels.Linear correlation analysis, partial correlation analysis and multiple linear regression analysis were conducted to evaluate the relationship between age, injected 18F-FDG dose, blood glucose, serum T3, T4, FT3, FT4, BMR, BMI and liver SUVmean.Results The SUVmean of the liver in males and females were 1.89±0.42 and 1.92±0.38 (t=0.693, P>0.05), but it was significantly different among BMI groups (F=3.056, P<0.05).Age, blood glucose and FT3 were significantly associated with liver SUVmean (r′ values: 0.108, 0.140 and 0.105, all P<0.05) and were independent factors that indicated variation of liver SUVmean (β values: 0.006, 0.070 and 0.088, all P<0.05).Blood glucose was the strongest powerful predicting variable of liver SUVmean (β′=0.154, P<0.001).Conclusions Blood glucose and its related metabolic factors can affect the liver 18F-FDG uptake.Age, FT3, blood glucose are independent factors predicting variation of liver SUVmean.The impact of glucose metabolism status should be considered when assessing liver 18F-FDG uptake.
8.Methyl Carboprost-NEGPF Administered at Different Time in the Prevention of Postpartum Hemorrhage

OBJECTIVE:To observe the efficacy of Methyl Carboprost-NEGPF administered per rectum at different time in preventing postpartum hemorrhage in patients with vaginal delivery.METHODS:A total of 300 parturients undergoing vagi-nal delivery with possible postpartum hemorrhage were enrolled:Methyl Carboprost-NEGPF 1 granule(1mg) per rectum (with a suitable inserting depth of 6 cm) were administered in Group A(n=100) when their uterine orifices were fully open and Group B(n=100) after fetal delivery,while Group C(n=100) were injected i.m with oxytocin 10IU after fetal delivery. 9.Effects of miRNA-181a on proliferation and migration of multiple myelo-ma cells

AIM:To investigate the effect of miRNA-181a inhibition on the proliferation, migration and cell cycle of the human multiple myeloma cell line RPMI 8226.METHODS:Real-time PCR was used to detect miRNA-181a expression in serum samples from multiple myeloma or healthy subjects .After transfection with miRNA-181a inhibitor, the cell viability was examined by CCK-8 assay and colony formation assay .The cell migration ability was analyzed by wound healing assay .The cell cycle was detected by flow cytometry .Moreover , the protein level of cyclin D 1 and the phosphoryla-tion of PI3K and Akt were determined by Western blot .RESULTS:The expression of miRNA-181a was significantly in-creased in the serum from multiple myeloma patients as compared with healthy group .Inhibition of miRNA-181a expression by transfection with miRNA-181a inhibitor remarkably decreased the cell viability , migratory ability, the population of G0/G1 phase and cyclin D1 protein expression in the RPMI8226 cells.However, the population of S phase and the phosphory-lation of PI3K and Akt were reduced .CONCLUSION:Down-regulation of miRNA-181a inhibits the viability and migra-tory ability in the RPMI8226 cells via inhibition of cell cycle and PI 3K/Akt signaling pathway .
